Electrical properties estimated from the electrical resistance of the human body can serve as indicators of muscle tissue status and the risk of developing sarcopenia; however, to date, at least to the best of our knowledge, no studies have performed such an assessment in older individuals with advanced dementia. The present study examined the associations between grip strength, body composition and electrical properties using bioimpedance spectroscopy (BIS) in women aged 77-97 years residing in dementia group homes. A total of 33 participants were enrolled with an average age of 88.1±5.2 years; 57.6% of the participants had moderate or severe dementia. The resistance values of the participants were measured in the whole body, upper limbs and lower limbs using BIS, and their body composition, muscle mass index and electrical properties were estimated as indicators of muscle quality. In addition, grip strength was measured and the participants were classified into three groups (high, low and non-measurable) according to their cognitive function. The effect size (partial eta-squared and Cohen's d) was also evaluated. The Shapiro-Wilk test was used to assess the distribution of each variable; variables with non-normal distributions were analyzed following log transformation. Continuous variables were analyzed using a one-way analysis of variance and the Tukey-Kramer post hoc test was used. The post hoc sample size (statistical power: 1-ß) analysis revealed a power of ~80% (i.e., 76.1-88.7%), considering the minimum power for sufficient participants. No differences were found in body composition or muscle mass index among the three grip strength groups. As regards the upper limbs, the electrical properties of the characteristic frequencies were significant (P=0.006; effect size, large), and the membrane capacitance (P=0.005; effect size, large) was significantly higher in the high-dose group than in the other groups. A significant association was detected among grip strength, upper limb characteristic frequency and membrane capacitance. Hence, electrical properties may be an indicator of muscle quality in older women identified as needing care for dementia.

BACKGROUND & AIMS: The quantity gap between daily and loaded carbohydrates may affects blood glucose response to carbohydrate intake; however, no study has investigated the difference in 24-h span. This study aimed to determine differences in the 24-h glucose levels and variability in response to single-day carbohydrate overfeeding based on daily carbohydrate intake in healthy Japanese men. METHODS: Twenty male college students completed a 3-day dietary record and were divided into two groups based on whether their daily carbohydrate intake exceeded the median intake (H-CHO) or not (L-CHO). Thereafter, the participants consumed a high-carbohydrate diet (carbohydrate 8.1 g/kg/d) for 1 day. The 24-h glucose levels and glucose variability (CONGA1) were measured using a continuous glucose monitoring system. RESULTS: The mean daily carbohydrate intakes in the L-CHO and H-CHO groups were 3.9 ± 0.5 and 5.8 ± 0.6 g/kg/d, respectively (p < 0.001). The peak 24-h glucose level was not differ between the L-CHO group and the H-CHO group (8.0 ± 0.8 vs. 8.0 ± 1.0; p = 0.886). The mean 24-h glucose level was higher in the L-CHO group than in the H-CHO group (6.0 ± 0.3 vs. 5.6 ± 0.3 mmol/L; p = 0.010). The CONGA1 was higher in the L-CHO group than in the H-CHO group (5.40 ± 0.41 vs. 4.95 ± 0.25; p = 0.008). CONCLUSIONS: Mean glucose level and glucose variability in response to carbohydrate overfeeding were high in the individuals with small daily carbohydrate intake. These findings suggest that the large quantity gap between daily and loaded carbohydrates cause worse glucose control during carbohydrate overfeeding.

