RESUMEN

BACKGROUND: Chronic intestinal pseudo-obstruction (CIPO) is characterized by dilatation of the bowel lumen and abnormal motility. In this study, we aimed to quantify small bowel dysmotility in CIPO using a validated pan-intestinal motility assessment technique based on motion capture magnetic resonance imaging (MRI) compared to normal controls. In addition, we explored if motility responses of CIPO patients to neostigmine challenge differed from healthy volunteers. METHODS: Twenty healthy volunteers (mean age 28, range 22-48) and 11 CIPO patients (mean age 47, range 19-90) underwent MRI enterography to capture global small bowel motility. Eleven controls and seven CIPO patients further underwent a randomized placebo-controlled crossover study of either intravenous neostigmine (0.5 mg) or saline with motility MRI repeated at a mean of 3 weeks. Motility was quantified in regions of interest placed to encompass the whole small bowel volume using a validated, postprocessing technique to give a global motility index in arbitrary units (AU). Baseline and stimulated motility was compared using Wilcoxon rank-sum paired T-tests. KEY RESULTS: Baseline global small bowel motility was significantly lower in CIPO patients compared to controls (mean 0.25 AU vs 0.35 AU, p < 0.001). Motility in both groups increased significantly after neostigmine (0.06 AU increase, p = 0.016 in CIPO and 0.06 AU increase, p = 0.002 in controls). Three patients with scleroderma had a reduced response to neostigmine. CONCLUSIONS & INFERENCES: Global small bowel motility in CIPO patients is significantly lower than controls and response to the pro-kinetic agent neostigmine may differ according to disease phenotype. Software-quantified bowel motility using cine MRI has potential as a future tool to investigate enteric dysmotility.

ABSTRACT The use of green manure and/or cover crops is a technique that has been widespread for the cultivation of medicinal species, since it ensures the sustainability of agricultural soil, is flexible in its use, and improves biological activity in the soil. The purpose of this study was to evaluate the use of cover crops to increase biomass production of roselle, Brazilian peppertree, and carobinha plants. Two species of tropical legumes (black velvet bean and jack bean) were evaluated as ground covers, and also assessed were one control plot (without cover crops) and three medicinal species (roselle, Brazilian peppertree, and carobinha). The experiment was arranged in a split-plot design, where the plots had cover crops and the subplots contained the medicinal plants. The plants for the cover crop were propagated by direct sowing in the growing site, while the medicinal ones were transplanted directly between the cover crop. The dry mass production of the velvet beans and jack beans were 8.37 t ha-1 and 14.37 t, respectively. The use of cover crops contributed to the increased microbial biomass of the soil. The production of dry mass of leaves, stems, and fruits of roselle and fruits of Brazilian peppertree were higher when grown in succession to the crop cover for the soil, regardless of the species. The production of carobinha was similar when grown in areas with or without the cover. To increase the yield of roselle and Brazilian peppertree, preceding crops of jack bean and black velvet bean are recommended.

RESUMEN

BACKGROUND: The wireless motility capsule (WMC) offers the ability to investigate luminal gastrointestinal (GI) physiology in a minimally invasive manner. AIM: To investigate the effect of testing protocol, gender, age and study country on regional GI transit times and associated pH values using the WMC. METHODS: Regional GI transit times and pH values were determined in 215 healthy volunteers from USA and Sweden studied using the WMC over a 6.5-year period. The effects of test protocol, gender, age and study country were examined. RESULTS: For GI transit times, testing protocol was associated with differences in gastric emptying time (GET; shorter with protocol 2 (motility capsule ingested immediately after meal) vs. protocol 1 (motility capsule immediately before): median difference: 52 min, P = 0.0063) and colonic transit time (CTT; longer with protocol 2: median 140 min, P = 0.0189), but had no overall effect on whole gut transit time. Females had longer GET (by median 17 min, P = 0.0307), and also longer CTT by (104 min, P = 0.0285) and whole gut transit time by (263 min, P = 0.0077). Increasing age was associated with shorter small bowel transit time (P = 0.002), and study country also influenced small bowel and CTTs. Whole gut and CTTs showed clustering of data at values separated by 24 h, suggesting that describing these measures as continuous variables is invalid. Testing protocol, gender and study country also significantly influenced pH values. CONCLUSIONS: Regional GI transit times and pH values, delineated using the wireless motility capsule (WMC), vary based on testing protocol, gender, age and country. Standardisation of testing is crucial for cross-referencing in clinical practice and future research.

