RESUMO

The present study is focused on the determination of low-volatile chemosignals excreted or secreted by mouse pups in their early days of life involved in maternal care induction in mice adult females. Untargeted metabolomics was employed to differentiate between samples collected with swabs from facial and anogenital area from neonatal mouse pups receiving maternal care (first two weeks of life) and the elder mouse pups in the weaning period (4th week old). The sample extracts were analysed by ultra-high pressure liquid chromatography (UHPLC) coupled to ion mobility separation (IMS) in combination with high resolution mass spectrometry (HRMS). After data processing with Progenesis QI and multivariate statistical analysis, five markers present in the first two weeks of mouse pups life and putatively involved in materno-filial chemical communication were tentatively identified: arginine, urocanic acid, erythro-sphingosine (d17:1), sphingosine (d18:1) and sphinganine. The four-dimensional data and the tools associated to the additional structural descriptor obtained by IMS separation were of great help in the compound identification. The results demonstrated the great potential of UHPLC-IMS-HRMS based untargeted metabolomics to identity putative pheromones in mammals.

Spain is a secondary centre of the diversification of the melon (Cucumis melo L.), with high diversity represented in highly appreciated landraces belonging to the Flexuosus and Ibericus groups. A collection of 47 accessions of Flexuosus, Chate, Piel de Sapo, Tendral, Amarillo, Blanco, and Rochet was analysed using a genotyping-by-sequencing (GBS) approach. A total of 66,971 quality SNPs were identified. Genetic analysis differentiated Ibericus accessions and exotic materials (Ameri, Momordica, Kachri, and Agrestis), while Flexuous accessions shared ancestry between them. Within the Ibericus group, no clear genomic distinction could be identified for the different landraces evaluated, with accessions of different landraces showing high genetic similarity. The morphological characterization confirmed that the external colour and fruit shape had been used as recognition patterns for Spanish melon landraces, but variability within a landrace exists. Differences were found in the sugars and acid and volatile profiles of the materials. Flexuosus and Chate melons at the immature commercial stage accumulated malic acid and low levels of hexoses, while Ibericus melons accumulated high contents of sucrose and citric acid. Specific trends could be identified in the Ibericus landraces. Tendral accumulated low levels of sugars and citric acid and high of malic acid, maintaining higher firmness, Rochet reached higher levels of sugars, and Amarillo tended to lower malic acid contents. Interestingly, high variability was found within landraces for the acidic profile, offering possibilities to alter taste tinges. The main volatile organic compounds (VOCs) in Flexuosus and Chate were aldehydes and alcohols, with clear differences between both groups. In the Ibericus landraces, general trends for VOC accumulation could be identified, but, again, a high level of variation exists. This situation highlights the necessity to develop depuration programs to promote on-farm in situ conservation and, at the same time, offers opportunities to establish new breeding program targets and to take advantage of these sources of variation.

Very long-chain (> C24) polyunsaturated fatty acids (VLC-PUFA) play an important role in the development of nervous system, retinal function and reproductive processes in vertebrates. Their presence in very small amounts in specific lipid classes, the lack of reference standards and their late elution in chromatographic analyses render their identification and, most important, their quantification, still a challenge. Consequently, a sensitive and feasible analytical methodology is needed. In this work, we have studied the effect of chain length, as well as the number and position of unsaturations (or double bonds) on the response of GC-APCI-(Q)TOF MS, to establish an analytical method for VLC-PUFA quantification. The developed methodology allows the quantification of these compounds down to 2.5 × 10-3 pmol/mg lipid. The reduction of VLC-PUFA levels in lipid fractions of the organs from the herein sampled farmed fish suggesting a yet undetected effect on these compounds of high vegetable oil aquafeed formulations, that currently dominate the market.

In this study, we identify 11 mouse pup volatiles putatively involved in maternal care induction in adult females. For this purpose, we have adapted the dynamic headspace methodology to extract the volatolome of whole alive animals. Untargeted metabolomic methodology was used to compare the volatolome of neonatal (4-6 days) with elder pups until the age of weaning (21-23 days old). Pup volatolome was analyzed by gas chromatography (GC) coupled to single quadrupole mass spectrometry (MS) using automated thermal desorption for sample introduction. After data processing and multivariate statistical analysis, comparison with NIST spectral library allowed identifying compounds secreted preferentially by neonatal pups: di(propylen glycol) methyl ether, 4-nonenal, di(ethylene glycol) monobutyl ether, 2-phenoxyethanol, isomethyl ionone, tridecanal, 1,3-diethylbenzene, 1,2,4,5-tetramethylbenzene, 2-ethyl-p-xylene and tri(propylene glycol) methyl ether. Palmitic acid was enriched in the volatolome of fourth week youngsters compared to neonatal pups. The results demonstrated the great potential of the new sampling procedure combined with GC-MS based untargeted volatolomics to identify volatile pheromones in mammals.

