RESUMEN

Cavitation damage on a mercury target vessel for a pulsed spallation neutron source is induced by a proton beam injection in mercury. Cavitation damage is one of factors affecting the allowable beam power and the life time of a mercury target vessel. The prediction method of the cavitation damage using Monte Carlo simulations was proposed taking into account the uncertainties of the core position of cavitation bubbles and impact pressure distributions. The distribution of impact pressure attributed to individual cavitation bubble collapsing was assumed to be Gaussian distribution and the probability distribution of the maximum value of impact pressures was assumed to be three kinds of distributions: the delta function and Gaussian and Weibull distributions. Two parameters in equations describing the distribution of impact pressure were estimated using Bayesian optimization by comparing the distribution of the cavitation damage obtained from the experiment with the distribution of the accumulated plastic strain obtained from the simulation. Regardless of the distribution type, the estimated maximum impact pressure was 1.2-2.9 GPa and existed in the range of values predicted by the ratio of the diameter and depth of the pit. The estimated dispersion of the impact pressure distribution was 1.0-1.7 µm and corresponded to the diameter of major pits. In the distribution of the pits described by the accumulated plastic strain, which was assumed in three cases, the delta function and Gaussian and Weibull distributions, the Weibull distribution agreed well with the experimental results, particularly including relatively large pit size. Furthermore, the Weibull distribution reproduced the depth profile, i.e., pit shape, better than that using the delta function or Gaussian distribution. It can be said that the cavitation erosion phenomenon is predictable by adopting the Weibull distribution. This prediction method is expected to be applied to predict the cavitation damage in fluid equipment such as pumps and fluid parts.

Purification is essential before differentiating human induced pluripotent stem cells (hiPSCs) into cells that fully express particular differentiation marker genes. High-quality iPSC clones are typically purified through gene expression profiling or visual inspection of the cell morphology; however, the relationship between the two methods remains unclear. We investigated the relationship between gene expression levels and morphology by analyzing live-cell, phase-contrast images and mRNA profiles collected during the purification process. We employed these data and an unsupervised image feature extraction method to build a model that predicts gene expression levels from morphology. As a benchmark, it was confirmed that the method can predict the gene expression levels from tissue images for cancer genes, performing as well as state-of-the-art methods. We then applied the method to iPSCs and identified two genes that are well predicted from cell morphology. Although strong batch (or possibly donor) effects resulting from the reprogramming process preclude the ability to use the same model to predict across batches, prediction within a reprogramming batch is sufficiently robust to provide a practical approach for estimating expression levels of a few genes and monitoring the purification process.

We propose an image analysis method for quality evaluation of human pluripotent stem cells based on biologically interpretable features. It is important to maintain the undifferentiated state of induced pluripotent stem cells (iPSCs) while culturing the cells during propagation. Cell culture experts visually select good quality cells exhibiting the morphological features characteristic of undifferentiated cells. Experts have empirically determined that these features comprise prominent and abundant nucleoli, less intercellular spacing, and fewer differentiating cellular nuclei. We quantified these features based on experts' visual inspection of phase contrast images of iPSCs and found that these features are effective for evaluating iPSC quality. We then developed an iPSC quality evaluation method using an image analysis technique. The method allowed accurate classification, equivalent to visual inspection by experts, of three iPSC cell lines.

A lead bismuth eutectic (LBE) spallation target will be installed in the Target Test Facility (TEF-T) in the Japan Proton Accelerator Research Complex (J-PARC). The spallation target vessel filled with LBE is made of type 316L stainless steel. However, various damages, such as erosion/corrosion damage and liquid metal embrittlement caused by contact with flowing LBE at high temperature, and irradiation hardening caused by protons and neutrons, may be inflicted on the target vessel, which will deteriorate the steel and might break the vessel. To monitor the target vessel for prevention of an accident, an ultrasonic technique has been proposed to establish off-line evaluation for estimating vessel material status during the target maintenance period. Basic R&D must be carried out to clarify the dependency of ultrasonic wave propagation behavior on material microstructures and obtain fundamental knowledge. As a first step, ultrasonic waves scattered by the grains of type 316L stainless steel are investigated using new experimental and numerical approaches in the present study. The results show that the grain size can be evaluated exactly and quantitatively by calculating the attenuation coefficient of the ultrasonic waves scattered by the grains. The results also show that the scattering regimes of ultrasonic waves depend heavily on the ratio of wavelength to average grain size, and are dominated by grains of extraordinarily large size along the wave propagation path.

The Cyclotron and Radioisotope Center (CYRIC) of Tohoku University is a joint-use institution for education and research in a wide variety of fields ranging from physics to medicine. Accelerator facilities at the CYRIC provide opportunities for implementing a broad research program, including medical research using positron emission tomography (PET), with accelerated ions and radioisotopes. At the Great East Japan Earthquake on March 11, 2011, no human injuries occurred and a smooth evacuation was made in the CYRIC, thanks to the anti-earthquake measures such as the renovation of the cyclotron building in 2009 mainly to provide seismic strengthening, fixation of shelves to prevent the falling of objects, and securement of the width of the evacuation route. The preparation of an emergency response manual was also helpful. However, the accelerator facilities were damaged because of strong shaking that continued for a few minutes. For example, two columns on which a 930 cyclotron was placed were damaged, and thereby the 930 cyclotron was inclined. All the elements of beam transport lines were deviated from the beam axis. Some peripheral devices in a HM12 cyclotron were broken. Two shielding doors fell from the carriage onto the floor and blocked the entrances to the rooms. The repair work on the accelerator facilities was started at the end of July 2011. During the repair work, the joint use of the accelerator facilities was suspended. After the repair work was completed, the joint use was re-started at October 2012, one and a half years after the earthquake.
