RESUMEN

Physiological stress responses have been associated with lesser pregnancy rates in beef cattle. Alternative cattle restraint methods have been developed to reduce this negative effect. The objective was to determine if there was a difference in pregnancy rate as a result of fixed-time artificial insemination (FTAI) in Bos taurus females when using a breeding box compared with a squeeze chute for restraining cows when conducting FTAI. There were FTAI treatment regimens imposed on cows and heifers at two separate locations (Virginia Tech University, VT; and R.A. Brown Ranch, RAB) with the FTAI being conducted while animals were restrained in a squeeze chute (n = 169) or a breeding box (n = 162). There was imposing of a 7-day Co-Sync + CIDR pre-breeding treatment regimen. Females were randomly assigned to treatments just prior to breeding. Immediately following breeding, blood was collected using coccygeal venipuncture procedures for assessment of plasma cortisol concentration. Chute and exit scores (1-5) were determined at the time of FTAI. Pregnancy rate, cortisol, chute score, and exit score were analyzed. There was no difference in pregnancy rate between treatments (57 ± 26% and 69 ± 26 %, respectively P = 0.59).There were differences in chute (1.52 ± 0.05 and 1.27 ± 0.05, respectively P < 0.05) and exit (1.26 ± 0.04 and 1.08 ± 0.04, respectively P < 0.05) scores between the two treatment groups. Results from the present study are not conclusive in ascertaining effects of the two types of restraint during FTAI on reproductive efficiency.

The objective of this study was to determine if transportation and exercise stress in horses affect the microflora populations in the equine hindgut. Four horses were subjected to three transport periods (0, 3, and 6 hours) with a 7-d rest period between each transport. Horses were fed 0.91 kg/day of Purina Impact All Stages 12% and had ad libitum access to Cynodon dactylon (Coastal Bermudagrass) hay. Fecal samples were collected before (0 hours) and after (48 hours) transport. In addition, three horses underwent a different standardized exercise test with a 7-d rest period between each exercise. Standardized exercise test intensity was determined by heart rate to validate if the horse was in aerobic or anaerobic work. The protocol for fecal sample collection after exercise was the same as for transport. Prokaryotic community profiling was conducted by 16S metagenomic analysis. After DNA evaluation, differences were found in the microbiome at transport 0 hours and grouped transport 3 hours time 48 and transport 6 hours time 48 (PERMANOVA P = .037) where Bacteroidetes increased 48 hours after transport and Firmicutes decreased 48 hours after transport. Exercise microbial communities showed no difference in either alpha or beta diversity when compared with controls (0 hours). In the present study, difference in microflora may have resulted from stress duration of transport rather than stress duration of exercise.

The use of embryo transfer (ET) in cattle is important for profitability and improved genetic gains. The advent of the commercial embryo collection and transfer industry has led to advancements in multiple techniques and practices. Specific variables, however, have historically affected pregnancy rates but an understanding of the magnitude of these effects in the current industry is limited. Transfer location (cranial, middle, or caudal third of the uterine horn ipsilateral to the ovary with a CL), transfer score (range of 1-3 with 1 being excellent and 3 poor, based on difficulty of accessing the site of embryo deposition), and amount of time to complete a transfer, therefore, were recorded. These variables were collected in a setting designed to mimic commercial production practices as well as exaggerated time (due to data collection) to assess effects on pregnancy rates. Fresh and frozen in vivo-derived embryos (n = 256) from Bos taurus cows were transferred to Bos taurus recipients. There tended to be more pregnancies when embryos were deposited in the cranial part of the uterus (P = 0.08) compared to the middle and caudal third of the uterus. With a lesser degree of difficulty in transfers (score 1), there tended to be more pregnancies established (P = 0.07). When lesser time was needed for transferring embryos and collecting data, there were greater pregnancy rates (P = 0.03). Thus, these traditionally accepted variables of influence (site of embryo placement in uterus, difficulty, and time) continue to influence ET pregnancy success.
