RESUMEN

BACKGROUND: Coronary artery lesions (CALs) are the most common and major complication of Kawasaki disease (KD) in developed countries. However, the underlying immunologic mechanisms of CAL development in KD remain unclear. METHODS: Here, we conducted single-cell transcriptome analyses of 212 210 peripheral blood mononuclear cells collected from a cross-sectional cohort of 16 children, including 4 patients with KD with CALs, 5 patients with KD without CALs, 4 healthy controls, and 3 febrile controls. RESULTS: KD altered the proportion of peripheral blood mononuclear cells, including an increasing trend in inflammatory cells (megakaryocytes and monocytes) and a decreasing trend in lymphocytes (eg, CD4+ T, CD8+ T, mucosal-associated invariant T, natural killer, and γδ T cells), highlighting the potential presence of lymphopenia phenomenon in KD. Our data indicated the presence of inflammatory cytokine storm in patients with KD with CALs, caused by systemic upregulation of TNFSF13B (tumor necrosis factor superfamily member 13b), CXCL16 (C-X-C motif chemokine ligand 16), TNFSF10 (tumor necrosis factor superfamily member 10), and IL1RN (interleukin 1 receptor antagonist), mainly produced by monocytes (especially for the Mono_CD14-CD16 cluster) and megakaryocytes. We also found that myeloid cells of patients with KD, particularly in those with CALs, might play a role in vascular injury (eg, increased MMP [matrix metalloproteinase] 9, MMP17, and MMP25) and immune cell recruitment. The immune landscape of patients with KD with CALs was featured by lower exhaustion levels in natural killer cells, a high cytotoxic state in the CD8_Pro cluster, and activation of the complement system in monocytes. Additionally, the activation of B cells was more pronounced in the early stage of KD. CONCLUSIONS: Collectively, this study provides a comprehensive understanding of the roles of various immune cells and inflammatory cytokine storms in the development of CALs in KD and offers a valuable resource for identifying novel therapeutic targets for patients with KD with CALs.

The features of 16p11.2 deletion phenotype is developmental delay, intellectual disability, and autism spectrum disorder. Seizures are observed in approximately 20% of individuals with the microdeletion. Induced pluripotent stem cells (iPSCs) were generated from erythroblasts obtained from a child diagnosed with benign familial infantile epilepsy, caused by 16p11.2 deletion. These iPSCs exhibited stable amplification, expressed pluripotent markers, and differentiated spontaneously into three germ layers in vitro.

Hindgut microorganisms in newborn calves play an important role in the development of immunity and metabolism, and optimization of performance. However, knowledge of the extent to which microbiome colonization of the calf intestine is dependent on maternal characteristics is limited. In this study, placenta, umbilical cord, amniotic fluid, colostrum, cow feces, and calf meconium samples were collected from 6 Holstein cow-calf pairs. Microbial composition was analyzed by 16S rRNA gene high-throughput sequencing, and maternal transfer characteristics assessed using SourceTracker based on Gibbs sampling to fit the joint distribution using the mean proportions of each sample with meconium as the "sink" and other sample types as different "sources." Alpha and beta diversity analyses revealed sample type-specific microbiome features: microbial composition of the placenta, umbilical cord, amniotic fluid, colostrum, and calf feces were similar, but differed from cow feces (p < 0.05). Compared with profiles of meconium vs. placenta, meconium vs. umbilical cord, and meconium vs. colostrum, differences between the meconium and amniotic fluid were most obvious. SourceTracker analysis revealed that 23.8 ± 2.21% of the meconium OTUs matched those of umbilical cord samples, followed by the meconium-placenta pair (15.57 ± 2.2%), meconium-colostrum pair (14.4 ± 1.9%), and meconium-amniotic fluid pair (11.2 ± 1.7%). The matching ratio between meconium and cow feces was the smallest (10.5 ± 1%). Overall, our data indicated that the composition of the meconium microflora was similar compared with multiple maternal sites including umbilical cord, placenta, colostrum, and amniotic fluid. The umbilical cord microflora seemed to contribute the most to colonization of the fecal microflora of calves. Bacteria with digestive functions such as cellulose decomposition and rumen fermentation were mainly transmitted during the maternal transfer process.

Ultrasonic vocalizations (USVs) play an important role in parent-infant interactions during the neonatal period of rodents because of the pups' need to be suckled and protected by their parents. However, studies on the effects of USVs on parental care are focused on maternal behavior, and little data on paternal care are available, with especially few investigations in midday gerbils. In this study, we examined the effects of early social deprivation (ED) on offspring due to USVs and parental behavior responses in midday gerbils (Meriones meridianus). The results indicated that the number of USVs in ED gerbils increased on postnatal day nine (40 calls/minute), whereas the number of gerbils in the parental care (PC) group remained high for up to 2 weeks (46.6 calls/minute), before decreasing. The number of USVs and duration of single syllables from postnatal day 3 to 21 in ED pups were significantly lower than those in PC pups. ED increased maternal licking, grooming behavior, and nest-building, but decreased huddling and exploring behavior. Similarly, ED increased paternal behavior, like nest-building, but reduced exploring and self-grooming. Overall, gerbil pups displayed high levels of USVs and various syllable types before weaning. ED significantly reduced the number of USVs and syllable types, but increased maternal licking, grooming, and paternal nest-building behavior.

In Tibet, China, wheat stripe rust (caused by Puccinia striiformis f. sp. tritici) has recently become one of the most destructive diseases on winter wheat. To identify races of the pathogen in Tibet, 261 isolates were obtained in 2010 and tested on seedlings of a standard set of 19 wheat indicator genotypes. Of the 261 isolates, 248 were identified as members of 19 known races (CYR17, CYR20, CYR21, CYR22, CYR23, CYR29, CYR31, CYR32, CYR33, Lov13-6, Su11-1, Su11-2, Su11-3, Su11-4, Su11-5, Su11-6, Su11-7, Su11-8, and Su11-13), and 13 identified as representatives of 4 new races. CYR32 and CYR33 were the most predominant. The number of races and their frequencies in Tibet were more similar to the fungal populations in Sichuan and Gansu provinces than to those in Yunnan, Qinghai, and Shaanxi provinces. The results suggest that Tibet is also a possible center of inoculum source and genetic variation for the stripe rust pathogen in addition to Sichuan and Gansu.

This study was based on the isolation and identification of arbuscular mycorrhizal (AM) fungi collected from the alpine Stipa steppe of north Tibet, and focused on the influence of soil texture, pH, organic matter, and available P on the spore density, isolation frequency, relative abundance, importance value, species diversity, and species evenness of the AM fungi. In the rhizosphere soil of the steppe, a total of 15 species AM fungi were isolated and identified, including 9 species of Glomus, 6 species of Acaulospora, and 1 species of Scutellospora. Among them, Glomus and Acaulospora were the dominant genera, and Glomus claroideum and Acaulospora laevis were the dominant species. In the soils with different texture, the spore density, isolation frequency, relative abundance, and importance value of the AM fungi all showed a trend of Glomus > Acaulospora > Scutellospora. Soil pH value had no significant effects on the species composition of AM fungi. However, the isolation frequency, relative abundance, and importance value of Glomus and Acaulospora showed an increasing trend with increasing soil pH, while Scutellospora showed the contrary trend. In the soils with different organic matter content, the spore density of AM fungi all showed a trend of Glomus > Acaulospora > Scutellospora, while the distribution of AM fungi had no definite pattern. The species richness and spore density of AM fungi were less affected by the soil available P content, but the species diversity and evenness showed an increasing trend with the increasing content of soil available P.