BACKGROUND: Bioelectrical impedance analysis (BIA) is a minimally invasive, safe, easy, and quick technology used to determine body composition. OBJECTIVES: We compared the relationship among impedance indices obtained using single-frequency BIA, multi-frequency BIA, bioelectrical impedance spectroscopy (BIS), and skeletal muscle mass (SMM) of physically active young men and athletes using the creatine (methyl-d3) dilution method. We also compared the SMM and intracellular water (ICW) of athletes and active young men measured using a reference stable isotope dilution and BIS method, respectively. METHODS: We analyzed data from 28 men (mean age, 20 ± 2 y) who exercised regularly. Single-frequency BIA at 5 kHz and 50 kHz (R5 and R50), multi-frequency BIA (R250-5), and BIS (RICW) methods of determining the SMM were compared. The deuterium and sodium bromide dilution methods of obtaining the total body water, ICW, and extracellular water measurements were also used, and the results were compared to those acquired using bioimpedance methods. RESULTS: The correlation coefficients between SMM and L2/R5, L2/R50, L2/R250-5, and L2/RICW were 0.738, 0.762, 0.790, and 0.790, respectively (P < 0.01). The correlation coefficients between ICW and L2/R5, L2/R50, L2/R250-5, and L2/RICW were 0.660, 0.687, 0.758, and 0.730, respectively (P < 0.001). However, the correlation coefficients of L2/R50, L2/R250-5, and L2/RICW for SMM and ICW were not significantly different. CONCLUSIONS: Our findings suggest that single-frequency BIA at L2/R50, multi-frequency BIA, and BIS are valid for assessing the SMM of athletes and active young men. Additionally, we confirmed that the SMM and ICW were correlated with single-frequency BIA, multi-frequency BIA, and BIS. Bioimpedance technologies may be dependable and practical means for assessing SMM and hydration compartment status of active young adult males; however, cross-validation is needed.

Muscle fiber composition is associated with physical performance, with endurance athletes having a high proportion of slow-twitch muscle fibers compared to power athletes. Approximately 45% of muscle fiber composition is heritable, however, single nucleotide polymorphisms (SNP) underlying inter-individual differences in muscle fiber types remain largely unknown. Based on three whole genome SNP datasets, we have shown that the rs236448 A allele located near the cyclin-dependent kinase inhibitor 1A (CDKN1A) gene was associated with an increased proportion of slow-twitch muscle fibers in Russian (n = 151; p = 0.039), Finnish (n = 287; p = 0.03), and Japanese (n = 207; p = 0.008) cohorts (meta-analysis: p = 7.9 × 10−5. Furthermore, the frequency of the rs236448 A allele was significantly higher in Russian (p = 0.045) and Japanese (p = 0.038) elite endurance athletes compared to ethnically matched power athletes. On the contrary, the C allele was associated with a greater proportion of fast-twitch muscle fibers and a predisposition to power sports. CDKN1A participates in cell cycle regulation and is suppressed by the miR-208b, which has a prominent role in the activation of the slow myofiber gene program. Bioinformatic analysis revealed that the rs236448 C allele was associated with increased CDKN1A expression in whole blood (p = 8.5 × 10−15) and with greater appendicular lean mass (p = 1.2 × 10−5), whereas the A allele was associated with longer durations of exercise (p = 0.044) reported amongst the UK Biobank cohort. Furthermore, the expression of CDKN1A increased in response to strength (p < 0.0001) or sprint (p = 0.00035) training. Accordingly, we found that CDKN1A expression is significantly (p = 0.002) higher in the m. vastus lateralis of strength athletes compared to endurance athletes and is positively correlated with the percentage of fast-twitch muscle fibers (p = 0.018). In conclusion, our data suggest that the CDKN1A rs236448 SNP may be implicated in the determination of muscle fiber composition and may affect athletic performance.

