RESUMEN

Acetophenones are naturally occurring phenolic compounds which have found in over 24 plant families and also fungi strains. They are exist in both free or glycosides form in nature. The biological activities of these compounds have been assayed and reported including cytotoxicity, antimicrobial, antimalarial, antioxidant and antityrosinase activities. Herein, we review the chemistry and biological activity of natural acetophenone derivatives that have been isolated and identified until January 2024. Taken together, it was reported 252 acetophenone derivatives in which the genera Melicope (69) and Acronychia (44) were the principal species as producers of acetophenones.

The current study assessed the risk posed to Iranian consumers by oral exposure to a mixture of ten mycotoxins in 138 packaged and unpackaged spices collected from the Iran market. Concentrations of mycotoxins in samples were quantified by liquid chromatography, tandem mass spectrometry with triple quadrupole, and ion trap. Probabilistic health risks of oral exposure to these mycotoxins for Iranians were assessed under percent tolerable daily intake (TDI) and cancer risk scenarios. Mean concentrations of mycotoxins in both packaged and unpackaged spice samples showed statistically significant variation among different spice samples. Based on a Monte Carlo simulation model, at the 50th, 80th, and 95th centiles, oral consumption of the analyzed samples poses no carcinogenic risk for exposure to aflatoxin. Moreover, in both packaged and unpackaged samples, while the percent TDIs for ochratoxin A, deoxynivalenol, zearalenone, patulin, fumonisin B1, and fumonisin B2 were below 1.0 at the 50th, 80th, and 95th centiles, the value was above 1.0 for aflatoxin B1, aflatoxin B2, aflatoxin G1, and aflatoxin G2 at each of these centiles.

In the current study, we assessed health risk posed to Iranian consumers through exposure to metals via oral consumption of coffee, tea, and herbal tea of various trademarks collected from Iran market. Level of As, Cd, Cr, Cu, Fe, Hg, Ni, and Pb in 243 samples was quantified by inductively coupled plasma-optical emission spectrometry (ICP-OES). The metal levels in coffee samples from different trademarks of a specific country had statistically similar levels of metals; however, metal levels differed significantly among brand names form different countries. Metal levels in tea samples differed significantly between domestic and imported products, while different trademarks of similar countries did not show significant variations in this respect. Metal level in herbal tea samples did not show significant variations among different trademarks. Nevertheless, it should be highlighted that mean concentrations of metals statistically differed among different herbal tea samples. Deterministic hazard quotients (HQs) were <1.0 for all non-carcinogenic metals and total hazard index (HI) values indicated no risk; however, probabilistic assessment calculated HI values >1. In both deterministic and probabilistic scenarios, carcinogenic metals As and Ni had an estimated incremental lifetime cancer risk (ILCR) of medium level while that of Pb indicated no cancer risk. Sensitivity analysis showed that the concentration of metals had the most significant effect on non-carcinogenic and carcinogenic risks.

The current study assessed the risk posed to Iranian consumers by oral exposure to a mixture of 20 pesticides and six metals in 96 fruit juice (FJ) samples (3 batches × 4 brands × 8 types of FJs) collected from Iran market. Concentrations of metals and pesticides in FJs were quantified by inductively coupled plasma-optical emission spectrometry (ICP-OES) and chromatography-mass spectrometry (GC-MS), respectively. The mean concentration of all pesticides was below the maximum residue limits (MRLs) set by the European Union (EU). The calculated target hazard quotients (THQs) and total hazard index (HI) were <1.0 for all pesticides residue, indicating no risk. For the carcinogenic metals (As, Ni, and Pb), estimated incremental lifetime cancer risks (ILCRs) at the 50th and 95th centiles were respectively 4.25 × 10-5 and 5.30 × 10-5 (for As), 2.85 × 10-5 and 3.71 × 10-5 (for Ni), and 2.84 × 10-8, and 3.97 × 10-8 (for Pb), indicating no risk. At the 50th and 95th centiles, HI for non-carcinogenic metals (Cd, Hg, and Cr) was <1.0, indicating no risk. Based on sensitivity analyses of the input variables, the concentration of metals and pesticides, and the FJs ingestion rate had significant influential impacts on the calculated THQ and HI.

