RESUMEN

Parasitism of Colossoma macropomum is of particular concern because it is the most commonly farmed native fish species in Brazil. Nevertheless, the parasitic fauna of this species in the state of Acre has been little studied. For this reason, an evaluation was made of the parasitic fauna of farmed C. macropomum in the municipality of Rio Branco in southwestern Amazon. Four monogenean species were found in the 122 fish examined: Anacanthorus spathulatus, Linguadactyloides brinkmanni, Notozothecium janauachensis and Mymarothecium boegeri. The most prevalent species was A. spathulatus (50%), followed by N. janauachensis (44.3%), M. boegeri (20.5%) and L. brinkmanni (9.0%). These results are the first data on the ecological indices of monogeneans in tambaqui in the state of Acre and will be useful for future comparisons of the influence of environmental factors on the parasite-host relationship.

Perna perna mussels, abundant throughout the Brazilian coast, are routinely applied as bioindicators in environmental monitoring actions due to their sessile and filter-feeding characteristics. In addition, they are noteworthy for their food importance, especially for coastal populations. In this context, the aim of this study was to investigate elemental contamination in commercially marketed and highly consumed P. perna samples from the highly impacted Guanabara Bay, Rio de Janeiro, Brazil. A total of 30 mussels were sampled, and elemental concentrations (As, Cd, Cr, Cu, Fe, Hg, Mn, Ni, Pb, Se, V, and Zn) were determined in adductor muscle samples by inductively coupled plasma mass spectrometry (ICP-MS). Human consumption risks were assessed by comparisons to Brazilian and international legislations. No significant differences between sex were observed for all analyzed elements. Even when analyzing only the adductor muscle, all mussel samples exceeded the Brazilian limit for Cr, while 12 samples exceeded the limit for Se. When compared to other regulatory agencies, As and Zn levels were higher than the limits set by China, New Zealand, and the USA. Estimated daily dietary intake values were not above limits imposed by the Food and Agriculture Organization of the United Nations/World Health Organization for any of the assessed elements, although it is important to note that only the adductor muscle was assessed. Therefore, continuous metal and metalloid monitoring in bivalves in the study region is suggested, as metal transport and bioavailability, especially in coastal estuaries such as Guanabara Bay, which are currently undergoing significant changes due to anthropogenic activities.

Structure of the helminth community and analyses of helminth population parameters of Pimelodus blochii collected in the Xapuri River in comparison with those in the Acre River were evaluated. Eight adult helminth species were found parasitizing P. blochii in the Acre River: the nematodes Orientatractis moraveci, Rondonia rondoni, Philometroides acreanensis, Cucullanus (Cucculanus) pinai pinai, Procamallanus (Spirocamallanus) pimelodus, Rhadochona acuminata, and Brasilnema sp., and the trematode Dadaytrema oxycephala. For Xapuri's fishes, nine helminth species were found: the nematodes O. moraveci, R. rondoni, C. (C.) pinai pinai, Procamallanus (Spirocamallanus) rarus, P. (S.) pimelodus, R. acuminata, Brasilnema sp., and Cystidicolidae gen. sp., and the trematode D. oxycephala. Nematode and Acanthocephala larvae were also reported. Helminth abundance, prevalence, and diversity were influenced by seasonality and locality (river). The helminth parasites from Acre's fishes formed a subset of the helminth community of the Xapuri's. The results indicate an influence of the environmental characteristics of the rivers on the helminth community structure and diversity. This is the first study of the parasite community of P. blochii in the Xapuri River. The paretheses of (Spirocamallanus) and (S.) should not be in italics all along the text and tables.

RESUMEN

Fish-borne heterophyid trematodes are known to have a zoonotic potential, since at least 30 species are able to infect humans worldwide, with a global infection of around 7 million people. In this paper, a 'state-of-the-art' review of the South American heterophyid species is provided, including classical and molecular taxonomy, parasite ecology, host-parasite interaction studies and a list of species and their hosts. There is still a lack of information on human infections in South America with undetected or unreported infections probably due to the information shortage and little attention by physicians to these small intestinal flukes. Molecular tools for specific diagnoses of South American heterophyid species are still to be defined. Additional new sequences of Pygidiopsis macrostomum, Ascocotyle pindoramensis and Ascocotyle longa from Brazil are also provided.