Daily muscle glycogen recovery after training is important for athletes. Few studies have reported a continuous change in muscle glycogen for 24 h. We aimed to investigate the changes in carbohydrate intake amount on muscle glycogen recovery for 24 h after exercise using 13C-magnetic resonance spectroscopy (13C-MRS). In this randomized crossover study, eight male participants underwent prolonged high-intensity exercise, and then consumed one of the three carbohydrate meals (5 g/kg body mass (BM)/d, 7 g/kg BM/d, or 10 g/kg BM/d). Glycogen content of thigh muscle was measured using 13C-MRS before, immediately after, and 4 h, 12 h and 24 h after exercise. Muscle glycogen concentration decreased to 29.9 ± 15.9% by exercise. Muscle glycogen recovery 4−12 h after exercise for the 5 g/kg group was significantly lower compared to those for 7 g/kg and 10 g/kg groups (p < 0.05). Muscle glycogen concentration after 24 h recovered to the pre-exercise levels for 7 g/kg and 10 g/kg groups; however, there was a significant difference for the 5 g/kg group (p < 0.05). These results suggest that carbohydrate intake of 5 g/kg BM/d is insufficient for Japanese athletes to recover muscle glycogen stores 24 h after completing a long-term high-intensity exercise.

Human skeletal muscle fiber is heterogenous due to its diversity of slow- and fast-twitch fibers. In human, slow-twitched fiber gene expression is correlated to MOTS-c, a mitochondria-derived peptide that has been characterized as an exercise mimetic. Within the MOTS-c open reading frame, there is an East Asian-specific m.1382A>C polymorphism (rs111033358) that changes the 14th amino acid of MOTS-c (i.e., K14Q), a variant of MOTS-c that has less biological activity. Here, we examined the influence of the m.1382A>C polymorphism causing MOTS-c K14Q on skeletal muscle fiber composition and physical performance. The myosin heavy chain (MHC) isoforms (MHC-I, MHC-IIa, and MHC-IIx) as an indicator of muscle fiber composition were assessed in 211 Japanese healthy individuals (102 men and 109 women). Muscular strength was measured in 86 physically active young Japanese men by using an isokinetic dynamometer. The allele frequency of the m.1382A>C polymorphism was assessed in 721 Japanese athletes and 873 ethnicity-matched controls. The m.1382A>C polymorphism genotype was analyzed by TaqMan SNP Genotyping Assay. Individuals with the C allele of the m.1382A>C exhibited a higher proportion of MHC-IIx, an index of fast-twitched fiber, than the A allele carriers. Men with the C allele of m.1382A>C exhibited significantly higher peak torques of leg flexion and extension. Furthermore, the C allele frequency was higher in the order of sprint/power athletes (6.5%), controls (5.1%), and endurance athletes (2.9%). Additionally, young male mice were injected with the MOTS-c neutralizing antibody once a week for four weeks to mimic the C allele of the m.1382A>C and assessed for protein expression levels of MHC-fast and MHC-slow. Mice injected with MOTS-c neutralizing antibody showed a higher expression of MHC-fast than the control mice. These results suggest that the C allele of the East Asian-specific m.1382A>C polymorphism leads to the MOTS-c K14Q contributes to the sprint/power performance through regulating skeletal muscle fiber composition.

Muscle glycogen is a crucial energy source for exercise, and assessment of muscle glycogen storage contributes to the adequate manipulation of muscle glycogen levels in athletes before and after training and competition. Muscle biopsy is the traditional and gold standard method for measuring muscle glycogen; alternatively, 13C magnetic resonance spectroscopy (MRS) has been developed as a reliable and non-invasive method. Furthermore, outcomes of ultrasound and bioimpedance methods have been reported to change in association with muscle glycogen conditions. The physiological mechanisms underlying this activity are assumed to involve a change in water content bound to glycogen; however, the relationship between body water and stored muscle glycogen is inconclusive. In this review, we discuss currently available muscle glycogen assessment methods, focusing on 13C MRS. In addition, we consider the involvement of muscle glycogen in changes in body water content and discuss the feasibility of ultrasound and bioimpedance outcomes as indicators of muscle glycogen levels. In relation to changes in body water content associated with muscle glycogen, this review broadens the discussion on changes in body weight and body components other than body water, including fat, during carbohydrate loading. From these discussions, we highlight practical issues regarding muscle glycogen assessment and manipulation in the sports field.