Sweet fennel (Foeniculum vulgare Mill. var. dulce) and thyme (Zataria multiflora Boiss.) are regarded as the important supplies for pharmaceutical, food, cosmetic, and perfume industries. The major components trans-anethole and thymol are represented in fennel and thyme, respectively. The essential oils (EOs) content and the value of their related constituents should be given in strict quality control due to the storage conditions, source, and adulterations. In this study, we compared the validation of quantitative 1H NMR (qH NMR) method with the gas chromatography with flame ionization detection (GC-FID) to quantify the trans-anethole and thymol in fennel and thyme EOs and their related supplements. The current results showed that the quantification of trans-anethole and thymol by qH NMR method was successfully achieved from their EOs and supplements. All the validation parameters including linearity, robustness, repeatability, and stability were authenticated for thymol and trans-anethole quantification. Similar results were obtained in both qH NMR and conventional GC-FID methods. Therefore, according to the measured values, the qH NMR method was adequate to determine the constituents of the EOs, with the results being roughly comparable to those obtained by GC-FID, with the advantage of being simple, repeatable, rapid (8-10 min, while for GC-FID 55 min) and essential for quality control of commercial samples.

Dracocephalum moldavica L. is a significant component in the Iranian food basket. This study aimed to investigate the bioactive compounds and biological activities of different extracts obtained from D. moldavica aerial parts. From the aerial parts, a crude methanolic (MeOH) extract and its four sub-fractions, that is, petroleum ether (Pet), ethyl acetate (EtOAc), n-butanol (n-BuOH), and aqueous (water) extracts were obtained. The total phenolic and flavonoid contents as well as the antioxidant and cytotoxic activities of the extracts were determined. Moreover, the phytochemical profiles of the essential oil (EO) and of those extracts with the highest antioxidant activity measured by GC/MS and UPLC-PDA-ESI-QTOF-MS/MS. Results showed that the highest concentrations of phenols and flavonoids as well as the most potent antioxidant potential according to the DPPH method were determined in the EtOAc and MeOH extracts with IC50 values of 22.0 and 34.4 µg.ml-1, respectively. Quantitative analysis of these extracts was subsequently performed by UPLC-PDA-ESI-QTOF-MS/MS. Both extracts contained mainly rosmarinic acid, caffeic acid, and 2-hydroxycinnamic acid, which may be responsible for their high antioxidant activity. Moreover, none of the extracts showed cytotoxic effects against MCF7, SW48, and a normal cell line of mouse embryonic fibroblast cells (NIH/3T3) in the tested concentrations (up to 400 µg.ml-1). Additionally, GC-MS analysis showed that oxygenated monoterpenes (55.4%) were the main constituents of the EO of D. moldavica.

To assist the development of new therapeutic strategies for several disorders, biologically active peptides/proteins obtained from plant sources can be considered. Current study expected to determine the biological activities of peptide fractions of Mucuna pruriens against hepatocellular carcinoma cell lines (HepG2/ADM, HepG2, SMMC-7721, and QGY-7703), as well as normal cell line to prove their selectivity. Moreover, anti-genotoxicity and antiviral activity against the hepatitis C virus (HCV) were assessed. The methods of this study were to isolate the peptides of M. pruriens and hydrolysate fractionation via fractionated pepsin-pancreatin hydrolysates by ultrafiltration/high-performance ultrafiltration cell, identify anti-hepatoma activity of peptide fractions human liver cancer and normal cells by (3-(4,5-dimethylthiazole-2-yl)-2,5-biphenyl tetrazolium bromide) (MTT) assay, determine anti-HCV, and assess anti-genotoxic effect of peptide fractions against damage that induced via alkylating agent methyl methanesulphonate in human mononuclear cells. The results showed that the fraction 5-10 kDa has been reported to exhibit significant cytotoxic activity against HepG2 and QGY-7703. It was proven that both of 5-10 and >10 kDa fractions are active against HCV. The cytotoxic concentration (CC50) of 5-10 kDa against the cell line was 703.04 ± 5.21 µg/ml. Anti-genotoxic activities of the peptide fractions were evaluated as mean values for the analyzed comet images. In this regard, the highest activity of protecting DNA damages was observed by the peptide fraction of 5-10 kDa. This study revealed the potential ability of peptide fractions of M. pruriens for the treatment of liver cancer, HCV, and high activities of protecting DNA damages.

