RESUMEN

Prostate cancer remains the second most common malignancy in men worldwide, is a global health issue, and poses a huge health burden. Precision medicine provides more treatment options for prostate cancer patients, but its popularity, drug resistance, and adverse reactions still need to be focused on. Chinese herbal medicines (CHMs) have been widely accepted as an alternative therapy for cancer, with the advantages of multiple targets, multiple pathways, and low toxicity. We searched the experimental research and clinical practice of CHMs for prostate cancer treatment published in PubMed, Embase, and Web of Science in the last five years. We found five CHM formulas and six single CHM extracts as well as 12 CHM-derived compounds, which showed induction of apoptosis, autophagy, and cell cycle arrest, suppression of angiogenesis, proliferation, and migration of prostate cancer cells, reversal of drug resistance, and enhancement of anti-tumor immunity. The mechanisms of action include the PI3K/Akt/mTOR, AR, EGFR and Wnt/ß-catenin signaling pathways, which are commonly implicated in the development of prostate cancer. We also summarized the advantages of CHMs in patients with hormone-sensitive and castration-resistant prostate cancer and provided ideas for their further experimental design and application.

A comprehensive understanding of carbon emission reduction and decoupling in urban agglomerations of the Yellow River Basin (YRB) has significant theoretical and practical value for formulating precise carbon reduction policies and achieving ecological conservation and high-quality development in the region. This study utilized a generalized Divisia index decomposition model to identify the primary driving factors behind carbon emission changes in urban agglomerations of the YRB. Based on this, a model measuring decoupling efforts was constructed to systematically investigate the decoupling relationship between carbon emissions. The research findings indicate that technological progress and output scale are two primary drivers of carbon emission increases in the YRB and its urban agglomerations, whereas technological carbon intensity, output carbon intensity, and energy carbon intensity play key roles in reducing carbon emissions. Except for a few years, the YRB and Jiziwan metropolitan area (JWMA) did not exhibit decoupling effects on carbon emissions. The Shandong Peninsula Urban Agglomeration (SPUA) and Central Plains Urban Agglomeration (CPUA) showed strong decoupling effects from 2016 to 2019. The Guanzhong Plain Urban Agglomeration (GPUA) demonstrated a strong decoupling effect from 2013 to 2019 (except from 2016 to 2017). The Lanxi Urban Agglomeration (LXUA) exhibited a strong decoupling effect from 2014 to 2019. Technological carbon intensity plays a decisive role in the transition from non-decoupling to decoupling. Therefore, the government must increase investments in green and low-carbon technologies and strictly implement carbon reduction measures. Thus, the YRB and its urban agglomerations have considerable potential for carbon emission reduction and strong decoupling effects.

Purpose: The prevalence of benign prostatic hyperplasia (BPH) in the general Chinese adult male population has risen sharply over the past few decades. Increasing evidence suggests that inflammation plays an important role in the pathogenesis of BPH. To better understand the role of inflammation in the pathogenesis of BPH, we can use the neutrophil-to-lymphocyte ratio (NLR) because it is a simple and effective marker of inflammation and immunity. This study aims to prospectively investigate the association between NLR levels and the prevalence of BPH in a general Chinese adult male population. Patients and Methods: This study included a total of 15,783 male participants free from BPH at baseline. NLR was measured according to the complete blood count. BPH was defined as total prostate volume (TPV) ≥30 mL, and TPV was determined by transabdominal ultrasonography. Multivariable Cox proportional hazards models were fitted to calculate hazards ratios (HRs) and corresponding 95% confidence intervals (CIs) for BPH risk with NLR levels. Results: During a median follow-up of 2.7 years, 5078 BPH cases were documented. After adjusting for age, body mass index, smoking, alcohol, education, occupation, income, physical activity, total energy intake, personal and family history of disease, and inflammation markers, the multivariable-adjusted HRs of BPH were 1.00 (reference), 1.08 (95% CIs 0.99, 1.17), 1.10 (95% CIs1.02, 1.19), and 1.12 (95% CIs1.03, 1.21), respectively, for participants with NLR in the first, second, third, and fourth quartiles (P for trend <0.01). Conclusion: Higher NLR levels were associated with a higher risk of BPH in Chinese adult male population. Our findings support the notion that NLR levels may be an important target for BPH prevention and intervention.

