Dogs are highly susceptible to leptospirosis and are a public health concern due to their important role as a source of spreading disease, particularly in urban settings. In this study, we present the pathogenesis, serological characterization, and complete genome sequencing of a virulent Brazilian strain (NEG7) of L. interrogans serovar Copenhageni isolated from the urine of a dog that died due to acute leptospirosis. Clinical investigation showed that the dog was presented with icteric mucous membranes, weakness, dehydration, anorexia, and kidney and liver failures. Necropsy followed by histopathological evaluation revealed lesions compatible with liver and kidney leptospirosis. The leptospires recovered from the urine were further characterized by genome analysis, which confirmed that the isolate belonged to L. interrogans serogroup icterohaemorrhagiae serovar Copenhageni. Multiple bioinformatics tools were used to characterize the genomic features, and comparisons with other available Copenhageni strains were performed. Characterization based on absence of an INDEL in the gene lic12008, associated with phylogenetic and ANI (99.99% identity) analyses, confirmed the genetic relatedness of the isolate with L. interrogans serovar Copenhageni. A better understanding of the diversity of the pathogenic Leptospira isolates could help in identifying genotypes responsible for severe infections. Moreover, it can be used to develop control and prevention strategies for Leptospira serovars associated with particular animal reservoirs.

Essential oils (EO) are aromatic compounds from the plant secondary metabolism. Melaleuca alternifolia EO is well known for its medicinal properties and promising use as an antimicrobial agent. Pythiosis is a difficult-to-treat and emerging disease caused by the oomycete Pythium insidiosum. This study evaluated a nanoemulsion formulation of M. alternifolia (NEMA) in topical and intralesional application to treat experimental pythiosis. Dermal toxicity tests were performed on M. alternifolia EO in Wistar rats. Pythiosis was reproduced in rabbits (n = 9) that were divided into groups: group 1 (control), cutaneous lesions with daily topical application of a non-ionizable gel-based formulation and intralesional application of sterile distilled water every 48 h; group 2 (topical formulation), lesions treated daily with topical application of a non-ionizable gel-based formulation containing 5 mg/ml of NEMA; and group 3 (intralesional formulation), lesions treated with NEMA at 5 mg/ml in aqueous solution applied intralesionally/48 h. The animals were treated for 45 days, and the subcutaneous lesion areas were measured every 5 days. M. alternifolia EO showed no dermal toxicity. The lesion areas treated with intralesional NEMA reduced at the end of treatment, differing from groups 1 and 2 (P < 0.05). In the topically treated group, the lesion areas did not differ from the control group, although the number of hyphae significantly reduced (P < 0.05). Under the experimental conditions of this study, the NEMA formulations presented a favorable safety profile. However, further studies are required to evaluate if this safety applies to higher concentrations of NEMA and to validate its use in clinical pythiosis.

ABSTRACT: In this study, an outbreak of spontaneous poisoning by Dodonaea viscosa (D. viscosa) in a herd of dairy cattle in the municipality of Capão do Leão, Rio Grande do Sul, was investigated. Three deaths occurred in a batch of 16 Jersey cattle, aged between three and four years, kept in a native field. The clinical signs observed were apathy, decreased production, and anorexia, with death occurring within approximately 48 h after the onset of signs. The three cattle were necropsied, and tissue samples were sent for histopathological examination. Necropsy findings included serosanguineous fluid in the abdominal cavity, intestines with congested serosa, and marked mesenteric edema. The mucosa of the abomasum of two of the animals was hemorrhagic with bloody content, and among the ruminal content of a bovine, leaves with morphological characteristics compatible with D. viscosa were observed. The livers of the three animals were enlarged, with accentuation of the lobular pattern. Histologically, centrilobular coagulation necrosis with congestion and hemorrhage was observed in the liver. Vacuolization and degeneration of hepatocytes were observed in the mid-zonal and periportal regions. The diagnosis of poisoning by D. viscosa leaves was based on epidemiological data, necropsy findings, and histopathological alterations. The presence of the plant in the rumen and in the grazing site of the affected cattle was essential for the diagnosis.
RESUMO: Neste trabalho, é descrito um surto de intoxicação espontânea por Dodonaea viscosa (D. viscosa) ocorrido em um rebanho de bovinos leiteiros, no município de Capão do Leão, no Rio Grande do Sul. Ocorreram três mortes em um lote de 16 bovinos da raça Jersey com idades entre três e quatro anos, mantidos em campo nativo. Os sinais clínicos observados foram apatia, queda na produção e anorexia, com morte em aproximadamente 48 horas após o início dos sinais. Os três bovinos foram necropsiados, e amostras de tecidos foram encaminhadas para exame histopatológico. Os achados de necropsia incluíam líquido serossanguinolento na cavidade abdominal, intestinos com serosas congestas e marcado edema de mesentério. A mucosa do abomaso de dois animais apresentava-se hemorrágica com conteúdo sanguinolento e, em meio ao conteúdo ruminal de um bovino foram observadas folhas com caracteres morfológicos compatíveis com D. viscosa. O fígado dos três animais estava aumentado, com acentuação do padrão lobular. Histologicamente no fígado havia necrose de coagulação centrolobular com congestão e hemorragia. Nas regiões médio-zonal e periportal observou-se vacuolização e degeneração dos hepatócitos. O diagnóstico de intoxicação pelas folhas D. viscosa foi baseado nos dados epidemiológicos, nos achados de necropsia e nas alterações histopatológicas. A presença da planta no rúmen e no local de pastoreio dos bovinos afetados foi fundamental para o diagnóstico.

