RESUMEN

The Culicidae mosquito family breeds in various natural and artificial water bodies. Phytotelmata are plant structures that hold water and are used by many mosquito species during their life cycle. Mosquitoes are disease vectors, and phytotelmata play an important role in harbouring them. Investigating these phytotelmata will facilitate to develop appropriate site specific vector control strategy. Approximately 750 phytotelmata in 16 locations on Car Nicobar Island was examined. Mosquito larvae were collected from different habitats such as tree holes, root holes, leaf axils, and bamboo stumps. On average, the number of larvae per 10 ml of water ranged from 0.003 to 3.833. Among the 16 mosquito species identified, there were vectors of malaria (Anopheles sundaicus), dengue (Aedes aegypti and Aedes albopictus) and filariasis (Culex quinquefasciatus), belonging to seven genera. The most common phytotelmata were coconut trees (17.5%) followed by Java apple trees (10.3%) and bamboo stumps (7.1%). The most widespread mosquito species was Aedes edwardsi, followed by Aedes malayensis and Ae. albopictus, and the per cent of phytotelmata that contained these species were 42.6, 22.0, and 16.8% respectively. Anopheles mosquitoes had the highest container index (6.1%) in root holes, while Aedes and Culex mosquitoes were more frequently found in bamboo stumps. The alpha biodiversity indices indicated that the mosquito population in Car Nicobar Island was highly diverse (S = 16; DMg = 36.9). Tree holes showed the highest diversity, as all 16 species were collected from them. Assessment of phytotelmata, there is a significant relationship observed between mosquito genus, village, habitat, and tree families, on the remote island of Car Nicobar. Considering the presence of disease-transmitting mosquito vectors in the phytotelmata, these habitats should be considered when implementing control measures to eliminate mosquito-borne infections.

Theobaldia indica Edwards, 1920, classified as a subspecies of Culiseta (Culiseta) alaskaensis (Ludlow, 1906), was recently restored to its original species status, as Culiseta (Culiseta) indica (Edwards), based on comprehensive assessments of morphological, ecological and distributional data. Predating the recognition of indica as a separate species, we collected specimens of this nominal taxon in the Western Himalayan region of India during 20152017, and used morphological and molecular studies to assess its taxonomic status. Our detailed examination of adult morphology, male genitalia and larval characteristics revealed significant distinctions between putative Cs. alaskaensis alaskaensis and Cs. alaskaensis indica, and sequencing of the barcode region of the mitochondrial COI gene yielded a Kimura 2 parameter genetic distance of 5.9% between them, well above the standard 23% commonly accepted to indicate separate species. These results strongly support the formal recognition of Cs. indica as a separate species. Furthermore, the geographical distribution of Cs. indica provides additional evidence in support of its species status.

To glean more information on mosquito diversity and distribution in Auroville, a cross-sectional study was carried out by mapping the distribution of water bodies and habitats supporting immature stages on the one hand and the distribution of water bodies/habitats supporting mosquito immature stages on the other. A satellite image covering an area of 8.08 km2 was overlaid with a grid of 500 × 500 m. Fifteen modules were selected and the area of each module served as the sampling site for the entomological survey. Adult and larval stages were sampled. Diversity indices were analyzed to compare mosquito diversity. Rarefaction estimations were used to compare abundance and richness of the mosquito species between different zones. In total, 750 mosquito larvae and 84 resting adults were sampled. Eighteen species of mosquitoes belonging to 11 subgenera and 7 genera were documented. Genera included Aedes (Johann Wilhelm Meigen 1818, Diptera, Culicidae), Anopheles (Johann Wilhelm Meigen 1818, Diptera, Culicidae), Armigeres (Theobald 1901, Diptera, Culicidae), Culex (Carl Linnaeus 1758, Diptera, Culicidae), Lutzia (Theobald 1903, Diptera, Culicidae), and Mimomyia (Theobald 1903, Diptera, Culicidae). Of the 18 mosquito species identified, 8 species are new records for Auroville. The Alpha (α) biodiversity indices show that the mosquito fauna is diverse (S = 18; DMg = 2.732 [95% CI: 2.732-2.732]). The Shannon-Weiner (H' = 2.199 [95% CI: 2.133-2.276]) and Simpson indices (λ = 0.8619 [95% CI: 0.8496-0.8723]) measured species richness, evenness, and dominance. The values of these indices suggest high species richness, evenness, and dominance. Prevailing conditions can provide suitable environment for establishment of different mosquito species in this ecosystem. Given the sociodemographic characteristics of this area, research on mosquito diversity and risk of vector-borne diseases will be of great use.