ABSTRACT: The aim of this experiment was to evaluate the biomass yield and flavonoid contents of Achyrocline satureioides grown under different levels of phosphate fertilizer in association with broiler litter, during two successive harvests. Four levels of P2O5 (0, 100, 200 and 300 kg ha-1), as triple superphosphate, and two levels of broiler litter (0 and 10 t ha-1) were arranged as a 4x2 factorial design in randomized blocks with four replications. Two harvests of plants, considered as split-plot, were made. Seeds were sown in polystyrene trays then transplanted to the experimental field and harvested at 195 and 223 days after transplanting (DAT). The maximum heights of the plants were 1.14 m under the addition of broiler litter and 0.97 m without it, at 195 days after transplanting. The highest yields of total fresh weight (30.31 ton ha-1) and dry weight (11.38 ton ha-1) were obtained at 223 DAT, both with the concentration of 300 kg ha-1 P2O5. The fresh weight of the inflorescences was greater under the addition of broiler litter (4.08 ton ha-1) than without it (3.49 ton ha-1). The plants grew linearly in response to the use of phosphate, and the maximum yield was 4.65 ton ha-1 under the concentration of 300 kg ha-1 P2O5, regardless of the harvest season. The dry weight of the inflorescences reached maximum yield (2.38 ton ha-1) at 223 DAT, in response to the use of 300 kg ha-1 P2O5. The highest yield of dry weight and the highest contents of flavonoids from the inflorescences were obtained when A. satureioides was grown under the concentration of P2O5 (300 kg ha-1) with the addition of broiler litter (10 ton ha-1) and harvested at 223 DAT.

RESUMEN

The aim of this study was to evaluate the phenolic and flavonoids contents and the antioxidant and antitumoral activity of leaf and calyx methanolic extracts from Hibiscus sabdariffa (roselle) cultivated with poultry litter and organosuper® under three modes of application. The total phenolic content in the each extract was determined using the Folin-Ciocalteu reagent and for aluminium chloride flavonoids. The antioxidant parameters were analyzed using a 2, 2-diphenyl-1-picrylhydrazyl (DPPH.) free radical scavenging assay. An antitumor colorimetric assay using sulforhodamine B. The highest contents of phenolic and flavonoids were observed in leaf extracts (389.98 and 104.52 mg g-1, respectively) and calyx extracts (474.09 and 148.35 mg g-1, respectively) from plants cultivated with organosuper®, although these values did not differ significantly from those observed for the other treatments. The average IC50 of leaves (43.48 µg mL-1) and calyces (37.15 µg mL-1) demonstrated that both have substances that may contribute to free radical scavenging action. The methanol extract from calyces showed significant selective activity against a leukemia line (K-562), with IC50 values of 0.12 mg mL-1 (organosuper®) and 1.16 mg mL-1 (poultry litter), with concentration-dependent, cytotoxic and cytocidal effects.

RESUMEN

The aim of this experiment was to evaluate the biomass yield and chemical composition of the essential oil of clove basil in response to doses of broiler litter (0, 5, 10, 15 and 20 t ha-1) added in presence of triple superphosphate (200 kg ha-1) or without it. Before the first harvest, the height of the plants increased significantly (34.42 cm) when 15 tonnes ha-1 broiler litter were added to the soil. However, after resprouting, the plant heights ranged only as a function of the vegetative cycle, with maximum height (76.0 cm) observed at 60 days. Phosphate fertilization also significantly influenced the leaf area (1.771 cm2 plant-1) during the first harvest. The fresh weight of leaves increased linearly in response to the addition of broiler litter (20 ton ha-1) with maximum yields of 1,709 and 7,140 kg ha-1 during the first harvest and regrowth, respectively. On average, the oil content of the leaves was 0.7%, with maximum performance (3.8 L ha-1) when 20 tonnes ha-1 of broiler litter were added on the soil. Eugenol was found to be the major compound of the essential oil (71.65%).