BACKGROUND: To date, no study has used bibliometric analysis to review the most influential articles in lumbar spinal stenosis. The objective of this study was to identify and analyze the characteristics and the level of evidence of the 100 most cited articles on lumbar spinal stenosis METHODS: The Thomson Reuters Web of Science was accessed to find the 100 most cited articles on lumbar spinal stenosis. For each article, we recorded the number and density of citations, authors, country, journals and years, department, level of evidence, type of study, and if it was part of any multicenter studies. RESULTS: Until January 2017, the 100 most cited articles accumulated 11,136 citations (average: 259.05/y), ranging individually between 442 and 50 (average: 111.36). The first reference was published in 1974 in Clinical Orthopaedics and Related Research. Therapeutic studies (n = 40), the 1990s (n = 46), United States as country of origin (n = 51), Harvard University as institution (n = 16), Katz JN as author (n = 10), and Spine as journal (n = 48) have the hegemony. Many were multicenter (n = 42) and using level 2 evidence (n = 49). There is an inverse relationship between citation index and long-standing studies, maintenance of those most cited, and a temporary advance toward better levels of evidence. CONCLUSION: This bibliometric analysis reveals a good level of evidence in the published clinical series and includes 100 articles useful for the approach of lumbar spinal stenosis.

The characterization of very long-chain (>C24) polyunsaturated fatty acids (VLC-PUFAs), which are essential in the vision, neural function, and reproduction of vertebrates, is challenging because of the lack of reference standards and their very low concentrations in certain lipid classes. In this research, we have developed a new methodology for VLC-PUFA identification based on gas chromatography coupled to quadrupole/time-of-flight mass spectrometry with an atmospheric pressure chemical ionization source (GC-APCI-QTOF MS). The mass accuracy attainable with the innovative QTOF instrument, together with the soft ionization of the APCI source, provides valuable information on the intact molecule, traditionally lost with electron ionization sources due to the extensive fragmentation suffered. We have identified, for the first time, VLC-PUFAs with chains up to 44 carbons in eyes, brain, and gonads of gilthead sea bream, a commercially important fish in the Mediterranean. The added value of ion mobility-mass spectrometry (IMS), recently developed in combination with GC-QTOF MS, and the contribution of the collisional cross section (CCS) parameter in the characterization of novel VLC-PUFAs (for which reference standards are not available) have been also evaluated. The methodology developed has allowed assessing qualitative differences between farmed and wild fish, and opens new perspectives in a still scarcely known field of research.

BACKGROUND: Musculoskeletal Diseases (MSDs) are among the most prevalent health problems encountered in the workforce in Europe. Multiple risk factors contribute to their onset. In the present study, different individual risk factors for chronic tendinous pathology affecting the shoulder were analysed in a sample of workers from the automotive manufacturing sector. METHODS: An observational retrospective study was conducted with 73 cases of officially recognised and compensated occupational diseases and 94 aleatory cases of healthy workers from the same car assembly company. The experimental group comprised individuals with tendinous chronic pathology of the rotator cuff. Multiple variables that identified the risks present in the job were assessed along with participants clinical evaluation. Furthermore, two standardised guidelines for risk factors assessment were also used: the Spanish National Institute of Social Security (INSS) and the American Occupational Information Network (O*Net). Both descriptive statistical analysis and Odds ratios calculations considering the occupational disease as a dependent variable were performed. RESULTS: The use of hand tools, exposure to mechanical pressure in the upper limbs and awkward postures were the most prevalent risk factors. Pressure on the palm of the hand and the hand tool impacting the hand were also important risk factors. Some psychosocial factors such as lack of autonomy and mental workload were also associated shoulder tendinous diseases. The association of age, load handling, and awkward postures were the core risk factors responsible for most of the tendinous chronic injuries of the shoulder in this sample of car assembly workers. CONCLUSIONS: Both ergonomic and psychosocial factors were present and increased the risk of developing occupational chronic tendinopathies at the shoulder in this sample of workers. Aging, load handling, and awkward postures showed the strongest predictive values. Greater knowledge of how risk factors interact would facilitate the design of better preventive workplace strategies.

