RESUMEN

The cheese making and vegetable processing industries generate immense volumes of high-nitrogen wastewater that is often treated at rural facilities using land applications. Laboratory incubation results showed denitrification decreased with temperature in industry facility soils but remained high in soils from agricultural sites (75% at 2.1°C). 16S rRNA, phospholipid fatty acid (PLFA), and soil respiration analyses were conducted to investigate potential soil microbiome impacts. Biotic and abiotic system factor correlations showed no clear patterns explaining the divergent denitrification rates. In all three soil types at the phylum level, Actinobacteria, Proteobacteria, and Acidobacteria dominated, whereas at the class level, Nitrososphaeria and Alphaproteobacteria dominated, similar to denitrifying systems such as wetlands, wastewater resource recovery facilities, and wastewater-irrigated agricultural systems. Results show that potential denitrification drivers vary but lay the foundation to develop a better understanding of the key factors regulating denitrification in land application systems and protect local groundwater supplies. PRACTITIONER POINTS: Incubation study denitrification rates decreased as temperatures decreased, potentially leading to groundwater contamination issues during colder months. The three most dominant phyla for all systems are Actinobacteria, Proteobacteria, and Acidobacteria. The dominant class for all systems is Nitrosphaeria (phyla Crenarchaeota). No correlation patterns between denitrification rates and system biotic and abiotic factors were observed that explained system efficiency differences.

RESUMEN

Nitrogen (N) behavior in soil is a major component of the global N cycle. Climate scientists seek to accurately measure N flux to the atmosphere, farmers want to maximize plant N uptake and reduce input costs, and industries land-applying wastewater must mitigate potential N leaching to drinking water supplies. The need to quantify denitrification rates of wastewater disposed of by vegetable processing and cheese making industries in Wisconsin drove the development of an autonomous high-throughput in situ sampling and analysis system for soil N flux. The system was deployed to six unique industry sites with different soil types for 7 days once per quarter and data collected continuously. Additional seasonal data collection allowed for the determination of system N mass balances. The system can deliver quality data under challenging conditions where staffing would be impractical and provide detailed information about soil gas emissions under a range of environmental conditions.

Inputs of polycyclic aromatic hydrocarbons (PAHs) of regulatory interest from diffuse atmospheric sources within urban areas frequently elevate local soil concentrations to levels requiring remediation despite the lack of in-situ contamination. This research sought to determine the distribution and potential health effects of aerially deposited PAHs in soil within the urban core of metropolitan Milwaukee, Wisconsin, U.S.A. as part of a soil regulatory standards reevaluation. Park areas (n = 27) identified as undisturbed for 80+ years, containing no fill material, and receiving only atmospheric deposition were selected for composite surface and 92 cm core soil sample collection (n = 295). Samples were analyzed for the 16 USEPA priority PAHs, 1- and 2- methylnapthalene and ancillary soil properties. Soil core and ancillary data confirm lack of site disturbance. PAH diagnostic ratios and homologue summations indicate that diffuse multiple point source emissions contribute equally to PAH deposition throughout the area. Benzo(a)pyrene (BaP) and dibenz(a,h)anthracene mean concentrations exceed health-based clean up levels. Risk assessment shows only a worst-case exposure scenario (BaP at the 95% upper confidence limit) increasing cancer risk (1.67 × 10-6) over current regulatory thresholds (1.0 × 10-6). Health quotients show potential health risks from fluoranthene and pyrene for daily park users and from BaP for all others. Mean soil PAH values are similar to New Orleans, but lower than Chicago, Boston, and London reflecting industrial history and site selection protocols. The soil PAH results presented here for sites selected for non-manipulated soils combined with an almost 100-year uninterrupted atmospheric exposure effectively show the maximum potential PAH values that can be found at any given undisturbed location within the Milwaukee urban core due solely to atmospheric deposition.