BACKGROUND: High-resolution anorectal manometry (HRAM) is a relatively new method for collection and interpretation of data relevant to sphincteric function, and for the first time allows a global appreciation of the anorectum as a functional unit. Historically, traditional anal manometry has been plagued by lack of standardization and healthy volunteer data of variable quality. The aims of this study were: (i) to obtain normative data sets for traditional measures of anorectal function using HRAM in healthy subjects and; (ii) to qualitatively describe novel physiological phenomena, which may be of future relevance when this method is applied to patients. METHODS: 115 healthy subjects (96 female) underwent HRAM using a 10 channel, 12F solid-state catheter. Measurements were performed during rest, squeeze, cough, and simulated defecation (push). Data were displayed as color contour plots and analysed using a commercially available manometric system (Solar GI HRM v9.1, Medical Measurement Systems). Associations between age, gender and parity were subsequently explored. KEY RESULTS: HRAM color contour plots provided clear delineation of the high-pressure zone within the anal canal and showed recruitment during maneuvers that altered intra-anal pressures. Automated analysis produced quantitative data, which have been presented on the basis of gender and parity due to the effect of these covariates on some sphincter functions. In line with traditional manometry, some age and gender differences were seen. Males had a greater functional anal canal length and anal pressures during the cough maneuver. Parity in females was associated with reduced squeeze increments. CONCLUSIONS & INFERENCES: The study provides a large healthy volunteer dataset and parameters of traditional measures of anorectal function. A number of novel phenomena are appreciated, the significance of which will require further analysis and comparisons with patient populations.

RESUMEN

The aim of this study was to evaluate the production and chemical composition of the essential oil of Brazilian pepper fruits grown in single and double rows using different doses of semi decomposed poultry litter in two evaluation times. The experiment was carried out at the Federal University of Grande Dourados, in the city of Dourados, state o- Mato Grosso do Sul, Brazil, from October 2009 to November 2010. Brazilian pepper plants were grown in single and double rows in soil with incorporated poultry litter at the doses of 0, 5, 10, 15 and 20 t ha-1. Treatments were arranged as a 2 x 5 factorial experiment in a randomized block design with four replications. Fruits were harvested 180 and 390 days after transplant (DAT). There was a significant interaction for fresh weight of fruits and weight of 50 fruits, being the values higher at 180 DAT in the double rows with increasing poultry litter doses. Fruits harvested 390 DAT showed higher diameter compared with those harvested 180 DAT. The number of fruits per bunch was significantly influenced by the doses of poultry litter, presenting a linear increase with increasing doses. The essential oil of the Brazilian pepper fruits obtained by hydrodistillation and analyzed by Gas chromatography - mass spectrometry exhibited predominance of monoterpenes, highlighting α-pinene (20.14%) as the major constituent. The chemical composition of the essential oil was not influenced by the number of plant rows in the plot or by the doses of poultry litter in any evaluation time. Therefore, the cultivation of Brazilian pepper plants is recommended in double rows, with 13.59 t ha-1 of incorporated poultry litter in the soi, and with harvest of 180 DAT for higher fruit production.