In the present study, levels of 22 pesticides, eight metals, and 16 polycyclic aromatic hydrocarbons (PAHs) in 1800 Iranian olive samples (20 cultivars from six different cultivation zones), were determined; then, health risk posed by oral consumption of the olive samples to Iranian consumers was assessed. Quantification of PAHs and pesticides was done by chromatography-mass spectrometry (GC-MS), and metal levels were determined using inductively coupled plasma-optical emission spectrometry (ICP-OES). There were no significant differences among the cultivars and zones in terms of the levels of the tested compounds. Target hazard quotients (THQ) were <1.0 for all pesticides, and total hazard indices (HI) indicated di minimis risk. At the 25th or 95th centiles, Incremental Life Time Cancer Risks (ILCRs) for carcinogenic elements, arsenic, and lead and noncarcinogenic metals did not exhibit a significant hazard (HI <1.0 for both cases). At the 25th or 95th centiles, ILCR and margins of exposure (MoE) for PAHs indicated di minimis risk. Sensitivity analysis showed that concentrations of contaminants had the most significant effect on carcinogenic and noncarcinogenic risks.

Saffron (Crocus sativus L.) is a highly valuable plant. Iran provides nearly 90% of the world's total saffron and is the biggest global producer. The allelopathic effects of saffron corm (SC) and saffron field soil (SFS) have been hypothesized to play an important role in replanting. Recently, adsorbent materials have been used to neutralize the effects of allelochemicals. These materials, including activated carbon and zeolite, have large surface areas, pore volumes, as well as tremendous adsorptive capacity and complex chemical and physical properties. In this study, three independent experiments were conducted. In the first test, the allelopathic effects of aqueous and methanolic extracts of SC remnant and 9-year-old SFS as well as filtered aqueous extract of soil were investigated. In the second assay, the effects of SC remnants and SFS with different ages (i.e., 4, 6, and 9 years old) in combination with adsorbents were examined on the germination and growth of lettuce (Lactuca sativa L.) seedlings by the sandwich method. In the third experiment, we examined the effects of SC remnants combined with adsorbents on lettuce growth parameters. Our results showed that the allelopathic effects of aqueous and methanolic extracts of SC remnant were significantly superior to those of 9-year-old SFS. The aqueous extract of SC remnant reduced the root length of lettuce by 50%. The use of activated carbon and zeolites significantly decreased the observed allelopathic effect. Moreover, lettuce growth in rhizosphere soil was significantly inhibited by SC remnant and SFS extracts. The allelopathic effects of SC remnants caused a growth imbalance between the shoot and roots. Based on biochemical analyses, using the adsorbents increased the carotenoid content and chlorophyll index of lettuce by 23.33% and 5.25%, respectively. Adsorbents may play a role in treating soils contaminated by allelochemicals.

