RESUMEN

Functional pancreatic islet beta cells are essential to ensure glucose homeostasis across species from zebrafish to humans. These cells show significant heterogeneity, and emerging studies have revealed that connectivity across a hierarchical network is required for normal insulin release. Here, we discuss current thinking and areas of debate around intra-islet connectivity, cellular hierarchies and potential "controlling" beta-cell populations. We focus on methodologies, including comparisons of different cell preparations as well as in vitro and in vivo approaches to imaging and controlling the activity of human and rodent islet preparations. We also discuss the analytical approaches that can be applied to live-cell data to identify and study critical subgroups of cells with a disproportionate role in control Ca2+ dynamics and thus insulin secretion (such as "first responders", "leaders" and "hubs", as defined by Ca2+ responses to glucose stimulation). Possible mechanisms by which this hierarchy is achieved, its physiological relevance and how its loss may contribute to islet failure in diabetes mellitus are also considered. A glossary of terms and links to computational resources are provided.

The development of a non-invasive infection-specific diagnostic probe holds the potential to vastly improve early-stage detection of infection, enabling precise therapeutic intervention and potentially reducing the incidence of antibiotic resistance. Towards this goal, a commercially available bacteria-targeting Zinc(II)-dipicolylamine (ZnDPA)-derived fluorophore, PSVue794, was assessed as a photoacoustic (PA) imaging probe (PIP). A radiolabeled version of the dye, [99mTc]Tc-PSVue794, was developed to facilitate quantitative biodistribution studies beyond optical imaging methods, which showed a target-to-non-target ratio of 10.1 ± 1.1, 12 h post-injection. The ability of the PIP to differentiate between bacterial infection, sterile inflammation, and healthy tissue in a mouse model, was then evaluated via PA imaging. The PA signal in sites of sterile inflammation (0.062 ± 0.012 a.u.) was not statistically different from that of the background (0.058 ± 0.006 a.u.). In contrast, high PA signal was detected at sites of bacterial infection (0.176 ± 0.011 a.u.) as compared to background (0.081 ± 0.04 a.u., where P ≤ 0.03). This work demonstrates the potential of utilizing established fluorophores towards PAI and utilizing PAI as a modality in the distinction of bacterial infection from sites of sterile inflammation.

BACKGROUND: Anthropogenic threats are causing alteration of coastal areas worldwide. Most of the coastal biodiversity is endangered, taking a particular toll on island ecosystems, like the Azores. To better understand the biotic and abiotic factors constraining the distribution and conservation status of two endemic plants, Azorina vidalii (Campanulaceae) and Lotus azoricus (Fabaceae), we performed a global survey of coastal plant communities in the archipelago, also covering environmental descriptors, natural and anthropogenic threats. Moreover, we revised their IUCN conservation status and estimated the population fractions within protected areas. RESULTS: Non-indigenous plants were commonly found in plots with or without the target endemics, contributing to the absence of well-defined coastal plant communities. Nonetheless, indigenous taxa commonly occurred at the plots with L. azoricus. With a larger area of occurrence, A. vidalii ecological niche differed from that of L. azoricus, the latter being restricted to dry and rocky sea cliffs, mostly in Santa Maria Island. Besides the presence of invasive plants, signs of habitat destruction, trampling and grazing, and of natural threats, such as coastal erosion, were commonly observed. CONCLUSIONS: Occurrence data indicated an endangered status for both species, although this would change to critically endangered for L. azoricus when using smaller-sized occurrence cells. Both species are threatened since their habitat is restricted to a very narrow vegetation belt, strongly limited by sea influence and human pressure, and with the frequent presence of invasive plants. While focusing on two endemic plants, our study allowed a broader view of the impact of anthropogenic disturbance on Azorean coastal plant communities.

