RESUMEN

The tailwater from wastewater treatment plants serves as an important water resource in arid regions, alleviating the conflict between supply and demand. However, the effects of different tailwater discharge proportions on microbial community dynamics remain unclear. In this study, we investigated the effects of different tailwater discharge proportions on the water quality and microbial community characteristics of sediments in receiving water bodies under controlled conditions (WF-1, WF-2, WF-3, WF-4, and WF-5, containing 0% tailwater + 100% natural water, 25% tailwater + 75% natural water, 50% tailwater + 50% natural water, 75% tailwater + 25% natural water, and 100% tailwater + 0% natural water, respectively). Microbial co-occurrence networks and structural equation model were used to unveil the relationship between microbial communities and their shaping factors. Results showed that distinct microbial community compositions were found in the sediments with low- (< 50%) and high- (> 50%) proportions of tailwater. Specifically, WCHB1-41 and g_4-29-1, which are involved in organic degradation-related functions, were the key genera in the high-proportion cluster. A total of 21 taxa were more abundant in the low-proportion (< 50%) cluster than that in high-proportion (> 50%). Moreover, higher modularity was observed in the low-proportion. Total phosphorus directly affected while ammonia nitrogen indirectly affected the microbial community structure. Our findings support the distinct heterogeneity of microbial communities driven by tailwater discharge in receiving water bodies, and further confirmed that high-proportion tailwater depletes sensitive microbial communities, which may be avoided through scientific management.

Coal mine water is usually recycled as supplementary water for aquatic ecosystems in arid and semiarid mining regions of China. To ensure ecosystem health, the coal mine water is rigorously treated using several processes, including reverse osmosis, to meet surface water quality standards. However, the potential environmental impacts of this management pattern on the ecological function of receiving water bodies are unclear. In this study, we built several microcosm water ecosystems to simulate the receiving water bodies. High-quality treated coal mine drainage was mixed into the model water bodies at different concentrations, and the sediment bacterial community response and functional changes were systematically investigated. The results showed that the high-quality coal mine drainage could still shape bacterial taxonomic diversity, community composition and structure, with a concentration threshold of approximately 50%. Moreover, both the Mantel test and the structural equation model indicated that the salinity fluctuation caused by the receiving of coal mine drainage was the primary factor shaping the bacterial communities. 10 core taxa in the molecular ecological network influenced by coal mine drainage were identified, with the most critical taxa being patescibacteria and g_Geothermobacter. Furthermore, the pathway of carbohydrate metabolism as well as signaling molecules and interactions was up-regulated, whereas amino acid metabolism showed the opposite trend. All results suggested that the complex physical-chemical and biochemical processes in water ecosystems may be affected by the coal mine drainage. The bacterial community response and underlying functional changes may accelerate internal nutrient cycling, which may have a potential impact on algal bloom outbreaks.

Vanadium (V) and microplastics in soils draw increasing attention considering their significant threats to ecosystems. However, little is known about the vertical co-distribution of V and microplastics in soil profile and their combined effects on microbial community dynamics and assembly. This study investigated the spatial distribution of V and microplastics in the soils at a V smelting site and the associated microbial community characteristics along the vertical gradient. Both V and microplastics were found in the 50 cm soil profile with average concentrations of 203.5 ± 314.4 mg/kg and 165.1 ± 124.8 item/kg, respectively. Topsoil (0-20 cm) and subsoil (20-50 cm) displayed distinct microbial community compositions. Metal-tolerant (e.g., Spirochaeta, Rubellimicrobium) and organic-degrading (e.g., Bradyrhizobium, Pseudolabrys) taxa as biomarkers were more abundant in the topsoil layer. V and microplastics directly affected the microbial structure in the topsoil and had indirect influences in the subsoil, with direct impacts from organic matter. In topsoil, deterministic processes were more prevalent for community assembly, whereas stochastic processes governed the subsoil. The interspecific relationship was closer in topsoil with greater network complexity and higher modularity. These findings promote the understanding of distinct heterogeneity of microbial communities jointly driven by V and microplastics in soil environment.