The heavy metal levels in six walnut cultivars from five geographical zones of Iran were measured. An assessment of risks was conducted by calculating the Target Hazard Quotient (THQ) and Incremental Lifetime Cancer Risk (ILCR) by use of the Monte Carlo simulation method. The highest amounts of As and Pb were reported in Farouj samples. Also, the highest levels of Cr, Zn, Cu and Mn were determined in samples collected from Tuyserkan. Accordingly, 50th and 95th ILCRs for general population due to consumption of walnut were 1.03 × 10-4 and 3.11 × 10-4 (for As), 4.10 × 10-6 and 1.1 × 10-5 (for Cr) and 4.71 × 10-9 and 1.05 × 10-8 (for Pb), respectively. In addition, the 50th and 95th centiles of the HIs for walnut ingestion by Iranians were 1.02 and 2.05, respectively, indicating a minor chance of non-cancer effects. Based on the calculated 95% ILCR, dietary exposure to As through the consumption of walnut poses a risk to Iranian consumer health. However, ILCR values of other heavy metals (HMs) were in acceptable ranges (ILCR < 1 × 10-4), representing no toxicological concern for consumers. The most significantly influential parameters were determined by sensitivity analysis during the MCS. According to THQ and ILCR methods, concentration was the most sensitive parameters. For THQ method the concentration effects were ranged from 72.4 to 85.1%. Moreover, for ILCR method the effects of concentration in As, Cr, and Pb were 87.1, 79.1 and 83.54%, respectively.

Risks based on cancer and non-cancer endpoints, to Iranians from exposure to several mycotoxins (aflatoxin B1, ochratoxin, deoxynivalenol and T-2 toxin) following consumption of rice were evaluated. Point estimates of hazard were made for each mycotoxin and a hazard index (HI) and probabilistic estimates were based on results of Monte Carlo Simulations (MCS). All known 17 peer-reviewed studies, published in databases included in Science Direct, PubMed, Scopus and Web of Science, as well as grey literature published in Google Scholar from 2008 to 2017 were considered. The 95th and 50th centiles of Hazard Index (HI) in Iranians due to ingestion of rice were estimated to be 2.5 and 0.5, respectively. The 95th and 50th centiles of people with positive surface antigens for hepatitis B (HBsAg+) risk characterisation for AFB1 in Iranian consumers of rice were 81 and 79.1, respectively. The 95th and 50th centiles for risks of Iranians negative for the surface antigen of hepatitis B HBsAg (HBsAg-) were 4.4 and 2.6, respectively. Based on results of the MCS for risks to cancer effects, the 95th and 50th centiles of margins of exposure (MOE) were 233 and 231, respectively. Therefore, it is recommended to update agricultural approaches and storage methods and implement monitoring and regulations based on risks to health posed by consumption of rice by the Iranian population.

Twenty-seven samples of infant formulae and follow-on formulae and fifteen samples of baby food from Iranian markets were analyzed for concentrations of four polycyclic aromatic hydrocarbons (PAH4) determined by use of gas chromatography coupled to mass spectrophotometry. An assessment of risks posed to infants and toddlers was conducted by calculating the margin of exposure and incremental lifetime cancer risk (ILCR) by use of the Monte Carlo Simulation Method. Benzo (a) anthracene, was not detected in any of the samples, while approximately 64.3% samples contained detectable amounts of benzo (a) pyrene, while chrysene was observed in three samples and benzo (b) fluoranthene was detected in one sample. One of the samples contained 1.43 µg PAH4/kg, which was greater than the maximum tolerable limit (MTL; 1 µg/kg) stated in Commission Regulation (EU) 2015/1125. Accordingly, the 95% ILCRs in the infants/toddlers due to ingestion of milk powder and baby foods were determined to be 1.3 × 10-6 and 7.3 × 10-7, respectively. Also, the 95th centiles of the MOEs, due to ingesting milk powder or baby foods by infants/toddlers were estimated to be 3.6 × 104 and 7.2 × 104, respectively. In Iran, infants and toddlers are not at serious health risk (MOE ≥ 1 × 104 and ILCR < 1 × 10-4).