INTRODUCTION: A new monogenean, Heterocotyle margaritae n. sp. (Monocotylidae: Heterocotylinae), is described based on specimens collected from the gills of the diamond stingray, Hypanus dipterurus (Jordan & Gilbert, 1880) (Myliobatiformes: Dasyatidae) captured off the South Pacific coast, Lima Region, Peru. MATERIALS AND METHODS: Monogeneans were fixed in hot 4% formalin, observed and measured as permanent mounts stained with Gomori's trichrome, and mounted in Canada balsam. Drawings were made with the aid of a drawing tube. RESULTS: The new species is distinguished from all congeners mainly by having a funnel-shaped male copulatory organ, spatulate distally with lateral folds and by its club-shaped accessory piece. The most similar congener, H. americana Hargis, 1955, resemble H. margaritae n. sp. regarding the general structure of the male copulatory organ (funnel-shaped with an accessory piece). However, besides other features, the new species differ from H. americana by having an accessory piece without a dorsal button (vs an accessory piece with a dorsal button in H. americana). CONCLUSIONS: This is the first report of a member of Heterocotyle Scott, 1904 in the Southeastern Pacific Ocean. The present finding brings to 20 the number of known species of Heterocotyle, and represents the fifth described monogenean species from Hypanus dipterurus.

INTRODUCTION: Specimens of Rhinobatonchocotyle pacifica Oliva & Luque, 1995 (Hexabothriidae) were collected from the gill filaments of Pacific guitarfish, Pseudobatos planiceps (Garman, 1880) (Rhinopristiformes: Rhinobatidae), from off the coast of Chorrillos, Lima, Peru. Since the original description of R. pacifica was incomplete, we herein redescribe R. pacifica based on re-examination of the holotype, paratypes, and newly collected voucher specimens. Nucleotide sequences (18S, 28S) for R. pacifica are presented for the first time, and a phylogenetic analysis conducted. MATERIALS AND METHODS: Monogeneans were examined with light microscopy and partial sequences of 18S and 28S rDNA genes were obtained. RESULTS: The following taxonomically important morphological features are described: presence of seminal receptacle, spiniform papillae within oral cavity, haptoral sucker sclerites and anchors, hook surface striations, muscular pad associated with each haptoral sucker sclerite (except sclerite 1), muscular bulbs in appendix suckers, and oötype lacking longitudinal rows of large cells (oötype not côtelé). The genital complex is detailed. The diagnosis of Rhinobatonchocotyle is emended to include species with or without seminal receptacle. Representative nucleotide sequences of partial 18S and 28S rDNA genes of R. pacifica were obtained for the first time and used for reconstructing the phylogeny of Hexabothriidae. CONCLUSION: The phylogenetic reconstruction confirmed the species within the Hexabothriidae.

The taxonomy of species of Dollfusentis is rather confused due to the overlap of morphological traits. The aim of this study was to follow an integrative taxonomy approach over the acanthocephalans collected from Orthopristis ruber in Brazil. Dollfusentis lenti n. sp. is described and is characterised by having an elongate trunk with spines sparsely distributed (largest 60-85 µm long) extending from the neck to almost reach the end of proboscis receptacle; additionally, the new species possesses a long proboscis with 12-14 longitudinal rows of 16-17 hooks each; 3-4 posterior hooks reduced in size, well-spaced from the eight ventrolateral crescent hooks, and lemnisci longer than proboscis receptacle. New sequences of 18S rDNA, ITS1, 5.8S and ITS2, 28S rDNA and COI mtDNA are provided. Dollfusentis bravoae is morphologically similar because it possesses the same number of proboscis hooks, although it differs by the size of testes and uterus and by having a higher number of trunk spines; additionally, new scanning electron micrographs and genetic data for both species support its distinction. Phylogenetic analysis obtained either with two nuclear genes or mitochondrial COI gene showed that Dollfusentis spp. belong to Illiosentidae, and the new species is yielded as the sister species of D. bravoae, with D. chandleri as the sister species of the latter two.

