RESUMEN

The technique is developed including corresponding kit of reagents "AmpliSens Florocenose/Candida-FL", to be applied in laboratory diagnostic of vuvlvovaginal candidiasis. The technique is based on polymerase chain reaction in real-time and permits identifying and quantitatively estimate in biological material content of five most prevalent agents of candidiasis: Candida albicans, C. glabrata, C. krusei, C. paprapsilosis, C. tropicalis. The experimental determination of analytical characteristics of developed technique was carried out. The analytical sensitivity amounted to 100 GE/mL (genome equivalent in 1 mL), linear range -from 200 to 2 x 10 7 GE/mL for every specie of detected Candida. The comparison of quantitative results of detection of content of Candida spp using the developed technique with results of inoculation demonstrated their mutual matching. In logarithm presentation the relation coefficient of lg (GE/mL) to lg (CFU/mL) amounted to 1.2. The correlation of results characterized by coefficient R 2 equal 0.76.

Objective. To determine the pattern of lymphocyte composition changes in Peyer's patches and peripheral lymph nodes in the development of experimental allergic encephalomyelitis (EAE) in mice without treatment and during fingolimod therapy. Material and methods. The research was performed on 40 inbred mice C57BI/6 which were stratified into two groups: mice with EAE of group 1 received no treatment, mice with EAE of group 2 were treated with fingolimod. Results. The development of EAE was accompanied by relative density increasing of lymphocytes, CD4, CD8-cells in Peyer's patches and peripheral lymph nodes that indicated the lymphoid tissue's active systemic response to EAE development. The fingolimod treatment of laboratory animals with EAE resulted in a more significant and prolonged increase in the relative density of lymphocytes, CD4 and especially CD8 cells in the lymphoid formations and also the development of less severe neurological symptoms that confirmed the efficacy of therapy. Conclusion. The intestinal lymphoid tissue may be actively involved in the development of autoimmune demyelinating process in the central nervous system.

RESUMEN

The genital mycoplasma is an opportunistic bacteria and its detection is to be implemented in qualitative format. The study was organized to compare reagents kits "Mycoplasma Duo", "Ureaplasma Microtest", "Mycoplasma microtest" and "AmpliSens-Florocenosis-Mycoplasma-FL". The study resulted in high indicators of diagnostic sensitivity and diagnostic specificity for all kits. At that, the lowest indicators were registered under application of "Mycoplasma Duo" kit. The study reveled correlation of qualitative values detected by using cultural analysis and polymerase chain reaction. The reproducibility of qualitative values of cultural method occurred significantly lower in comparison with "AmpliSens-Florocenosis-Mycoplasma-FL " kit.

Localization and the morphometric characteristics of NADPH-diaphorase--(NADPH-d)-positive neurons were studied in the superior cervical ganglion (SCG), stellate ganglion (SG) and the celiac ganglia (CG) in newborn, 10-, 20-, 30-, 60 and 180-day-old rats, mice and cats. No NADPH-d-positive neurons were found in rats and mice of all the age groups studied. In cats, the largest proportion of NADPH-d-positive neurons was found in the SG, the smaller one--in the SCG, while only the individual neurons we detected in the CG, irrespective of the animal age. In the SG, the proportion of NADPH-d-positive neurons increased during the first 20 days of life and then decreased after 30 days till the end of the second month. The content of NADPH-d-positive neurons in the CG and SCG d-remained unchanged during the development. There were no significant differences in the cross-sectional area between the neurons located in the different ganglia of animals from the same age group. It is concluded that the age development of NADPH-d-positive neurons in different sympathetic ganglia occured heterochronously was finished by the end of the second month of life.

The aim of this study was to obtain the normative data on the age-dependent transformation of morphometric and histochemical characteristics of neurocytes in different ganglia in albino rats. Cell cross-sectional area, activities of cholinesterase (demonstrated with thioacetic acid method) monoamine oxidase (demonstrated with Glenner method) were measured in neurocytes of stellate, spinal, trigeminal and gastric ganglia in rats aged 2 to 360 days. Measurements were made with the help of "Bioscan" videoanalyzer. Informational analysis was used for the evaluation of the degree of maturation of neurocyte systems. General features, age- and organ-related peculiarities of morphometric and enzyme-histochemical characteristics were established for neurocytes of different ganglia, as well as a heterochronism of their definitive state attainment. The time of stabilization for neurocytes of stellate and I thoracic spinal ganglia was the age of 60 days, for those of trigeminal ganglion and intramural gastric ganglia -90 and 120 days, respectively. By this time, neurocyte systems turned from a determined state into a probabilistic-determined one, this transformation being considered as a population stabilization.