Glyphosate (GLY) exposure, both exogenous and endogenous, is a global concern. Multiple studies of model systems in vitro and in vivo have demonstrated the potential toxic effects of GLY exposure on human organs, particularly the liver and renal system. However, there is currently limited epidemiological evidence establishing a link between GLY exposure and hepatorenal function in the general population. In this study, a multivariable linear regression model and forest plots were employed to evaluate the connection between urinary GLY and biomarkers of hepatorenal function in 2241 participants from the National Health and Nutrition Examination Survey 2013-2016. Additionally, subgroup analyses were conducted based on age, gender, race, BMI, and chronic kidney disease (CKD). Alkaline phosphatase (ALP), alanine aminotransferase (ALT), aspartate aminotransferase (AST), AST/ALT and fibrosis 4 score (FIB-4) all increased with elevated urinary GLY concentrations after adjusting for potential confounders, while albumin (ALB) exhibited the opposite trend, particularly among younger, female, non-Hispanic white, overweight, and CKD participants. Furthermore, individuals in the third tertile had a greater risk of liver dysfunction than those in the first tertile after categorizing urinary GLY concentrations. However, our study showed no proof that GLY exposure affects the ratio of urine albumin to creatinine (ACR) or serum creatinine levels. Overall, these results imply that GLY exposure may have adverse effects on human liver function.

Mesenchymal stem cells (MSCs) exert beneficial therapeutic effects in acute kidney injury (AKI), while the detailed repair mechanism remains unclear. Herein, we probed the underlying mechanisms of MSC therapy in AKI by performing unbiased single-cell RNA sequencing in IRI model with/without MSC treatment. Our analyses uncovered the tubular epithelial cells (TECs) and immune cells transcriptomic diversity and highlighted a repair trajectory involving renal stem/progenitor cell differentiation. Our findings also suggested that profibrotic TECs expressing pro-fibrotic factors such as Zeb2 and Pdgfb promoted the recruitment of inflammatory monocytes and Th17 cells to injured kidney tissue, inducing TGF-ß1 secretion and renal fibrosis. Finally, in addition to activating the repair properties of renal progenitor/stem cells, we uncovered a role for MSC-derived miR-26a-5p in mediating the therapeutic effects of MSCs by inhibiting Zeb2 expression and suppressing pro-fibrotic TECs and its subsequent recruitment of immune cell subpopulations. These findings may help to optimize future AKI treatment strategies.

Purpose: This study aimed to prospectively investigate the association between mean platelet volume (MPV) levels and risk of benign prostatic hyperplasia (BPH) in a general Chinese adult male population, and assessed this association in metabolic syndrome (MetS) patients. Patients and methods: This study included a total of 14,923 male participants free from BPH at baseline. MPV was measured by the method of laser-based flow cytometric impedance according to the complete blood sample. BPH was defined as total prostate volume (TPV) ≥ 30 mL, TPV was determined by transabdominal ultrasonography. Multivariable Cox proportional hazards models were fitted to calculate hazards ratios (HRs) and corresponding 95% confidence intervals (CIs) for BPH risk with NLR levels. Results: During a median follow-up of 2.7 years, 4848 BPH cases were documented in total male participants, and 1787 BPH cases were documented in MetS participants. After adjusting for age, body mass index, smoking, alcohol and personal and family history of disease, the multivariable-adjusted HRs of BPH were 1.00 (reference), 1.03 (95% CIs 0.96, 1.11), 1.00 (95% CIs 0.92, 1.08) and 0.98 (95% CIs 0.90, 1.06), respectively, for participants with MPV in the 1st, 2nd, 3rd and 4th quartiles (P for trend = 0.47). In MetS patients, the multivariable-adjusted HRs of BPH were 1.00 (reference), 1.03 (95% CIs 0.90, 1.16), 0.99 (95% CIs 0.87, 1.14) and 1.01 (95% CIs 0.89, 1.15) (P for trend= 0.98), respectively. Conclusion: A non-significant association was observed between MPV levels and risk of BPH, and no association in this association in MetS patients. Our findings support the notion that MPV levels may not be a target for BPH prevention and intervention.