A new genus and species of monogenean belonging to Hexabothriidae, Hypanocotyle bullardi n. gen. n. sp., is described based on specimens collected from the gill filaments of the diamond stingray, Hypanus dipterurus (Jordan et Gilbert) (Myliobatiformes: Dasyatidae), a demersal chondrichthyan collected off the coast of Callao, Peru. Hypanocotyle n. gen. has the following combination of diagnostic features that differentiate it from other hexabothriid genera: haptor symmetrical; vasa efferentia having proximal (narrow, with thin glandular wall) and distal (expanded, interlaced, with thick glandular wall) portions, joining medially to form vas deferens; vas deferens having proximal (expanded, sinuous, with thick glandular wall) and distal (narrow, strongly sinuous, with thin glandular wall) portions; male copulatory organ unarmed, proximal portion slightly sinuous and tube-like, distal portion funnel-shaped; prostatic glands present, distributed around of the MCO; seminal receptacle present; ootype lacking longitudinal rows of large cells (no oötype côtelé); vaginae parallel, with well-differentiated proximal (glandular, narrow, tube-like, slightly sinuous) and distal (musculoglandular, convoluted) portions; gland cells surrounding the vaginal duct along the entire length of distal portion, densely clustered in middle portion; uterine eggs with 2 elongate filaments. Phylogenetic reconstructions by maximum-likelihood method, based on newly obtained partial 18S and 28S sequences, shows that H. bullardi n. gen. is included within the family Hexabothriidae, order Diclybothriidea. This is the second hexabothriid genus recorded from a diamond stingray (Dasyatidae), and the fourth hexabothriid species recorded from Peru. A key to hexabothriid genera is provided.

BACKGROUND: Schistosomiasis has been reported in 78 endemic countries and affects 240 million people worldwide. The digenetic parasite Schistosoma mansoni needs fresh water to compete its life cycle. There, it is susceptible to soluble compounds that can affect directly and/or indirectly the parasite's biology. The cercariae stage is one of the key points in which the parasite is vulnerable to different soluble compounds that can significantly alter the parasite's life cycle. Molluscicides are recommended by the World Health Organization for the control of schistosomiasis transmission and Euphorbia milii latex is effective against snails intermediate hosts. METHODOLOGY/PRINCIPAL FINDINGS: We used parasitological tools and electron microscopy to verify the effects of cercariae exposure to natural molluscicide (Euphorbia milii latex) on morphology, physiology and fitness of adult parasite worms. In order to generate insights into key metabolic pathways that lead to the observed phenotypes we used comparative transcriptomics and proteomics. CONCLUSIONS/SIGNIFICANCE: We describe here that the effect of latex on the adult is not due to direct toxicity but it triggers an early change in developmental trajectory and perturbs cell memory, mobility, energy metabolism and other key pathways. We conclude that latex has not only an effect on the vector but applies also long lasting schistosomastatic action. We believe that these results are of interest not only to parasitologists since it shows that natural compounds, presumably without side effects, can have an impact that occurred unexpectedly on developmental processes. Such collateral damage is in this case positive, since it impacts the true target of the treatment campaign. This type of treatment could also provide a rational for the control of other pests. Our results will contribute to enforce the use of E. milii latex in Brazil and other endemic countries as cheap alternative or complement to mass drug treatment with Praziquantel, the only available drug to cure the patients (without preventing re-infection).

Arapaima gigas, a fish with a high market value, has been farmed in different localities within Brazil. Among its parasites, adults of Goezia spinulosa are reported to cause ulcers in the stomach and to result in the death of farmed fingerlings. Due to the veterinary importance of this nematode in cultured arapaimas, an integrative taxonomic study is proposed, combining morphological, ultrastructural and genetic profiles of this parasite. The fish were obtained from semi-intensive fish farming in Acre State, Brazil. The fish measured 7-42cm in total length and the intensity of infection was 1-60 parasites per fish. The site of infection was mainly the stomach. Morphological and ultrastructural analyses of G. spinulosa showed the importance of its spiny body in firmly attaching the worm to the gastric mucosa, resulting in lesions, ulcers and deep gastric perforations of the stomach wall. New sequences for partial 18S rDNA, ITS1, 5.8S and ITS2 rDNA, partial 28S rDNA, cox1 mtDNA and for cox2 mtDNA are presented. Phylogenetic reconstructions based on the partial 18S and 28S rDNA shows species of Goezia occur in a clade well separated from other genera in both analyses. Both the partial 18S and 28S rDNA genes represented good genetic markers for distinguishing genera of the Raphidascarididae, with exception of Hysterothylacium. This integrated taxonomic study produced a robust profile for G. spinulosa that will aid the diagnosis of both adults and larval stages from arapaimas and possible intermediate hosts.