Iran is a major supplier of the world pistachio market. In this study, we collected five pistachio cultivars from four main pistachio-producing zones in August and September 2016, and determined the residues of 18 organophosphorus, carbamate, pyrethroid and nicotinoid pesticide in these samples using Gas Chromatography-Mass Spectrometry, as an efficient method for determination of pesticides' residues. Next, single-chemical and chronic cumulative risk assessment was done based on the new approaches of the food specific Hazard Quotient and adversity specific Hazard Index. Fifteen from eighteen food-specific Hazard Quotients were above 1 even in cases when the respective contamination was bellow MRLs. The adversity specific Hazard Indexes values were above 1 for five from six adversities indicating various risks in the resulted levels of pistachios' contamination from the pesticides' mixture. However, no risk for carcinogenicity was found. Our results indicate the necessity of taking appropriate measures to control/standardize pesticides practice in pistachio cultivation in Iran and the need to re-establish the MRLs based on cumulative exposure.

The present experiment evaluated how coronatine (COR) elicitation affects chemical and biological properties of cumin (Cuminum cyminum L.) seed essential oil (CSEO). Following isolation of the EO, its chemical composition was analyzed by gas chromatography-mass spectrometry; also, its bioactivities in terms of antimicrobial/antifungal, cytotoxic (measured by MTT assay) and antioxidant effects (evaluated by DPPH, ß-carotene bleaching (BCB) and TBARS methods) were evaluated. COR-elicitation significantly increased CSEO yield and the level of its chemical components, especially cumin aldehyde which is the main component of CSEO. Results showed that COR-elicitation significantly reduced the minimum inhibitory concentration (MIC) and the minimum bactericidal concentration (MBC) of CSEO against 4 Gram-positive and 3 Gram-negative bacteria and 2 fungi. Moreover, elicitation markedly enhanced the antioxidant and in vitro cytotoxic activity of CSEO. Therefore, COR may be regarded as a useful biotic elicitor for improving EO chemical and biological properties.

The use of some Stachys genus as herbal remedies is known and the aerial parts have a pharmaceutical interest, being used in Anatolia and Iran as wild tea. In this study, chemical composition, antimicrobial, antioxidant, and antiproliferative activities of the methanolic extract and essential oil (EO) of Stachys parviflora L. (S. parviflora) were evaluated. Qualitative analysis of metabolites of S. parviflora methanolic extract was studied using liquid chromatography coupled to high resolution mass spectrometry (LC-ESI/LTQOrbitrap/MS), evidencing the presence of phenolic acids and flavonoids derivatives. The EO was analyzed using gas chromatography coupled to mass spectrometry (GC/MS). Eighty-seven compounds were characterized in the EO of S. parviflora, of which α-terpenyl acetate (23.6%), ß-caryophyllene (16.8%), bicyclogermacrene (9.3%), spathulenol (4.9%) and α-pinene (4.2%) were found to be the major components. The highest antimicrobial effect of EO was found to S. aureus and B. cereus (MIC = 0.01 µg/ml), while the highest activity of extract was against B. cereus (MIC = 125 µg/ml). The methanolic extract exhibited strong antioxidant activity in DPPH (IC50 = 76.87 µg/ml) and ß-carotene/linoleic acid assay (BCB, IC50 = 188.47 µg/ml) methods. Furthermore, in vitro cytotoxicity evaluation against three cell lines namely human ovarian carcinoma (A2780), human colon carcinoma (HCT), and mouse melanoma cell line (B16F10), showed an anti-proliferative activity of the EO ranging from IC50 value 30.95 µg/ml to 16.55 µg/ml. The results from this study have demonstrated the promising cytotoxic, antibacterial, and antifungal properties of S. parviflora, which could have wide potential applications in food and pharmaceutical industries.

Chemical composition, antibacterial, antioxidant and cytotoxic activities of (Pistacia khinjuk) hull essential oil (EO) were evaluated in this study. The EO was isolated and analyzed by gas chromatography-mass spectrometry. The minimum inhibitory concentration (MIC) and the minimum bactericidal concentration (MBC) were determined using 6 strains of Gram-positive and negative bacteria. DPPH radical scavenging (DPPH) and ß-Caroten Bleaching (BCB) assays were used to measure antioxidant activity of the EO. In vitro cytotoxic activity was measured using MTT assay. Fifty-six compounds representing 99.5% of the total oil composition were identified. In the antibacterial results, Staphylococcus aureus was found to be the most susceptible strain (MIC and MBC = 16 µg/ml). Antioxidant IC50 values were respectively 19.03 ±â€¯0.001 and 49.22 ±â€¯0.005 µg/mL. The IC50 indexes of cytotoxic tests were 29.6, 37.3 and 41.1 µg/mL for MCF-7, PC3 and DU-145 cell lines, respectively.