Plastic pollution has become a global concern, affecting many species around the world. While well-documented for marine ecosystems, the impact of plastic pollution on terrestrial ecosystems is comparatively limited. In fact, only recently have some studies begun to explore the occurrence, pathways, and impacts of plastic in the atmosphere and on terrestrial species. Here, we assess the presence of synthetic material in nests of three swift species breeding in the Western Palearctic: the common swift (Apus apus), the pallid swift (Apus pallidus), and the alpine swift (Tachymarptis melba). Using data from 487 nests spanning 25 colonies and seven European countries, we show that 36.5 % of the examined nests contained anthropogenic materials, mainly plastic debris. Notably, Pallid swifts' nests, with 85 % of the total nests examined with plastic, rank among birds with the highest plastic content in nests. We also demonstrate that the probability of finding plastic in the nest increased substantially with the human footprint of the landscape. Last, we recorded four cases of swifts entangled in their own nest, a low proportion compared to other species studied previously. Our study provides compelling evidence that plastic pollution may also be considered a concern for other terrestrial species, particularly for birds with highly aerial lifestyles, such as other swifts. The correlation with the human footprint suggests that areas with higher human activity contribute more significantly. Moreover, the entanglement cases, although low, indicate a threat to bird health and welfare. To our knowledge, our study is the first to report a direct interaction between floating plastic debris in the atmosphere and any species. Understanding this interaction is key, not only due to the lack of research on the topic, but also because it highlights that plastic pollution is a multifaceted environmental issue affecting various ecosystem categories, and the broader implications of atmospheric plastic circulation on wildlife and ecosystems health.

BACKGROUND: Globally, internalizing problems disproportionately affect females in adolescence and adulthood, with limited research at earlier ages due to a focus on disruptive behaviors. Our study addresses this gap by exploring the structure of internalizing problems and gender differences in Brazilian preschoolers. METHODS: We analyzed data from the Child Behavioral Checklist 1.5-5 in the Preschool Mental Health Study, involving 1,292 children aged 4 to 5 in Embu das Artes, São Paulo, Brazil. Confirmatory factor analysis and mean comparisons explored internalizing problems and gender variations. RESULTS: A two-factor model best fit both internalizing and externalizing problems.The hierarchical model with four factors (Emotionally Reactive, Anxiety/Depression, Somatic Complaints, and Withdrawn) best fit internalizing problems, achieving partial invariance between boys and girls. Boys scored higher in Withdrawn Syndrome, while girls scored higher in Somatic Complaints Syndrome. CONCLUSIONS: Preschoolers' internalizing problems warrant attention beyond their link to externalizing problems. While the overall construct is similar in boys and girls, divergent syndrome scores indicate potential distinct risk patterns requiring further exploration.

Herein, graphene oxide was used as the highly efficient phenazopyridine adsorbent from aqueous medium, synthetic, and human urine. The nanoadsorbent was characterized by different instrumental techniques. The adsorption capacity (1253.17 mg g-1) was reached at pH 5.0, using an adsorbent dosage of 0.125 g L-1 at 298 K. The Sips and Langmuir described the equilibrium data well. At the same time, the pseudo-second order was more suitable for fitting the kinetic data. Thermodynamic parameters revealed the exothermic nature of adsorption with an increase in randomness at the solid-liquid interface. The magnitude of the enthalpy variation value indicates that the process involves the physisorption phenomenon. At the same time, ab initio molecular dynamics data corroborated with the thermodynamic results, indicating that adsorbent and adsorbate establish hydrogen bonds through the amine groups (adsorbate) and hydroxyl groups on the adsorbent surface (weak interactions). Electrostatic interactions are also involved. Additionally, the adsorption assays conducted in simulated medium and human urine showed the excellent performance of adsorbent material to remove the drug in real concentrations excreted by the kidneys (removal values higher than 60%).