RESUMEN

The influence of chemical deafferentation (capsaicin 50 mg/kg in dose) on age-related reorganisations of cervicothoracic ganglion neurocytes was studied. Disturbance of normal age dynamics of neurocyte area, change of neurocyte choline esterase and monoaminooxidase activity and ganglionic choroid plexus density within 6 months following neonatal administration of capsaicine were discovered. The changes were maximally manifested on d 30. Metric and histochemical parameters did not stabilise completely within observation terms.

RESUMEN

Single administration of 100 mg/kg of capsaicine to newborn rats causes 30% decline in neurocyte number, decrease of average area of section neurocytes and dystrophic changes of majority of cells developing in a month. In 3 and 6 months 60% decline of neurocyte number occur, increase of average neurocyte section area is retained, dystrophic changes in cells are less manifested. It was revealed through histogrammes that decline in neurocyte number was conditioned by reduction of fraction of small neurocytes. The results indicate the stable effect of chemical differentiation.

RESUMEN

Piperacillin/tazobactam (P/T) was used in the treatment of 14 patients at the age of 5 to 15 years: 5 patients with acute lymphoblastic leukemia, 3 with acute nonlymphoblastic leukemia, 4 with severe aplastic anemia, 1 with lymphoma and 1 with neuroblastoma. P/T was administered as intravenous infusions in a daily dose of 200-300/25-37.5 mg/kg body weight divided into 3 or 4 portions. All the patients were subjected to intestine selective antimicrobial decontamination. In 7 patients the afebrile condition was recorded before elimination of agranulocytosis without correction of the therapy i.e. without combination of P/T with some other antibiotics such as amphotericin B, vancomycin or aminoglycosides. In 5 patients the effect was stated after correction of the regimen by its supplementing with amphotericin B. 2 patients died. No side effects of P/T was observed. The afebrile condition was provided in 1 to 7 days. P/T is recommended for the treatment of fever of obscure etiology in patients with agranulocytosis and for effective control of infection in such patients.

Using histochemical method mitochondrial cytochrome oxydase (CO) in acoustic and somatosensory centers of rat brain has been studied to reveal CO activity distribution in norm and after impulse-modulated high-ultra-high frequency influence. After ultra-high frequency influence the increase of enzymic activity in a number of regions of rat brain centers with relationship to processing ecologically important sensory signals is revealed.

A relative density of acetylcholine esterase-reactive fibers in adventitium of the large subcutaneous vein changes in the postnatal ontogenesis in women: it is the lowest in newborns, the greatest in mature women, in old women it grows small. In newborns these fibers are situated in the compact layer of the adventitium, in mature and old women they are revealed in interlayers of the connective tissue that separate fasciculi of muscle fibers of the external longitudinal and internal circulatory layers of the middle sheath.

The authors studied the disinfection regimens of materials made of lavsan, capron, acetate, triacetate, viscose by the method of immersion into the disinfecting solutions of dichlor-I, chloramine calcium hypochlorite salt, potassium and sodium salt of dichlorisocianuric acid, sulfochloranthin, dichlordimethylhydantoin, catapin, amphocept, sodium metasilicate, hydrogen, peroxide, formalin, phenol, benzylphenol. As revealed, solutions of the following preparations could be recommended for the disinfection of linen made of chemical fibers at the foci of intestinal and droplet infection, except tuberculosis: sulfochloranthin (0.2%), amphocept and chloramine B (1%), sodium metasilicate, formalin, catapin, hydrogen peroxide with a 0.5% of washing solution (2%); these solutions gave a reliable effect in 60 minutes at room temperature, and in 20 to 30 minutes at a temperature of 40--45 degrees C.