BACKGROUND: Genetic factors and muscle strength both contribute to the risk of major depressive disorder (MDD), but whether high muscle strength can offset the risk of MDD with different genetic risk is unknown. This study aims to examine whether a higher muscle strength is associated with lower risk of MDD regardless of genetic risk among middle-aged and older adults. METHODS: This cohort study obtained data from the UK Biobank, which includes 345,621 individuals aged 40-69 years (mean (standard deviation): 56.7 (7.99) years) without baseline MDD. Polygenic risk score for MDD was categorised as low, intermediate or high. The mean of the right- and left-hand grip strength values was used in the analysis and was divided into three categories. RESULTS: 9,753 individuals developed MDD within 2,752,461 person-years of follow-up. The multivariable adjusted hazard ratios (HRs) (95% confidence intervals (CIs)) of MDD across increased grip strength categories were 1.00, 0.72 (0.68-0.75) and 0.56 (0.53-0.59) (P for trend <0.0001). The HRs (95% CIs) of incident MDD across the genetic risk categories were 1.00, 1.11 (1.05-1.17) and 1.20 (1.13-1.28) (P for trend <0.0001); 4.07% of individuals with a high genetic risk and low grip strength developed MDD, and 1.72% of individuals with a low genetic risk and high grip strength developed MDD, with an HR (95% CI) of 0.44 (0.39-0.50). CONCLUSIONS: Both muscle strength and genetic risk were significantly associated with incident MDD. A higher muscle strength was associated with a lower MDD risk among individuals with a high genetic risk. Improving muscle strength should be encouraged for all individuals, including individuals with high genetic risk for MDD.

CONTEXT: Thyroid-stimulating hormone (TSH), as the most sensitive and specific marker of thyroid status, is associated with multiple health outcomes, including mortality. However, whether TSH levels are causally associated with the risk of mortality remains unclear. OBJECTIVE: This study aims to investigate the causal association between TSH levels and all-cause mortality using Mendelian randomization (MR) analyses. METHODS: MR analyses using single-nucleotide polymorphisms (SNPs) associated with TSH levels (P < 5 × 10-8) as instruments. Mortality data were obtained from the UK Biobank, including 384 344 participants who were recruited from 22 assessment centers across the UK taken between 2006 and 2010. Cox proportional hazards regression was used to estimate the association of the TSH genetic risk score (GRS) with all-cause and cause-specific mortality. RESULTS: 15 557 individuals died during a median of 9.00 years of follow-up in the UK Biobank. A total of 70 SNPs were included in the MR analysis. The main MR analyses showed that 1 SD increase in TSH was associated with a decreased risk of all-cause mortality (OR 0.972, 95% CI 0.948-0.996), which may be largely attributed to respiratory disease mortality (OR 0.881, 95% CI 0.805-0.963). The multivariable hazard ratios (HRs) (95% CI) of all-cause mortality across 3 TSH GRS categories were 1.00 (reference), 0.976 (0.940-1.014), and 0.947 (0.911-0.985), respectively (P for trend < .01). Moreover, except digestive diseases mortality, genetically predicted TSH levels were negatively associated with mortality from CVD, cancer, noncancer diseases causes, and dementia, although not statistically significant. CONCLUSION: Higher TSH levels were causally associated with lower risk of all-cause mortality, which may be largely attributed to respiratory disease mortality.

