RESUMEN

This work is devoted to the description of the synthesis of hydrogels in the process of cryotropic gel formation based on copolymerization of synthesized potassium 3-sulfopropyl methacrylate and 2-hydroxyethyl methacrylate (SPMA-co-HEMA) and assessing the potential possibility of their use as substrates for growing plants in intensive light culture in a greenhouse. Gel substrates based on the SPMA-co-HEMA were created in two compositions, differing from each other in the presence of macro- and microelements, and their effects were studied on the plants' physiological state (content of chlorophylls a and b, activity of catalase and peroxidase enzymes, intensity of lipid peroxidation, elemental compositions) at the vegetative period of their development and on the plants' growth, productivity and quality of plant production at the final stages of development. Experiments were carried out under controlled microclimate conditions. Modern and standard generally accepted methods of gels were employed (ATR-FTIR and 13C NMR spectral studies, scanning electron microscopy, measurement of specific surface area and pore volume), as well as the methods of the physiological and chemical analysis of plants. The study demonstrated the swelling ability of the created gel substrates. Hydrogels' structure, their specific surface area, porosity, and pore volume were investigated. Using the example of representatives of leaf, fruit and root vegetable crops, the high biological activity of gel substrates was revealed throughout the vegetation period. Species specificity in the reaction of plants to the presence of gel substrates in the root-inhabited environment was revealed. Lettuce, tomato and cucumber plants were more responsive to the effect of the gel substrate, and radish plants were less responsive. At the same time, more pronounced positive changes in plant growth, quality and productivity were observed in cucumber and lettuce in the variant of gel substrates with macro- and microelements and in tomato plants in both variants of gel substrates. Further research into the mechanisms of the influence of gel substrates on plants, as well as the synthesis of new gel substrates with more pronounced properties to sorb and retain moisture is promising.

OBJECTIVE: Temporal lobe epilepsy (TLE) is a network disorder that alters the total organization of the language-related network. Task-based functional magnetic resonance imaging (fMRI) aimed at functional connectivity is a direct method to investigate how the network is reorganized. However, such studies are scarce and represented mostly by the resting-state analysis of the individual connections between regions. To fill this gap, we used a graph-based analysis, which allows us to cover the total language-related network changes, such as disruptions in an integration/segregation balance, during a language task in TLE. METHODS: We collected task-based fMRI data with sentence completion from 19 healthy controls and 28 people with left TLE. Using graph-based analysis, we estimated how the language-related network segregated into modules and tested whether they differed between groups. We evaluated the total network integration and the integration within modules. To assess intermodular integration, we considered the number and location of connector hubs-regions with high connectivity. RESULTS: The language-related network was differently segregated during language processing in the groups. While healthy controls showed a module consisting of left perisylvian regions, people with TLE exhibited a bilateral module formed by the anterior language-related areas and a module in the left temporal lobe, reflecting hyperconnectivity within the epileptic focus. As a consequence of this reorganization, there was a statistical tendency that the dominance of the intramodular integration over the total network integration was greater in TLE, which predicted language performance. The increase in the number of connector hubs in the right hemisphere, in turn, was compensatory in TLE. SIGNIFICANCE: Our study provides insights into the reorganization of the language-related network in TLE, revealing specific network changes in segregation and integration. It confirms reduced global connectivity and compensation across the healthy hemisphere, commonly observed in epilepsy. These findings advance the understanding of the network-based reorganizational processes underlying language processing in TLE.

This study aimed to obtain a recombinant chimeric protein named trx-NsW2 via theheterologous expression of the multifunctional antimicrobial peptide nigellothionin from black cumin (Nigella sativa L.) seeds in the Escherichia coli system. The protein was purified using a combination of Ni-NTA affinity chromatography and reversed-phase HPLC. Based on the HPLC calibration, the total yield of the protein was calculated to be 650 mg/L of bacterial culture. The fungistatic activity of trx-NsW2 against the food-spoiling fungus Aspergillus niger was demonstrated as itinhibited the maturation of conidiawithout affecting conidial germination or fungal growth. In contrast to mature nigellothionin NsW2, the fusion protein showeda low level of cytotoxicity towards both normal and tumor cell lines at concentrationsof up to 100-200 µM. Interestingly, at lower concentrations, it even stimulated cytokinesis. These findings are of critical importance for applying chimeric antimicrobial proteins obtained via microbiological synthesis in applied science.

The patterns of change in bioclimatic conditions determine the vegetation cover and soil properties along the altitudinal gradient. Together, these factors control the spatial variability of soil respiration (RS) in mountainous areas. The underlying mechanisms, which are poorly understood, shape the resulting surface CO2 flux in these ecosystems. We aimed to investigate the spatial variability of RS and its drivers on the northeastern slope of the Northwest Caucasus Mountains, Russia (1,260-2,480 m a.s.l.), in mixed, fir, and deciduous forests, as well as subalpine and alpine meadows. RS was measured simultaneously in each ecosystem at 12 randomly distributed points using the closed static chamber technique. After the measurements, topsoil samples (0-10 cm) were collected under each chamber (n = 60). Several soil physicochemical, microbial, and vegetation indices were assessed as potential drivers of RS. We tested two hypotheses: (i) the spatial variability of RS is higher in forests than in grasslands; and (ii) the spatial variability of RS in forests is mainly due to soil microbial activity, whereas in grasslands, it is mainly due to vegetation characteristics. Unexpectedly, RS variability was lower in forests than in grasslands, ranging from 1.3-6.5 versus 3.4-12.7 µmol CO2 m-1 s-1, respectively. Spatial variability of RS in forests was related to microbial functioning through chitinase activity (50% explained variance), whereas in grasslands it was related to vegetation structure, namely graminoid abundance (27% explained variance). Apparently, the chitinase dependence of RS variability in forests may be related to soil N limitation. This was confirmed by low N content and high C:N ratio compared to grassland soils. The greater sensitivity of grassland RS to vegetation structure may be related to the essential root C allocation for some grasses. Thus, the first hypothesis concerning the higher spatial variability of RS in forests than in grasslands was not confirmed, whereas the second hypothesis concerning the crucial role of soil microorganisms in forests and vegetation in grasslands as drivers of RS spatial variability was confirmed.

