RESUMEN

Objective.Brain injury is the leading cause of long-term disability worldwide, often resulting in impaired hand function. Brain-machine interfaces (BMIs) offer a potential way to improve hand function. BMIs often target replacing lost function, but may also be employed in neurorehabilitation (nrBMI) by facilitating neural plasticity and functional recovery. Here, we report a novel nrBMI capable of acquiring high-γ(70-115 Hz) information through a unique post-traumatic brain injury (TBI) hemicraniectomy window model, and delivering sensory feedback that is synchronized with, and proportional to, intended grasp force.Approach. We developed the nrBMI to use electroencephalogram recorded over a hemicraniectomy (hEEG) in individuals with TBI. The nrBMI empowered users to exert continuous, proportional control of applied force, and provided continuous force feedback. We report the results of an initial testing group of three human participants with TBI, along with a control group of three skull- and motor-intact volunteers.Main results. All participants controlled the nrBMI successfully, with high initial success rates (2 of 6 participants) or performance that improved over time (4 of 6 participants). We observed high-γmodulation with force intent in hEEG but not skull-intact EEG. Most significantly, we found that high-γcontrol significantly improved the timing synchronization between neural modulation onset and nrBMI output/haptic feedback (compared to low-frequency nrBMI control).Significance. These proof-of-concept results show that high-γnrBMIs can be used by individuals with impaired ability to control force (without immediately resorting to invasive signals like electrocorticography). Of note, the nrBMI includes a parameter to change the fraction of control shared between decoded intent and volitional force, to adjust for recovery progress. The improved synchrony between neural modulations and force control for high-γsignals is potentially important for maximizing the ability of nrBMIs to induce plasticity in neural circuits. Inducing plasticity is critical to functional recovery after brain injury.

Recent studies have shown the ability to record high-γ signals (80-160 Hz) in electroencephalogram (EEG) from traumatic brain injury (TBI) patients who have had hemicraniectomies. However, extraction of the movement-related high-γ remains challenging due to a confounding bandwidth overlap with surface electromyogram (EMG) artifacts related to facial and head movements. In our previous work, we described an augmented independent component analysis (ICA) approach for removal of EMG artifacts from EEG, and referred to as EMG Reduction by Adding Sources of EMG (ERASE). Here, we tested this algorithm on EEG recorded from six TBI patients with hemicraniectomies while they performed a thumb flexion task. ERASE removed a mean of 52 ± 12% (mean ± S.E.M) (maximum 73%) of EMG artifacts. In contrast, conventional ICA removed a mean of 27 ± 19% (mean ± S.E.M) of EMG artifacts from EEG. In particular, high-γ synchronization was significantly improved in the contralateral hand motor cortex area within the hemicraniectomy site after ERASE was applied. A more sophisticated measure of high-γ complexity is the fractal dimension (FD). Here, we computed the FD of EEG high-γ on each channel. Relative FD of high-γ was defined as that the FD in move state was subtracted by FD in idle state. We found relative FD of high-γ over hemicraniectomy after applying ERASE were strongly correlated to the amplitude of finger flexion force. Results showed that significant correlation coefficients across the electrodes related to thumb flexion averaged ~0.76, while the coefficients across the homologous electrodes in non-hemicraniectomy areas were nearly 0. After conventional ICA, a correlation between relative FD of high-γ and force remained high in both hemicraniectomy areas (up to 0.86) and non-hemicraniectomy areas (up to 0.81). Across all subjects, an average of 83% of electrodes significantly correlated with force was located in the hemicraniectomy areas after applying ERASE. After conventional ICA, only 19% of electrodes with significant correlations were located in the hemicraniectomy. These results indicated that the new approach isolated electrophysiological features during finger motor activation while selectively removing confounding EMG artifacts. This approach removed EMG artifacts that can contaminate high-gamma activity recorded over the hemicraniectomy.