BACKGROUND: Severe rapid weight loss (RWL) induces a decrease in muscle glycogen (mGly). Nevertheless, adequate carbohydrate intake after RWL has not been reported to optimize muscle glycogen following a weigh-in the evening until a wrestling tournament morning. The purpose of this study was to investigate the effect of an overnight high-carbohydrate recovery meal of 7.1 g kg-1 following RWL on mGly concentration. METHODS: Ten male elite wrestlers lost 6% of their body mass within 53 h and then subsequently ate three meals, within 5 h, containing total of 7.1 g kg-1 of carbohydrates. mGly was measured by 13C-magnetic resonance spectroscopy before (BL) and after RWL (R0) at 2 h (R2), 4 h (R4), and 13 h (R13) after initiating the meal. Body composition, muscle cross-sectional area, and blood and urine samples were collected at BL, R0, and R13. RESULTS: Body mass decreased by 4.6 ± 0.6 kg (p < 0.05) and did not recover to BL levels in R13 (- 1.7 ± 0.6 kg, p < 0.05). Likewise, mGly by 36.5% ± 10.0% (p < 0.05) and then did not reach BL levels by R13 (p < 0.05). CONCLUSION: A high-carbohydrate meal of 7.1 g kg-1 after 6% RWL was not sufficient to recover mGly during a 13 h recovery phase. Participating in high-intensity wrestling matches with an mGly concentration below normal levels is maybe undesirable.

Human muscle fiber composition is heterogeneous and mainly determined by genetic factors. A previous study reported that experimentally induced iron deficiency in rats increases the proportion of fast-twitch muscle fibers. Iron status has been reported to be affected by genetic factors. As the TMPRSS6 rs855791 T/C and HFE rs1799945 C/G polymorphisms are strongly associated with iron status in humans, we hypothesized that the genotype score (GS) based on these polymorphisms could be associated with the muscle fiber composition in humans. Herein, we examined 214 Japanese individuals, comprising of 107 men and 107 women, for possible associations of the GS for iron status with the proportion of myosin heavy chain (MHC) isoforms (I, IIa, and IIx) as markers of muscle fiber composition. No statistically significant correlations were found between the GS for iron status and the proportion of MHC isoforms in all participants. When the participants were stratified based on sex, women showed positive and negative correlations of the GS with MHC-IIa (age-adjusted p = 0.020) and MHC-IIx (age-adjusted p = 0.011), respectively. In contrast, no correlation was found in men. In women, a 1-point increase in the GS was associated with 2.42% higher MHC-IIa level and 2.72% lower MHC-IIx level. Our results suggest that the GS based on the TMPRSS6 rs855791 T/C and HFE rs1799945 C/G polymorphisms for iron status is associated with muscle fiber composition in women.

PURPOSE: To replicate previous genome-wide association study identified sprint-related polymorphisms in 3 different cohorts of top-level sprinters and to further validate the obtained results in functional studies. METHODS: A total of 240 Japanese, 290 Russians, and 593 Brazilians were evaluated in a case-control approach. Of these, 267 were top-level sprint/power athletes. In addition, the relationship between selected polymorphisms and muscle fiber composition was evaluated in 203 Japanese and 287 Finnish individuals. RESULTS: The G allele of the rs3213537 polymorphism was overrepresented in Japanese (odds ratio [OR]: 2.07, P = .024) and Russian (OR: 1.93, P = .027) sprinters compared with endurance athletes and was associated with an increased proportion of fast-twitch muscle fibers in Japanese (P = .02) and Finnish (P = .041) individuals. A meta-analysis of the data from 4 athlete cohorts confirmed that the presence of the G/G genotype rather than the G/A+A/A genotypes increased the OR of being a sprinter compared with controls (OR: 1.49, P = .01), endurance athletes (OR: 1.79, P = .001), or controls + endurance athletes (OR: 1.58, P = .002). Furthermore, male sprinters with the G/G genotype were found to have significantly faster personal times in the 100-m dash than those with G/A+A/A genotypes (10.50 [0.26] vs 10.76 [0.31], P = .014). CONCLUSION: The rs3213537 polymorphism found in the CPNE5 gene was identified as a highly replicable variant associated with sprinting ability and the increased proportion of fast-twitch muscle fibers, in which the homozygous genotype for the major allele (ie, the G/G genotype) is preferable for performance.

