RESUMEN

OBJECTIVE: To investigate the anti-oxidative effect of ethyl pyruvate (EP) and taurine (TAU) on the quality of red blood cells stored at 4±2 ℃, hemolysis, energy metabolism and lipid peroxidation of the red blood cells in the preservation solution were studied at different intervals. METHODS: At 4±2 ℃, the deleukocyte red blood cells were stored in the citrate-phosphate-dextrosesaline-adenine-1 (CPDA-1) preservation (control group), preservation solution with EP (EP-AS), and TAU (TAU-AS) for long-term preservation. The enzyme-linked immunoassay and automatic blood cell analyzer were used to detect hemolysis and erythrocyte parameters. Adenine nucleoside triphosphate (ATP), glycerol 2,3-diphosphate (2,3-DPG) and malondialdehyde (MDA) kits were used to test the ATP, 2,3-DPG and MDA concentration. RESULTS: During the preservation, the rate of red blood cell hemolysis in EP-AS and TAU-AS groups were significantly lower than that in CPDA-1 group (P<0.01). The MCV of EP-AS group was increased with the preservation time (r=0.71), while the MCV of the TAU-AS group was significantly lower than that in the other two groups (P<0.05). The concentration of ATP and MDA in EP-AS and TAU-AS groups were significantly higher than that in CPDA-1 group at the 14th day (P<0.01). The concentrations of 2,3-DPG in the EP-AS and TAU-AS groups were significantly higher than that in the CPDA-1 group from the 7th day (P<0.01). CONCLUSION: EP and TAU can significantly reduce the red blood cell hemolysis rate, inhibit the lipid peroxidation level of red blood cells, and improve the energy metabolism of red blood cells during storage. The mechanism of EP and TAU may be related to their antioxidation and membrane protection effect, so as to improve the red blood cell quality and extend the preservation time.

End-stage liver disease (ESLD) is characterized by the deterioration of liver function and a subsequent high mortality rate. Studies have investigated the use of adult stem cells to treat ESLD. Here, a systematic review and meta-analysis was conducted to determine the efficacy of a combination therapy with adult stem cell transplantation and traditional medicine for treating ESLD. Four databases-including PubMed, Web of Science, Embase, and Cochrane Library-were investigated for studies published before January 31, 2021. The main outcome indicators were liver function index, model for end-stage liver disease (MELD) scores, and Child‒Turcotte‒Pugh (CTP) scores. Altogether, 1604 articles were retrieved, of which eight met the eligibility criteria; these studies included data for 579 patients with ESLD. Combination of adult stem cell transplantation with conventional medicine significantly improved its efficacy with respect to liver function index, CTP and MELD scores, but this effect gradually decreased over time. Moreover, a single injection of stem cells was more effective than two injections with respect to MELD and CTP scores and total bilirubin (TBIL) and albumin (ALB) levels, with no significant difference in aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels. With respect to the TBIL levels, patients receiving mononuclear cells (MNCs) experienced a significantly greater therapeutic effect-starting from twenty-four weeks after the treatment-whereas with respect to ALB levels, CD34+ autologous peripheral blood stem cells (CD34+ APBSCs) and MNCs had similar therapeutic effects. Severe complications associated with adult stem cell treatment were not observed. Although the benefits of combination therapy with respect to improving liver function were slightly better than those of the traditional treatment alone, they gradually decreased over time.Systematic review registration: PROSPERO registration number: CRD42021238576.