RESUMEN

We provide a summary overview of the diversity of trematode parasites in freshwater fishes of the 'New World', i.e. the Americas, with emphasis on adult forms. The trematode fauna of three regions, South America, Middle America, and USA and Canada (North America north of Mexico), are considered separately. In total, 462 trematode species have been reported as adults from the Americas. The proportion of host species examined for parasites varies widely across the Americas, from a high of 45% in the Mexican region of Middle America to less than 5% in South America. North and South America share no adult species, and one exclusively freshwater genus, Creptotrema Travassos, Artigas & Pereira, 1928 in the Allocreadiidae Looss, 1902 is the most widely distributed. Metacercariae of strigeiforms maturing in fish-eating birds (e.g. species of the Diplostomidae Poirier, 1886) are common and widely distributed. The review also highlights the paucity of known life-cycles. The foreseeable future of diversity studies belongs to integrative approaches and the application of molecular ecological methods. While opportunistic sampling will remain important in describing and cataloguing the trematode fauna, a better understanding of trematode diversity and biology will also depend on strategic sampling throughout the Americas.

An anisakid nematode larva found in cod sold in the state of Minas Gerais, Brazil was studied by light and scanning electron microscopy and by a molecular approach. Mitochondrial cytochrome c-oxidase subunit 2 (mtDNA cox-2), 28S rRNA and ITS1, 5.8S and ITS2 regions were amplified using the polymerase chain reaction and sequenced to evaluate the phylogenetic relationships of the larva. The genetic profile confirmed that this larva belongs to the species Pseudoterranova decipiens (sensu stricto). This is the first molecular and ultrastructural study of Pseudoterranova decipiens (sensu stricto) in imported cod sold in Brazil. The health implications of these findings are discussed.

Uma larva de Nematoda anisakídeo encontrada em bacalhau comercializado no Estado de Minas Gerais, Brasil foi estudada por microscopias de luz e eletrônica de varredura e por uma abordagem molecular. As regiões da subunidade 2 da citocromo c-oxidase mitocondrial (mtDNA cox-2), 28S rRNA e ITS1, 5.8S e ITS2 foram amplificadas usando a reação em cadeia da polimerase e sequenciadas para avaliar as relações filogenéticas da larva. O perfil genético confirmou que esta larva pertence à espécie Pseudoterranova decipiens (sensu stricto). Esse é o primeiro estudo molecular e ultraestrutural de Pseudoterranova decipiens (sensu stricto) de bacalhau importado vendido no Brasil. As implicações destes resultados para a saúde são discutidas.

Although the Chiroptera represents a significant proportion (c.20%) of the mammalian fauna and South America has the highest diversity of bat species, only about a third of the known species in this region have had helminth parasites reported from them. This work represents the first comprehensive checklist of the helminth parasites (nematodes, acanthocephalans, trematodes and cestodes) of South American bats. The data were extracted from more than 120 references and are presented as a key to each group of parasites down to the generic level, with an indication of how the bats become infected, accompanied by a list of the species recorded for each genus. This is followed, in tabular form, by parasite-host and host-parasite checklists. The parasite-host list also includes their geographical distribution in South America (at the country level) and site data, plus the references in which the parasite records occur. The host-parasite list is arranged according to the classification of the hosts. In all, c.370 host-parasite associations are recorded, involving 114 nominal species of helminths from 92 named chiropteran taxa.

