RESUMO

Increasing soil organic carbon (SOC) confers benefits to soil health, biodiversity, underpins carbon sequestration and ameliorates land degradation. One recommendation is to increase SOC such that the SOC to clay ratio (SOC/clay) exceeds 1/13, yet normalising SOC levels based on clay alone gives misleading indications of soil structure and the potential to store additional carbon. Building on work by Poeplau & Don (2023) to benchmark observed against predicted SOC, we advance an alternative indicator: the ratio between observed and "typical" SOC (O/T SOC) for pan-European application. Here, "typical" SOC is the average concentration in different pedo-climate zones, PCZs (which, unlike existing SOC indicators, incorporate land cover and climate, alongside soil texture) across Europe, determined from mineral (<20 % organic matter) topsoils (0-20 cm) sampled during 2009-2018 in LUCAS, Europe's largest soil monitoring scheme (n = 19,855). Regression tree modelling derived 12 PCZs, with typical SOC values ranging 5.99-39.65 g kg-1. New index classes for comparison with SOC/clay grades were established from the quartiles of each PCZ's O/T SOC distribution; these were termed: "Low" (below the 25th percentile), "Intermediate" (between the 25th and 50th percentiles), "High" (between the 50th and 75th percentiles), and "Very high" (above the 75th percentile). Compared with SOC/clay, O/T SOC was less sensitive to clay content, land cover, and climate, less geographically skewed, and better reflected differences in soil porosity and SOC stock, supporting 2 EU Soil Health Mission objectives (consolidating SOC stocks; improving soil structure for crops and biota). These patterns held for 2 independent datasets, and O/T SOC grades were sensitive enough to reflect land management differences across several long-term field experiments. O/T SOC used in conjunction with several other physical, chemical and biological soil health indicators can help support the EU Soil Monitoring Law and achieve several United Nations Sustainable Development Goals.

Land degradation directly affects around 25% of land globally, undermining progress on most of the UN Sustainable Development Goals (SDG), particularly target 15.3. To assess land degradation, SDG indicator 15.3.1 combines sub-indicators of productivity, soil carbon and land cover. Over 100 countries have set Land Degradation Neutrality (LDN) targets. Here, we demonstrate application of the indicator for a well-established agricultural landscape using the case study of Great Britain. We explore detection of degradation in such landscapes by: 1) transparently evaluating land cover transitions; 2) comparing assessments using global and national data; 3) identifying misleading trends; and 4) including extra sub-indicators for additional forms of degradation. Our results demonstrate significant impacts on the indicator both from the land cover transition evaluation and choice or availability of data. Critically, we identify a misleading improvement trend due to a trade-off between improvement detected by the productivity sub-indicator, and 30-year soil carbon loss trends in croplands (11% from 1978 to 2007). This carbon loss trend would not be identified without additional data from Countryside Survey (CS). Thus, without incorporating field survey data we risk overlooking the degradation of regulating and supporting ecosystem services (linked to soil carbon), in favour of signals from improving provisioning services (productivity sub-indicator). Relative importance of these services will vary between socioeconomic contexts. Including extra sub-indicators for erosion or critical load exceedance, as additional forms of degradation, produced a switch from net area improving (9%) to net area degraded (58%). CS data also identified additional degradation for soil health, including 44% arable soils exceeding bulk density thresholds and 35% of CS squares exceeding contamination thresholds for metals.

Efforts to improve soil health require that target values of key soil properties are established. No agreed targets exist but providing population data as benchmarks is a useful step to standardise soil health comparison between landscapes. We exploited nationally representative topsoil (0-15 cm) measurements to derive soil health benchmarks for managed and semi-natural environments across Great Britain. In total, 4587 soil organic matter (SOM), 3860 pH, 2908 bulk density (BD), and 465 earthworm abundance (EA) datapoints were used. As soil properties are sensitive to site-specific characteristics, data were stratified by habitat, soil type, and mean annual precipitation, with benchmarks defined as the middle 80 % of values in each distribution - yielding 135 benchmarks. BD and pH decreased with land management intensity (agriculture > semi-natural grasslands > woodlands > heathlands > wetlands), and vice versa for SOM and EA. Normalising benchmark ranges by medians revealed soil health indicator benchmark widths increased in the order: pH < BD < SOM < EA, while width increased with decreasing land management intensity. Arable and horticulture and improved grassland exhibited narrow benchmarks for SOM, pH and BD, yet the widest EA benchmark, suggesting additional drivers impact EA patterns. Upland wetlands had the widest BD benchmarks, important when determining carbon stocks. East Anglia currently possesses the largest proportions of atypical soils, including below typical SOM (19.2 %), above typical BD (17.4 %) and pH (39.1 %), and the smallest proportions of above typical SOM (2.4 %), and below typical BD (5.8 %) and pH (2.3 %). This is found even after land use, soil type and rainfall have been considered, underscoring how urgently soil health should be addressed here. Our benchmarking framework allows landowners to compare where their measured soil health indicators fall within expected ranges and is applicable to other biomes, national and multinational contexts.