Background: Aplastic anemia (AA), a disease of bone marrow failure, is caused by CD8+T mediated apoptosis of hematopoietic cells. However, traditional immunosuppressive therapy (IST) has severe liver and kidney toxicity and even cannot achieve the expected therapeutic effect in some patients. Purpose: Our study is aimed to investigate the effect and mechanism of dioscin (DNS) for treating AA. Methods: Briefly, we established and evaluated the AA mouse model, DNS and positive control drugs were used for intervention treatment. After 14 days of intervention, femoral bone marrow pathology, bone marrow mononuclear cells (BMMCs) apoptosis rate, bone marrow CD34+ cell surface Fas (CD95) expression and Fas signaling pathway key proteins were detected. Results: After the establishment of the AA mouse model, the number of peripheral blood cells including granulocytes, erythrocytes, hemoglobin, platelets and reticulocytes in the AA group model was significantly decreased compared with the group control (P < 0.01). The degree of bone marrow hyperplasia in the sternum and femur is extremely low. After different drug interventions, compared with the group model, the number of peripheral blood cells in the AA mice rebounded significantly in group DNS (P < 0.01). Not only that the apoptosis rate of BM-MCs decreased (P < 0.01), meanwhile, the CD95 molecule expressed on the CD34+ bone marrow cells had a significant decline (P < 0.01), and the expression level of the key proteins of Fas signaling pathway was also significantly decreased (P < 0.01). Conclusion: DNS recovered the peripheral pancytopenia and bone marrow failure in AA mice. DNS reduced the key protein of Fas signaling pathway level to inhibit apoptosis of bone marrow cells to treat AA.

OBJECTIVES: The immune-induced aplastic anemia (AA) mouse model has been used for the study of AA. However, there were no uniform conditions for establishing a model and no assessment of immunological homeostasis. Our study aimed to identify the conditions of establishing a model and assess the AA model in immunology and pathology. METHODS: We induced an AA mouse model by the combination between sublethal irradiation and spleen-thymus lymphocyte infusion. The success of establishing the AA model was identified by blood routine tests and pathology of bone marrow. The frequency of Th17 and Treg cells was measured by flow cytometry. The frequency of CD34+ and CD41+ cells was detected by immunohistochemical technique.IL-6, IL-8, IL-17, TNF-α and IFN-γ were evaluated by ELISA. RESULTS: The 137Cs sublethal irradiation (5 Gy) and spleen-thymus lymphocyte infusion (5 × 106) induced the AA mouse model successfully. The AA mice had a long lifetime and manifested pancytopenia and bone marrow failure. The percentage of Th17 cells increased and the percentage of Treg cells decreased distinctly in AA mice. The area of hematopoietic tissues and count of CD34+ cells and CD41+ cells were significantly reduced in AA mice.The level of cytokines, IL-6, IL-8, IL-17, TNF-α and IFN-γ, was increased significantly in peripheral blood and bone marrow. CONCLUSION: Our data suggest that the improved AA mouse model conforms to the diagnosis standard of AA and simulates the immune internal environment of human AA. The AA mouse model has a longer lifetime and unbalances of Th17/Treg cells caused the destruction of CD34+ cells and CD41+ cells, which was immune-mediated pathogenesis to adapt to long-term research.

To achieve China's carbon peak and carbon neutrality goals, conducting systematic research on the energy, economy, and emission factors that affect the sustainable development of society is of great significance. This paper first uses the vector error correction model (VECM)-based Granger causality test to analyze the joint causal relations and feedback correction mechanisms among energy consumption, economic growth, and CO2 emissions in China from 1980 to 2019 at the energy heterogeneity level; then, analyzes the decoupling effect of China's four major energy sources (coal, oil, natural gas, and electricity) and economic growth from the perspective of energy heterogeneity; finally, the Tapio decoupling elastic model is decomposed into the emission reduction elasticity and energy saving elasticity to analyze the decoupling causality chain of the economy, energy, and carbon emissions. The research results show that there is a long-term, two-way causal relation between coal consumption and CO2; coal consumption has a one-way causal relation with economic growth; and long-term, two-way causal relations exist between oil and CO2, natural gas and CO2, electricity and CO2, electricity, and economic growth. In addition, when energy consumption, economic growth, and CO2 emissions deviate from their equilibrium states in the short term, various energy consumption, economic growth, and CO2 emissions in the previous year will be adjusted by 19.5%, 0.6%, …, 7.7%, and 3.4% to bring the nonequilibrium states back to the long-term equilibrium states. Furthermore, the energy-saving elasticity of China's total energy, coal, oil, and natural gas is the main factor affecting the corresponding decoupling elasticity, but the emission reduction elasticity of electricity has a stronger impact on the decoupling elasticity than the emission reduction elasticity does.

