RESUMEN

Rubus, the largest genus in Rosaceae, contains over 1400 species that distributed in multiple habitats across the world, with high species diversity in the temperate regions of Northern Hemisphere. Multiple Rubus species are cultivated for their valuable fruits. However, the intrageneric classification and phylogenetic relationships are still poorly understood. In this study, we sequenced, assembled, and characterized 17 plastomes of Rubus, and conducted comparative genomics integrating with 47 previously issued plastomes of this genus. The 64 plastomes of Rubus exhibited typical quadripartite structure with sizes ranging from 155,144 to 156,700 bp, and contained 132 genes including 87 protein-coding genes, 37 tRNA genes and eight rRNA genes. All plastomes are conservative in the gene order, the frequency of different types of long repeats and simple sequence repeats (SSRs), the codon usage, and the selection pressure of protein-coding genes. However, there are also some differences in the Rubus plastomes, including slight contraction and expansion of the IRs, a variation in the numbers of SSRs and long repeats, and some genes in certain clades undergoing intensified or relaxed purifying selection. Phylogenetic analysis based on whole plastomes showed that the monophyly of Rubus was strongly supported and resolved it into six clades corresponding to six subgenera. Moreover, we identified 12 highly variable regions that could be potential molecular markers for phylogenetic, population genetic, and barcoding studies. Overall, our study provided insight into plastomic structure and sequence diversification of Rubus, which could be beneficial for future studies on identification, evolution, and phylogeny in this genus.

OBJECTIVE: An important factor during pituitary adenoma surgery is to preserve pituitary stalk (PS) as this plays a role in reduction of the risk of postoperative diabetes insipidus. The hypothalamic-hypophyseal tract (HHT) projects through the PS to the posterior pituitary gland. To reconstruct white matter fiber pathways, methods like diffusion tensor imaging (DTI) tractography have been widely used. In this report we attempted to predict the position of PS using DTI tractography and to assess its intraoperative correlation during surgery of pituitary adenomas. METHODS: DTI tractography was used to tract the HHT in nine patients before craniotomy for pituitary adenomas. The DTI location of the HHT was compared with the PS position identified at the time of surgery. DTI fiber tracking was carried out in nine patients prior to the planned craniotomy for pituitary adenomas. In one patient, the PS could not be identified during the surgery. In the other eight patients, a comparison was made between the location of the HHT identified by DTI and the position of the PS visualized at the time of surgery. RESULTS: The position of the HHT identified by DTI showed consistency with the intraoperative position of the PS in seven patients (88.9% concordance). CONCLUSION: This study shows that DTI can identify the position of the HHT and thus the position of the PS with a high degree of reliability.

KEY MESSAGE: Cytokinin might be an important factor to regulate floral sex at the very early stage of flower development in sacha inchi. Sacha inchi (Plukenetia volubilis, Euphorbiaceae) is characterized by having female and male flowers in a thyrse with particular differences. The mechanisms involved in the development of unisexual flowers are very poorly understood. In this study, the inflorescence and flower development of P. volubilis were investigated using light microscopy and scanning electron microscopy. We also investigated the effects of cytokinin on flower sex determination by exogenous application of 6-benzyladenine (BA) in P. volubilis. The floral development of P. volubilis was divided into eight stages, and the first morphological divergence between the male and female flowers was found to occur at stage 3. Both female and male flowers can be structurally distinguished by differences in the shape and size of the flower apex after sepal primordia initiation. There are no traces of gynoecia in male flowers or of androecia in female flowers. Exogenous application of BA effectively induced gynoecium primordia initiation and female flower development, especially at the early flower developmental stages. We propose that flower sex is determined earlier and probably occurs before flower initiation, either prior to or at inflorescence development due to the difference in the position of the female and male primordia in the inflorescence and in the time of the female and male primordia being initiated. The influence of cytokinin on female primordia during flower development in P. volubilis strongly suggests a feminization role for cytokinin in sex determination. These results indicate that cytokinin could modify the fate of the apical meristem of male flower and promote the formation of carpel primordia in P. volubilis.
