RESUMEN

Cancer remains a leading cause of mortality and poses a substantial threat to public health. Studies have revealed that Long noncoding RNA DANCR is a cytoplasmic lncRNA whose aberrant expression plays a pivotal role in various cancer types. Within tumour biology, DANCR exerts regulatory control over crucial processes such as proliferation, invasion, metastasis, angiogenesis, inflammatory responses, cellular energy metabolism reprogramming, and apoptosis. By acting as a competitive endogenous RNA for miRNAs and by interacting with proteins and mRNAs at the molecular level, DANCR contributes significantly to cancer progression. Elevated DANCR levels have also been linked to heightened resistance to anticancer drugs. Moreover, the detection of circulating DANCR holds promise as a valuable biomarker for aiding in the clinical differentiation of different cancer types. This article offers a comprehensive review and elucidation of the primary functions and molecular mechanisms through which DANCR influences tumours.

OBJECTIVE: To investigate the protective effect of Xuebijing injection on acute lung injury (ALI) associated with cardiopulmonary bypass (CPB) by regulating the apoptosis of polymorphonuclear neutrophils (PMN). METHODS: Thirty male Sprague-Dawley (SD) rats were randomly divided into sham operation group (Sham group), CPB model group (CPB group) and Xuebijing pretreatment group (XBJ group) according to the random number table method, with 10 rats in each group. Rats in the CPB group and XBJ group undergoing CPB procedures for 60 minutes. Rats in the Sham group did not undergo CPB. Rats in the XBJ group received intraperitoneal injection of 4 mL/kg Xuebijing injection 2 hours before CPB. Rats in the Sham group and CPB group were injected with an equal amount of normal saline. 4 hours after CPB, arterial blood was collected for blood gas analysis to calculate respiratory index (RI), and lung tissue of rats was collected for determination of lung index (LI) and pulmonary water containing rate. PMN in bronchoalveolar lavage fluid (BALF) were collected and the activity of caspase-3 was detected. The apoptosis rate was detected by flow cytometry. The expressions of microRNA-142-3p (miR-142-3p) and FoxO1 mRNA were detected by real-time fluorescence quantitative polymerase chain reaction (RT-qPCR). The protein expression of FoxO1 was detected by Western blotting. In addition, HL-60 cells were divided into control oligonucleotide transfection group, miR-142-3p mimics transfection group, and miR-142-3p inhibitor transfection group. After 48 hours of transfection, the activity of miR-142-3p binding to FoxO1 was detected using dual luciferase reporter genes. RESULTS: Compared with Sham group, RI, LI and pulmonary water containing rate were significantly increased in CPB group. The caspase-3 activity and apoptosis rate of PMN obtained from BALF were significantly decreased, the expression of miR-142-3p was decreased, and the expression of FoxO1 protein was increased. However, compared with CPB group, RI, LI and pulmonary water containing rate were significantly decreased in XBJ group [RI: 0.281±0.066 vs. 0.379±0.071, LI: 4.50±0.26 vs. 5.71±0.42, pulmonary water containing rate: (80.31±32.50)% vs. (84.59±3.41)%, all P < 0.01]. The caspase-3 activity and apoptosis rate of PMN obtained from BALF were significantly increased [caspase-3 activity: 0.350±0.021 vs. 0.210±0.014, apoptosis rate: (15.490±1.382)% vs. (8.700±0.701)%, both P < 0.01], the expression of miR-142-3p was significantly up-regulated (2-ΔΔCt: 2.61±0.17 vs. 0.62±0.05, P < 0.01), and the protein expression of FoxO1 was decreased [FoxO1/GAPDH (relative expression level): 0.81±0.04 vs. 1.22±0.06, P < 0.01]. However, there was no statistically significant difference in FoxO1 mRNA expression among the three groups. The bioinformatics analysis results showed that miR-142-3p can bind to the FoxO1 3'untranslated region (3'UTR). In HL-60 cells, compared with control oligonucleotide transfection group, the transfection of miR-142-3p mimics could reduce the expression of FoxO1 protein [FoxO1/GAPDH (relative expression level): 0.48±0.06 vs. 1.00±0.05, P < 0.01], however, the transfection of miR-142-3p inhibitor increased the expression of FoxO1 protein [FoxO1/GAPDH (relative expression level): 1.37±0.21 vs. 1.00±0.05, P < 0.05]. But, transfection with miR-142-3p mimics or inhibitor had no effect on FoxO1 mRNA expression. The luciferase reporter gene showed that miR-142-3p could bind to the FoxO1 3'UTR to inhibit FoxO1 expression. CONCLUSIONS: Xuebijing injection may promote the apoptosis of pulmonary alveolar PMN through the miR-142-3p/FoxO1 axis, and play a role in the prevention and treatment of CPB-induced ALI.