STUDY DESIGN: Bibliometric review of current literature. OBJECTIVE: To identify and analyze the characteristics of the 100 most cited articles on idiopathic scoliosis focusing on the level of evidence. SUMMARY OF BACKGROUND DATA: The scientific literature on idiopathic scoliosis has been constantly evolving, but many aspects of its true etiology, natural history, and response to treatment continue to be discussed. To date, no study has used bibliometric analysis to review the most influential articles about idiopathic scoliosis. METHODS: The Thompson Reuters Web of Science was accessed to find the 100 most cited articles on idiopathic scoliosis. The number and citation density, authorship, institutions, country of origin, year of publication, source journals, type of study, topic, study design, and level of evidence were analyzed. RESULTS: The 100 most frequently cited articles accumulated 13,749 citations. The number of citations ranged between 616 and 80. The 10 most cited articles represent 24.6% of all citations. The treatment of idiopathic scoliosis is the most commonly studied issue (n = 46), and specifically surgical correction (n = 36). Most studies originated in the United States (n = 62) and were published in Spine (n = 56). Almost half of the papers (n = 49) were published between 2000 and 2008. The majority of studies have a case series design (n = 35). Most of the cited articles have low levels of evidence (Level III = 36; Level IV = 35). CONCLUSION: This bibliometric analysis includes the 100 most cited articles on idiopathic scoliosis, recognizing its importance as a basic milestone in today's spine knowledge. The results indicate that the evolution of the knowledge on idiopathic scoliosis has been through case reports and case series, which analyzed retrospectively today are considered to have a poor level of evidence. This observation seems to be paradoxical because they are the most influential articles on IS and had been published in the top, leading journals of spine surgery.

A sensitive method for the simultaneous quantification of dechloranes, polybrominated diphenyl ethers (PBDEs) and novel brominated flame retardants (NBFRs) has been developed for gas chromatography (GC) coupled to tandem mass spectrometry operating in electron capture negative ionization (ECNI) mode. The major advance has been achieved by combining selected ion monitoring (SIM) and multiple reaction monitoring (MRM) modes in well-defined time windows, to determine dechloranes, PBDEs and NBFRs at picogram per gram level in one single analysis in complex matrix biological samples. From the chromatographic point of view, efforts were devoted to study several injection modes using multimode inlet (MMI) in order to obtain low instrumental detection limits, necessary for trace compounds such as Dechlorane Plus (DP) isomers. Method performance was also evaluated: calibration curves were linear from 20 fg µL-1 to 100 pg µL-1 for the studied compounds, with method detection limits at levels of 50 fg g-1 for DPs. Repeatability and reproducibility, expressed as relative standard deviation, were better than 5% even in solvent vent mode for the injection of standards. The application to a wide range of complex samples (including food, human and animal serum samples) indicated a sensitive and reliable way to quantify at the picogram per gram level 4 halogenated norbornenes (HNs), Dechlorane Plus (anti-DP and syn-DP) and 2 of their homologues (Dechlorane-602 and Dechlorane-603), 11 PBDE congeners (no. 28, 47, 49, 66, 85, 99, 100, 153, 154, 183 and 209) and 5 novel BFRs, i.e. decabromodiphenyl ethane (DBDPE), 1,2-bis(2,4,6-tribromophenoxy)ethane (BTBPE), hexabromobenzene (HBB), 2,3,4,5-tetrabromo-ethylhexyl-benzoate (TBB) and tetrabromophthalate (TBPH). Acetaldehyde, the main biological metabolite of ethanol, is nowadays considered to mediate some ethanol-induced effects. Previous studies on alcohol effect attenuation have shown that D-(-)-penicillamine (3-mercapto-D-valine), a thiol amino acid, acts as an effective agent for the inactivation of acetaldehyde. In the study reported here, laboratory rats were treated with ethanol and D-(-)-penicillamine at different doses looking for the interaction (in vivo) of D-(-)-penicillamine with metabolically formed acetaldehyde following a condensation reaction to form the stable adduct (4S,2RS)-2,5,5-trimethylthiazolidine-4-carboxylic acid (TMTCA). A novel and rapid procedure based on liquid chromatography coupled to tandem mass spectrometry (LC/MS/MS) was developed for quantification and reliable identification of TMTCA in different rat tissues, including plasma, liver and brain. Firstly, plasma was obtained from whole blood. Then, proteins were precipitated from plasma, brain and liver extracts with acetonitrile and the clarified extracts diluted 10-fold. A 20 microL aliquot of the final extracts was then analyzed using an Atlantis C18 5 microm, 100x2 mm column which was connected to the electrospray source of a LC/triple quadrupole mass spectrometer. The analyte was detected in positive ion mode acquiring four MS/MS transitions in selected reaction monitoring (SRM) mode. The method has been validated and it has proved to be fast, reliable and sensitive. The accuracy and precision were evaluated by means of recovery experiments from plasma, liver and brain samples fortified at two concentration levels obtaining satisfactory recoveries in all cases: 95 and 105% in plasma (at 10 and 100 ng/mL, respectively), 79 and 89% in brain (100 and 1000 ng/g), 85 and 99% in liver (100 and 1000 ng/g). Precision, expressed as repeatability, was in all tissues analyzed lower than 17% at the two concentrations tested. The estimated detection limits were 1 ng/mL in plasma, 4 ng/g in brain and 5 ng/g in liver. The limit of quantitation objective (the lowest concentration that was validated with acceptable results) was set up at 10 ng/mL for plasma and 100 ng/g for brain and liver tissue. The reliable identification of the analyte was ensured by the acquisition of four transitions and by their ion abundance ratio measurement. Due to its excellent selectivity and sensitivity, the method developed in this work provides an excellent tool for the specific determination of this cyclic amino acid in biological samples.