Five unique Keggin-type polyoxometalate (POM)-based metal-organic frameworks (POMFs), namely [CH2L1]2[(CuL12)(PMoVI9MoV3O40)] (1), [Zn0.5(H2O)(L1)][Zn(L1)1.5Cl][H2L1]0.5[PMo12O40]·1.25H2O (2), (TBA)[Cu(H2O)2L12][PMo12O40] (3), [Cu2(H2O)2(L1)3][PMVI11MoVO40] (4), (H2L2)0.5[(CuIL2)2(PMo12O40)]·H2O (5), L1 = 4,4'-bis((1H-1,2,4-triazol-1-yl)methyl)-biphenyl, L2 = 1,4-bis((1H-1,2,4-triazol-1-yl)methyl)-benzene, have been synthesized under ionothermal conditions. According to single-crystal data, 1 exhibits a novel mechanically interlocked molecular architecture (MIMA) constructed by two-dimensional (2D) interpenetrating polyrotaxane layers with unique cyclophanes (tetra-cationic viologen macrocycle cyclobis(paraquat- p-phenylene) (CBPQT4+ system)), resulting in an H-bonded 3D supermolecule, and is the first synthesis of self-assembled cyclophane-PMOFs under ionothermal conditions. 2 shows a novel 2D three-fold interpenetrating polyrotaxane host and guest network. 1 and 2 are presented as the first MIMA polyrotaxane structures to have been synthesized under ionothermal conditions. Electrochemical impedance spectroscopy (EIS) reveals that 2, 4, and 5 show high proton conductivity owing to their unique structural properties. Solid-state diffuse reflection UV-vis-NIR measurements show the title compounds are potentially semiconductor materials. Photocatalytic investigations indicate that 1-5 possess high and stable photocatalytic H2 evolution and rhodamine B (RhB) degradation under visible-light irradiation.

Herein, we report a novel thermal/photoresponsive shape-memory polyurethane network with a pendant azobenzene group by utilizing its anisotropic-isotropic phase transitions and photoresponsive feature concurrently. To achieve this goal, the side-chain liquid crystalline polyurethane networks based on the pendant azobenzene group [SCLCPU(AZO)-Ns] were developed in a well-defined architecture. The smectic C nature of an LC phase in the polyurethane networks was confirmed by differential scanning calorimetry, polarized optical microscopy, and one-dimensional and two-dimensional wide-angle X-ray diffraction. The well-defined architecture-made SCLCPU(AZO)-N displays two distinct transition temperatures (Ttrans) (Tg and Tcl), with a difference of about 40 °C. Consequently, the excellent triple-shape-memory effect in this network was demonstrated by cyclic thermomechanical analysis. By making full use of the trans-cis photoisomerization of azobenzene, the reversible bending and unbending behaviors were realized under the light irradiation with wavelengths of 450 and 550 nm, respectively.

OBJECTIVE: To observe the changes in the expressions of somatomedins in the prostate epithelial cells in anoxic condition. METHODS: We cultured prostate epithelial cell line RWPE-1 in vitro. At 4, 8, 12, 24, 48 hours after seeding of the cells, we determined the gene and protein expressions of the epidermal growth factor (EGF), basic fibroblast growth factor (bFGF), transforming growth factor-beta (TGF-beta), insulin-like growth factor-1 (IGF-1) and vascular endothelial growth factor (VEGF) in the prostate epithelial cells by RT-PCR and ELISA, respectively. RESULTS: With the increase of time, the expressions of the EGF, bFGF, TGF-beta, IGF-1 and VEGF genes were obviously up-regulated, more significantly in the anoxic than in the normoxic prostate epithelial cells. Take FGF mRNA, its expression level was 0.14 +/- 0.01 in the anoxic and 0. 12 +/- 0.01 in the normoxic prostate epithelial cells at 8 hours (P = 0.01), but increased to 0.29 +/- 0.01 and 0.14 +/- 0.01, respectively, at 48 hours (P < 0.001). The expression of the TGF-beta protein was also more significantly increased in the anoxic than in the normoxic prostate epithelial cells, 0.32 +/- 0.01 versus 0.26 +/- 0.01 at 4 hours (P = 0.017) and 1.56 +/- 0.13 versus 0.87 +/- 0.06 at 48 hours (P < 0.001). The other 4 somatomedins showed no significant differences in their protein expressions between anoxic and normoxic conditions. CONCLUSION: Anoxia can up-regulate the gene expressions of somatomedins and increase the secretion of TGF-beta in prostate epithelial cells.

Production of recombinant human-like collagen (RHLC) by thermoinduction of recombinant Escherichia coli BL 21 during high cell density cultivation was investigated in a 30 L bioreactor. The effects of induction temperature (T), pH, and carbon-to-nitrogen molar ratio of the nutrient medium (C/N) were examined. The optimal thermoinduction protocol for RHLC production was determined by using a model coupling genetic algorithm and artificial neural networks. The optimal operating conditions were as follows: maintenance of induction temperature at 42°C for 3 H and then at 39.4°C until the end, induction pH at 7.03, and C/N at 4.8 (mol/mol). The theoretical maximum concentration of RHLC was 12.5 g/L, whereas the experimental value was 12.1 g/L under the optimal induction conditions.