The development of cost-effective non-noble metal electrocatalysts for the oxygen reduction reaction (ORR) and oxygen evolution reaction (OER) opens up the possibility for sustainable energy systems. Herein, we report a surface overcoating strategy with lanthanum organic complex (La-OC) as the precursor to prepare lanthanum species (La-SPc) encapsulated in nitrogen, fluorine, and sulfur self-doped porous carbon (NFS-PC) composites (La-SPc@NFS-PC) for efficient ORR and OER. The La-SPc is introduced not only as a promoter to increase the electrochemical stability of the La-SPc@NFS-PC catalysts but also to tailor the electronic structure of NFS-PC due to the unique electrochemical properties of La-SPc. In addition, the integration of La-SPc and NFS-PC can improve the electronic conductivity of composites by inducing electron redistribution and lowering the band gap, which is advantageous in enhancing the kinetics of charge transfer. Simultaneously, benefiting from the optimized porous structure and positive cooperation of La-SPc with NFS-PC shells, the obtained La-SPc@NFS-PC-3 delivers robust bifunctional ORR/OER activities and stabilities. More importantly, the Zn-air battery (ZAB) assembled with La-SPc@NFS-PC-3 demonstrates an outstanding power density (181.1 mW cm-2) and long cycling life, outperforming the commercial Pt/C. This work offers a rational approach to preparing high-efficiency rare-earth-based catalysts and provides potential applications in ZABs.

Background: Tuberculosis (TB) remains a significant challenge for public health and is closely associated with malnutrition; however, few studies have attempted to screen malnutrition among TB patients. The study aimed to evaluate the nutrition status and build a new nutritional screening model for active TB. Methods: A retrospective, multicenter, large cross-sectional study was conducted in China from 1 January 2020 to 31 December 2021. All included patients diagnosed with active pulmonary TB (PTB) were evaluated both by Nutrition Risk Screening 2002 (NRS 2002) and Global Leadership Initiative on Malnutrition (GLIM) criteria. Univariate and multivariate analyses were conducted to screen the risk factors associated with malnutrition, and a new screening risk model, mainly for TB patients, was constructed. Results: A total of 14,941 cases meeting the inclusion criteria were entered into the final analysis. The malnutrition risk rate among PTB patients in China was 55.86% and 42.70%, according to the NRS 2002 and GLIM, respectively. The inconsistency rate between the two methods was 24.77%. A total of 11 clinical factors, including elderly, low body mass index (BMI), decreased lymphocyte cells, taking immunosuppressive agents, co-pleural TB, diabetes mellitus (DM), human immunodeficiency virus (HIV), severe pneumonia, decreased food intake within a week, weight loss and dialysis were identified as independent risk factors of malnutrition based on multivariate analyses. A new nutritional risk screening model was constructed for TB patients with a diagnostic sensitivity of 97.6% and specificity of 93.1%. Conclusions: Active TB patients have severe malnutrition status according to screening by the NRS 2002 and GLIM criteria. The new screening model is recommended for PTB patients as it is more closely tailored to the characteristics of TB.

Mutual phase coherence is the utmost crucial factor in an asynchronous optical sampling system. The enhancement is commonly achieved by phase-locking dual femtosecond lasers to a shared narrow-linewidth cavity-stabilized laser. Here we report such a system with a low residual optical phase jitter at a level of 0.04 rad in a Fourier frequency band from 1 Hz to 5 MHz around 1.55 µm, corresponding to a timing jitter of 30 as. The residual phase jitter reaches 1 rad at a Fourier frequency between 0.06 Hz and 0.1 Hz, from which the phase-coherence time is inferred to be at least 10 s. The multi-heterodyne beats experimentally reveal a hardware-limited phase coherence time of ∼8.155 s throughout the direct lasing spectral band.

Broadband, coherent narrow-linewidth optical frequency synthesis is of crucial importance in dual-comb interferometric measurement. Here we present a detailed description of the construction and performance characterization of a hertz-level linewidth coherent optical frequency synthesizer across the optical telecommunication band. A narrow-linewidth cavity-stabilized laser at 1565.00 nm is built and coherently transferred through a fiber link with an additional fractional frequency instability of 2.0×10-16 at 1 s averaging time. Broadband, coherent optical frequency synthesis is then achieved by steering one mode of a laser frequency comb with the transferred optical frequency oscillation. By beating with a 1542.14 nm ultra-stable cavity-stabilized laser, the evaluated fractional frequency stability and absolute linewidth of the nearest synthesized optical oscillation are 3.5×10-15 at 1 s averaging time and 1.8 Hz, respectively. According to the ultra-low-noise feature of the utilized laser frequency comb of 4.7×10-17 at 1 s averaging time, the synthesized optical frequency oscillations could maintain the high coherence across the comb's output bandwidth.

In this work, 3-Phenyl-N-[4-(10,15,20-triphenyl-porphyrin-5-yl)-phenyl]- acrylamide (TPPCA) was synthesized for the determination of metronidazole (MTZ). It was found that the type of fluorescence quenching was static quenching determined by Stern-Volmer plot and UV absorption spectroscopy, and thermodynamic related parameters were also obtained. Furthermore, the corresponding measurement conditions: the acidity of the system, the type of surfactant, the concentration of TPPCA and the sequence of reagent addition were optimized. Under the optimal experimental conditions, the linear range of MTZ was determined to be 0.01-0.20 µg mL-1, and the limit of detection (LOD) was 0.004 µg mL-1. Importantly, this report provides a simple, fast, and sensitive probe for the determination of MTZ in pharmaceutical practice.