The objective of this work was to conduct a retrospective study of causes of diarrhea in calves in southern Rio Grande do Sul between 1980 and 2014. All necropsy protocols and materials received in the Regional Diagnostic Laboratory of the Veterinary School of the Federal University of Pelotas were reviewed. Ninety-four cases of calves with diarrhea were diagnosed, of these, 44.7% were caused by parasitic infections and 34.0% by bacterial infections. Parasites as cause of diarrhea was the most important diagnostic in southern Rio Grande do Sul and affected calves from three to 12 months of age. The occurrence of diarrhea in animals even infants may be associated with environmental factors and the form of management that these animals are kept. Bacterial diseases are also important causes of diarrhea in calves up to three months of age in the region.
O objetivo deste trabalho foi descrever os dados de um estudo retrospectivo nos registros de 34 anos atividade de um laboratório de diagnóstico sobre as causas de diarreia em bezerros na região sul do Rio Grande do Sul. Foram revisados os protocolos de necropsia e materiais desta espécie encaminhados ao Laboratório Regional de Diagnóstico da Faculdade de Veterinária da Universidade Federal de Pelotas. Foram diagnosticados 94 casos de bezerros com diarreia, destes, 44,7% foram de causas parasitárias e 34,0% de causas bacterianas. Diarreia de causa parasitária foi a mais importante na região sul do Rio Grande do Sul e afetou bezerros de três a 12 meses de idade. A ocorrência de diarreia em animais mesmo lactentes pode estar associada a fatores ambientais e a forma de manejo no qual esses animais são mantidos. As enfermidades bacterianas foram, também, importantes causas de diarreia em bezerros de até três meses de idade na região.

Pythiosis is a rapidly progressing disease that can be lethal to affected individuals due to resistance to available therapeutic protocols. The disease affects mammals, with the largest number of reports in horses and humans. The present study investigated the activity of biogenic silver nanoparticles (bioAgNP) in the treatment of experimental pythiosis. The disease was reproduced in nine female 90-day-old New Zealand rabbits. Animals were divided into three groups: group1 (control, n = 3) daily and topically treated with a nonionized gel-based formulation and 1 ml of sterile distilled water intralesion administered every 48 hours; group 2 (n = 3), daily and topically treated with gel-based formulation containing 1 µg/ml bio-AgNP; group 3 (n = 3), treated with 1 ml bio-AgNP in 1 µg/ml aqueous solution intralesion administered every 48 hours. Animals were treated for 45 days, and the area of subcutaneous lesions was measured every 5 days. Results showed that groups 2 and 3 differed from control group (P < .05) in the lesion area, as well as the amount of hyphae within the lesions. It was observed that lesions of treated animals (groups 2 and 3) did not differ from each other, showing that the application route did not influence the regression of lesions. However, it was observed that one animal from group 2 reached clinical cure at 35 days of treatment. This research is pioneer in the application of nanocomposites for the treatment of experimental pythiosis and showed that bio-AgNP can be powerful allies of integrative medicine and can be included in pythiosis therapeutic protocols.