O objetivo desse estudo é traçar o perfil epidemiológico das notificações de violência contra a mulher no estado da Paraíba, no período de 2009 a 2019. Trata-se de um estudo transversal, descritivo, realizado por meio de consulta online ao Sistema de Informação TabNet na seção de Doenças e Agravos de Notificação através do campo violência interpessoal. Como parte dos resultados, identificamos que a violência mais frequente é a física, 45,2%. Foi observado que o maior número de queixas envolve mulheres negras, com 60,11% dos casos; na faixa etária de 20 a 29 anos, 22,71%; com ensino fundamental incompleto, 14,21%. Ademais, 59,2% das situações de violência contra a mulher ocorrem, predominantemente, em locais residenciais. Quanto aos encaminhamentos ao setor de saúde, os achados revelam que, nesse item, em 54,15% dos registros é definido como ignorado. O estudo apontou uma grande fragilidade nos registros de dados sobre violência, tanto pela falta de informações quanto pelo preenchimento inadequado das fichas. Por isso, é crucial reforçar a importância do preenchimento adequado das fichas de notificação compulsória e a qualificação dos profissionais para fornecer evidências precisas sobre o problema e subsidiar a gestão para os enfrentamentos.


This study aims to trace the epidemiological profile of notifications of violence against women in the state of Paraíba from 2009 to 2019. This cross-sectional, descriptive study conducted an online consultation of the TabNet Information System in the Diseases and Notifiable Diseases section through the field of interpersonal violence. As part of the results, we identified that the most frequent form of violence was physical violence (45.2%). We found that the highest number of complaints involved black women, with 60.11% of cases; in the 20-29 age group, 22.71%; and with incomplete primary education, 14.21%. In addition, 59.2% of situations of violence against women occur predominantly in residential areas. As for referrals to the health sector, the findings reveal that 54.15% of the records were defined as ignored. The study pointed to a significant weakness in the recording of data on violence, both due to the lack of information and the inadequate filling out of forms. For this reason, it is crucial to reinforce the importance of correctly filling out compulsory notification forms and training professionals to provide accurate evidence of the problem and support management in dealing with it.

Animals can adjust their diet to maximize energy or nutritional intake. For example, birds often target fruits that match their beak size because those fruits can be consumed more efficiently. We hypothesized that pressure to optimize diet-measured as matching between fruit and beak size-increases under stressful environments, such as those that determine species' range edges. Using fruit-consumption and trait information for 97 frugivorous bird and 831 plant species across six continents, we demonstrate that birds feed more frequently on closely size-matched fruits near their geographic range limits. This pattern was particularly strong for highly frugivorous birds, whereas opportunistic frugivores showed no such tendency. These findings highlight how frugivore interactions might respond to stressful conditions and reveal that trait matching may not predict resource use consistently.

The sustainable agri-food system is an important sector recognized for promoting the United Nations' Sustainable Development Goals on food security, resource conservation and climate change mitigation. However, the increasing food loss and waste (FLW) along the supply chains has continued to hinder these goals. This study evaluates the trend of FLW research from 1975 to 2022 and how it promotes the achievement of resource and environmental sustainability in agri-food systems. The salient research themes and hotspots that are of interest to researchers were identified. Bibliometric and network analyses were carried out on scholarly research articles from the Scopus database using bibliometrix and VOSviewer. Furthermore, the content analysis was conducted on the selected highly influential articles containing relevant data to understand the role of FLW in promoting sustainable agri-food systems. The results showed disaggregate and unbalanced research distribution on the impacts of FLW among the countries, with China and the United States having the highest contributions. The identified major research themes relating to sustainable agri-food systems are food waste and sustainable systems, food waste management and food waste impact assessment. Moreover, the circular economy was found to be a relatively new approach being explored in agri-food systems to promote FLW reduction and ensure sustainability of resource use. This study highlights the critical role of the impact of FLW in addressing the grand challenge of food security, resource use efficiency and environmental sustainability.

Natural native forests are rapidly being replaced by anthropogenic forests often with a strong presence of invasive alien plant species. Eucalypt species are widely planted worldwide, with Eucalyptus globulus plantations being particularly expressive in Portugal. Poor forestry practices often lead to the associated expansion of invasive species, such as Acacia dealbata. However, we still know relatively little about the functioning of anthropogenic forests, such as seed and pollen dispersal services. Here, we compared bird abundance and richness and the seed and pollen dispersal networks in both forest types. Anthropogenic forests presented lower bird abundance, and smaller, more simplified, and more random (abundance-based) seed dispersal services than those of natural forests. Interestingly, the pollen dispersal network was more similar than the seed dispersal network for both forest types and dominated by opportunistic and neutral processes, given the absence of specialized nectarivorous. The proportion of birds transporting seeds decreased, while those carrying pollen significantly increased in the anthropogenic forest compared to the native forest. Our work highlights the impact of anthropogenic forests on bird abundance, with consequences for seed dispersal services and forest regeneration.