Soil lead (Pb) hazard level is contingent on bioavailability, but existing assays that estimate Pb bioavailability for human health risks are too expensive or otherwise inaccessible to many people that are impacted by Pb-contaminated soil. This study investigated the use of routine soil nutrient tests to estimate soil-Pb bioaccessibility as a surrogate measure of Pb bioavailability. A silt loam soil was spiked to a target concentration of 2000 mg Pb kg-1 with Pb(NO3)2 and amended with H3PO4 (varying P-to-Pb molar ratios) and KCl (Cl-to-P molar ratio of 2:5) to generate soils with similar total Pb concentrations but a range of Pb bioavailability (and bioaccessibility). Soils were extracted using Mehlich 3, Mehlich 1, Bray P1, Olsen, and  micronutrient (DTPA) methods, and the results were compared to U.S. Environmental Protection Agency method 1340 data as well as to extended X-ray absorption fine structure (EXAFS) spectroscopy. The Mehlich 3 and method 1340 treatment effect ratios were well-correlated ( r2 = 0.88, p ≤ 0.05), whereas Bray P1, DTPA, and Olsen results were more reflective of EXAFS data. Preliminary animal-feeding trials suggest that the Mehlich 3 is as effective as method 1340 at predicting the impact of P treatment on Pb relative bioavailability; however, both methods over-estimated the Pb hazard to mice in P-amended soil. Other routine soil tests that have heightened sensitivity to P amendment (e.g., Bray P1) may be promising candidates for Pb bioaccessibility assessment.

A series of legal decisions culminated in 2002 with the California State Water Resources Control Board funding the San Francisco Estuary Institute to develop and implement a 3-year monitoring program to determine the potential environmental impacts of aquatic herbicide applications. The monitoring program was intended to investigate the behavior of all aquatic pesticides in use in California, to determine potential impacts in a wide range of water-body types receiving applications, and to help regulators determine where to direct future resources. A tiered monitoring approach was developed to achieve a balance between program goals and what was practically achievable within the project time and budget constraints. Water, sediment, and biota were collected under "worst-case" scenarios in close association with herbicide applications. Applications of acrolein, copper sulfate, chelated copper, diquat dibromide, glyphosate, fluridone, triclopyr, and 2,4-D were monitored. A range of chemical analyses, toxicity tests, and bioassessments were conducted. At each site, risk quotients were calculated to determine potential impacts. For sediment-partitioning herbicides, sediment quality triad analysis was performed. Worst-case scenario monitoring and special studies showed limited short-term and no long-term toxicity directly attributable to aquatic herbicide applications. Risk quotient calculations called for additional risk characterizations; these included limited assessments for glyphosate and fluridone and more extensive risk assessments for diquat dibromide, chelated copper products, and copper sulfate. Use of surfactants in conjunction with aquatic herbicides was positively associated with greater ecosystem impacts. Results therefore warrant full risk characterization for all adjuvant compounds.

Estrogenic potencies of four herbicides (triclopyr, 2,4-dichlorophenoxyacetic acid (2,4-D), diquat dibromide, glyphosate), two alkylphenol ethoxylate-containing surfactants (R-11 and Target Prospreader Activator (TPA)), and the binary mixture of surfactants with the herbicides were evaluated using an in vivo rainbow trout vitellogenin assay. Juvenile rainbow trout exposed to 2,4-D (1.64 mg/l) for 7 days had a 93-fold increase in plasma vitellogenin (Vtg) levels compared with untreated fish, while rainbow trout exposed to other pesticides alone did not show elevated vitellogenin levels compared to the control fish. When combined with surfactants, trends indicated enhanced estrogenicity for all combinations, but only 2,4-D and triclopyr caused significant induction of Vtg. Concentration-response studies demonstrated that the lowest observed effect concentrations (LOECs) for 2,4-D and triclopyr were 0.164 mg/l and 1 mg/l, respectively. In terms of measured 4-nonylphenol (4-NP), the LOECs of R-11 and TPA were 20 micro/l and 9.5 microg/l, respectively. Binary mixtures of TPA and 2,4-D showed a greater than additive estrogenic response at the lowest concentrations tested, but a less than additive response at the highest combined concentrations. Binary mixtures of TPA with triclopyr also caused greater than additive Vtg responses in two middle concentrations when compared to TPA or triclopyr alone. When trout were exposed to water collected from a site where triclopyr was used in combination with TPA, a concentration-dependent increase in Vtg expression was observed. Measured values of 4-NP were 3.7 microg/l, and triclopyr concentrations were below detection (<5 ng/l). Estradiol equivalents (EEQs) of the lake water were calculated from an estradiol concentration-response curve and were similar (8.5 +/- 7.7 ng/l) to the mean values for the combined triclopyr + TPA treatments (9.9-12.2 ng/l) in the laboratory, suggesting the estrogenicity of the water may have been due to the treatment. These results demonstrated the binary mixture of alkylphenol ethoxylate-containing surfactants with two aquatic pesticides possessed greater than additive estrogenic responses in fish under laboratory conditions and in a field setting.