The gut microbiota is closely associated with the health and production performance of livestock. Partial studies on ruminant microbiota are already in progress in the Qinghai-Tibetan Plateau Area (QTPA) in China, but large-scale and representative profiles for the QTPA are still lacking. Here, 16S rRNA sequencing was used to analyze 340 samples from yak, cattle, yak-cattle hybrids, and Tibetan sheep, which lived in a shared environment from 4 eco-regions of the QTPA during the same season, and aimed to investigate the fecal microbiota community composition, diversity, and potential function. All samples were clustered into 2 enterotypes, which were derived from the genera Ruminococcaceae UCG-005 and Acinetobacter, respectively. Environment, human activity, species, and parasitization all affected the fecal microbiota. By assessing the relationship between the fecal microbiota and the above variables, we identified a scattered pattern of fecal microbiota dissimilarity based more significantly on diet over other factors. Additionally, gastrointestinal nematode infection could reduce the capacity of the bacterial community for biosynthesis of other secondary metabolites, carbohydrate metabolism, and nucleotide metabolism. Ultimately, this study provided a fecal microbiota profile for ruminants living in 4 eco-regions of the QTPA and its potential future applications in developing animal husbandry regimes. IMPORTANCE Cattle, yak, and sheep reside as the main ruminants distributed throughout most regions of Qinghai-Tibetan Plateau Area (QTPA) in China. However, there is a lack of large-scale research in the QTPA on their fecal microbiota, which can regulate and reflect host health as an internalized "microbial organ." Our study depicted the fecal microbiota community composition and diversity of yak, cattle, yak-cattle hybrids, and Tibetan sheep from 4 eco-regions of the QTPA. Additionally, our results demonstrated here that the ruminant samples could be clustered into 2 enterotypes and that diet outweighed other factors in shaping fecal microbiota in the QTPA. This study provided a basis for understanding the microbiota characteristic of ruminants and its possible applications for livestock production in the QTPA.

Myogenic factor 5 plays active roles in the regulation of myogenesis. The aim of this study is to expose the genetic variants of the MYF5 and its association with growth performance and ultrasound traits in grassland short-tailed sheep (GSTS) in China. The combination technique of sequencing and SNaPshot revealed seven SNPs in ovine MYF5 from 533 adult individuals (male 103 and female 430), four of which are novel ones located at g.6838G > A, g.6989 G > T, g.7117 C > A in the promoter region and g.9471 T > G in the second intron, respectively. Genetic diversity indexes showed the seven SNPs in low or intermediate level, but each of them conformed HWE (p > 0.05) in genotypic frequencies. Association analysis indicated that g.6838G > A, g.7117 C > A, g.8371 T > C, g.9471 T > G, and g.10044 C > T had significant effects on growth performance and ultrasound traits. The diplotypes of H1H3 and H2H3 had higher body weight and greater body size, and haplotype H3 had better performance on meat production than the others. In addition, the dual-luciferase reporter assay showed that there are two active regions in the MYF5 promoter located at −1799~−1197 bp and −514~−241 bp, respectively, but g.6838G > A and g.7117 C > A were out of the region, suggesting these two SNPs influence the phenotype by other pathway. The results suggest that the MYF5 gene might be applied as a promising candidate of functional genetic marker in GSTS breeding.

Microscopic structure and size are important metrics for estimating aggregates environmental behaviors during water and wastewater treatment. However, in-situ determination of these characteristics is still a challenge. Here, we drew inspiration from a block disassembly process to propose an electrical impedance spectroscopy (EIS) method and constructed a generalized framework to associate macroscale electrical properties with microscopic structure and size-related characteristics of aggregates of different hierarchies. Extracted via EIS, the proposed models were verified to be capable of describing the self-similarity of aggregates and capturing the fractal and size information. Further, the proposed models exhibited a wide range of applications, which agrees well with the data gathered from various activated sludges, other colloids, and microgels in water and wastewater treatment. Finally, the EIS method was achieved online monitoring of fractal dimension and floc size during a sludge pre-oxidation conditioning process, which was elected as an example to illustrate the potential online applications of this EIS method in real water and wastewater environment. The obtained on-line data were used to indicate the potential suitable oxidation time during sludge pre-oxidation conditioning. These observations may inspire new methods of quantifying the aggregate structure and promote intelligent and dynamic decision-making during water and wastewater treatment.