Bioimpedance spectroscopy (BIS) is an easy tool to assess hydration status and body composition. However, its validity in athletes remains controversial. We investigated the validity of BIS on total body water (TBW) and body composition estimation in Japanese wrestlers and untrained subjects. TBW of 49 young Japanese male subjects (31 untrained, 18 wrestlers) were assessed using the deuterium dilution method (DDM) and BIS. De Lorenzo's and Moissl's equations were employed in BIS for TBW estimation. To evaluate body composition, Siri's 3-compartment model and published TBW/fat-free mass (FFM) ratio were applied in DDM and BIS, respectively. In untrained subjects, DDM and BIS with de Lorenzo's equation showed consistent TBW estimates, whereas BIS with Moissl's equation overestimated TBW (p < 0.001 vs. DDM). DDM and BIS with de Lorenzo's equation estimated FFM and percent of fat mass consistently, whereas BIS with Moissl's equation over-estimated and under-estimated them (p < 0.001 vs. DDM). In wrestlers, BIS with de Lorenzo's and Moissl's equations assessed TBW similarly with DDM. However, the Bland-Altman analysis revealed a proportional bias for TBW in BIS with de Lorenzo's equation (r = 0.735, p < 0.001). Body composition assessed with BIS using both equations and DDM were not different. In conclusion, BIS with de Lorenzo's equation accurately estimates the TBW and body composition in untrained subjects, whereas BIS with Moissl's equation is more valid in wrestlers. Our results demonstrated the usefulness of BIS for assessing TBW and body composition in Japanese male wrestlers.

PPARGC1A rs8192678 G/A (Gly482Ser) and NRF1 rs6949152 A/G polymorphisms have been associated with endurance athlete status, endurance performance phenotypes, and certain health-related markers of different pathologies such as metabolic syndrome, diabetes, and dyslipidemia. We hypothesized that they could be considered interesting candidates for explaining inter-individual variations in muscle fiber composition in humans. We aimed to examine possible associations of these polymorphisms with myosin heavy-chain (MHC) isoforms as markers of muscle fiber compositions in vastus lateralis muscle in a population of 214 healthy Japanese subjects, aged between 19 and 79 years. No significant associations were found in men for any measured variables. In contrast, in women, the PPARGC1A rs8192678 A/A genotype was significantly associated with a higher proportion of MHC-I (p = 0.042) and with a lower proportion of MHC-IIx (p = 0.033), and the NRF1 rs6949152 AA genotype was significantly associated with a higher proportion of MHC-I (p = 0.008) and with a lower proportion of MHC IIx (p = 0.035). In women, the genotype scores of the modes presenting the most significant results for PPARGC1A rs8192678 G/A (Gly482Ser) and NRF1 rs6949152 A/G polymorphisms were significantly associated with MHC-I (p = 0.0007) and MHC IIx (p = 0.0016). That is, women with combined PPARGC1A A/A and NRF1 A/A genotypes presented the highest proportion of MHC-I and the lowest proportion of MHC-IIx, in contrast to women with combined PPARGC1A GG+GA and NRF1 AG+GG genotypes, who presented the lowest proportion of MHC-I and the highest proportion of MHC-IIx. Our results suggest possible associations between these polymorphisms (both individually and in combination) and the inter-individual variability observed in muscle fiber composition in women, but not in men.