Bovine tuberculosis (BTB) is an infectious and zoonotic disease with socioeconomic importance, responsible for economic losses in livestock due to the decrease in animal productivity, mortality, bovine carcass condemnation and restriction on exportations. The objective of the present study was to evaluate the epidemiological aspects of this disease through a descriptive analysis and time series of cases of bovine tuberculosis diagnosed at the Laboratório Regional de Diagnóstico (LRD) of the Faculdade de Medicina Veterinária, Universidade Federal de Pelotas (UFPel) from 2000 to 2015. A survey was carried out in the LRD database of cases diagnosed as tuberculosis in cattle during this period. The incidence rate of BTB ([cases/population] x 100000) was calculated. Qualitative variables (sex, age, race, origin, type of material and presumptive diagnosis) were analyzed in a descriptive way, aiming to establish the profile of affected animals, characterize the municipalities in which the disease is originated and also fix the main referral organs for diagnosis. The time series analysis was performed using the Gretl statistical software 1.9.12 (GNU Regression, Econometric and Time-series Library). To verify the existence of the stationary distribution, the test of Dickey-Fuller was used, considering a value of p<0.05. During this period, 331 cases of BTB were diagnosed in the LRD. The present study identified that the predominant profile of the bovines affected by tuberculosis was female bovine aged more than two years old and crossbreed; the main organ sent for histopathological examination was lymph nodes. The microregion of Pelotas presented the highest incidence of the disease. There was also a tendency of a decrease in the incidence rate of this disease over the years, absence of seasonal influence in the occurrence of the disease and no cases of epidemics occurred from 2002 to 2015 in the area covered by the LRD-UFPel.(AU)
Tuberculose bovina é uma enfermidade infectocontagiosa e zoonótica, de importância socioeconômica, responsável por prejuízos econômicos na pecuária em decorrência da diminuição na produtividade animal, mortalidades, condenações de carcaças e restrições nas exportações. O objetivo do presente estudo foi avaliar os aspectos epidemiológicos, por meio de uma análise descritiva e de séries temporais, dos casos de tuberculose bovina diagnosticados no Laboratório Regional de Diagnóstico da Faculdade de Veterinária, Universidade Federal de Pelotas (LRD-UFPel), no período de 2000 a 2015. Foi realizado um levantamento no banco de dados do LRD dos casos diagnosticados como tuberculose em bovinos, entre os anos de 2000 e 2015. Foi calculada a taxa de incidência da tuberculose bovina ([casos/população] x 100000). As variáveis qualitativas (sexo, idade, raça, procedência, tipo de material e diagnóstico presuntivo) foram analisadas de forma descritiva, tendo como finalidade estabelecer o perfil dos animais acometidos, caracterizar os municípios de origem, e estabelecer os principais órgãos remetidos para diagnóstico. A análise de série temporal foi feita através do software estatístico Gretl 1.9.12 (GNU Regression, Econometricand Time-series Library) e para verificar a existência da estacionariedade utilizou-se o teste de Dickey-Fuller aumentado, considerando um valor de p<0,05. Durante este período foram diagnosticados 331 casos de tuberculose bovina no LRD. O presente estudo permitiu concluir que o perfil predominante dos bovinos acometidos por tuberculose foi de fêmeas, com mais de dois anos de idade e mestiças; o principal órgão encaminhado para exame histopatológico foram linfonodos e a microrregião de Pelotas apresentou a maior incidência da doença. Foi constatada, ainda, uma tendência de queda na taxa de incidência da doença ao longo dos anos analisados, ausência de influência sazonal na ocorrência da enfermidade e, não foram registrados casos de epidemias nos anos de 2002 a 2015, na área de abrangência do LRD-UFPel.(AU)

Hydatidosis and cysticercosis are parasitoses caused by the larval forms of the cestodes Equinococcus spp. and Taenia spp., which belong to the Taeniidae family. Their definitive hosts are canids and humans, respectively, with ruminants as the intermediate hosts and humans as an accidental host of both diseases. These parasites are responsible for large economic losses in slaughterhouses due to condemnation of carcasses and by-products. The present study reports the mean incidence rates of hydatidosis and cysticercosis in cattle slaughtered in the southern region of Rio Grande do Sul state, Brazil. The incidence rates observed between 2013 and 2016 were 19.96% and 0.9%, respectively, with decreased tendency of occurrence of both diseases in those years. Despite the downward tendency of the diseases, hydatidosis presented high incidence. Hydatidosis performance over the years was characterized by significant increase in the number of cases, followed by marked decrease. Cysticercosis presented a decrease in number of cases at the beginning and the end of each year. These diseases have a significant socioeconomic impact as they are responsible for large losses in the livestock industry, due to reduced productivity and carcass condemnation, and represent a risk to public health.(AU)
Hidatidose e cisticercose são parasitoses causadas pelas formas larvais dos cestódeos Equinococcus spp. e Taenia spp., pertencentes a família Taeniidae, possuem como hospedeiros definitivos canídeos e o homem respectivamente, sendo os ruminantes os hospedeiros intermediários, e o homem um hospedeiro acidental, em ambas as doenças. Estas parasitoses são responsáveis por grandes perdas econômicas em frigoríficos, devido a condenações de carcaças e subprodutos. O presente artigo relata que a incidência média de hidatidose e cisticercose em bovinos abatidos na região sul do Rio Grande do Sul, entre os anos de 2013 e 2016, foi de 19,96% e 0,9% respectivamente, sendo avaliada uma tendência de queda no número de casos de ambas as doenças ao longo dos anos analisados. Apesar da tendência de queda das doenças, a hidatidose apresentou uma alta incidência. O comportamento da hidatidose ao longo dos anos foi caracterizado por aumentos significativos no número de casos da doença, seguidos por acentuadas quedas. Já a cisticercose apresentou uma diminuição no número de casos no início e no final de cada ano. Estas doenças apresentam grande impacto socioeconômico, pois são responsáveis por grandes perdas na pecuária, por redução na produtividade e condenações, além de representarem um risco à saúde pública.(AU)