The present work investigated the differences in the composition and internal microstructure of four types gypsum rock-fiber gypsum, transparent gypsum, alabaster, and ordinary gypsum by X-ray fluorescence spectrometry, X-ray diffraction, scanning electron microscope and Brazilian split test, and analyzed its effects on the tensile strength and fracture characteristics of gypsum rock. For alabaster, fiber gypsum, transparent gypsum, and ordinary gypsum, CaSO4·2H2O is the main component with 72.78%, 72.72%, 72.57%, and 71.51% content, and tensile strength of 1.79, 2.22, 3.22, and 4.35 MPa, respectively. In addition, the fracture line is arc-shaped, vertical, and zigzag for fiber gypsum, ordinary and transparent gypsums, and alabaster, respectively. On the microscopic level, fiber gypsum has an evident striated structure while the gradual increased pore development for alabaster, transparent gypsum, and ordinary gypsum. Gypsum rock has an obvious layered crystal structure with the increase of CaSO4·2H2O, contributing to the phenomenon with a larger grain size and lower tensile strength. In addition, the number of particles for alabaster, transparent gypsum, and ordinary gypsum increased in turn, while their particle size decreased uniformly, indicating that the lower CaSO4·2H2O content, the more sufficient energy accumulation and release. This paper can provide a theoretical basis for the analysis of the mechanical properties of rocks with different mineral composition and contribute to the design for different ore grades mining.

Carbon emissions from tourism are an important indicator to measure the impact of tourism on environmental quality. As the world's largest industry, tourism has many related industries and is a strong driver of energy consumption. The emission reductions it can achieve will directly determine whether China's overall carbon emission reduction target can be met. This paper analyzes the drivers of the evolution of carbon emissions from the tourism industry in China over the period 2000-2017 as a research sample using the Generalized Dividing Index Method (GDIM), and on this basis, it uses scenario analysis and Monte Carlo simulation to predict the carbon peak in tourism for the first time. The research results show that the scale of industry and energy consumption are the key factors leading to increased tourism carbon emissions, and the carbon intensity of tourism industry, energy consumption carbon intensity, investment efficiency, and energy intensity are the main factors leading to reduced carbon emissions from tourism. The scale of investment and the carbon intensity of investment have a dual effect; the scenario analysis and Monte Carlo simulation used to predict peak carbon in China's tourism industry show that the peak carbon will occur approximately in 2030. The government needs to further guide and encourage the tourism industry to increase investment activities targeting energy conservation and emission reduction. Under the conditions of strictly implementing energy conservation and emission reduction measures and vigorous promotion of the transformation and upgrading of tourism development methods, the tourism industry will have considerable potential to reduce carbon emissions.

A comprehensive understanding of the relationships between greenhouse gas (GHG) emissions and industrial structure and economic growth holds great significance for China to realize the development of a green economy. This paper calculates GHG emissions based on China's energy consumption, divides the industrial structure in detail, and uses the extended Stochastic Impacts by Regression on Population, Affluence, and Technology model that is realized by PLS method and Tapio decoupling model to study the relationship of GHG emissions to industrial structure and economic growth. The results show that (1) China's total GHG emissions showed a year-on-year growth trend from 2000 to 2017. For CO2, CH4, and N2O, only N2O emission showed a significant downward trend, while CO2 and CH4 emissions showed a slow growth trend. (2) The proportions of added value of industry and construction are positively correlated with GHG emissions, while those of farming, forestry, animal husbandry, and fishery; wholesale and retail trade; transport; and accommodation and catering are negatively correlated with GHG emissions. (3) China's GHG emissions and overall economic growth are in a decoupling state, but in the energy field, N2O emission reduction control has the best effect. Additionally, the overall economic growth of China's industrial sector and GHG emissions have experienced the process of decoupling-link-negative decoupling-link-decoupling. Graphical abstract.

