RESUMEN

Rhipicephalus microplus, is one parasites that cause severe productivity losses in the cattle industry of Brazil and shows increasing resistance to conventional pesticides. This research aims to study the chemical composition, and acaricidal activity of the essential oil from Ocotea notata leaves, a brazilian endemic species, against R. microplus. The effect on R. microplus engorged adult females was evaluated using the immersion test. The oil reduced the survival by 90% after incubation for 15 days and there was 100% reduction for posture inhibition and reproductive capacity. These results suggest that the O. notata essential oil has activity on the R. microplus.


Rhipicephalus microplus, es un parásito que causa graves pérdidas de productividad en la industria ganadera de Brasil y muestra una creciente resistencia a los pesticidas convencionales. Esta investigación tiene como objetivo estudiar la composición química y la actividad acaricida del aceite esencial de las hojas de Ocotea notata, una especie endémica brasileña, contra R. microplus. El efecto sobre las hembras adultas engordadas de R. microplus se evaluó mediante la prueba de inmersión. El aceite redujo la supervivencia en 90% después de la incubación durante 15 días y hubo una reducción del 100% para la inhibición de la postura y la capacidad reproductiva. Estos resultados sugieren que el aceite esencial de O. notata tiene actividad contra R. microplus.

BACKGROUND: This work describes a chemical study of the essential oil from leaves of Xylopia ochrantha, an endemic Annonaceae species from Brazil, and its activity against Biomphalaria species. Considering its poor solubility in aqueous medium, the essential oil was nanoemulsified to evaluate its action on controlling some mollusc species of genus Biomphalaria, snail hosts of Schistosoma mansoni that causes schistosomiasis, which mainly affects tropical and subtropical countries. OBJECTIVES: The main aims of this work were to analyse the chemical composition of essential oil from X. ochrantha, and to evaluate the effect of its nanoemulsion on molluscs of genus Biomphalaria and their oviposition. METHODS: Chemical analysis was performed by gas chromatography coupled to mass spectrometry. Nanoemulsions were prepared by a low energy method and characterised by particle size and polydispersity index. Biological assays evaluating the mortality of adult species of B. glabrata, B. straminea and B. tenagophila and their ovipositions upon contact with the most stable nanoemulsion during 24 and 48 h were performed. FINDINGS: Chemical analysis by mass spectrometry revealed the majority presence of bicyclogermacrene and germacrene D in the essential oil. The formulation with a hydrophilic-lipophilic balance (HLB) of 9.26 was the most suitable for the oil delivery system. This nanoemulsion caused the mortality in B. tenagophila, B. straminea and B. glabarata of different sizes at levels ranging from 50 to 100% in 48 h. Additionally, the formulation could inhibit the development of deposited eggs. CONCLUSION: Thus, these results suggest the use of nanoemulsified essential oil from X. ochrantha as a possible alternative in controlling some Biomphalaria species involved in the schistosomiasis cycle.

Chagas disease is caused by the Trypanosoma cruzi affecting millions of people, and widespread throughout Latin America. This disease exhibits a problematic chemotherapy. Benznidazole, which is the drug currently used as standard treatment, lamentably evokes several adverse reactions. Among other options, natural products have been tested to discover a novel therapeutic drug for this disease. A lot of plants from the Brazilian flora did not contain studies about their biological effects. Restinga de Jurubatiba from Brazil is a sandbank ecosystem poorly studied in relation to plant biological activity. Thus, three plant species from Restinga de Jurubatiba were tested against in vitro antiprotozoal activity. Among six extracts obtained from leaves and stem parts and 2 essential oils derived from leave parts, only 3 extracts inhibited epimastigote proliferation. Substances present in the extracts with activity were isolated (quercetin, myricetin, and ursolic acid), and evaluated in relation to antiprotozoal activity against epimastigote Y and Dm28 Trypanosoma cruzi strains. All isolated substances were effective to reduce protozoal proliferation. Essentially, quercetin and myricetin did not cause mammalian cell toxicity. In summary, myricetin and quercetin molecule can be used as a scaffold to develop new effective drugs against Chagas's disease.

Zanthoxylum caribaeum Lamarck (Rutaceae) is plant species with a variety of medical applications, including insecticidal activity. This study determined the bioacaricidal activity of the essential oil from Z. caribaeum leaves against engorged Rhipicephalus (Boophilus) microplus (Canestrini, 1887) females using the adult immersion test. For this purpose, three serial concentrations (5.0, 2.5, and 1.25%, vol:vol, in 1% dimetilsulfoxide) of the essential oil were used. Essential oil 5% caused 65% mortality on the first day after treatment, 85% on the second day, and 100% mortality by the fifth day. To our knowledge, this is the first demonstration of the acaricidal activity of the essential oil from Z. caribaeum leaves against cattle ticks.

BACKGROUND: Plants have been recognized as a good source of insecticidal agents, since they are able to produce their own defensives to insect attack. Moreover, there is a growing concern worldwide to develop pesticides with low impact to environment and non-target organisms. Hexane-soluble fraction from ethanolic crude extract from fruits of Manilkara subsericea and its triterpenes were considered active against a cotton pest (Dysdercus peruvianus). Several natural products with insecticidal activity have poor water solubility, including triterpenes, and nanotechnology has emerged as a good alternative to solve this main problem. On this context, the aim of the present study was to develop an insecticidal nanoemulsion containing apolar fraction from fruits of Manilkara subsericea. RESULTS: It was obtained a formulation constituted by 5% of oil (octyldodecyl myristate), 5% of surfactants (sorbitan monooleate/polysorbate 80), 5% of apolar fraction from M. subsericea and 85% of water. Analysis of mean droplet diameter (155.2 ± 3.8 nm) confirmed this formulation as a nanoemulsion. It was able to induce mortality in D. peruvianus. It was observed no effect against acetylcholinesterase or mortality in mice induced by the formulation, suggesting the safety of this nanoemulsion for non-target organisms. CONCLUSIONS: The present study suggests that the obtained O/A nanoemulsion may be useful to enhance water solubility of poor water soluble natural products with insecticidal activity, including the hexane-soluble fraction from ethanolic crude extract from fruits of Manilkara subsericea.