Red blood cells (RBCs) possess intact cyto-architectures while haemoglobin (Hb) is a cell-free, homogeneous solution. Both RBCs and Hb are generalized oxygen carriers. In this paper, kinetic studies on oxygen-releasing of high concentration of Hb and RBCs under various conditions were carried out regarding Hb and RBCs as fluids. Among them, Hb under specific conditions was seen as the simplest Hb-based oxygen carrier (HBOC), Also, factors affecting the oxygen releasing of Hb and RBCs, including osmotic pressure, viscosity and allosteric agent, have been well studied. Analysis of the results from the measurement above showed that kinetics of oxygen releasing of either pure Hb or the simplest HBOCs was obviously different from that of RBCs. The oxygen-releasing time of Hb was shorter and the oxygen-releasing rates of Hb were quicker than those of RBCs under various conditions. Therefore, as fluids, only by changing the milieus it exists in, Hb could not achieve the expected oxygen-releasing effect on the microcirculation so well as RBCs do in the same system, irrespective of the interaction between the fluids and blood vessels. Furthermore, kinetic properties of HBOCs must be considered and matched with those of RBCs in the study of HBOCs.

Dual-comb system with simplified integration acts a crucial role in the next stage of field-deployable application. Here we demonstrate a home-made dual-comb spectrometer that meets this requirement based on optically stabilized erbium-fiber frequency combs. The simplified integration and operability are highlighted by the embedded large motion-span automatic optical delay line that resulted in 1.7 MHz of continuous repetition rate tuning range and avoid multiple fiber cutbacks. This relaxes the second similar frequency comb duplication, therefore simplifies dual-comb system integration. The mutual coherence was enhanced by optical phase lock to a common narrow-linewidth reference laser via pump current modulation. We acquired the absorption feature of acetylene 12C2H2 molecule in 1.5 µm region with an apodized instrument linewidth of 0.076 cm-1. The mean signal-to-noise ratio within a spectral bandwidth of 90 cm-1 achieved 280, and the corresponding noise equivalent absorption coefficient was 3.7 × 10-6 cm-1·Hz-1/2 for an absorption path of 7.5 cm and a pressure of 100 Torr. The accuracy of the retrieved spectrum was further confirmed by comparing with HITRAN database.

The chemokine (C-X3-C motif) ligand 1 (CX3CL1), also named fractalkine (FKN), has been implicated in psychiatric disorders and functions as a novel adipocytokine. However, no attention has been paid to the role of FKN in anorexia nervosa (AN). The current study was performed to explore FKN levels in AN to determine its role in the involvement of AN. A total of 96 girls aged 11-18 years with AN (n=34), healthy controls (HC; n=32) and simple obesity (OB, n=30) were enrolled in the cross-sectional study. Blood samples were collected during the fasting state. Serum FKN concentrations were determined using ELISA. The skinfold thickness (TSF) of the biceps and triceps as well as mid-arm muscle circumference (MAMC) were used to determine the nutritional status. Our results showed that serum FKN levels were significantly lower in the AN group than in the control and OB groups. After adjusting for body mass index (BMI), FKN concentrations in the AN group were statistically higher than in the HC and OB groups. Significant correlations between serum FKN and body weight, BMI, Cole index and serum insulin were observed. In addition, serum FKN levels were positively related to TSF and MAMC in all subjects. Serum FKN concentrations are attenuated in girls with AN compared with healthy adolescents and are positively related to nutritional status. The lower FKN levels may be regulated by nutrition status and response to starvation. After adjusting for BMI, higher FKN levels may reflect that persistent inflammation is present in patients with AN.

UNLABELLED: The SH2B1 adaptor protein is recruited to multiple ligand-activated receptor tyrosine kinases that play important role in the physiologic and pathologic features of many cancers. The purpose of this study was to assess SH2B1 expression and to explore its contribution to the non-small cell lung cancer (NSCLC). METHODS: SH2B1 expression in 114 primary NSCLC tissue specimens was analyzed by immunohistochemistry and correlated with clinicopathological parameters and patients' outcome. Additionally, 15 paired NSCLC background tissues, 5 NSCLC cell lines and a normal HBE cell line were evaluated for SH2B1 expression by RT-PCR and immunoblotting, immunofluorescence being applied for the cell lines. RESULTS: SH2B1 was found to be overexpressed in NSCLC tissues and NSCLC cell lines. More importantly, high SH2B1 expression was significantly associated with tumor grade, tumor size, clinical stage, lymph node metastasis, and recurrence respectively. Survival analysis demonstrated that patients with high SH2B1 expression had both poorer disease- free survival and overall survival than other patients. Multivariate Cox regression analysis revealed that SH2B1 overexpression was an independent prognostic factor for patients with NSCLC. CONCLUSIONS: Our findings suggest that the SH2B1 protein may contribute to the malignant progression of NSCLC and could offer a novel prognostic indicator for patients with NSCLC.
