RESUMEN

Process analytical technologies are widely used to inform process control by identifying relationships between reagents and products. Here, we present a novel process analytical technology system for operando XAS on multiphase multicomponent synthesis processes based on the combination of a conventional lab-scale agitated reactor with a liquid-jet cell. The preparation of sulfonate-stabilized CaCO3 particles from polyphasic Ca(OH)2 dispersions was monitored in real time by Ca K-edge XAS to identify changes in Ca speciation in the bulk solution/dispersion as a function of time and process conditions. Linear combination fitting of the spectra quantitatively resolved composition changes from the initial conversion of Ca(OH)2 to the Ca(R-SO3)2 surfactant to the ultimate formation of nCaCO3·mCa(R- SO3)2 particles. The system provides a novel tool with strong chemical specificity for probing multiphase synthesis processes at a molecular level, providing an avenue to establishing the relationships between critical quality attributes of a process and the quality and performance of the product.

RESUMEN

It is important to be able to identify the precise position of H-atoms in hydrogen bonding interactions to fully understand the effects on the structure and properties of organic crystals. Using a combination of near-edge X-ray absorption fine structure (NEXAFS) spectroscopy and density functional theory (DFT) quantum chemistry calculations, we demonstrate the sensitivity of core-level X-ray spectroscopy to the precise H-atom position within a donor-proton-acceptor system. Exploiting this sensitivity, we then combine the predictive power of DFT with the experimental NEXAFS, confirming the H-atom position identified using single-crystal X-ray diffraction (XRD) techniques more easily than using other H-atom sensitive techniques, such as neutron diffraction. This proof of principle experiment confirms the H-atom positions in structures obtained from XRD, providing evidence for the potential use of NEXAFS as a more accurate and easier method of locating H-atoms within organic crystals.

RESUMEN

A detailed understanding of the interactions between drugs and their targets is crucial to develop the best possible therapeutic agents. Structure-based drug design relies on the availability of high-resolution structures obtained primarily through X-ray crystallography. Collecting and analysing quickly a large quantity of structural data is crucial to accelerate drug discovery pipelines. Researchers from academia and industry can access the highly automated macromolecular crystallography (MX) beamlines of Diamond Light Source, the UK national synchrotron, to rapidly collect diffraction data from large numbers of crystals. With seven beamlines dedicated to MX, Diamond offers bespoke solutions for a wide variety of user requirements. Working in synergy with state-of-the-art laboratories and other life science instruments to provide an integrated offering, the MX beamlines provide innovative and multidisciplinary approaches to the determination of structures of new pharmacological targets as well as the efficient study of protein-ligand complexes.

RESUMEN

Structure-based drug design has become a key tool for the development of novel drugs. The process involves elucidating the three-dimensional structure of the potential drug molecule bound to the target protein that has been identified as playing a key role in the disease state. Using this three-dimensional information facilitates the process of making improvements to the potential drug molecule by highlighting existing and possible new interactions within the binding site. This knowledge is used to inform increases in potency and selectivity of the molecules as well as to help improve other drug-like properties. The speed and numbers of samples that can be studied, combined with the improved resolution of the structures that can be obtained using synchrotron radiation, have had a significant impact on the utilization of crystallography in the drug discovery process.

RESUMEN

Heterobimetallic complexes of a vanadium(V) and rhenium(VII) combination connected by a mu-oxido bridge [LVO(mu-O)ReO 3].H 2O [H 2L = N, N'-ethylene bis(salicylideneimine) (H 2salen) and its methoxy derivative] ( 1, 2) are reported. The compounds have been prepared by a single-pot synthesis in which the precursor [V (IV)OL] complexes are allowed to be oxidized aerially in the presence of added perrhenate. The oxidized [V (V)OL] (+) species accommodate the ReO 4 (-) anion in their vacant coordination site, trans to the terminal oxido group, providing the complexes 1 and 2. The later generates a binuclear oxovanadium(V) compound [H 2en][(TBC)VO(mu-TBC) 2OV(TBC)].5H 2O ( 3) when treated with tetrabromocatechol. Single crystal X-ray diffraction analysis and (1)H NMR spectroscopy have been used to establish their identities. In compound 2, the Re(1)-O(11)-V(1) bridge angle is barely linear [170.2(3) degrees ] with a Re...V separation of 3.9647(9) A. The redox behavior of 1 and 2 are quite interesting, each undergoing two reductions both in the positive potential range at E 1/2 = 0.59 (process I) and E 1/2 = 0.16 V (process II) versus Ag/AgCl reference (corresponding potentials are 0.59 and 0.18 V for 2). Process I has a single-electron stoichiometry involving the [VO(salen)] part of the complexes as established by combined coulometry-Electron Paramagnetic Resonance (EPR) experiments which provide an eight-line isotropic EPR pattern at room temperature ( = 1.967; = 87 x 10 (-4) cm (-1)), characteristic of an unpaired electron being coupled to a vanadium nuclear spin ( (51)V, I = 7/2). The almost linear V-O-Re bridge in 1 and 2 allows this unpaired electron to interact effectively with the neighboring Re nuclear spin, leading to familiar " two-line pattern" superhyperfine coupling ( A ( (185,187)Re) = 20.7 x 10 (-4) cm (-1)). Process II, on the other hand, is based on a Re(VII/VI) electron transfer as confirmed by differential pulse and normal pulse voltammetric experiments.

The polydentate ligand 2,4,6-tris(dipyridin-2-ylamino)-1,3,5-triazine (dpyatriz) in combination with the Cu(ClO 4) 2/CuX 2 salt mixtures (X (-) = Cl (-), Br (-), or N 3 (-)) leads to the formation of molecular coordination aggregates with formulas [Cu 3Cl 3(dpyatriz) 2](ClO 4) 3 ( 2), [Cu 3Br 3(dpyatriz) 2](ClO 4) 3 ( 3), and [Cu 4(N 3) 4(dpyatriz) 2(DMF) 4(ClO 4) 2](ClO 4) 2 ( 4). These complexes consist of two dpyatriz ligands bridged via coordination to Cu (II) and disposed either face-to-face in an eclipsed manner ( 2 and 3) or parallel and mutually shifted in one direction. The copper ions complete their coordination positions with Cl (-) ( 2), Br (-) ( 3), or N 3 (-), ClO 4 (-), and N, N-dimethylformamide (DMF) ( 4) ligands. All complexes crystallize together with noncoordinate ClO 4 (-) groups that display anion...pi interactions with the triazine rings. These interactions have been studied by means of high level ab initio calculations and the MIPp partition scheme. These calculations have proven the ClO 4 (-)...[C 3N 3] interactions to be favorable and have revealed a synergistic effect from the combined occurrence of pi-pi stacking of triazine rings and the interaction of these moieties with perchlorate ions, as observed in the experimental systems.