Inflammation plays a crucial role in the formation of benign breast disease. Given the limited study to explore the association between leukocyte as an indicator of immune system and benign breast disease, we used data from a large cross-sectional study to investigate association between leukocyte and its subtypes and benign breast disease among women in the general population. The data were derived from baseline data of the Tianjin chronic low-grade systemic inflammation and health (TCLSIH) cohort study during 2014 and 2016. Breast thickness and nodules status were assessed by using ultrasonography. Leukocyte and its subtype counts were carried out using the automated hematology analyzer. Multiple logistic regression analysis was used to examine the association between leukocyte and its subtypes and prevalence of benign breast disease. In the present study, the prevalence of benign breast disease was 20.9%. After adjustments for potentially confounding factors, the odds ratios (95% confidence interval) for benign breast disease across lymphocyte quintiles were as follows: 1.00 (reference), 0.99 (0.82, 1.2), 0.85 (0.69, 1.04), 0.84 (0.68, 1.02), and 0.75 (0.61, 0.92) (P for trend = 0.002). An inverse association between lymphocyte counts and benign breast disease was found, but leukocyte and other subtypes have nothing to do with benign breast disease. Further prospective studies are needed to determine the findings.

A comprehensive understanding of the relationships among carbon emissions, the industrial structure and economic growth holds great significance for China's transition to a low-carbon economy, industrial structure optimization, and achievement of energy conservation and emission reduction targets. We selected six major industrial sectors (agriculture, industry, construction, transportation, retail and accommodation and other industries) as research objects, introduced the extended STIRPAT decomposition model, Tapio decoupling model and the grey relation analysis to discuss the relationship among the three. Results showed that (i) since 2000, the proportions of value added of agriculture, manufacturing, and transportation are negatively correlated with carbon emissions, while those of construction, retail and accommodation, and other industries are positively correlated with carbon emissions. (ii) The overall economic growth and carbon emissions of these six major industries have experienced the process of decoupling-coupling-decoupling-coupling-decoupling. (iii) The relevance of these six industries to GDP is ranked as follows: transportation > manufacturing > retail andaccommodation > agriculture > construction > other industries. Additionally, accelerating the achievement of a clean energy structure, strengthening the strength and speed of industrial structure adjustment and reducing the dependence on fossil energy are the key steps for China to reach carbon emissions peak goal.

BACKGROUND: Patients with coronary heart disease (CHD) angina pectoris are in critical condition, which can cause sudden death, myocardial infarction, and other adverse events, and bring serious burden to families and society. Timely treatment should be given to improve the condition. Western medicine treatment of angina pectoris failed to meet the demand of angina symptom control. OBJECTIVE: It is hoped that the research method with higher evidential value will be adopted to compare the short-term, medium-term, and long-term effects of Chinese patent medicine combined with conventional western medicine and conventional western medicine alone in the treatment of CHD angina pectoris, so as to tap the clinical efficacy advantages of traditional Chinese medicine (TCM) and provide reliable data support for its clinical application. METHODS: A prospective cohort study was conducted among patients with CHD angina pectoris who were treated with oral Chinese patent medicine and conventional western medicine. The patients were divided into exposed group and nonexposed group according to whether or not the patients with CHD angina pectoris were treated with Chinese patent medicine. The exposed group was treated with TCM combined with conventional western medicine, while the nonexposed group was treated with conventional western medicine alone. Patients need to be hospitalized for 2 weeks as the introduction period and whether to enter the group is determined according to the treatment and medication conditions of the patients. The follow-up time points were 0th, 4th, 12th, 24th, and 48th weeks. The main events and secondary events were used as the evaluation criteria for clinical efficacy of CHD angina pectoris. In the experimental study, we will use strict indicators to detect standard operation procedure for multinomics and bacterial flora detection. CONCLUSION: This study will provide evidence for the clinical efficacy advantages of Chinese patent medicine and reliable support for its clinical application through test data.

