RESUMEN

Shenling Baizhu San(SLBZS) is a commonly used medicine for the treatment of ulcerative colitis(UC). This study aims to explore the mechanism of SLBZS in treating UC by using colonic metabolomics and network pharmacology. BALB/c mice were randomly divided into four groups: a blank group, a model group, an SLBZS group, and a sulfasalazine group. UPLC-Q-TOF-MS/MS technology was utilized to analyze the metabolic profiles of colonic tissue in mice, and differential metabolites and related metabolic pathways were screened. Based on the online database, active ingredients, action targets, and UC disease targets of SLBZS were screened. The protein-protein interaction(PPI) network of core targets of SLBZS in treating UC was constructed using STRING and Cytoscape 3.9.1. Gene Ontology(GO) functional and Kyoto Encyclopedia of Genes and Genomes(KEGG) pathway enrichment analyses were performed using the DAVID database. A "metabolite-reaction-enzyme-gene" network was constructed to conduct a combined analysis of metabolomics and network pharmacology. SLBZS reversed the levels of 25 metabolites involved in various pathways such as D-glutamine and D-glutamate metabolism, caffeine metabolism, sphingolipid metabolism, arginine biosynthesis, lysine degradation, alanine, aspartate, and glutamate metabolism, glycerophospholipid metabolism, and pyrimidine metabolism in UC colonic tissue. 47 core targets of SLBZS in treating UC were involved in pathways including the MAPK signaling pathway, TNF signaling pathway, Toll-like receptor signaling pathway, lipid and atherosclerosis, inflammatory bowel disease, and Th17 cell differentiation. Integrated analysis showed that glycerophospholipid metabolism and pyrimidine metabolism were key metabolic pathways in the treatment of UC with SLBZS. The results suggested that SLBZS improved colonic mucosal morphology by regulating colonic metabolites, down-regulated the expression of inflammation-related core target genes to reduce inflammation levels, and alleviated lipid metabolism disorders, thereby exerting a therapeutic effect on UC.

BACKGROUND: The advanced first-line regimen for advanced gastric cancer is based on a combination of fluoropyrimidine and platinum and/or paclitaxel (PTX), forming a two- or three-drug regimen. Compared to conventional PTX, nanoparticle albumin-bound PTX (Nab-PTX) has better therapeutic effects and fewer adverse effects reported in studies. Nab-PTX is a great option for patients presenting with advanced gastric cancer. Herein, we highlight an adverse event (hemorrhagic cystitis) of Nab-PTX in advanced gastric cancer. CASE SUMMARY: A 55-year-old male was diagnosed with lymph node metastasis after a laparoscopic-assisted radical gastrectomy for gastric cancer that was treated by Nab-PTX and S-1 (AS). On the 15th day after treatment with AS, he was diagnosed with hemorrhagic cystitis. CONCLUSION: Physicians should be aware that hemorrhagic cystitis is a potential adverse event associated with Nab-PTX treatment.

An acid-promoted cyclization of α-azidobenzyl ketones has been developed for the synthesis of 6-substituted quinoline derivatives. A variety of synthetically useful 6-OTf or -OMs quinoline derivatives were obtained in moderate to good yields. The reaction proceeds via C═N bond formation without organophosphine, providing convenient access to structurally interesting and synthetically important 6-substituted quinoline derivatives in moderate to good yields. A mechanistic perspective that is different from the traditional intramolecular Schmidt reaction has been proposed.

OBJECTIVES: Based on peripheral blood lymphocyte subsets and common laboratory test indexes, this study aimed to construct a predictive scoring system for intravenous immunoglobulin (IVIG)-resistant Kawasaki disease (KD). METHODS: Children hospitalized in Tianjin Children's Hospital from January 2021 to March 2023 were included in the study (185 cases of IVIG-sensitive KD and 41 cases of IVIG -resistant KD). Forty-six healthy children matched for age and gender were selected as controls. The relative percentage and absolute counts of peripheral lymphocyte subsets were measured by flow cytometry. Multivariate logistic regression was used to identify the predictive factors for IVIG-resistant KD and to construct a predictive scoring system for predicting IVIG-resistant KD. RESULTS: The multivariate logistic regression analysis showed that CD4+ T cell absolute count, natural killer cell absolute count, serum sodium level, globulin level, and total bilirubin level were identified as predictive factors for IVIG-resistant KD (P<0.05). The predictive scoring system based on these factors achieved a sensitivity of 70.7% and a specificity of 83.8% in predicting IVIG-resistant KD. CONCLUSIONS: Peripheral blood lymphocyte subsets can serve as predictive indicators for IVIG-resistant KD in children. The introduction of this indicator and the establishment of a scoring system based on it can provide a higher accuracy in predicting IVIG-resistant KD in children.