Electroencephalographic (EEG) recordings are often contaminated by electromyographic (EMG) artifacts, especially when recording during movement. Existing methods to remove EMG artifacts include independent component analysis (ICA), and other high-order statistical methods. However, these methods can not effectively remove most of EMG artifacts. Here, we proposed a modified ICA model for EMG artifacts removal in the EEG, which is called EMG Removal by Adding Sources of EMG (ERASE). In this new approach, additional channels of real EMG from neck and head muscles (reference artifacts) were added as inputs to ICA in order to "force" the most power from EMG artifacts into a few independent components (ICs). The ICs containing EMG artifacts (the "artifact ICs") were identified and rejected using an automated procedure. ERASE was validated first using both simulated and experimentally-recorded EEG and EMG. Simulation results showed ERASE removed EMG artifacts from EEG significantly more effectively than conventional ICA. Also, it had a low false positive rate and high sensitivity. Subsequently, EEG was collected from 8 healthy participants while they moved their hands to test the realistic efficacy of this approach. Results showed that ERASE successfully removed EMG artifacts (on average, about 75% of EMG artifacts were removed when using real EMGs as reference artifacts) while preserving the expected EEG features related to movement. We also tested the ERASE procedure using simulated EMGs as reference artifacts (about 63% of EMG artifacts removed). Compared to conventional ICA, ERASE removed on average 26% more EMG artifacts from EEG. These findings suggest that ERASE can achieve significant separation of EEG signal and EMG artifacts without a loss of the underlying EEG features. These results indicate that using additional real or simulated EMG sources can increase the effectiveness of ICA in removing EMG artifacts from EEG. Combined with automated artifact IC rejection, ERASE also minimizes potential user bias. Future work will focus on improving ERASE so that it can also be used in real-time applications.

Although IL-15 has been implicated in the pathogenic hyperimmune activation that drives progressive HIV and SIV infection, as well as in the generation of HIV/SIV target cells, it also supports NK and T cell homeostasis and effector activity, potentially benefiting the host. To understand the role of IL-15 in SIV infection and pathogenesis, we treated two cohorts of SIVmac239-infected rhesus macaques (RM; Macaca mulatta), one with chronic infection, the other with primary infection, with a rhesusized, IL-15-neutralizing mAb (versus an IgG isotype control) for up to 10 wk (n = 7-9 RM per group). In both cohorts, anti-IL-15 was highly efficient at blocking IL-15 signaling in vivo, causing 1) profound depletion of NK cells in blood and tissues throughout the treatment period; 2) substantial, albeit transient, depletion of CD8+ effector memory T cells (TEM) (but not the naive and central memory subsets); and 3) CD4+ and CD8+ TEM hyperproliferation. In primary infection, reduced frequencies of SIV-specific effector T cells in an extralymphoid tissue site were also observed. Despite these effects, the kinetics and extent of SIV replication, CD4+ T cell depletion, and the onset of AIDS were comparable between anti-IL-15- and control-treated groups in both cohorts. However, RM treated with anti-IL-15 during primary infection manifested accelerated reactivation of RM rhadinovirus. Thus, IL-15 support of NK cell and TEM homeostasis does not play a demonstrable, nonredundant role in SIV replication or CD4+ T cell deletion dynamics but may contribute to immune control of oncogenic γ-herpesviruses.

Brain-machine interfaces (BMIs) translate brain signals into control signals for an external device, such as a computer cursor or robotic limb. These signals can be obtained either noninvasively or invasively. Invasive recordings, using electrocorticography (ECoG) or intracortical microelectrodes, provide higher bandwidth and more informative signals. Rehabilitative BMIs, which aim to drive plasticity in the brain to enhance recovery after brain injury, have almost exclusively used non-invasive recordings, such electroencephalography (EEG) or magnetoencephalography (MEG), which have limited bandwidth and information content. Invasive recordings provide more information and spatiotemporal resolution, but do incur risk, and thus are not usually investigated in people with stroke or traumatic brain injury (TBI). Here, in this paper, we describe a new BMI paradigm to investigate the use of higher frequency signals in brain-injured subjects without incurring significant risk. We recorded EEG in TBI subjects who required hemicraniectomies (removal of a part of the skull). EEG over the hemicraniectomy (hEEG) contained substantial information in the high gamma frequency range (65-115 Hz). Using this information, we decoded continuous finger flexion force with moderate to high accuracy (variance accounted for 0.06 to 0.52), which at best approaches that using epidural signals. These results indicate that people with hemicraniectomies can provide a useful resource for developing BMI therapies for the treatment of brain injury.

In recent years, many studies examined if EEG signals from traumatic brain injury (TBI) patients can be used for new rehabilitation technologies, such as BCI systems. However, extraction of the high-gamma band related to movement remains challenging due to the presence of surface electromyogram (sEMG) caused by unconscious facial and head movement of patients. In this paper, we proposed a modified independent component analysis (ICA) model for EMG artifact removal in the EEG data from TBI patients with a hemicraniectomy. Here, simulated EMG was generated and added to the raw EEG data as the extra channels for independent components calculation. After running ICA, the independent components (ICs) related to artifacts were identified and rejected automatically through several criteria. EEG data underlying hand movement from one healthy subject and one TBI patient with a hemicraniectomy were conducted to verify the efficacy of this algorithm. Results showed that the proposed algorithm removed sEMG artifacts from the EEG data by up to 86.72% while preserving the associated brain features. In particular, the high-gamma band (80 to 160 Hz) was found to arise principally from the hemicraniectomy area after this technique was applied. Meanwhile, we found that the magnitude of gamma power during movement improved after removal of sEMG artifacts.