Pimelea poisoning of cattle causes distinct symptoms and frequently death, attributable to the toxin simplexin. Pimelea poisoning was induced via addition of ground Pimelea trichostachya plant to the daily feed in a three-month trial with Droughtmaster steers. The trial tested four potential mitigation treatments, namely, biochar, activated biochar, bentonite, and a bacterial inoculum, and incorporated negative and positive control groups. All treatments tested were unable to prevent the development of simplexin poisoning effects. However, steers consuming a bentonite adsorbent together with Pimelea showed lesser rates-of-decline for body weight (P < 0.05) and four hematological parameters (P < 0.02), compared to the positive control group fed Pimelea only. Microbiome analysis revealed that despite displaying poisoning symptoms, the rumen microbial populations of animals receiving Pimelea were very resilient, with dominant bacterial populations maintained over time. Unexpectedly, clinical edema developed in some animals up to 2 weeks after Pimelea dosing was ceased.

The presence of phenazopyridine in water is an environmental problem that can cause damage to human health and the environment. However, few studies have reported the adsorption of this emerging contaminant from aqueous matrices. Furthermore, existing research explored only conventional modeling to describe the adsorption phenomenon without understanding the behavior at the molecular level. Herein, the statistical physical modeling of phenazopyridine adsorption into graphene oxide is reported. Steric, energetic, and thermodynamic interpretations were used to describe the phenomenon that controls drug adsorption. The equilibrium data were fitted by mono, double, and multi-layer models, considering factors such as the numbers of phenazopyridine molecules by adsorption sites, density of receptor sites, and half saturation concentration. Furthermore, the statistical physical approach also calculated the thermodynamic parameters (free enthalpy, internal energy, Gibbs free energy, and entropy). The maximum adsorption capacity at the equilibrium was reached at 298 K (510.94 mg g-1). The results showed the physical meaning of adsorption, indicating that the adsorption occurs in multiple layers. The temperature affected the density of receptor sites and half saturation concentration. At the same time, the adsorbed species assumes different positions on the adsorbent surface as a function of the increase in the temperature. Meanwhile, the thermodynamic functions revealed increased entropy with the temperature and the equilibrium concentration.

The goals of the "dual carbon" program in China are to implement a series of air pollution policies to reduce the emission of carbon-bearing particulate matter (PM). Following improvements in the reduction in carbon emissions in Handan City, China, fine particulate matter (PM2.5) was collected in the winters from 2016 to 2020 to characterize the concentrations and sources of carbonaceous components in PM2.5. Trend analysis revealed that both organic carbon (OC) and elemental carbon (EC) concentrations significantly decreased. The proportion of total carbon aerosol (TCA) in PM2.5 decreased by 47.0%, highlighting the effective reduction in carbon emissions. Secondary organic carbon (SOC) concentrations increased from 2016 (12.86 ± 14.10 µg·m-3) to 2018 (36.76 ± 21.59 µg·m-3) and then declined gradually. SOC/OC was larger than 67.0% from 2018 to 2020, implying that more effective synergistic emission reduction measures for carbonaceous aerosol and volatile organic compounds (VOCs) were needed. In the winters from 2016 to 2020, primary organic carbon (POC) concentrations reduced by 76.1% and 87.6% under a light/moderate pollution period (LP) and heavy/severe pollution periods (HPs), respectively. The TCA/PM2.5 showed a decreasing trend under LP and HP conditions, decreasing by 42.1% and 54.7%, respectively. Source analysis revealed that carbonaceous components were mainly from biomass burning, coal combustion and automotive exhaust emissions in the winters of 2016 and 2020. OC/EC and K+/EC analysis pointed out that air pollutant reduction measurements should focus on rectification biomass fuels in the next stage. Compared with 2016, the contributions of automotive exhaust emissions decreased in 2020. OC and EC concentrations decreased due to control measures on automotive exhaust emissions.