To explore the change features of PM2.5-bound metals in a background site of North China in the past ten years, 71 and 160 samples were collected from December 2011 to January 2013 (period Ⅰ) and from September 2019 to November 2021 (period Ⅱ) in Tuoji Island National Atmospheric Monitoring Station, respectively.The concentration of metals sampled was determined using ICP-MS, and the concentrations, sources, and health risks of heavy metals were compared. The results revealed that the average concentration of PM2.5 was (54.06±39.71) µg·m-3during period Ⅱ, which was 3.53 ng·m-3 lower than that during period Ⅰ. The concentrations of Zn, Mn, As, Pb, and V in stage Ⅱ decreased by 54.53, 172.63, 0.8, 79.06, and 3.81 ng·m-3, respectively, whereas the concentrations of Cr, Cu, Cd, and Ni increased by 2.01, 5.42, 3.03, and 3.55 ng·m-3, respectively. The PMF model results indicated that the biggest contributor to PM2.5-bound metal was industrial emissions (32.32%), followed by coal combustion (27.47%), vehicle emissions (23.70%), ship emissions (9.69%), and dust sources (6.83%) during period Ⅱ. The contribution ratio of dust sources and ship emissions decreased by 20.73% and 8.83%, respectively, whereas for coal combustion and industrial emissions it increased by 2.50% and 13.52%, respectively, when compared with that during period Ⅰ. The total carcinogenic risk induced by PM2.5-bound heavy metals of period Ⅱ increased, with the highest contributions by Cr and Cd. The total non-carcinogenic risk decreased, with Mn contributing the most. Therefore, in the process of air pollution control, the control of pollution sources of heavy metals such as Cr and Mn should be reinforced.

AIM: To access the agreement of intraocular pressure (IOP) values obtained from biomechanically corrected tonometer [Corvis ST (CST)], non-contact tonometer (NCT), and Goldmann applanation tonometer (GAT) in children with NCT measured-IOP (NCT-IOP) values of 22 mm Hg or more, and related factors. METHODS: A total of 51 eyes with NCT-IOP≥22 mm Hg in children aged 7 to 14y were examined and IOP was measured by CST, NCT, and GAT. Based on GAT measured IOP (GAT-IOP), ocular hypertension (OHT) group (≥22 mm Hg, 24 eyes) and the non-OHT group (<22 mm Hg, 27 eyes) were defined. We compared the agreement of the three measurements, i.e., CST measured IOP (CST-IOP), GAT-IOP, and NCT-IOP, and further analyzed the correlation between the differences in tonometry readings, central corneal thickness (CCT), axial length (AL), optic disc rim volume, and age. RESULTS: Compared with the OHT group, thicker CCT, larger rim volume, and higher differences between NCT-IOP and GAT-IOP, were found in the non-OHT group. The differences between CST-IOP and GAT-IOP were lower than the differences between NCT-IOP and GAT-IOP in both groups. The mean differences in CST-IOP and GAT-IOP were 1.26 mm Hg (95% limit of agreement ranged from 0.1 to 2.41 mm Hg, OHT group) and 1.20 mm Hg (95% limit of agreement ranged from -0.5 to 3.00 mm Hg, non-OHT group), and the mean differences in NCT and GAT were 3.90 mm Hg (95% limit of agreement ranged from -0.19 to 9.70 mm Hg, OHT group) and 6.00 mm Hg (95% limit of agreement ranged from 1.50 to 10.50 mm Hg, non-OHT group). The differences between CST-IOP and GAT-IOP were not related to CCT, age, and AL in both groups; while the differences between NCT-IOP and GAT-IOP were related to CCT in the OHT group (r=0.93, P<0.001) and to CCT and AL in the non-OHT group (r=0.66, P<0.001, r=-0.81, P<0.001). CONCLUSION: The accuracy of NCT in the diagnosis of pediatric OHT is low. The agreement of CST-IOP and GAT-IOP was significantly higher in children with and without OHT than in those with NCT-IOP and GAT-IOP. Therefore, CST can be used as a good alternative for IOP measurement in children. The impacts of CCT and AL on NCT measurement need to be fully considered when managing childhood IOP.

End-functionalization is an effective strategy for constructing functional materials. A method for chain-end functionalization of helical polycarbenes is herein developed that relied on Sonogashira coupling reaction. In this work, a family of helical polycarbenes with controlled molecular mass (Mn ) and low polydispersity (Mw /Mn ) is readily prepared using Pd(II) and the Wei-Phos ligand as initiator. The Pd(II) complex is confirmed to remain at the chain end of polycarbene. Subsequently, a series of terminal alkyne derivatives with interesting functional groups, including the F atom, aldehyde, or anthracene groups, are synthesized. They could be installed at the chain end of polycarbene through Sonogashira coupling reaction catalyzed by the Pd(II) complex at the chain end. Moreover, a couple of hybrid block copolymers are easily obtained by installing terminal alkynes modified by another type of polymer. The structures of the isolated polymers are confirmed by 1 H nuclear magnetic resonance (1 H NMR), 19 F nuclear magnetic resonance (19 F NMR), 31 P nuclear magnetic resonance (31 P NMR), and Fourier transform infrared spectroscopy (FT-IR), respectively. The self-assembly properties of the hybrid block copolymers are also investigated by atomic force spectroscopy analysis. By the hereby developed method, various functional groups can be introduced at the chain end of helical polycarbenes for constructing functional polymer materials, moreover, the transition metal residues at the end of polymer chains can be easily removed.

RESUMEN

PURPOSE: Thyroid cancers with unsatisfactory curative effect nowadays are the most common malignant tumors of the endocrine system. Apoptosis evasion, a hallmark of cancer, has driven the search of stimulating novel cell death way in cancer therapy. This review aims to explore the relationship between autophagy and thyroid cancer, especially the chemotherapy agents which are based on autophagy in treating thyroid cancers. METHODS: A computerized literature search of MEDLINE was performed using the following search terms: autophagy and thyroid cancer. RESULTS: Recent studies have found that several chemotherapeutic agents and knockdown of specific microRNA may contribute to autophagic tumor cell death in most thyroid cancer types. CONCLUSIONS: Stimulating autophagy may be an effective alternative treatment to most types of thyroid cancer.