Vanadium (V) and microplastics have been respectively detected in environmental media, posing threats to ecosystem and human health. However, their co-existence situations in environment with microbial adaptation have been poorly understood. In this study, water and sediments collected from potential V polluted marine and riverine ecosystems were analyzed to reveal the distribution of V and microplastics with microbial responses. High concentrations of V (1.65-6.92 µg/L in water and 6.16-347.92 mg/kg in sediment) and microplastics (800-15600 item/m3 in water and 20-700 item/kg in sediment) co-occurred in aquatic environment. Less rich and diverse bacterial communities were colonized on microplastics compared to surrounding environment. Plastic-degrading taxa (e.g., Ralstonia, Rhodococcus) and V(V) reducers (e.g., Bacillus, Pseudomonas) were enriched in microplastic biofilms. Redundancy analysis showed that V, together with nutrients, ambient conditions and Cr, contributed significantly to the compositions of microbial community on microplastics. Besides directly acting on microbial community, V could also alter it by influencing environmental factors (e.g., pH), as indicated through structural equation model. This study advances understanding the previously ignored interactions of biogeochemical processes of V and microplastics in aquatic environment.

Protozoans of Entamoeba spp. are globally distributed zoonotic parasites that infect diverse animal hosts and humans. Prevalence and species/genotypes distribution of Entamoeba spp. in domestic animals are not fully investigated on Qinghai-Tibetan Plateau (QTP), an animal husbandry and agriculture region of China. In a survey, 528 fecal samples were collected from 7 species of domestic animals on multiple locations across QTP region and analyzed by PCR and sequencing analysis. The overall prevalence of Entamoeba spp. infection in all examined animals was 97.9 %. Four Entamoeba species, E. bovis, E. moshkovskii, E. ecuadoriensis and E. histolytica were found, and majority (94.2 %) of Entamoeba-infected animals harbored E. bovis. Six Entamoeba genotypes, Entamoeba ribosomal lineages (RL) 1, 2, 3, 4, 8 and 9 were identified by sequencing analysis. Two zoonotic species, E. moshkovskii and E. histolytica, were present in horses, while E. ecuadoriensis and E. bovis were found in horses and all species of seven farm animals, respectively. It was also observed that six Entamoeba genotypes were distributed in animals in specific pattern. The results revealed high prevalence of Entamoeba spp. infection in livestock, broad range of hosts as well as diversity and species/genotype distribution of Entamoeba spp. in farm animals inhabiting on the high altitude QTP region.

Blastocystis is a common intestinal protozoan parasite of humans and a variety of animal species. To date, the prevalence of Blastocystis and major subtypes distribution in the domestic animals inhabiting in the Qinghai-Tibetan Plateau Area (QTPA) of China is yet poorly studied. In this study, we investigated the distribution and genetic diversity of Blastocystis in seven animal species in QTPA in China. Four hundred and five fresh fecal samples were collected from domestic animals in Qinghai, Yunnan, and Tibet of China and analyzed using nested PCR and SSU rRNA gene sequencing. It was found that the overall prevalence of Blastocystis infection was 40.2% (163/405) in the animals studied. The most predominant subtype of Blastocystis was ST10 (57.7%) followed by ST14 (28.8%) and ST2 (13.5%). These results reveal the epidemiological features of Blastocytis infection in animals in the high altitude plateau area. The finding of presence of ST2 in a number of animal species suggests a zoonotic nature of Blastocystis and might be of importance of public health.