Background & objectives: Malaria is an important public health problem in Andaman & Nicobar archipelago. Among the three districts, Nicobar is the most endemic district where API is >2. In this district, the malaria incidence in Car Nicobar Tehsil has declined steadily over the past 10 years. A renewed initiative to consolidate this gain is being made with the ultimate objective of achieving zero indigenous transmission of malaria in Car Nicobar. So, the present study undertook a close environmental monitoring of water bodies for assessing changes in the risk potential of mosquito vector breeding habitats which can augment the elimination programme. Methods: The breeding habitats of anopheline mosquitoes were sampled in 16 areas of Car Nicobar Island for eight time periods during 2017-2020. Along with anophelines, various associated water parameters (n=60) were estimated, viz. physicochemical (n=13), and biological, which included culicine mosquito immatures, insect predators (n=5), phytoplanktons (n=31) and zooplanktons (n=10). Results: In the 16 study sites, overall 1126 surface water stagnating bodies constituting 21 different habitat types were surveyed. Of these, 17 were positive for anopheline breeding. Water bodies from three villages were consistently found to be positive for anopheline breeding. However, early instars of anopheline larvae were more abundant compared to the late instars. Four anopheline species were recorded, including Anopheles sundaicus, A. barbirostris, A. insulaeflorum and A. subpictus, in which 48 per cent were A. sundaicus. Multivariable analysis indicated that anopheline density was significantly higher in permanent water bodies than in temporary habitats (P<0.05) (high risk of anophelines). The highest pH (≥8.2), dissolved solids (≥0.39) levels showed significantly (P<0.05) decreased larval densities (lower risk of breeding), adjusted with breeding sites and season. Nitrite levels increased (P=0.022) larval densities. Interpretation & conclusions: The present study facilitated estimating the productive period of a larval habitat enabling target larval sources to reduce adult populations. Implementing larviciding strategy before monsoon season is presumably the most cost-effective strategy. The output can be utilized for environmental monitoring of mosquito breeding risk in other malaria endemic areas, particularly where medium/large water bodies are the predominant breeding sites for malaria vectors.

OBJECTIVES: Following the Public Health Emergency of International Concern declared on Zika by the World Health Organization during 2016, the Indian Council of Medical Research carried out nationwide vector surveillance for Zika and Dengue viruses (ZIKV and DENV) in India as a preparedness measure in 2016-19. METHODS: High-risk zones distributed to 49 Districts in 14 states/union territories were included in the study. Seven ICMR institutions participated, following a standard operating protocol. Aedes specimens sampled weekly were processed by multiplex reverse transcriptase-polymerase chain reaction (RT-PCR) for ZIKV/DENV and random samples crosschecked with real-time RT-PCR for ZIKV. RESULTS: Altogether, 79 492 Aedes specimens in 6492 pools were processed; 3 (0.05%) and 63 (0.97%) pools, respectively, were found positive for ZIKV and DENV. ZIKV infections were recorded in Aedes aegypti sampled during the 2018 sporadic Zika outbreak in Jaipur, Rajasthan. However, these belonged to the Asian lineage of the virus, already circulating in the country. Both Ae. aegypti and Aedes albopictus distributed to 8 states/union territories were found to be infected with DENV. Both sexes of Ae. albopictus were infected, indicating transovarial transmission. CONCLUSION: This investigation evinced no active transmission of the American lineage-pandemic Zika virus in India during the pandemic period.

