RESUMEN

Azobenzene (azo)-based photothermal energy storage systems have garnered great interest for their potential in solar energy conversion and storage but suffer from limitations including rely on solvents and specific wavelengths for charging process, short storage lifetime, low heat release temperature during discharging, strong rigidity and poor wearability. To address these issues, an azo-based fabric composed of tetra-ortho-fluorinated photo-liquefiable azobenzene monomer and polyacrylonitrile fabric template is fabricated using electrospinning. This fabric excels in efficient photo-charging (green light) and discharging (blue light) under visible light range, solvent-free operation, long-term energy storage (706 days), and good capacity of releasing high-temperature heat (80-95 °C) at room temperature and cold environments. In addition, the fabric maintains high flexibility without evident loss of energy-storage performance upon 1500 bending cycles, 18-h washing or 6-h soaking. The generated heat from charged fabric is facilitated by the Z-to-E isomerization energy, phase transition latent heat, and the photothermal effect of 420 nm light irradiation. Meanwhile, the temperature of heat release can be personalized for thermal management by adjusting the light intensity. It is applicable for room-temperature thermal therapy and can provide heat to the body in cold environments, that presenting a promising candidate for wearable personal thermal management.

In this study, the polysaccharide (RHCP) extracted from Houttuynia cordata rhizome was acetylated through the acetic anhydride method. The physicochemical properties of RHCP and its acetylated derivatives (Ac-RHCP) were determined by infrared spectra, scanning electron microscopy, and Congo red test. Meanwhile, the α-glucosidase inhibition mechanism of RHCP and Ac-RHCP was analyzed by inhibition kinetics, and circular dichroism and fluorescence spectroscopy. Ac-RHCP resulted in a more porous surface structure and 1.83-fold higher solubility compared with RHCP. At a concentration of 6 mg/mL, the α-glucosidase inhibition rate of Ac-RHCP was 75.40%, while that of RHCP was 44.68%. RHCP and Ac-RHCP inhibited α-glucosidase in a mixed-type manner, reduced the endogenous fluorescence of α-glucosidase, affected the microenvironment of amino acid residues, and changed the conformation of α-glucosidase. The study indicates that Ac-RHCP exhibits a certain level of α-glucosidase inhibition, demonstrating its potential as a functional food for glycemic control.

Mulberry pomace is rich in phytochemicals, but there are few studies on its utilization as a by-product. Natural foods containing phytochemicals can alleviate the toxic effects of excessive glucose intake. In this study, we investigated the protective effect of Lactobacillus plantarum-fermented mulberry pomace extract (FMPE) under hyperglycemic conditions. The phenolic compounds and α-glucosidase inhibition of FMPE were determined using UPLC-MS and chemical models. Furthermore, Caenorhabditis elegans was a model system to study the hypoglycemic effects. The results showed that the polyphenolics and α-glucosidase inhibition were improved during fermentation. Three phenolic components (cyanidin, 2,4,6-trihydroxybenzaldehyde, and taxifolin) were important variables for α-glucosidase inhibition. FMPE and the three key compound treatments reduced the glucose content and reactive oxygen species (ROS) level in Caenorhabditis elegans. The protective mechanism occurred by activating DAF-16/FOXO and SKN-1/Nrf2. This study suggests that Lactobacillus plantarum-fermentation was a potential way to utilize mulberry pomace polyphenols as hypoglycemic food ingredients.

The aim of this study was to investigate the modulation of gut microbiota by fermented raspberry juice (FRJ) both in vitro and in vivo. Results showed that total phenolic content and antioxidant activities of FRJ reached the highest after fermentation for 42 h. Seventeen phenolic compounds were contained in FRJ, mainly including ellagic acid (496.64 ± 2.91 µg/g) and anthocyanins (total concentration: 387.93 µg/g). FRJ modulated the gut microbiota into a healthy in vitro status, with increase of valeric and isovaleric acids production. In healthy mice, all FRJ treatments improved the production of acetic, butyric and isovaleric acids as well as the gene expression of ZO-1, Claudin-1, Claudin-4, Ocdudin, E-cadherin and Muc-2. Moreover, variable gut microbial compositions were found among the groups fed diet-supplemented the different doses of FRJ, within low and median doses of FRJ may regulate the microbiota to a healthier state compared to the high dose supplementation. This study indicated that fermentation is a potential way to produce plant-based juices, which could reshape the gut microbiota and improve the host health.

Intestinal secretory immunoglobulin A (sIgA)-improving function of Lactobacillus casei-fermented blueberry pomace (FBP) was investigated in this study. Male C57BL/6 mice were fed with control diet (CD) or high-fat diet (HFD) with or without FBP supplementation. Expressions of sIgA-associated genes/proteins were evaluated by quantitative polymerase chain reaction (qPCR), western blot and enzyme-linked immunosorbent assay (ELISA). Commensal microbiota in Peyer's patches (PPs) and caecal contents were analyzed by 16S rRNA Illumina sequencing and qPCR, respectively. FBP improved sIgA production in HFD mice at mRNA and protein levels. Akkermansia and Lactobacillus in PPs of HFD mice were statistically increased by FBP. Beneficial microbiota and short-chain fatty acids (SCFAs) in caecal contents were positively correlated with caecal immunoglobulins in HFD mice. FBP showed an ability to modulate intestinal microbiota, which improved sIgA production in HFD mice, warranting the potential use of berry by-products as functional ingredients in improving the intestinal immune barrier of HFD individuals.