Prophylactic vaccination of rhesus macaques with rhesus cytomegalovirus (RhCMV) vectors expressing simian immunodeficiency virus (SIV) antigens (RhCMV/SIV) elicits immune responses that stringently control highly pathogenic SIV infection, with subsequent apparent clearance of the infection, in ~50% of vaccinees. In contrast, here, we show that therapeutic RhCMV/SIV vaccination of rhesus macaques previously infected with SIV and given continuous combination antiretroviral therapy (cART) beginning 4-9 d post-SIV infection does not mediate measurable SIV reservoir clearance during over 600 d of follow-up on cART relative to RhCMV/control vaccination. However, none of the six animals started on cART on day four or five, across both RhCMV/SIV- and RhCMV/control-vaccinated groups, those rhesus macaques with SIV reservoirs most closely resembling those of prophylactically RhCMV/SIV-vaccinated and protected animals early in their course, showed post-cART viral rebound with up to nine months of follow-up. Moreover, at necropsy, these rhesus macaques showed little to no evidence of replication-competent SIV. These results suggest that the early SIV reservoir is limited in durability and that effective blockade of viral replication and spread in this critical time window by either pharmacologic or immunologic suppression may result in reduction, and potentially loss, of rebound-competent virus over a period of ~two years.

The development of coordinated reach-to-grasp movement has been well studied in infants and children. However, the role of motor cortex during this development is unclear because it is difficult to study in humans. We took the approach of using a brain-machine interface (BMI) paradigm in rhesus macaques with prior therapeutic amputations to examine the emergence of novel, coordinated reach to grasp. Previous research has shown that after amputation, the cortical area previously involved in the control of the lost limb undergoes reorganization, but prior BMI work has largely relied on finding neurons that already encode specific movement-related information. In this study, we taught macaques to cortically control a robotic arm and hand through operant conditioning, using neurons that were not explicitly reach or grasp related. Over the course of training, stereotypical patterns emerged and stabilized in the cross-covariance between the reaching and grasping velocity profiles, between pairs of neurons involved in controlling reach and grasp, and to a comparable, but lesser, extent between other stable neurons in the network. In fact, we found evidence of this structured coordination between pairs composed of all combinations of neurons decoding reach or grasp and other stable neurons in the network. The degree of and participation in coordination was highly correlated across all pair types. Our approach provides a unique model for studying the development of novel, coordinated reach-to-grasp movement at the behavioral and cortical levels. NEW & NOTEWORTHY Given that motor cortex undergoes reorganization after amputation, our work focuses on training nonhuman primates with chronic amputations to use neurons that are not reach or grasp related to control a robotic arm to reach to grasp through the use of operant conditioning, mimicking early development. We studied the development of a novel, coordinated behavior at the behavioral and cortical level, and the neural plasticity in M1 associated with learning to use a brain-machine interface.

Studies on neural plasticity associated with brain-machine interface (BMI) exposure have primarily documented changes in single neuron activity, and largely in intact subjects. Here, we demonstrate significant changes in ensemble-level functional connectivity among primary motor cortical (MI) neurons of chronically amputated monkeys exposed to control a multiple-degree-of-freedom robot arm. A multi-electrode array was implanted in M1 contralateral or ipsilateral to the amputation in three animals. Two clusters of stably recorded neurons were arbitrarily assigned to control reach and grasp movements, respectively. With exposure, network density increased in a nearly monotonic fashion in the contralateral monkeys, whereas the ipsilateral monkey pruned the existing network before re-forming a denser connectivity. Excitatory connections among neurons within a cluster were denser, whereas inhibitory connections were denser among neurons across the two clusters. These results indicate that cortical network connectivity can be modified with BMI learning, even among neurons that have been chronically de-efferented and de-afferented due to amputation.