Feasibility of implementing a DEC-fortified (DEC at 0.2% w/w and iodine) salt strategy to hasten elimination of diurnally sub-periodic Wuchereria bancrofti (DspWB) from the lone foci in Nancowry islands, Nicobar district, India, was assessed. This is a two-arm community-based study: one arm (12 villages, population 2936) received double fortified salt along with annual mass drug administration (MDA) of DEC plus albendazole (DEC-salt+MDA-arm), and another (14 villages; population 4840) received MDA under the National Filaria Elimination Programme. DEC salt was distributed on camp mode supplemented by door delivery. Monthly survey was carried out in fixed and random households to assess the coverage, usage of DEC salt and DEC content. The impact on prevalence of mf at community level and antigenaemia among children was assessed. A total of 21 metric tonnes of free-flow DEC salt manufactured by Tamil Nadu Salt Corporation, India, was distributed for 1 year. In the DEC-salt+MDA-arm, > 90% of the households received and used the DEC salt. DEC was within therapeutic range (0.2-0.32% w/w) in the samples collected from kitchens. Community mf prevalence reduced from 2.27 to 0.14% in the DEC-salt-arm (< 1% in all the villages) and 1.26 to 0.74% (> 1% in 4 out of 14 villages) in the MDA-arm. Ag prevalence reduced to zero from 1.0 (DEC-salt+MDA-arm) and 6.3% (MDA-arm) in 2-3 years old, 1.2 and 3.6% from 2.9 in the DEC-salt-arm and 4.5% in the MDA-arm among 6-7 years old. It was feasible to deliver DEC-fortified salt covering > 90% of the households with compliance reaching the elimination target in the islands.

Aedes albopictus (Skuse 1894) is prevalent in the urban/peri-urban Port Blair, posing a public health threat, during past outbreaks of chikungunya (2006) and dengue (2010). Despite its vector potential, information on the biology is scanty. Therefore, impact of temperature on survival of immature stages, under laboratory conditions, was studied on F1 population of Andamans. Ae. albopictus larvae were exposed to static temperatures viz. 37°C, 39°C, 41°C, 43°C and 45°C, and the lethal time to cause 50% (LT50) and 90% mortality (LT90) was computed. To assess adaptive thermotolerance, larvae exposed (37°C and 39°C) were re-exposed to higher temperatures (43°C and 45°C). All larvae survived at 37°C and 39°C for the entire exposure period of 420 min, while variable mortality was observed at 41°C, 43°C and 45°C. Larvae re-exposed to 43°C and 45°C showed an increase in thermotolerance with respect to non-adapted larvae. The results are discussed in the context of survival, development and distribution.

Onges, an indigenous vulnerable tribe, inhabit the Little Andaman Island. Study undertaken during 1997, recorded high prevalence of undernutrition among children. Subsequently, food rationing was modified by Andaman and Nicobar administration. In the present study, health and nutritional status of this tribe was assessed in 2013 after a gap of 15 years. All individuals in the tribe were included. Various health parameters, viz, nutritional anthropometry, haemoglobin level, fasting sugar, lipids and intestinal parasites were investigated. Prevalence of stunted growth and underweight among children of 0-5 years was 86.4%. Severe underweight were more prevalent (40%), as compared to those recorded 15 years ago (10%), and the difference was statistically significant (P < 0.05). There has been a substantial improvement in the overall nutritional status of children in the age group of <18 years. The prevalence of wasting has halved and that of stunting and wasting has come down to one-third of that observed in 1997. Prevalence of anaemia significantly declined from 87% in 1997 to 51% 15 years later.

The study was undertaken in South Andaman district, comprising three tehsils, viz. Port Blair, Ferrargunj and Little Andaman Tehsils, respectively. Intensive pupal infestation surveys were carried out along the National Highway (NH 223), the main passenger and trade route, referred to as Great Andaman Trunk Road. Sampling locations at every 3 km were geo-referenced with global positioning system unit. A total of 17314 water collections were examined from 29 locations across the South Andaman district, among which 1021 (5.9%) were colonized by immature stages of Aedes albopictus, Aedes aegypti and other mosquito species. Ae. aegypti were found in 12 locations, showing higher infestation in the densely built Aberdeen Bazaar. Breeding populations of Ae. albopictus were observed in 27 sampling locations. Both the species were not recorded in two Northern localities. In the areas where both the species are present, they were often found in the same developmental sites, suggesting convergent habitat selection. The most frequently encountered man-made, artificial and natural developmental sites were fixed cement tanks, plastic drums, plastic cans, metal drums, metal pots, discarded tires, coconut shells, leaf axils and tree holes. Ae. aegypti and Ae. albopictus were observed in varying proportions in Port Blair and Ferrargunj Tehsils, while the former species appeared to be absent in Little Andaman. This study elucidates the spatial distribution of Ae. aegypti and Ae. albopictus with preponderance of the latter species, pointing towards arboviral transmission and assumes public health importance in South Andaman district, endemic for dengue.

