RESUMO

OBJECTIVE: To evaluate the effect of near infra-red gallium-aluminium-arsenide (GaAlAs) diode laser (805 nm) irradiation on proliferation and differentiation of rat femoral bone marrow-derived mesenchymal stem cells (BMSCs) cultured in osteogenic medium. MATERIALS AND METHODS: BMSCs were obtained from femurs of 60 Sprague Dawley rats (200 gm). The control group comprised isolated BMSCs supplemented with an osteogenic differentiation medium. On the other hand, in the experimental group, the BMSCs were irradiated with a near-infrared laser in addition to an osteogenic differentiation medium. The experimental group was irradiated with a soft tissue laser comprising of allium-aluminium-arsenic (Ga-Al-Ar) Diode at a near-infrared wavelength of 805 nm in continuous mode. The different output powers applied were 0.5 W, 1.0 W, 1.5 W and 2.0 W respectively. Various energy levels of 1, 4, 7 and 10 J were used for irradiation. Alkaline phosphatase (ALP) assay and Alizarin staining were performed to confirm osteogenic differentiation. Statistical analysis was done using a one-way ANOVA and a p-value of <0.05 was considered significant. RESULTS: According to our findings, 1.27 J/cm2 was the optimal energy density value that significantly increased the BMSC proliferation at the output of 1.5 W with the power density of 1.27 W/cm2. On 1.27 J/cm2, there was a significant difference compared to the control group on the first day, and the osteogenic differentiation increased significantly on the 4th day compared to the 1st day. CONCLUSIONS: According to our findings, 1.27 J/cm2 was the optimal energy density value that significantly increased the BMSC proliferation at the output of 1.5 W with the power density of 1.27 W/cm2.

In order to determine the prevalence of fetal mortality and investigate hypothesized risk factors associated with its occurrence in goats, 962 female goats were studied using ultrasound. To diagnose pregnancy and to identify dead fetuses, ultrasound scanning was conducted using real-time machines equipped with a transabdominal curvilinear probe. A questionnaire was supplied for collection of signalment and sampling data. Ultrasound examination revealed that out of 962 female goats, 431 (44.8%) goats were diagnosed as nonpregnant, 88 (9.14%) were pseudopregnant, 4 (0.42%) were diagnosed as having pyometra, and 439 were diagnosed as pregnant (45.63%). Of the 439 pregnant goats, 36 were diagnosed as bearing dead fetuses (8.2%). Season of the year, locality, breed of the dam, age of the dam, parity number, breed of the buck, and feeding type were all found not to be significantly associated with fetal death. It is concluded that ultrasound is a reliable method for diagnosis of fetal death and documenting the prevalence of its occurrence in goats.

Thirteen Saanen and Saanen crossbred female goats, between the ages of 6 months and 7, years were presented to the clinic, College of Veterinary Medicine, Sudan University of Science and Technology, for sonographic pregnancy diagnosis. Transabdominal ultrasound was performed using 3.5 MHz probe which revealed non-viable fetuses as judged by absence of heart beats and movements. Twelve goats were given single i/m injection of PGF2α analogue and 5% oxytetracycline. Ten goats responded to the treatment and six of them became pregnant and gave birth within the normal gestational period. One goat was diagnosed as non-pregnant, one goat developed hydrometra, and the subsequent fertility of two goats was unknown. Two full-term goats did not respond to treatment. Another dose of PGF2α was administered to them and again they did not respond. Manual attempts were done to deliver the full-term goat with dilated cervix and they were unsuccessful. Cesarean section and hysterectomy were then performed for the three full-term goats with unfavorable outcome. It can be concluded that ultrasound is a rapid, reliable, and nonhazardous procedure for the diagnosis of fetal mortality in goats and PGF2α treatment in conjunction with oxytetracycline is an efficient treatment.