The barrier-improving functions of fermented blueberry pomace (FBP) and its potential mechanism were investigated in this study. Polyphenols and the approximate composition of FBP were evaluated according to the National Standard of the People's Republic of China and the UPLC-MS system. Male C57BL/6 mice were fed a control diet (CD) or a high-fat diet (HFD) with or without FBP supplementation. Oxidative stress, inflammation, histological morphology and the expression of functional proteins in the small intestine of mice were evaluated using the enzyme linked immunosorbent assay (ELISA), quantitative polymerase chain reaction (qPCR) and western blotting. The content of protein, fat, soluble dietary fiber, insoluble dietary fiber and carbohydrates (non-dietary fiber) was 114.5 ± 1.5 g kg-1, 5.0 ± 0.2 g kg-1, 48.0 ± 0.1 g kg-1, 360.3 ± 2.2 g kg-1 and 423 g kg-1 (by difference), respectively. Thirty-six polyphenols were identified in FBP. FBP improved the growth of mice and attenuated hepatic and intestinal oxidative stress. Intestinal inflammation was significantly reduced through the decrease of tumor necrosis factor-alpha (TNF-α) and myeloperoxidase (MPO) as well as an increase of interleukin-10 (IL-10). FBP supplementation significantly improved the intestinal morphology and barrier function, potentially by mediating the NF-κB-MLCK signaling pathway. The supplementation of FBP in HFD mice enhanced the intestinal barrier function. This suggested that polyphenol-rich by-products might provide a similar health effect in HFD individuals.

This work aims to study the pathogenesis of learning and memory impairment in offspring rats resulting from maternal enflurane anesthesia by focusing on the expression of the N-methyl-d-aspartic acid receptor subunit 2B (NR2B) in the hippocampus of the offspring. Thirty female Sprague-Dawley rats were randomly divided into three groups: control (C group), 4 h enflurane exposure (E1 group), and 8 h enflurane exposure (E2 group) groups. Eight to ten days after the initiation of pregnancy, rats from the E1 and E2 groups were allowed to inhale 1.7% enflurane in 2 L/min oxygen for 4 h and 8 h, respectively. Rats from the C group were allowed to inhale 2 L/min of oxygen only. The Morris water maze was used to assay the learning and memory function of the offspring on postnatal days 20 and 30. RT-PCR and immunohistochemistry assays were then used to measure the mRNA levels and protein expression of NR2B, respectively. Relative to offspring rats from the C group, those from the E1 and E2 groups exhibited longer escape latencies, lesser number of crossings over the platform, and less time spent in the target quadrant in the spatial exploration test (P < 0.05). In addition, the mRNA and protein expression levels of NR2B in the hippocampus of offspring rats in the E1 and E2 groups were down-regulated (P < 0.05). No significant differences between the E1 and E2 groups were observed (P > 0.05) in terms of mRNA levels and protein expression of NR2B. The cognitive function of the offspring is impaired when maternal rats are exposed to enflurane during early pregnancy. A possible mechanism of this effect is related to the down-regulation of NR2B expression.


Este trabalho objetiva o estudo da patogênese de deficiência no aprendizado e memória de prole de ratos resultante da anestesia maternal por enflurano, por meio da expressão da subunidade 2B do receptor do ácidoN-metil-D-aspártico (NR2B) no hipocampo dos filhotes. Dividiram-se, aleatoriamente, 30 fêmeas de ratos Sprague-Dawley em três grupos: controle (grupo C), exposição ao enflurano por 4 h (grupo E1) e por 8 h (grupo E2). De oito a 10 dias após o início da gravidez, os ratos dos grupos E1 e E2 inalaram enflurano 1,7% em 2 L/min de oxigênio, por 4 h e 8 h, respectivamente. Ratos do grupo C inalaram apenas 2 L/min de oxigênio. O labirinto de água de Morris foi empregado para analisar as funções de aprendizado e memória da cria em 20 e 30 dias após o nascimento. Utilizaram-se ensaios de RT-PCR e de imuno-histoquímica para medir os níveis de mRNA e expressão da proteína do NR2B, respectivamente. Em comparação com os ratos controle do grupo C, aqueles dos grupos E1 e E2 exibiram latências de escape mais longas, menor número de travessias na plataforma e menos tempo gasto no quadrante alvo no teste de exploração espacial (P < 0,05). Adicionalmente, os níveis de expressão de mRNA e de proteína do NR2B no hipocampo dos filhotes nos grupos E1 e E2 estavam reduzidos (P < 0,05). Não se observaram diferenças significativas entre os grupos E1 e E2 (P < 0,05) quanto aos níveis de mRNA e à expressão de proteína de NR2B. A função cognitiva dos filhotes é prejudicada quando as mães são expostas ao enflurano durante o início da gravidez. O mecanismo possível para esse efeito está relacionado à diminuição na expressão de NR2B.