From May to June 2014, an outbreak of dengue virus (DENV) illness occurred in the Havelock Island, South Andaman. Entomological investigations were undertaken during the peak of the outbreak, from 26th May-4th June, to identify the primary vector(s) involved in the transmission so that appropriate public health measures could be implemented. Adult mosquitoes were collected by BG-Sentinel traps in houses and neighborhoods of clinically ill patients. Water holding containers were inspected for the presence of mosquito larvae and pupae. Adult mosquitoes were analyzed by RT-PCR for the presence of nucleic acids of DENV and CHIKV. A total of 498 mosquitoes were collected and processed in 27 pools. The species composition comprised of 58.3% Aedes albopictus, 7.5% Aedes aegypti and 4.2% Aedes edwardsi and 3.1% constituted others. Two A. albopictus pools were found to be positive for DENV RNA. Sequencing of the RT PCR 511 base pair amplicon positive samples showed homology with DENV-3, suggesting that serotype-3 was responsible for the outbreak and A. albopictus was the primary vector responsible. This was supported by high container (10.1%), premise (25.4%) and Breteau (27.9) indices, with miscellaneous receptacles (2.4%), tree holes (1.2%) and discarded tires (1.2%) registering relatively higher container indices. This is the first report of detection of DENV in A. albopictus from Andaman and Nicobar Islands.

Dengue and chikungunya are important arboviral infections in the Andaman Islands. Competent vectors viz. Aedes aegypti and Aedes albopictus are widely prevalent. The most effective proven method for interrupting the transmission of these arboviruses is vector control, mediated through insecticides. Currently, DDT and temephos are the insecticides used for vector control in these islands. Lack of information on susceptibility necessitated assessing the susceptibility profile of A. aegypti and A. albopictus. F1 generation of adult and larvae were assayed, and LT50 and LT90 values were interpreted following the World Health Organization (WHO) protocol. Adults were found resistant to DDT-4 % while susceptible to dieldrin-0.4 %. Against organophosphates, both showed resistance to fenitrothion but susceptible to malathion-5 %. Both species showed resistance to carbamate and bendiocarb-0.1 % while susceptible to propoxur-0.1 %. Of the four synthetic pyrethroids, both were susceptible to deltamethrin-0.05 %, while resistant to permethrin-0.75 %, lambdacyhalothrin-0.05 % and cyfluthrin-0.15 %. Larvae of both species showed resistance to temephos at 0.02 mg/L but susceptible to malathion at 1 mg/L and fenthion at 0.05 mg/L. Currently, there is no prescribed WHO dose for adult-insecticide susceptibility testing. The emergence of resistance to DDT and temephos in the vector population poses a challenge to the on-going vector control measures. The results highlight the need for monitoring resistance to insecticides in the vector population. Impetus for source reduction and alternative choices of control measures are discussed for tackling future threat of arboviral infections in these islands.

In India diurnally subperiodic filariasis (DspWB) is prevalent only in the Nicobar district of Andaman and Nicobar Islands. Studies undertaken at different points of time indicate that this form of filariasis is restricted to a small region in Nancowry group of islands where it is transmitted by mosquito Downsiomyia nivea, a day biting mosquito. Studies on prevalence, distribution, and assessment of endemicity status, vector incrimination, bioecology, host seeking behaviour, population dynamics of the vector, transmission dynamics and clinical epidemiology indicate the prevalence and persistence of this infection in the Nancowry group of islands with perennial transmission. There was no control programme in these islands, until the National programme to eliminate filariasis was launched in 2004. Eight rounds of annual mass drug administration (MDA) with diethyl carbamazine (DEC) + albendazole have been completed. Despite this, microfilaria prevalence remains at above one per cent, the level identified for initiating transmission assessment survey to decide on continuation of MDA further. This necessitates adjunct measures to the ongoing MDA programme in these islands. The vector control options could be an adjunct measure, but the vector is a forest dweller with a unique bio-ecology, therefore, not a technically feasible option. Use of DEC fortified salt for six months to one year could hasten the process of elimination. Although administration of DEC-fortified salt is simple, rapid, safe, and cost-effective, challenges are to be tackled for evolving operationally realistic strategy. Such a strategy requires commitment of all sections of the society, a distribution mechanism that ensures the use of DEC-fortified salt in the Nancowry islands. Here we discuss the plan of action to serve the indigenous communities and operationalizing DEC fortified salt strategy through an inter-sectoral approach involving multiple stakeholders.