We investigated the effect of muscle damage and inflammation on electrical resistance and the body composition assessment by using bioimpedance spectroscopy (BIS). Twenty-two subjects completed 30 repetitions of maximal eccentric contractions of the elbow flexors with one arm. Whole-body resistance of extracellular and intracellular components (Re and Ri, respectively) on the exercised and non-exercised sides were measured using BIS. Body composition was calculated from both sides of resistance at baseline and 96 h after exercise. Re decreased only on the exercised side at 96 h after exercise (P < 0.05). Fat-free and fat mass values estimated from resistance on the exercised side were altered by 3.1% and - 15.6%, respectively, at 96 h after exercise (P < 0.05); those estimated from the non-exercised side were unaltered. Eccentric exercise-induced muscle damage and inflammation reduce Re and induce non-negligible estimation error in the body composition assessment using BIS.

We investigated the effect of rapid weight loss (RWL) and weight regain (WR) on thoracic gas volume (VTG) and body composition assessment using air displacement plethysmography (ADP) in male wrestlers. Eight male elite collegiate wrestlers completed a RWL regimen (6% of body mass) over a 53-h period, which was followed by a 13-h WR period. ADP was used at three time points (baseline (T1), post-RWL (T2) and post-WR (T3)) according to the manufacturer's testing recommendations. The total body water and bone mineral content were estimated using the stable isotope dilution method and dual energy X-ray absorptiometry, respectively, at the same time points. Body composition was assessed with two-component (2C) or four-component (4C) models using either the measured VTG (mVTG) or predicted VTG (pVTG). Measured VTG increased from T1 to T2 (0.36 ± 0.31 L, p < 0.05) and decreased from T2 to T3 (-0.29 ± 0.15 L, p < 0.01). However, the changes in fat mass and fat free mass, which were calculated by both 2C and 4C models, were not significantly different when compared between calculations using mVTG and those using pVTG. Our results indicate that VTG significantly changes during RWL and WR, but both measured and predicted VTG can be used to assess changes in body composition during RWL and WR.

To determine energy density for rapid weight loss (RWL) of weight-classified sports, eight male elite wrestlers were instructed to lose 6% of body mass (BM) within 53 h. Energy deficit during the RWL was calculated by subtracting total energy expenditure (TEE) determined using the doubly labeled water method (DLW) from energy intake (EI) assessed with diet records. It was also estimated from body composition change estimated with the four-component model (4C) and other conventional methods. BM decreased significantly by 4.7 ± 0.5 kg (6.4 ± 0.5%). Total body water loss was the major component of the BM loss (71.0 ± 7.6%). TEE was 9446 ± 1422 kcal, and EI was 2366 ± 1184 kcal during the RWL of 53-h; therefore, the energy deficit was 7080 ± 1525 kcal. Thus, energy density was 1507 ± 279 kcal/kg ∆BM during the RWL, comparable with values obtained using the 4C, three-component model, dual energy X-ray absorptiometry, and stable isotope dilution. Energy density for RWL of wrestlers is lower than that commonly used (7400 or 7700 kcal/kg ΔBM). Although RWL is not recommended, we propose that commonly practiced extreme energy restriction such as 7400 or 7700 kcal/kg ΔBM during RWL appears to be meaningless.