Basil and lettuce were planted at field in joined experiment, which established five treatments: two rows of basil in monocrop system (M2) spaced 0.50 m; three rows of basil in monocrop system spaced 0.33 m (M3); four rows of lettuce in monocrop system spaced 0.25 m (A4); two rows of basil in monocrop system and intercropped with four rows of lettuce (M2A4) and three rows of basil intercropped with four rows of lettuce (M3A4). Used design was randomized blocks with eight replications. Lettuce plants were harvested on 48 days after transplanting and basil plants were harvested in two dates, on 96 and on 113 days after transplanting (DAT). Intercrop did not influence on foliar area neither on yields of leaves of inflorescences of basil neither on commercial or non-commercial lettuce heads. Although, regarding plant arrangement, on second harvest date, basil plants that were cultivated under two rows had the greatest foliar area (2,076.99 cm² plant-1) and the greatest fresh masses of leaves (5,012.53 kg ha-1). Considering that the values of Equivalent Area Ration in the first and the second harvest of basil were higher than 1.0. It was concluded that to intercrop basil with lettuce is viable. It is recommended the arrangement of three rows of basil alternated with four rows of lettuce, with harvested at 96 DAT, because it had EAR of 2.01.

The aimed of this study was to evaluate the effect of of chicken manure and phosphorus incorporated to the soil on growth and yield of fáfia. Treatments were five doses of chicken manure (0, 5, 10, 15 and 20 t ha-1), with and without the use of phosphorus (200 kg ha-1 of P2O5, in triple superphosphate form), arranged in 5 x 2 factorial scheme in randomized block design with four replications. Plant heights varied as a function of chicken manure doses, which the highest (138.50 cm) under 20 t ha-1. Yields of fresh and dried weight of leaves, stems and roots were influenced significantly by the interaction chicken manure x phosphorus. The highest yields of fresh and dried leaves (597 kg ha-1 and 100 kg ha-1, respectively) were obtained with the dose of 20 t ha-1 of chicken manure, without the use of phosphorus. For stems, the highest yields of fresh and dried weight (7.757 kg ha-1 and 2.633 kg ha-1, respectively) were obtained with the dose of 20 t ha-1 of chicken manure, with the use of phosphorus. The highest yields of fresh and dried weight of roots (9.065 kg ha-1 and 377 kg ha-1, respectively) were obtained with the dose of 20 t ha-1 of chicken manure, without the use of phosphorus. Average contents of N, P and K of leaves were 8.60; 0.88 and 4.46 g kg-1, respectively. Contents of Ca and Mg were maximum (7.56 and 5.12 g kg-1, respectively) with the use of 20 t ha-1 of chicken manure, independent on the use of phosphorus. In roots, average contents of N, P and Mg were 2.74, 0.27 and 0.96 g kg-1, respectively. K content reduced linearly with chicken manure doses and of Ca was maximum with the use of 20 t ha-1. For obtaining the highest yields of fresh and dried weight of leaves and roots, the fáfia plants must be grown with chicken manure (20 t ha-1) incorporated to the soil within the use of phosphorus.

This study aimed to evaluate the morphometry of fruits and seeds of Jacaranda decurrens subsp. symmetrifoliolata ("carobinha") and the effect of temperature on the germination of seeds that were stored in environment without controlled temperature and relative humidity. The study was carried out at Seed Laboratory of Federal University of Grande Dourados, Mato Grosso do Sul, Brazil. For a sample of 120 fruits and 550 seeds, the following variables were determined: length, width and thickness, besides the number of seeds per fruit and fruit biomass. Germination was studied after three seed storage periods (120, 480 and 720 days) at environment temperature and four germination temperatures: three constant temperatures (18, 25 and 30ºC) with continuous light, and one alternating temperature, 20-30ºC, with 12-hour photoperiod. Experimental design was completely randomized, in 3 x 4 factorial schemes, with four replicates. The variations in the measures of fruits and seeds evidence a high genetic variability in the population. The seeds stored for 120 days and subjected to germination test at 25ºC resulted in 69.4% germination and germination velocity index - GVI of 0.983. At 30ºC, constant temperature, and 20-30ºC, alternating temperature, germination was 63.9% and 63.8% while GVI was 0.486 and 0.413, respectively. At 18ºC, germination was slow and lower, 27.7%, and GVI was 0.119. Seeds stored for 480 and 720 days did not germinate. Based on the physiological behavior of seeds, it is possible to classify Jacaranda decurrens subsp. symmetrifoliolata as an intermediate species (between orthodox and recalcitrant).