In today's digital landscape, organizations face significant challenges, including sensitive data leaks and the proliferation of hate speech, both of which can lead to severe consequences such as financial losses, reputational damage, and psychological impacts on employees. This work considers a comprehensive solution using a microservices architecture to monitor computer usage within organizations effectively. The approach incorporates spyware techniques to capture data from employee computers and a web application for alert management. The system detects data leaks, suspicious behaviors, and hate speech through efficient data capture and predictive modeling. Therefore, this paper presents a comparative performance analysis between Spring Boot and Quarkus, focusing on objective metrics and quantitative statistics. By utilizing recognized tools and benchmarks in the computer science community, the study provides an in-depth understanding of the performance differences between these two platforms. The implementation of Quarkus over Spring Boot demonstrated substantial improvements: memory usage was reduced by up to 80% and CPU usage by 95%, and system uptime decreased by 119%. This solution offers a robust framework for enhancing organizational security and mitigating potential threats through proactive monitoring and predictive analysis while also guiding developers and software architects in making informed technological choices.

Given the growing scarcity of water and the continuous increase in emerging pollutants detected in water bodies, there is an imperative need to develop new, more effective, and sustainable treatments for wastewater. Advanced oxidation processes (AOPs) are considered a competitive technology for water treatment. Specifically, ozonation has received notable attention as a promising approach for degrading organic pollutants in wastewater. However, different groups of pollutants are hardly degradable via single ozonation. With continuous development, it has been shown that using engineered nanomaterials as nanocatalysts in catalytic ozonation can increase efficiency by turning this process into a low-selective AOP for pollutant degradation. Nanocatalysts promote ozone decomposition and form active free radicals responsible for increasing the degradation and mineralization of pollutants. This work reviews the performances of different nanomaterials as homogeneous and heterogeneous nanocatalysts in catalytic ozonation. This review focuses on applying metal- and carbon-based engineered nanomaterials as nanocatalysts in catalytic ozonation and on identifying the main future directions for using this type of AOP toward wastewater treatment.

The COVID-19 pandemic lockdowns affected the lifestyles of children and adolescents, leading to an increase in childhood obesity. Paediatric patients with familial hypercholesterolemia (FH) may be more susceptible to lockdown effects due to their increased cardiovascular risk. However, data are lacking. We investigated the effect of lockdowns on the metabolic profile of paediatric patients with FH. Blood lipids and anthropometry measured in September 2021-April 2022 were retrospectively compared with pre-pandemic values. Thirty participants were included (1-16 years; 57% female). From baseline to post-pandemic, median [P25, P75] blood LDL-C concentration was 125 [112, 150] mg/dL vs. 125 [100, 147] mg/dL (p = 0.894); HDL-C was 58 [52, 65] mg/dL vs. 56 [51, 61] mg/dL (p = 0.107); triglycerides were 64 [44, 86] mg/dL vs. 59 [42, 86] mg/dL (p = 0.178). The BMI z-score did not change significantly (0.19 [-0.58, 0.89] vs. 0.30 [-0.48, 1.10], p = 0.524). The lack of deterioration in metabolic profiles during lockdowns is positive, as some deterioration was expected. We speculate that patients and caregivers were successfully educated about healthy lifestyle and dietary habits. Our results should be interpreted with caution since the study sample was small and heterogeneous. Multicentre research is needed to better understand the impact of lockdowns on this population.