The infection and development of a parasite may cause physiological, morphological and behavioral changes in its host. Changes in the locomotory activity of a host induced by their parasites may also influence the life-cycles of both host and parasite in the environment. The aim of the present work was to evaluate the locomotory activities of Biomphalaria glabrata before and after an experimental infection with Schistosoma mansoni relating to the shedding of cercaria. In addition, the reproductive parameters of infected B. glabrata were analyzed during the prepatent and patent periods of the infection. The locomotory activity was recorded using an image analysis biomonitoring system based on a Videomex V. Five parameters were analyzed: 'Distance traveled', 'Ambulatory time', 'Stereotypic time', 'Resting time' and 'Average speed'. The number of shed cercariae was counted twice at 45 and 52 days post-infection. The reproductive parameters of infected B. glabrata analyzed were the numbers of egg masses, eggs and hatched snails. All statistical analyses were performed using the R program. Of the 69 snails infected with S. mansoni, 33 (47.8%) shed cercariae ('positive') and 36 (52.2%) ('exposed') failed to exhibit any cercarial shedding prior to the end of the experiment. The locomotory activity of the all snails increased significantly after infection with S. mansoni. However, when the 'positive' and 'exposed' snails were compared, the former, shedding cercariae, were less motile. With regard to reproduction, 84.8% (28/33) of the 'positive' and 27.7% (10/36) of the 'exposed' snails failed to lay egg masses during patent period. The number of cercariae individually shed by each 'positive' snail presented a positive relation with 'Stereotypic time' and a negative relation with egg laying. Our findingshighlight the way in which infection with S. mansoni affects the locomotory and the reproductive behavior of B. glabrata. The number of cercariae shed is directly associated with the reduction/interruption in egg-laying and with an increase in random movement.

The sequences presented 99 and 98% of similarity with C. pelagicum sequences from Argentina, deposited on GenBank for the same genetic region and with a strong statistical support inferred from the phylogenetic tree. The morphological and ultrastructural studies that were carried out confirmed the genetic analysis.

Foram determinadas três novas sequências da região do Citocromo c-oxidase da subunidade II do DNA mitocondrial (cox-2 mtDNA) de Contracaecum pelagicum, parasito de Spheniscus magellanicus, pinguim Magalhães, de águas brasileiras. As sequências apresentaram 99 e 98% de similaridade com sequências de C. pelagicum da Argentina depositadas no GenBank da mesma região genética com forte suporte estatístico inferido pela arvore filogenética. Estudos morfológicos e ultraestruturais realizados confirmaram a identidade genética.

RESUMEN

This article presents an improvement to the Kato-Katz (KK) method, making it faster and more efficient for the visualisation of fertile eggs in stool samples. This modified KK method uses sodium acetate formalin as a fixative and reveals the intensity of infection in less than 1 h, reducing the diagnostic time without increasing the cost. This modified method may contribute to future epidemiological studies in both hospitals and the field due to its rapid and precise diagnostic, which allow for immediate treatment.

BACKGROUND: Melanoides tuberculatus (Müller, 1774) (Thiaridae), an introduced gastropod mollusc with a wide geographical distribution in the Neotropics, is the intermediate host of the trematode Centrocestus formosanus (Nishigori, 1924) (Heterophyidae). This parasite is considered to be pathogenic to humans. The aim of the present work was to evaluate the locomotory activity of uninfected M. tuberculatus compared with those naturally infected with C. formosanus. FINDINGS: The locomotory activity of each mollusc was recorded using an image analysis biomonitoring system, Videomex-V, to evaluate and quantify the parameters of 'Stereotypic' and 'Resting time'. The Generalized Estimating Equation analysis of locomotory activity of M. tuberculatus infected with C. formosanus revealed significant differences compared with uninfected molluscs for the parameters 'Stereotypic time' and 'Resting time' with a reduction of movement. The variations in the values of the monitoring intervals recorded showed a significant difference for the infected molluscs in the case of Stereotypic time, with an irregular locomotory activity pattern, as compared to that of uninfected molluscs. The analysis of the standard length of all molluscs did not exhibit any correlation with locomotory activity, showing that C. formosanus is able to alter the locomotory activity of its snail host regardless of the standard length. CONCLUSIONS: The trematode C. formosanus affects the locomotory activity of the mollusc M. tuberculatus by reducing its movement and causing it to exhibit an irregular pattern of activity, both of which are independent of the snail's standard length.

RESUMEN

Brasicystis bennetti Thatcher, 1979 was first described from specimens obtained from the subcutaneous tissues of the mouth and operculum of Plagioscyion squamosissimus from the Amazon River in Brazil, however since 2008, Brasicystis has been considered a genus inquirendum. This study reviews some of the diagnostic characters from the original description of B. bennetti from the Amazon Delta, and redescribes the genus and species with a discussion of their taxonomic status. Ultrastructural and molecular approaches complement the data presented on this monotypic genus. The diagnosis of the tribe Didymozoini Monticelli, 1888 is amended to incorporate the genus Brasicystis, which is redescribed and revalidated here, with the proposal of an amended key.