Iran is one of the main suppliers of pistachio for the European market accounting for over 90% of its demands; hence, efficient analytical methods are required for detection of mycotoxins contamination in pistachio kernels before exporting them. In this study, aflatoxin B1 (AFB1) and ochratoxin A (OTA) levels in five pistachio cultivars collected from four sites of Iran, were measured by HPLC. Based on the results, risk assessment for AFB1 and OTA residues was done. The highest mean concentrations of AFB1 and OTA were found in Ahmad-aghaei (4.33 and 2.19 ng/g, respectively) and Akbari (4.08 and 1.943 ng/g, respectively) cultivars from Rafsanjan, Iran. Even the highest concentrations of AFB1 and OTA in analyzed samples were lower than the corresponding maximum limits set by EU authorities. The hazard index (HI) value for consumers of Iranian pistachio is below one. It could be concluded that consumption of pistachio cultivated in these regions poses no health risk of mycotoxins exposure.

OBJECTIVE: In this study, the levels and antioxidant activities of some secondary metabolites isolated from five pistachio (Pistacia vera) cultivars collected from four different geographical regions of Iran, were studied. MATERIALS AND METHODS: Total phenolic compounds levels were determined by Folin-Ciocalteu method. Total flavonoid content was determined as AlCl3 complex and expressed as mg of quercetin equivalents (QE)/g dry extract and total proantocyanidins content was expressed as mg of catechin equivalents (CA)/g dry extract. In order to evaluated the antioxidant activity of the compounds, DPPH and FRAP assays were used. RESULTS: The highest level of total phenols (156.42 mg GA/g DE), total flavonoids (130.94 mg QE/g DE) and total proantocyanidins (152.816 mg CA/g DE) were obtained in Akbari cultivar from Rafsanjan, followed by Badami-e-sefid and Ahmad aghaei. The lowest amount of total phenolic content (TPC), total flavonoid content (TFC) and total proanthocyanidin content (TPrAC) were found in Badami-e-sefid from Feizabad (128.140 mg GA/g DE, 93.176 mg QE/g DE and 118.870 mg CA/g DE, respectively). Also, a positive correlation (r2=0.9834) was found between antioxidant activity and total phenolic compounds. CONCLUSION: Pistachio increased their phytochemical compounds to contrast with abiotic stress. Our data could be useful for introducing special characteristics to the plants, and can be considered when planning a new breeding program or choosing a specific cultivar for a particular use.

Pistachio is an important horticultural product and Iran is considered as a main pistachio producing country. Assessment of heavy metals in this export fruit is crucial for protecting public health against toxic heavy metals. The concentration of selected heavy metals in soil, water and five pistachio cultivars from four geographical regions of Iran were measured. Although none of the elements were detected in water irrigation, infield metal content in the soil had good correlation with that of pistachio. The highest amounts of Al, As, Co, Ni and Se were reported in samples collected from Sarakhs, Iran. Considering both cultivar and region effects on selected heavy metals concentration, Kaleghoochi cultivar from Sarakhs site showed the highest amount of Al, As, Ni and Se. The maximum concentration of Hg was found in Akbari cultivar collected from Damghan. In the Akbari and the Ahmad aghaei cultivars collected from Sarakhs and Damghan cultivation zones, respectively, the highest amount of Co were observed. Based on our results, the HI value for the consumers of Iranian pistachio was 0.066. It seems that the levels of heavy metals in these pistachio samples pose no risk to consumers.