IL-15 has been implicated as a key regulator of T and NK cell homeostasis in multiple systems; however, its specific role in maintaining peripheral T and NK cell populations relative to other γ-chain (γc) cytokines has not been fully defined in primates. In this article, we address this question by determining the effect of IL-15 inhibition with a rhesusized anti-IL-15 mAb on T and NK cell dynamics in rhesus macaques. Strikingly, anti-IL-15 treatment resulted in rapid depletion of NK cells and both CD4(+) and CD8(+) effector memory T cells (TEM) in blood and tissues, with little to no effect on naive or central memory T cells. Importantly, whereas depletion of NK cells was nearly complete and maintained as long as anti-IL-15 treatment was given, TEM depletion was countered by the onset of massive TEM proliferation, which almost completely restored circulating TEM numbers. Tissue TEM, however, remained significantly reduced, and most TEM maintained very high turnover throughout anti-IL-15 treatment. In the presence of IL-15 inhibition, TEM became increasingly more sensitive to IL-7 stimulation in vivo, and transcriptional analysis of TEM in IL-15-inhibited monkeys revealed engagement of the JAK/STAT signaling pathway, suggesting alternative γc cytokine signaling may support TEM homeostasis in the absence of IL-15. Thus, IL-15 plays a major role in peripheral maintenance of NK cells and TEM However, whereas most NK cell populations collapse in the absence of IL-15, TEM can be maintained in the face of IL-15 inhibition by the activity of other homeostatic regulators, most likely IL-7.

When reaching to grasp, we coordinate how we preshape the hand with how we move it. To ask how motor cortical neurons participate in this coordination, we examined the interactions between reach- and grasp-related neuronal ensembles while monkeys reached to grasp a variety of different objects in different locations. By describing the dynamics of these two ensembles as trajectories in a low-dimensional state space, we examined their coupling in time. We found evidence for temporal compensation across many different reach-to-grasp conditions such that if one neural trajectory led in time the other tended to catch up, reducing the asynchrony between the trajectories. Granger causality revealed bidirectional interactions between reach and grasp neural trajectories beyond that which could be attributed to the joint kinematics that were consistently stronger in the grasp-to-reach direction. Characterizing cortical coordination dynamics provides a new framework for understanding the functional interactions between neural populations.

In the study of population coding in neurobiological systems, tracking unit identity may be critical to assess possible changes in the coding properties of neuronal constituents over prolonged periods of time. Ensuring unit stability is even more critical for reliable neural decoding of motor variables in intra-cortically controlled brain-machine interfaces (BMIs). Variability in intrinsic spike patterns, tuning characteristics, and single-unit identity over chronic use is a major challenge to maintaining this stability, requiring frequent daily calibration of neural decoders in BMI sessions by an experienced human operator. Here, we report on a unit-stability tracking algorithm that efficiently and autonomously identifies putative single-units that are stable across many sessions using a relatively short duration recording interval at the start of each session. The algorithm first builds a database of features extracted from units' average spike waveforms and firing patterns across many days of recording. It then uses these features to decide whether spike occurrences on the same channel on one day belong to the same unit recorded on another day or not. We assessed the overall performance of the algorithm for different choices of features and classifiers trained using human expert judgment, and quantified it as a function of accuracy and execution time. Overall, we found a trade-off between accuracy and execution time with increasing data volumes from chronically implanted rhesus macaques, with an average of 12 s processing time per channel at ~90% classification accuracy. Furthermore, 77% of the resulting putative single-units matched those tracked by human experts. These results demonstrate that over the span of a few months of recordings, automated unit tracking can be performed with high accuracy and used to streamline the calibration phase during BMI sessions. Our findings may be useful to the study of population coding during learning, and to improve the reliability of BMI systems and accelerate their deployment in clinical applications.

Recordings from chronically implanted multielectrode arrays have become prevalent in both neuroscience and neural engineering experiments. To date, however, the extent to which populations of single-units remain stable over long periods of time has not been well characterized. In this study, neural activity was recorded from a Utah multielectrode array implanted in the primary motor cortex of a rhesus macaque during 18 recording sessions spanning nine months. We found that 67% of the units were stable through the first 15 days, 31% of units were stable through 47 days, 21% of units were stable through 106 days, and 8% of units were stable over 9 months. Thus not only were units stable over a timescale of several months, but units stable over 2 months were more likely to remain stable in the next 2 months.

Operant conditioning with biofeedback has been shown to be an effective method to modify neural activity to generate goal-directed actions in a brain-machine interface. It is particularly useful when neural activity cannot be mathematically mapped to motor actions of the actual body such as in the case of amputation. Here, we implement an operant conditioning approach with visual feedback in which an amputated monkey is trained to control a multiple degree-of-freedom robot to perform a reach-to-grasp behavior. A key innovation is that each controlled dimension represents a behaviorally relevant synergy among a set of joint degrees-of-freedom. We present a number of behavioral metrics by which to assess improvements in BMI control with exposure to the system. The use of non-human primates with chronic amputation is arguably the most clinically-relevant model of human amputation that could have direct implications for developing a neural prosthesis to treat humans with missing upper limbs.