The aim of this study was to evaluate the effect of five spacing between plants and the use or not of semi-decomposed chicken manure incorporated into the soil on the biomass production of plants and on the levels of phenols, flavonoids and antioxidant activity of the extract of C. adamantium fruits. The experiment was carried out in the Medicinal Plant Garden - HPM, of Federal University of Grande Dourados/UFGD, in Dourados-Mato Grosso do Sul State, Brazil. C. adamantium was studied under five spacing between plants in a row (0.30; 0.35; 0.40; 0.45 and 0.50 m) and with and without semi-decomposed chicken manure incorporated in the soil, at a dose of 10 t ha-1. Treatments were arranged as 5 x 2 factorial, in randomized block design, with four replicates. At 390 days after transplanting, the plants had 54.89 cm height, 10.01 mm stem diameter and 178.27 leaves per plant. The longitudinal (17.34 mm) and transverse (18.07 mm) diameters of the fruits did not vary with the spacing or with the use of chicken manure. The largest number (28.56 fruits plant-1) and greatest fresh weight (83.65 g plant-1) of fruits were obtained for plants cultivated under spacing of 0.32 and 0.35 m between plants, respectively, independent of the use of chicken manure. The chicken manure induced a significant increase in phenol content and reduced flavonoid content and the antioxidant activity of the fruit pulp. The chicken manure did not influence the productivity of C. adamantium or the chemical composition of fruits. The use of spacing of 0.35 m between plants and 1.50 m between rows is recommended, with the aim of producing fruits.

RESUMEN

The lethal and sublethal effects of the ecdysone agonist methoxyfenozide on the fall armyworm, Spodoptera frugiperda (J. E. Smith), were investigated by feeding a methoxyfenozide-treated diet to fifth instars until pupation in doses corresponding to the LC10 and LC25 for the compound. Larval mortality reached 8% and 26% in the low and high concentration groups, respectively, on the seventh day of the experiment. A progressive larval mortality of 12% for the LC10 and 60% for the LC25 was observed before pupation. Treated larvae exhibited lower pupal weights, higher pupal mortality, presence of deformed pupae, and more deformed adults than untreated larvae. The incorporation of methoxyfenozide into the diet had a significant effect on the timing of larval development. The development period for males and females was about seven days longer than the controls for both concentrations tested. In contrast, the compound affected neither pupae nor adult longevity. Finally, S. frugiperda adults that resulted from fifth instars treated with methoxyfenozide were not affected in their mean cumulative number of eggs laid per female (fecundity), nor percentages of eggs hatched (fertility), or the sex ratio. Our results suggest that the combination of lethal and sublethal effects of methoxyfenozide may have important implications for the population dynamics of the fall armyworm.

RESUMEN

BACKGROUND: Colonic propagating sequences (PS) are important for the movement of colonic content and defecation, and aberrant PS patterning has been associated with slow transit constipation. However, because these motor patterns are typically recorded over long periods (24 h +), the visualization of PS spatiotemporal patterning is difficult. Here, we develop a novel method for displaying pan-colonic motility patterns. METHODS: A 3D mesh representing the geometry of the human colon was created as follows: (i) Human colon images from the Visible Human Dataset were digitized to create a 3D data cloud, and (ii) A surface mesh was fitted to the cloud using a least-squares minimization technique. Colonic manometry catheters were placed in the ascending colon of healthy controls and patients with slow transit constipation (STC), with the aid of a colonoscope. The colonic manometry data were interpolated and mapped to the model according to the following anatomical landmarks: cecum, hepatic flexure, splenic flexure, sigmoid-descending junction, and anus. KEY RESULTS: These 3D images clearly and intuitively communicate characteristics of normal and abnormal colonic motility. Specifically we have shown the reduced amplitude of the antegrade propagating pressure waves (PPW) throughout the colon and reduced frequency of PPWs at the mid-colon in patients with STC. CONCLUSIONS AND INFERENCES: A novel method for the 3D visualization of PS is presented, providing an intuitive method for representing a large volume of physiological data. These techniques can be used to display frequency, amplitude or velocity data, and will help to convey regions of abnormally in patient populations.