Although each gram of glycogen is well known to bind 2.7-4.0 g of water, no studies have been conducted on the effect of muscle glycogen depletion on body water distribution. We investigated changes in extracellular and intracellular water (ECW and ICW) distribution in each body segment in muscle glycogen-depletion and glycogen-recovery condition using segmental bioimpedance spectroscopy technique (BIS). Twelve male subjects consumed 7.0 g/kg body mass of indigestible (glycogen-depleted group) or digestible (glycogen-recovered group) carbohydrate for 24 h after a glycogen-depletion cycling exercise. Muscle glycogen content using 13C-magnetic resonance spectroscopy, blood hydration status, body composition, and ECW and ICW content of the arm, trunk, and leg using BIS were measured. Muscle glycogen content at the thigh muscles decreased immediately after exercise (glycogen-depleted group, 71.6 ± 12.1 to 25.5 ± 10.1 mmol/kg wet wt; glycogen-recovered group, 76.2 ± 16.4 to 28.1 ± 16.8 mmol/kg wet wt) and recovered in the glycogen-recovered group (72.7 ± 21.2 mmol/kg wet wt) but not in the glycogen-depleted group (33.2 ± 12.6 mmol/kg wet wt) 24 h postexercise. Fat-free mass decreased in the glycogen-depleted group ( P < 0.05) but not in the glycogen-recovered group 24 h postexercise. However, no changes were observed in ECW and ICW content at the leg in both groups. Our results suggested that glycogen depletion per se does not alter body water distribution as estimated via BIS. This information is valuable in assessing body composition using BIS in athletes who show variable glycogen status during training and recovery. NEW & NOTEWORTHY Segmental bioimpedance spectroscopy analysis reveals the effect of muscle glycogen depletion on body segmental water distribution in controlled conditions. Despite the significant difference in the muscle glycogen levels at the leg, no difference was observed in body resistance and the corresponding water content of the extracellular and intracellular compartments.

Genetic polymorphisms and sex differences are suggested to affect muscle fiber composition; however, no study has investigated the effects of genetic polymorphisms on muscle fiber composition with respect to sex differences. Therefore, the present study examined the effects of genetic polymorphisms on muscle fiber composition with respect to sex differences in the Japanese population. The present study included 211 healthy Japanese individuals (102 men and 109 women). Muscle biopsies were obtained from the vastus lateralis to determine the proportion of myosin heavy chain (MHC) isoforms (MHC-I, MHC-IIa, and MHC-IIx). Moreover, we analyzed polymorphisms in α-actinin-3 gene ( ACTN3; rs1815739 ), angiotensin-converting enzyme gene ( ACE; rs4341 ), hypoxia-inducible factor 1 α gene ( rs11549465 ), vascular endothelial growth factor receptor 2 gene ( rs1870377 ), and angiotensin II receptor, type 2 gene ( rs11091046 ), by TaqMan single-nucleotide polymorphism genotyping assays. The proportion of MHC-I was 9.8% lower in men than in women, whereas the proportion of MHC-IIa and MHC-IIx was higher in men than in women (5.0 and 4.6%, respectively). Men with the ACTN3 RR + RX genotype had a 4.8% higher proportion of MHC-IIx than those with the ACTN3 XX genotype. Moreover, men with the ACE ID + DD genotype had a 4.7% higher proportion of MHC-I than those with the ACE II genotype. Furthermore, a combined genotype of ACTN3 R577X and ACE insertion/deletion (I/D) was significantly correlated with the proportion of MHC-I ( r = -0.23) and MHC-IIx ( r = 0.27) in men. In contrast, no significant correlation was observed between the examined polymorphisms and muscle fiber composition in women. These results suggest that the ACTN3 R577X and ACE I/D polymorphisms independently affect the proportion of human skeletal muscle fibers MHC-I and MHC-IIx in men but not in women. NEW & NOTEWORTHY In men, the RR + RX genotype of the α-actinin-3 gene ( ACTN3) R577X polymorphism was associated with a higher proportion of myosin heavy chain (MHC)-IIx. The ID + DD genotype of the angiotensin-converting enzyme gene ( ACE) insertion/deletion (I/D) polymorphism, in contrast to a previous finding, was associated with a higher proportion of MHC-I in men. In addition, the combined genotype of these polymorphisms was correlated with the proportion of MHC-I and MHC-IIx in men. Thus ACTN3 R577X and ACE I/D polymorphisms influence the muscle fiber composition in Japanese men.