Polybrominated diphenyl ethers (PBDEs) in the feather and muscle of common kestrels (Falco tinnunculus), eagle owls (Bubo bubo) and little owls (Athene noctua) collected from Beijing, China were investigated. The concentrations of ∑23PBDEs in the muscle and feather of all the birds of prey ranged from 46 to 7.77 × 104 ng/g lipid weight (lw; median 241 ng/g lw) and 1.50-191 ng/g dry weight (dw; median 21.1 ng/g dw), respectively. Higher brominated congeners, e.g., BDE-209, -153, -207 and -196 were the dominant congeners in both feather and muscle. However, the concentrations of lower brominated congeners in feather were significantly correlated with those in muscle (p < 0.05), which suggested that feather could efficiently reflect lower brominated BDEs in the internal tissue of birds of prey. The calculated hazard quotients (HQs) > 10 in common kestrel and little owl suggested that the high levels of PBDEs posed potentially high risk to some birds of prey living in Beijing area.

In order to understand the characteristics of PM2.5 pollution in the atmosphere of Chengdu southwest suburb, PM2.5 particles in Chengdu southwest suburb were collected and analyzed from March 18 to March 31st, 2015. The results showed that the daily average concentration of PM2.5 in the southwest suburb of Chengdu reached 121.21 µg · m⁻³, and the average daily concentration of 24 samples in 31 PM2.5 samples was over 75 µg · m⁻³, the daily excessive rate was 77%, indicating the PM2.5 pollution in the study area was serious in March. When studying the relationship between atmospheric and meteorological factors, it was found that there was a significant index correlation between PM2.5 concentration and atmospheric visibility, and it had a positive correlation with temperature and humidity, but the correlation was not obvious. NH4⁺ (16.24%), SO4²- (12.58%) and NO3⁻ (9.91%) were dominant in PM2.5 The ratio of NO3⁻/SO4²â» was 0.77, which indicated that the pollution of stationary sources in the southwest suburb was more severe than that of mobile sources. Organic carbon (OC)/elemental carbon (EC) ratios were higher than 2, which indicated the existence of second organic carbon (SOC). Using OC/EC ratio method to estimate the concentration of SOC, it was found that the average concentration of SOC in the southwest suburb of Chengdu in March was 3.49 µ · m⁻³, and the contribution rate of OC was 20.6%, which showed that the main source of OC in the southwest suburb of Chengdu was primary discharge. The correlation analysis of OC and EC showed that the correlation coefficient reached 0.95, indicating that the OC and EC sources were similar and relatively stable, and there was a great impact of local source emissions on Chengdu southwest suburb in spring, and primary discharge played a dominant role, while the contribution of SOC to OC was relatively small, which was consistent with the SOC characteristics obtained by estimation. Using principal component analysis method to analyze the sources of PM2.5 in the southwest of Chengdu, it was found that the main pollution sources of PM2.5 in southwest suburb of Chengdu were coal burning and biomass burning, secondary nitrate/sulfate, soil and dust, vehicle emissions, electronic production source, and mechanical processing source.

The aim of this study was to determine levels of awareness of basic knowledge concerning control and prevention of chronic obstructive pulmonary disease (COPD) among general practitioners (GPs) in a Shanghai suburb and to evaluate adherence to GOLD (COPD) guidelines in primary care. All 593 practitioners, including rural and urban community GPs at 15 primary care centers in the Songjiang district of Shanghai, were questioned, comprising 331 from urban communities and 262 from rural. Only 115 GPs (19.4%) understood that COPD is categorized into acute exacerbation and stable stages, and 328 GPs (55.3%) recognized that treatment for COPD patients was still required during stable stages. A total of 235 GPs (39.6%) knew that oxygen therapy should be administered for >15 h/d as regular home therapy. In 97 cases of stage III and IV COPD, only 1 patient had accepted long-term home oxygen therapy. Prescriptions that conformed with the GOLD recommendations occurred in approximately 8% of patients according to GOLD COPD severity staging. Basic knowledge of prevention and treatment of COPD was seriously lacking in GPs. Additional training is clearly needed.