BACKGROUND: 123Iodine-meta-iodobenzylguanidine scintigraphy is useful for assessing cardiac autonomic dysfunction and predict outcomes in heart failure (HF). The relationship of cardiac sympathetic function with myocardial remodeling and diffuse fibrosis remains largely unknown. We aimed to evaluate the cardiac sympathetic function of patients with HF and its relation with myocardial remodeling and exercise capacity. METHODS AND RESULTS: Prospectively enrolled patients with HF (New York Heart Association class II-III) were stratified into HF with preserved left ventricular ejection fraction [LVEF] ≥45%) and reduced LVEF. Ventricular morphology/function and myocardial extracellular volume (ECV) fraction were quantified by cardiovascular magnetic resonance, global longitudinal strain by echocardiography, cardiac sympathetic function by heart-to-mediastinum ratio from 123iodine-meta-iodobenzylguanidine scintigraphy. All participants underwent cardiopulmonary exercise testing. The cohort included 33 patients with HF with preserved LVEF (LVEF, 60±10%; NT-proBNP [N-terminal pro-B-type natriuretic peptide], 248 [interquartile range, 79-574] pg/dL), 28 with HF with reduced LVEF (LVEF, 30±9%; NT-proBNP, 743 [interquartile range, 250-2054] pg/dL) and 20 controls (LVEF, 65±5%; NT-proBNP, 40 [interquartile range, 19-50] pg/dL). Delayed (4 hours) 123iodine-meta-iodobenzylguanidine heart-to-mediastinum ratio was lower in HF with preserved LVEF (1.59±0.25) and HF with reduced LVEF (1.45±0.16) versus controls (1.92±0.24; P<0.001), and correlated negatively with diffuse fibrosis assessed by ECV (R=-0.34, P<0.01). ECV in segments without LGE was increased in HF with preserved ejection fraction (0.32±0.05%) and HF with reduced left ventricular ejection fraction (0.31±0.04%) versus controls (0.28±0.04, P<0.05) and was associated with the age- and sex-adjusted maximum oxygen consumption (peak oxygen consumption); (R=-0.41, P<0.01). Preliminary analysis indicates that cardiac sympathetic function might potentially act as a mediator in the association between ECV and NT-proBNP levels. CONCLUSIONS: Abnormally low cardiac sympathetic function in patients with HF with reduced and preserved LVEF is associated with extracellular volume expansion and decreased cardiopulmonary functional capacity.

Owing to its exposed nature, the skin can be injured by various factors, including by Staphylococcus aureus, which inhabits its innate microbiota. Treatment of infected wounds presents an important challenge, making it imperative to develop new treatment options. Plant-derived formulations, such as those containing Melaleuca alternifolia essential oil (MaEO), are used for wound treatment because of their healing, anti-inflammatory, and antimicrobial properties. This study presents a cream containing 2% MaEO (2% CMa) and evaluates its effects in an S. aureus-infected wound murine model. The 2% CMa was subjected to quality control testing and pH and analysis of density, organoleptic characteristics, and microbiological effects. The quality control parameters all revealed the good stability of the 2% CMa. The formulation strongly reduced the S. aureus ATCC 6538 colony-forming unit (CFU) count in an ex vivo porcine skin model. In the murine model, daily topical application of 2% CMa reduced the severity and size of S. aureus-infected wounds and the bacterial load. These effects may be due to the presence of terpinen-4-ol, which exhibits anti-inflammatory activity. Based on these findings, the formulation exhibits good quality and safety. We suggest the topical application of this formulation, which exhibited an antimicrobial effect, as an interesting treatment strategy for wound healing.

Chronic spinal pain (CSP) is a prevalent condition, and prolonged sitting at work can contribute to it. Ergonomic factors like this can cause changes in motor variability. Variability analysis is a useful method to measure changes in motor performance over time. When performing the same task multiple times, different performance patterns can be observed. This variability is intrinsic to all biological systems and is noticeable in human movement. This study aims to examine whether changes in movement variability and complexity during real-time office work are influenced by CSP. The hypothesis is that individuals with and without pain will have different responses to office work tasks. Six office workers without pain and ten with CSP participated in this study. Participant's trunk movements were recorded during work for an entire week. Linear and nonlinear measures of trunk kinematic displacement were used to assess movement variability and complexity. A mixed ANOVA was utilized to compare changes in movement variability and complexity between the two groups. The effects indicate that pain-free participants showed more complex and less predictable trunk movements with a lower degree of structure and variability when compared to the participants suffering from CSP. The differences were particularly noticeable in fine movements.
