RESUMEN

Glucocorticoid-induced osteonecrosis of the femoral head (GONFH) is a common refractory joint disease characterized by bone damage and the collapse of femoral head structure. However, the exact pathological mechanisms of GONFH remain unknown. Here, we observed abnormal osteogenesis and adipogenesis associated with decreased ß-catenin in the necrotic femoral head of GONFH patients. In vivo and in vitro studies further revealed that glucocorticoid exposure disrupted osteogenic/adipogenic differentiation of bone marrow mesenchymal cells (BMSCs) by inhibiting ß-catenin signaling in glucocorticoid-induced GONFH rats. Col2+ lineage largely contributes to BMSCs and was found an osteogenic commitment in the femoral head through 9 mo of lineage trace. Specific deletion of ß-catenin gene (Ctnnb1) in Col2+ cells shifted their commitment from osteoblasts to adipocytes, leading to a full spectrum of disease phenotype of GONFH in adult mice. Overall, we uncover that ß-catenin inhibition disrupting the homeostasis of osteogenic/adipogenic differentiation contributes to the development of GONFH and identify an ideal genetic-modified mouse model of GONFH.

This study systematically investigated the performance and microstructure characterization of cryogenic rolling (CR) and room-temperature rolling (RTR) Al-Mg-Si alloys. The result showed that the hardness of the CR alloys decreased at the early aging stage, but that the hardness of the RTR alloys increased at the early aging stage. Retrogression phenomena were apparent in the CR alloys at the early aging stage. Despite undergoing the same solid solution treatment, a few substructures were still observed in the CR alloys, and the degree of recrystallization in the CR alloys was significantly inferior to that in the RTR alloys. After aging for 50 h, the strength and precipitates' density in the CR 75 alloy were higher than that in the other alloys; this indicated that the substructures were beneficial to precipitation and precipitate growth. A precipitation strength model was employed to illustrate the precipitation contribution at different aging stages. The results showed that the CR 75 alloy obtained the strongest precipitation strengthening.

In this study, quinoid structures, semiquinone radical structures, and electron affinity energies (EAEs) of many polycyclic quinones containing heteroatoms (O, B, and F) or heterocycles (pyrrole, imidazole, and pyrazine) were calculated. Quinones with unstable quinoid structures and stable semiquinone radical structures had high EAEs. The main factors of quinoid structural instability were spatial repulsion and antiaromaticity, and the stability factors of the semiquinone radical structure comprised inductive effects, hydrogen bonds, electrostatic interactions, and orbital interactions. Compound 11 had both the antiaromaticity of the quinoid structure and the orbital interactions of the semiquinone radical structure, thus having the highest EAE. The crystal structure of compound 8 was obtained, and it confirmed the reliability of the calculated results of this work.

Alkaptonuria (AKU) is a rare autosomal recessive disorder caused by homogentisc acid (HGA) accumulation, the deposition of which in the joints usually causes ochronotic arthropathy. With no specific therapy for AKU currently, total joint arthroplasty in ochronotic arthropathy is applied to relieve the symptoms. A 63-year-old female patient came to our Orthopedic Surgery Department in 2019, complaining of severe limitation of movement and pain in the right hip for more than one year. A right total hip arthroplasy (THA) was performed due to the ineffective conservative therapy. At a follow-up of more than 15 months, the woman had full mobility with no complaining of pains. Since there is no relevant case reported about THA therapy for Chinese AKU patients, this report provides a feasible scheme, which makes clinical data more comprehensive.

Drought is one of the most common abiotic stressors in plants. Melatonin (MT) is a high-efficiency and low-toxicity growth regulator that plays an important role in plant responses to drought stress. As a wild relative of wheat, Agropyron mongolicum has become an important species for the improvement of degraded grasslands and the replanting of sandy grasslands. However, the physiological and molecular mechanisms by which exogenous MT regulates drought stress in A. mongolicum remain unclear. To assess the effectiveness of MT intervention (100 mg·L-1), polyethylene glycol 6000 was used to simulate drought stress, and its ameliorating effects on drought stress in A. mongolicum seedlings were investigated through physiology, transcriptomics, and metabolomics. Physiological analysis indicated that MT treatment increased the relative water content and chlorophyll content and decreased the relative conductivity of A. mongolicum seedlings. Additionally, MT decreased malondialdehyde (MDA) and reactive oxygen species (ROS) accumulation by enhancing antioxidant enzyme activities. The transcriptome and metabolite profiling analysis of A. mongolicum seedlings treated with and without MT under drought stress identified the presence of 13,466 differentially expressed genes (DEGs) and 271 differentially expressed metabolites (DEMs). The integrated analysis of transcriptomics and metabolomics showed that DEGs and DEMs participated in diverse biological processes, such as flavonoid biosynthesis and carbohydrate metabolism. Moreover, MT may be involved in regulating the correlation of DEGs and DEMs in flavonoid biosynthesis and carbohydrate metabolism during drought stress. In summary, this study revealed the physiological and molecular regulatory mechanisms of exogenous MT in alleviating drought stress in A. mongolicum seedlings, and it provides a reference for the development and utilization of MT and the genetic improvement of drought tolerance in plants from arid habitats.

Hyperspectral imaging that obtains the spatial-spectral information of a scene has been extensively applied in various fields but usually requires a complex and costly system. A single-pixel detector based hyperspectral system mitigates the complexity problem but simultaneously brings new difficulties on the spectral dispersion device. In this work, we propose a low-cost compressive single-pixel hyperspectral imaging system with RGB sensors. Based on the structured illumination single-pixel imaging configuration, the lens-free system directly captures data by the RGB sensors without dispersion in the spectral dimension. The reconstruction is performed with a pre-trained spatial-spectral dictionary, and the hyperspectral images are obtained through compressive sensing. In addition, the spatial patterns for the structured illumination and the dictionary for the sparse representation are optimized by coherence minimization, which further improve the reconstruction quality. In both spatial and spectral dimensions, the intrinsic sparse properties of the hyperspectral images are made full use of for high sampling efficiency and low reconstruction cost. This work may introduce opportunities for optimization of computational imaging systems and reconstruction algorithms towards high speed, high resolution, and low cost future.

In this paper, we propose a convolutional neural network based on epipolar geometry and image segmentation for light-field depth estimation. Epipolar geometry is utilized to estimate the initial disparity map. Multi-orientation epipolar images are selected as input data, and the convolutional blocks are adopted based on the disparity of different-direction epipolar images. Image segmentation is used to obtain the edge information of the central sub-aperture image. By concatenating the output of the two parts, an accurate depth map could be generated with fast speed. Our method achieves a high rank on most quality assessment metrics in the HCI 4D Light Field Benchmark and also shows effectiveness in estimating accurate depth on real-world light-field images.