Near-infrared spectroscopy (NIRS) shows two types of tissue blood volume (BV) parameters: the total hemoglobin concentration (cHb; assessed by continuous-wave NIRS) and the normalized tissue hemoglobin index (nTHI; evaluated by spatially resolved NIRS). This study was performed to investigate the differences between cHb and nTHI, estimated by calibration using acute reduction of BV at the onset of exercise. Seven active male volunteers (age: 22  ±  4 years) performed a prolonged (60-min) cycling exercise test at 65% or 70% of the peak oxygen uptake. During the tests, cHb and nTHI from the vastus lateralis muscle were monitored. The two parameters were expressed relative to the resting value of 100% and the minimum value of 0% at the onset of exercise. The value of cHb started to increase after arriving at the minimum value and continued until the end of exercise. Similar to cHb, nTHI increased acutely from the lowest level, but the parameter remained almost unchanged from ~5 min throughout the test. Therefore, the two parameters interacted significantly. Moreover, in comparison with the same exercise duration, cHb was significantly higher than nTHI from the 28th min to the end of the test. These results suggest that cHb and nTHI in working muscle are not synchronized during prolonged exercise, especially after ~30 min. The differences between the two BV parameters are likely partly attributable to other factors, such as the increase in skin blood flow.

Oxygenation, measured by near-infrared spectroscopy (NIRS), in inactive muscles decreases during high-intensity exercise because of the decrease of oxygen supply. However, there have been few reports regarding recovery of inactive muscle oxygenation after exercise. This study was performed to examine reoxygenation in the biceps brachii muscle (BB) after supramaximal cycling exercise. Six active young male volunteers (age: 22  ± â€…3 years, peak oxygen uptake (VO2peak): 53.8  ± â€…5.4 mL/kg/min, mean  ± â€…S.D.) performed warm-up exercise, followed by cycling exercise at 140% of VO2peak for 30 s and then at 0 watt for 4 min (recovery exercise). Tissue oxygen saturation (StO2) in the vastus lateralis muscle (VL) and BB was monitored by spatially resolved NIRS throughout the test. The decrease rate of StO2 during exercise was 24.7  ± â€…7.5 p.p. in VL and 15.1  ± â€…8.2 p.p. in BB (N.S.). StO2 in VL increased immediately after the end of exercise, but StO2 in BB decreased continuously for 12.7  ± â€…7.8 s after exercise. Moreover, the time of half-recovery from the minimum to peak values after exercise was significantly (P â€…< â€…0.05) longer at StO2 in BB (39.5  ± â€…12.2 s) than VL (25.2  ± â€…6.0 s). These results suggest that the recovery of microvascular oxygen supply and blood flow in inactive muscles does not start immediately after exercise.

Estimated energy requirements (EERs) are important for sports based on body weight classifications to aid in weight management. The basis for establishing EERs varies and includes self-reported energy intake (EI), predicted energy expenditure, and measured daily energy expenditure. Currently, however, no studies have been performed with male wrestlers using the highly accurate and precise doubly labeled water (DLW) method to estimate energy and fluid requirement. The primary aim of this study was to compare total energy expenditure (TEE), self-reported EI, and the difference in collegiate wrestlers during a normal training period using the DLW method. The secondary aims were to measure the water turnover and the physical activity level (PAL) of the athletes, and to examine the accuracy of two currently used equations to predict EER. Ten healthy males (age, 20.4±0.5 y) belonging to the East-Japan college league participated in this study. TEE was measured using the DLW method, and EI was assessed with self-reported dietary records for ~1 wk. There was a significant difference between TEE (17.9±2.5 MJ•d-1 [4,283±590 kcal•d-1]) and self-reported EI (14.4±3.3 MJ•d-1 [3,446±799 kcal•d-1]), a difference of 19%. The water turnover was 4.61±0.73 L•d-1. The measured PAL (2.6±0.3) was higher than two predicted values during the training season and thus the two EER prediction equations produced underestimated values relative to DLW. We found that previous EERs were underestimating requirements in collegiate wrestlers and that those estimates should be revised.