Mosquito foraging behavior is a determinant of host-vector contact and has an impact on the risk of arboviral epidemics. Therefore, blood-feeding patterns is a useful tool for assessing the role in pathogen transmission by vector mosquitoes. Competent vectors of dengue and chikungunya viz. Aedes aegypti and Aedes albopictus are widely prevalent in the Andaman and Nicobar archipelago. Considering the vector potential, medical importance of both these mosquito species and lack of information on host-feeding patterns, blood meal analysis of both these vector mosquitoes was undertaken. Biogents Sentinel traps were used for sampling blooded mosquitoes, for identifying the source of blood meal by agar gel-precipitin test. We identified vertebrate source of 147 and 104 blood meals in Ae. aegypti and Ae. albopictus from heterogeneous landscapes in South Andaman district. Results revealed that Ae. aegypti (88 %) and Ae. albopictus (49 %) fed on human and a small proportion on mammals and fowls, indicative of predominance of anthropophilism. Ae. aegypti predominantly fed on human blood (94.2 %-densely built urban, 89.8 %-low vegetation coverage, and 78.3 %-medium vegetation coverage). Anthropophilism in Ae. albopictus was maximal in densely built urban (90.5 %) and progressively decreased from low vegetation-vegetation/forested continuum (66.7, 36.4, and 8.7 %), indicating plasticity in feeding across these landscapes. Epidemiological significance of the findings is discussed.

Leaves of an endemic plant Glyptopetalum calocarpum are used by Nicobarese tribes of Andaman and Nicobar Islands, India, to prepare traditional medicine for treating fever. In the present investigation, pharmacologically active compounds were isolated from this plant and their antimicrobial efficacy was evaluated against the leptospiral strains. The anti-leptospiral activity of six plant-derived compounds was determined by both microdilution and macrodilution methods. Two out of six compounds, namely lupenone and stigmasterol, showed anti-leptospiral activity. The minimum inhibitory concentrations of the two compounds tested against pathogenic leptospiral strains belonging to 10 serovars were in the range of 100-200 µg/mL. The range of minimum bactericidal concentrations was 400-800 µg/mL. Compounds lupenone, stigmasterol, lupeol, ß-amyrin and ß-amyrin acetate had negligible or no haemolytic activity, exhibiting IC50 values of greater than 5 mg/mL. Further in vivo studies are needed to investigate the pharmacological and toxicological properties of G. calocarpum before it can be considered as a new anti-leptospiral agent.

BACKGROUND & OBJECTIVES: In India, diurnally sub periodic Wuchereria bancrofti transmitted by Downsiomyia nivea is prevalent only in the Nicobar district of Andaman and Nicobar Islands. The ongoing LF elimination programme aims at transmission interruption by bringing down the microfilarie (mf) load in the community, which has implication on the parasite load in mosquito vector. Therefore, understanding density dependent constraints on transmission assumes significance from control perspective. The present study was undertaken in Teressa Island to understand the density dependent parasite mortality and survival probability of the parasite Do. nivea. METHODS: The entomological data collected from Teressa Island, endemic for the diurnally sub periodic form of W. bancrofti were used to examine the parasite loss and its survival up to infectivity. Patterns of parasite distribution in Do. nivea were examined. RESULTS: Distribution patterns of microfilariae were found to be over dispersed in Do. nivea. The later stages of the parasite in the vector were randomly distributed. Distribution pattern of various filarial larval stages suggested that the loss of parasites occurred as development progressed and was maximal between the first and second stages. Further, both the prevalence of infection and the degree of parasite aggregation in the vector population have fallen significantly with development of parasite stage. INTERPRETATION & CONCLUSIONS: Results indicate the operation of parasite density dependent mortality of vectors or parasite loss or combination of both. The present study with Aedes transmitted filariasis conducted before launching LF elimination programme in the study area indicates a comparable level of parasite regulation in the vector which has similar implications on the transmission threshold. Thus, the consideration of Aedes with Culex in deriving the critical level of antigen positive for making decisions on cessation of mass drug administration (MDA) can be justified. However, with MDA aiming at reducing parasite load in the community, the operation of density dependent factor in the transmission becomes less pronounced in the subsequent rounds of MDA.