The aim of this study was to evaluate the development and yield of "carobinha" (Jacaranda decurrens subsp. symmetrifoliolata) cultivated ex situ under two plant arrangements, with or without semi-decomposed chicken manure. This study was carried out under field conditions at the Medicinal Plant Garden (HPM), Federal University of Grande Dourados (UFGD), in Distroferric Red Latossol. Arrangements with simple and double rows and the use or not of semi-decomposed chicken manure as covering were studied as 2 x 2 factorial arrangement in a randomized block experimental design, with six replicates. The highest maximum heights (61.3 cm plant-1) at 555 days after transplant were obtained by plants cultivated under double rows with chicken manure. The mean numbers of leaves and branches per plant during the cycle were eleven and two, respectively. The mean stem diameter of plants that were cultivated in simple rows was 8.3 mm, while that of plants cultivated in double rows was 10.8 mm, both at 552 days of the cycle. The evaluated characteristics, in the harvest at 555 days after transplant, were not influenced by the interaction between plant arrangements and use of chicken manure or by factors in an isolated way. Thus, cultivation in double rows was favorable to "carobinha" yield for allowing the highest yield per area.

The aim of this trial was to evaluate the initial development of Campomanesia adamantium grown under different phosphorus and nitrogen rates. The experiment was conducted in a greenhouse at the Universidade Federal da Grande Dourados, Dourados-MS. It was studied five phosphorus rates (20, 120, 200, 280 and 380 kg ha-1) in the form of superphosphate and five nitrogen rates (6, 36, 60, 84 and 114 kg ha-1) in the form of ammonium sulfate. Treatments were defined using the Plan Puebla experimental matrix, resulting in nine combinations, respectively, of phosphorus and nitrogen (kg ha-1): 280 and 84, 280 and 36, 120 and 6, 120 and 36, 120 and 84, 200 and 60, 380 and 84, 20 and 36 and 280 and 114. The experimental design was randomized block with four replications. Experimental unit consisted of five vessels, with one plant per pot. The trial harvest was carried out 270 days after transplanting (DAT). Highest rates of N and P resulted on the greatest plant height (38.12 cm) reached after 261 DAT, maximum number of leaves per plant (54) at 186 DAT, highest dry weight of leaves (13.99 and 5.68 g plant-1) and root (15.9 and 6.2 g plant-1), greatest leaf area (610 cm² plant-1) and number of branches (3 planta-1), all at 270 DAT. The average of Chlorophyll levels was 36 ICF. Thus, it's recommend the supply of high doses of phosphorus (380 kg ha-1) and nitrogen (114 kg ha-1) for guavira cultivation.

RESUMEN

BACKGROUND: The morphology, motor responses and spatiotemporal organization among colonic propagating sequences (PS) have never been defined throughout the entire colon of patients with slow transit constipation (STC). Utilizing the technique of spatiotemporal mapping, we aimed to demonstrate 'manometric signatures' that may serve as biomarkers of the disorder. METHODS: In 14 female patients with scintigraphically confirmed STC, and eight healthy female controls, a silicone catheter with 16 recording sites spanning the colon at 7.5 cm intervals was positioned colonoscopically with the tip clipped to the cecum. Intraluminal pressures were recorded for 24 h. KEY RESULTS: Pan-colonic, 24 h, spatiotemporal mapping identified for the first time in STC patients: a marked paucity of propagating pressure waves in the midcolon (P = 0.01), as a consequence of a significant (P < 0.0001) decrease in extent of propagation of PS originating in the proximal colon; an increase in frequency of retrograde PS in the proximal colon; a significant reduction in the spatiotemporal organization among PS (P < 0.001); absence of the normal nocturnal suppression of PS. CONCLUSIONS & INFERENCES: Pancolonic, 24 h, spatiotemporal pressure mapping readily identifies characteristic disorganization among consecutive PS, regions of diminished activity and absent or deficient fundamental motor patterns and responses to physiological stimuli. These features are all likely to be important in the pathophysiology of slow transit constipation.