The benefits of active music participation and training for cognitive development have been evidenced in multiple studies, with this link leveraged in music therapy approaches with clinical populations. Although music, rhythm, and movement activities are widely integrated into children's play and early education, few studies have systematically translated music therapy-based approaches to a nonclinical population to support early cognitive development. This study reports the follow-up effects of the Rhythm and Movement for Self Regulation (RAMSR) program delivered by generalist preschool teachers in low socioeconomic communities. This randomized control trial (RCT) involved 213 children across eight preschools in disadvantaged communities in Queensland, Australia. The intervention group received 16-20 sessions of RAMSR over 8 weeks, while the control group undertook usual preschool programs. Primary outcome measures included executive function (child assessment of shifting, working memory, and inhibition) and self-regulation (teacher report), with secondary outcomes of school readiness and visual-motor integration. Data were collected pre- and post-intervention, and again 6 months later once children had transitioned into school. Results demonstrated significant intervention effects across the three time points for school readiness (p = 0.038, ηp 2  = 0.09), self-regulation (p < 0.001, ηp 2  = 0.08), and inhibition (p = 0.002 ηp 2  = 0.23). Additionally, the feasibility of building capacity in teachers without any music background to successfully deliver the program was evidenced. These findings are important given that children from low socioeconomic backgrounds are more likely to need support for cognitive development yet have inequitable access to quality music and movement programs. RESEARCH HIGHLIGHTS: Initial effects of self-regulation from a rhythm and movement program were sustained following transition into school for children from disadvantaged backgrounds. Delayed effects of inhibition and school readiness from a rhythm and movement program appeared 6 months post-intervention as children entered school. Generalist teachers can successfully implement a rhythm and movement program, which boosts critical developmental cognitive skills.

Global land use change has resulted in more pasture and cropland, largely at the expense of woodlands, over the last 300 years. How this change affects soil hydraulic function with regard to feedbacks to the hydrological cycle is unclear for earth system modelling (ESM). Pedotransfer functions (PTFs) used to predict soil hydraulic conductivity (K) take no account of land use. Here, we synthesize >800 measurements from around the globe from sites that measured near-saturated soil hydraulic conductivity, or infiltration, at the soil surface, on the same soil type at each location, but with differing land use, woodland (W), grassland (G) and cropland (C). We found that texture based PTFs predict K reasonably well for cropland giving unbiased results, but increasingly underestimate K in grassland and woodland. In native woodland and grassland differences in K can usually be accounted for by differences in bulk density. However, heavy grazing K responses can be much lower indicating compaction likely reduces connectivity. We show that the K response ratios (RR) between land uses vary with cropland (C/W = 0.45 [W/C = 2.2]) and grassland (G/W = 0.63 [W/G = 1.6]) having about half the K of woodland.

This study contributes to understandings of early childhood self-regulation and executive function, and their components, through taking a person-centered approach to investigating how these skills cluster together in children aged 4-5years. A sample of children (N=206) from preschools in low socioeconomic communities were assessed through teacher report of self-regulation and three executive function tasks at the commencement of the preschool year. Outcome variables included teacher report of social skills and behavioral problems, and children's school readiness and visual motor integration skills were directly assessed. When the scores from this low-income sample were compared to available norms, over 70% of children scored below the 50th percentile in executive function measures, approximately 20% were below average in self-regulation skills, 48% were delayed in school readiness scores, 36% had above average levels of internalizing problems, and 25% were above average in externalizing problems. A series of four latent profile models each used different measurement approaches and combinations of self-regulation and executive function components. In three of the four models (two which combined self-regulation and executive function measures and one with teacher report of self-regulation only), a high skill and low skill profile were found with 31 to 42% of children in the low profile depending on the model. Children were very similarly classified across all three models. When three executive function scores were modeled alone, a more complex three-profile solution emerged (low, moderate, and high) with 52% in the low profile. Children identified in the low profiles across all models were at greater risk of poorer school readiness, visual motor integration and social skills, and increased behavioral problems. Taken together, the findings suggest that self-regulation and executive function skills tend to cluster together at this age and in this low-income sample. Composite scores of teacher report of self-regulation are somewhat sufficient in identifying children who also have poorer executive function skills and are at risk of poorer motor, social, and school readiness outcomes. These children are an important target group for additional supports prior to school entry.

Forest regeneration and expansion are occurring in many countries, with 80 million ha established from 2000 to 2012 under the Bonn accord and 17.5 million ha established from 1990 to 2005 according to the Food and Agriculture Organisation. Multiple reviews have linked increasing forest cover with reduced river flow and potentially detrimental effects downstream. Previous reviews have investigated trends in river flow response over time, but the influence of forest age remains uncertain. Partial river flow recovery (towards non-forested conditions) has been reported in decades following forest establishment, but the role of climate in driving these trends has not been explored. Here, we evaluate river flow trends in 43 studies following forest establishment, which provide sufficient information to distinguish the effects of ageing forests from variable climate. Our meta-analysis supports previous findings showing that forestation reduces annual river flow (by 23% after 5 years and 38% after 25 years) with greater reductions in catchments with higher mean annual precipitation, larger increases in forest cover, and which were idle, rather than agricultural land, prior to forestation. The impact of forests on river flow is sensitive to annual precipitation and potential evapotranspiration, but responses are highly variable. Forests affect river flow less when annual precipitation is low, and sensitivity to precipitation decreases as catchment aridity increases. The majority of catchments demonstrated persistent river flow declines after forest establishment. However, nine catchments showed partial flow recovery after an initial decrease, with peak flow reductions at an average age of 15 and across a range of tree species. The mean rate of recovery was 34 mm/year over 5 years. Partial flow recovery with forest age cannot be commonly expected, however, and forestation programmes should take into account that changes to annual river flow are likely to persist for up to five decades.

RESUMO

The need for large quantities of purified plasmid DNA has increased as the applications of DNA vaccines continue to expand. This chapter describes a simple, scaleable procedure based on the fed-batch cultivation of various Escherichia coli clones, which can be easily implemented and scaled-up to large bioreactors. Although some clones may require minor modifications to the feeding strategy, in general, this procedure, implemented as described, is likely to support the production of milligram to gram quantities of plasmid DNA.