A rapid, sensitive and selective method has been developed for the direct determination of ethephon residues in vegetables (apple, cherry and tomato). Given the anionic character of ethephon, the use of ion-pairing liquid chromatography (LC) in combination with tandem mass spectrometry (MS/MS, triple quadrupole) allowed its direct determination in these matrices avoiding a derivatisation step and favouring the automation of the method. Samples were extracted with a mixture of dichloromethane/aqueous formic acid (pH 3) (1:1). Then, tetrabutylammonium acetate (TBA) was added as an ion-pairing reagent, and an aliquot of the aqueous extract was directly injected into the LC/MS/MS system. Quantification was performed with matrix-matched standards prepared from blank sample extracts. MS/MS measurements were made in the selected reaction monitoring (SRM) mode, using the most sensitive transition (m/z 107 > 79) for quantification, and up to four additional transitions for confirmation. Quantitative recoveries were obtained for all matrices (between 83% and 96%) at two concentration levels tested (0.05 and 0.5 mg/kg), with relative standard deviations lower than 9% in all cases. The addition of TBA directly into the sample extract contained in the injection vial was found sufficient to obtain satisfactory LC retention for the analyte. Under these conditions, the absence of ion-pairing reagent in the mobile phase minimised the ionisation suppression for ethephon in the MS source, leading to an increase in the sensitivity of the method and reaching limits of detection of 0.02 mg/kg for all matrices investigated. The acquisition of five specific MS/MS transitions for ethephon allowed the simultaneous and reliable quantification and confirmation of the analyte in the samples.

The potential of gas chromatography coupled to tandem mass spectrometry (GC/MS/MS) with a triple quadrupole analyzer (QqQ) has been investigated for the accurate and sensitive determination of xenoestrogens in human breast tissues. Special emphasis has been given to the confirmation of the identity of compounds detected in the samples analyzed in order to avoid the reporting of false positives. The work has been focused on the determination of approximately 30 organochlorine compounds (PCBs and pesticides) and organobromine compounds (polybrominated diphenyl ethers) in adipose breast tissue and in tumoral fragment. Analytes were extracted by dissolving the samples in hexane, and the extracts were purified by automated normal-phase HPLC prior to GC/MS/MS analysis. Three isotopically labeled standards were added before extraction as surrogates for the quality control of the analyses. Accuracy and precision were evaluated by means of recovery experiments using adipose breast tissue spiked at three concentration levels, with satisfactory results for most analytes. The excellent selectivity and sensitivity of QqQ in selected reaction monitoring mode allowed us satisfactory quantification and confirmation at levels as low as 5-25 ng/g, i.e., the lowest concentration level for which the method was fully validated. Two MS/MS transitions were selected for each analyte, using the concentration ratio obtained from them as a confirmatory parameter. The developed methodology was applied to the analysis of 51 breast samples (26 adipose tissues and 25 tumoral fragments), giving as a result the detection and confirmation of several organochlorine compounds in both types of samples. Due to its adequate analytical characteristics, the optimized method fits with the requirements of accurate quantification and reliable confirmation of the identity of compounds detected according to the most recent European Guidelines. As an ultimate unequivocal confirmation, several selected samples were reanalyzed by gas chromatography coupled to mass spectrometry with a time-of-flight (TOF) analyzer. Confirmation of analytes present at higher concentrations was successful with mass error less than 5 mDa. However, confirmation by TOF MS was not possible al low concentrations (i.e., at the few ng/g level) as a consequence of its lower sensitivity compared with that of triple quadrupole in selected reaction monitoring mode.