BACKGROUND: As a recognized risk factor for cardiovascular disease (CVD), hyperlipidemia (HLP) has developed into a high incidence disease that seriously threatens human health. Finding a new target for effective treatment of HLP will be a powerful way to reduce the incidence of CVD. The purpose of this study was to find potential biomarkers in urine of HLP patients and analyze their metabolic pathways to study the pathogenesis of HLP. METHODS: An UPLC-Q-TOF/MS technology was used to detect the metabolites in urine of 60 HLP patients and 60 normal controls. Based on PLS-DA pattern recognition, potential biomarkers related to HLP were screened out. RESULTS: 22 potential biomarkers related to HLP were identified, which involved amino acid metabolism, fatty acid metabolism, nucleotide metabolism, steroid hormone metabolism and intestinal flora metabolism, and their possible pathogenesis was found to be related to inflammatory reaction and oxidative stress. CONCLUSION: The non-targeted metabolomic method based on UPLC-Q-TOF/MS technology can effectively identify potential biomarkers in the urine of HLP patients and explore the possible pathogenesis. Our research will lay a foundation for finding new targets for the treatment of HLP and provide a basis for clinical research on the treatment of HLP.

Coronary heart disease (CHD) threatens human health. The discovery and assessment of potential biometabolic markers for different syndrome types of CHD may contribute to decipher pathophysiological mechanisms and identify new targets for diagnosis and treatment. On the basis of UPLC-Q-TOF/MS metabolomics technology, urine samples of 1072 participants from nine centers, including normal control, phlegm and blood stasis (PBS) syndrome and Qi and Yin deficiency (QYD) syndrome, and other syndromes of CHD, were conducted to find biomarkers. Among them, the discovery set ( n = 125) and the test set ( n = 337) were used to identify and validate biomarkers, and the validation set ( n = 610) was used for the application and evaluation of the support vector machine (SVM) prediction model. We discovered 15 CHD-PBS syndrome biomarkers and 12 CHD-QYD syndrome biomarkers, and the receiver-operator characteristic (ROC) area-under-the-curve (AUC) values of them were 0.963 and 0.990. The established SVM model has a good diagnostic ability and can well distinguish the two syndromes of CHD with a high predicted accuracy >98.0%. The discovery of biomarkers and metabolic pathways in different syndrome types of CHD provides a basis for the diagnosis and evaluation of CHD, thereby improving the accurate diagnosis and precise treatment level of Chinese medicine.

BACKGROUND: In recent years, using metabolomics technology to study hypertension has made some progress. However, in actual clinical studies, there are few studies on hypertension related metabonomics with human urine as samples. In this study, the urine samples of patients with essential hypertension (EH) were studied by comparing with healthy people to explore the changes of urine metabolites between hypertensive patients and healthy people in order to find potential biomarkers and metabolic pathways. METHODS: An ultra performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF/MS) technology was used to analyze the urine metabolites of 75 cases of essential hypertension group (EH) and 75 cases of healthy control group (HC). RESULTS: According to the PLS-DA pattern recognition analysis, substances with significant differences (P < .05) between the EH group and the HC group were screened out, including 10 potential biomarkers such as L-methionine. The metabolic pathways involved were amino acid metabolism, fatty acid metabolism steroid hormone, biosynthesis and oxidative stress. CONCLUSION: The non-targeted metabolomics based on UPLC-Q-TOF/MS technology can effectively identify the differential metabolites of potential biomarkers in the urine of essential hypertensive patients and provide a theoretical basis for the treatment of clinical hypertension.