Restoring the anatomical and functional characteristics of the cornea using various biomaterials is especially relevant in the context of a global shortage of donor tissue. Such biomaterials must be biocompatible, strong, and transparent. Here, we report a Viscoll collagen membrane with mechanical and optical properties suitable for replacing damaged stromal tissue. After removing a portion of the stroma, a Viscoll collagen membrane was implanted into the corneas of rabbits. After 6 months, the active migration of host cells into Viscoll collagen membranes was noted, with the preservation of corneal transparency in all experimental animals. Effective integration of the Viscoll collagen membrane with corneal tissue promoted nerve regeneration in vivo, as confirmed by in vivo confocal microscopy. We also demonstrated the safety and efficacy of the Viscoll collagen membrane for corneal stroma regeneration. Thus, in combination with the proposed packaging format that provides long-term storage of up to 10 months, this material has great potential for replacing and regenerating damaged stromal tissues.

Rising air temperatures caused by global warming affects microbial decomposition rate of soil organic matter (SOM). The temperature sensitivity of SOM decomposition (Q10) may depend on SOM quality determined by vegetation type. In this study, we selected a long transect (3.6 km) across the five ecosystems and short transects (0.1 km) from grazed and ungrazed meadows to forests in the Northwest Caucasus to consider different patterns in Q10 changes at shift of the vegetation belts. It is hypothesized that Q10 will increase along altitudinal gradient in line with recalcitrance of SOM according to kinetics-based theory. The indicators of SOM quality (BR:C, respiration per unit of soil C; MBC:C, ratio of microbial biomass carbon to soil carbon; soil C:N ratio) were used for checking the hypothesis. It was shown that Q10 did not differ across vegetation types within long and short transects, regardless differences in projective cover (14-99%) and vegetation species richness (6-12 units per plot). However, Q10 value differed between the long and short transects by almost two times (on average 2.4 vs. 1.4). Such a difference was explained by environmental characteristics linked with terrain position (slope steepness, microclimate, and land forms). The Q10 changes across studied slopes were driven by BR:C for meadows (R2 = 0.64; negative relationship) and pH value for forests (R2 = 0.80; positive relationship). Thus, proxy of SOM quality explained Q10 variability only across mountain meadows, whereas for forests, soil acidity was the main driver of microbial activity.

To avoid post-neurosurgical language deficits, intraoperative mapping of the language function in the brain can be complemented with preoperative mapping with functional magnetic resonance imaging (fMRI). The validity of an fMRI "language localizer" paradigm crucially depends on the choice of an optimal language task and baseline condition. This study presents a new fMRI "language localizer" in Russian using overt sentence completion, a task that comprehensively engages the language function by involving both production and comprehension at the word and sentence level. The paradigm was validated in 18 neurologically healthy volunteers who participated in two scanning sessions, for estimating test-retest reliability. For the first time, two baseline conditions for the sentence completion task were compared. At the group level, the paradigm significantly activated both anterior and posterior language-related regions. Individual-level analysis showed that activation was elicited most consistently in the inferior frontal regions, followed by posterior temporal regions and the angular gyrus. Test-retest reliability of activation location, as measured by Dice coefficients, was moderate and thus comparable to previous studies. Test-retest reliability was higher in the frontal than temporo-parietal region and with the most liberal statistical thresholding compared to two more conservative thresholding methods. Lateralization indices were expectedly left-hemispheric, with greater lateralization in the frontal than temporo-parietal region, and showed moderate test-retest reliability. Finally, the pseudoword baseline elicited more extensive and more reliable activation, although the syllable baseline appears more feasible for future clinical use. Overall, the study demonstrated the validity and reliability of the sentence completion task for mapping the language function in the brain. The paradigm needs further validation in a clinical sample of neurosurgical patients. Additionally, the study contributes to general evidence on test-retest reliability of fMRI.

The iron-oxidizing system of a moderately thermophilic, extremely acidophilic, gram-positive mixotroph, Sulfobacillus sibiricus N1(T), was studied by spectroscopic, high-performance liquid chromatography and inhibitory analyses. Hemes B, A, and O were detected in membranes of S. sibiricus N1(T). It is proposed that the electron transport chain from Fe²(+) to O2 is terminated by 2 physiological oxidases: aa3-type cytochrome, which dominates in the early-exponential phase of growth, and bo3-type cytochrome, whose role in iron oxidation becomes more prominent upon growth of the culture. Both oxidases were sensitive to cyanide and azide. Cytochrome aa3 was more sensitive to cyanide and azide, with K(i) values of 4.1 and 2.5 µmol·L⁻¹, respectively, compared with K(i) values for cytochrome bo3, which were 9.5 µmol·L⁻¹ for cyanide and 7.0 µmol·L⁻¹ for azide. This is the first evidence for the participation of a bo3-type oxidase in ferrous iron oxidation. The respiratory chain of the mixotroph contains, in addition to the 2 terminal oxidases, a membrane-bound cytochrome b573.