Babesiose cerebral é uma enfermidade causada pelo protozoário Babesia bovis. O agente faz parte do complexo Tristeza Parasitária Bovina, uma das mais importantes doenças parasitárias em bovinos. O presente estudo relata um surto causado por B. bovis em vinte bezerros de aproximadamente 7 a 25 dias de idade. O surto ocorreu entre março e junho de 2015, na região sul do Brasil, área de instabilidade enzoótica para a Tristeza Parasitária Bovina. O diagnóstico foi realizado pela epidemiologia, lesões macroscópicas e pela presença de numerosas formas parasitárias de Babesia bovis em capilares encefálicos, observados em imprints corados por Giemsa. Surtos de babesiose por B. bovis cerebral nos primeiros dias de vida de bezerros é incomum, porém não pode ser desconsiderada em surtos com alta letalidade em áreas de instabilidade enzoótica.(AU)
Cerebral babesiosis is a protozoan disease caused by Babesia bovis. This parasite belongs to the bovine parasitic complex of tick-borne diseases that affect livestock worldwide. The present study reports an outbreak caused by B. bovis affected twenty 7-25 day-old calves. Outbreak occurred from May to July 2015 in the south of Brazil, where there is an area of enzootic instability for cattle tick fever. The macroscopic lesions were anemia, hemoglobinuria, splenomegaly, hepatomegaly, yellow liver and cherry-pink discoloration of cerebral and cerebellar cortex. The diagnosis was based on epidemiology, necropsy and microscopic findings in the brain that showed B. bovis in the capillary vessels of the brain in imprints stained by Giemsa. Cases of cerebral babesiosis by Babesia bovis in such young calves are uncommon but should be considered as a diagnosis possibility when there is high mortality rate in areas ofen zootic instability.(AU)

Descrevem-se os aspectos clínico patológicos de quatro casos de tuberculose em felinos domésticos no Sul do Rio Grande do sul. Foi realizado um estudo retrospectivo dos casos diagnosticados como tuberculose em gatos necropsiados no Laboratório Regional de Diagnóstico da Universidade Federal de Pelotas (LRD/UFPel) no período de janeiro de 2000 a dezembro de 2014. Em dois casos a amplificação da sequência genética IS6110 específica para Mycobacterium tuberculosis demonstrou que a doença era de origem humana. Os outros dois foram positivos para Mycobacterium spp. Os sinais clínicos caracterizaram-se por emagrecimento progressivo, anorexia e dispneia. As alterações macroscópicas evidenciadas eram de caquexia, aumento de volume dos linfonodos submandibulares com áreas focais de aspecto caseoso ao corte. Nos pulmões havia áreas multifocais a coalescentes amareladas de aspecto caseoso, discretamente elevadas e firmes na superfície pleural. Histologicamente havia broncopneumonia e linfadenite granulomatosas. A coloração de Ziehl-Neelsen evidenciou a presença de bacilos álcool-ácidos resistentes. Houve imunomarcação positiva para micobactéria pela técnica de imuno-histoquímica. Alerta-se para a importância da tuberculose felina como um problema de saúde pública pois gatos infectados podem servir de fonte para a disseminação das micobactérias no ambiente, além de serem sentinelas para a ocorrência da enfermidade em seres humanos.(AU)
Clinical pathological aspects of four cases of tuberculosis in domestic cats in southern Rio Grande do Sul is described. A retrospective study was conducted of cats diagnosed with tuberculosis and necropsied at the Regional Diagnostic Laboratory of the Federal University of Pelotas (LRD/UFPel) from January 2000 to December 2014. In two cases, the amplification of genetic sequence IS6110 specific to Mycobacterium tuberculosis demonstrated that the disease was of human origin. The other two were positive for Mycobacterium spp. The clinical signs were characterized by progressive weight loss, anorexia, and dyspnea. The evident macroscopic changes were cachexia, swelling of submandibular lymph nodes with focal areas of caseous aspect on cut surface. The lungs had multifocal caseous areas at the pleural surface. Histologically, the cats had pneumonia and granulomatous lymphadenitis. The Ziehl-Neelsen staining revealed the presence of alcohol-acid resistant bacteria. There was a positive immunostaining for mycobacteria by immunohistochemistry technique. We alert to the importance of feline tuberculosis as a public health problem, because infected cats may serve as source for dissemination of mycobacteria in the environment, and be sentinels for the occurrence of the disease in humans.(AU)
