RESUMO

Background: Distributional data on planktonic, benthic and sympagic copepods collected in the framework of the XXXIVth Expeditions of the Italian National Antarctic Programme (PNRA) to the Ross Sea sector from 2018-2019 are here provided. These occurrences correspond to specimens collected from the 25 µm filters used in the desalination plant of the Italian research station "Mario Zucchelli" (MZS), located in the Terra Nova Bay area (TNB; Ross Sea, Antarctica). This dataset is a contribution to the Antarctic Biodiversity Portal, the thematic Antarctic node for both the Ocean Biogeographic Information System (AntOBIS) and the Global Biodiversity Information Facility Antarctic Biodiversity Information Facility (ANTABIF). The dataset was uploaded and integrated with the SCAR-AntOBIS database (the geospatial component of SCAR-MarBIN). Please follow the guidelines from the SCAR Data Policy (ISSN 1998-0337) when using the data. New information: We describe the diversity of marine copepods Terra Nova Bay sampled by the filters installed in the desalination unit (DU) of the Italian research station "Mario Zucchelli" described in recent work. The opening of the intake pipe of the DU is positioned at a depth of 4 m and allowed a total of 2,116 specimens to be sampled and recognised. In addition, new occurrence records of copepod genera and species are reported in the same zone. We provide an overview of the marine copepod diversity reported for TNB. The total of 2,116 individuals corresponds to 14 genera and 15 species and is represented by 136 occurrence records in this dataset. Around 52% of the total number of species are new records for the TNB area. The publication of this data paper was funded by the Belgian Science Policy Office (BELSPO, contract n°FR/36/AN1/AntaBIS) in the Framework of EU-Lifewatch as a contribution to the SCAR Antarctic biodiversity portal (biodiversity.aq).

Recent surveys of Antarctic waters in the Terra Nova Bay (Ross Sea) revealed numerous bryozoan species including ctenostome bryozoans. Whereas cheilostome bryozoans are well-studied in these latitudes, ctenostomes remain highly neglected. Large ctenostomes are easily recognized by their lack of calcified skeletons, but this lack also renders them difficult and tedious to identify. As a result, histology and reconstructions of internal soft tissues are required to classify this group of bryozoans. Thanks to the availability of new specimens from Terra Nova Bay, a detailed analysis of growth form, gut morphology and tentacle number of two colonies, initially ascribed to the ctenostome bryozoan genus Alcyonidum Lamouroux, 1813, turned out to be a new species, Alcyonidium kuklinskii sp. nov., which we described in this study. These specimens were also barcoded (COI) and sequences compared to available ones. Together with the new species described here, a total of ten species of Alcyonidium is now known for the Southern Ocean, accounting for one eighth of the entire genus diversity. All Southern Ocean species appear to be endemic. In order to speed the identification of the Antarctic Alcyonidium species, we provide an identification key and a distribution map of all type species. In brief, colony morphology, zooidal size and, in particular tentacle number represent the most suitable characters for identifying species within this genus. The Antarctic marine environment hosts diversified and highly endemic benthos owing to its unique geologic and climatic history. Current warming trends have increased the urgency of understanding Antarctic species history to predict how environmental changes will impact ecosystem functioning. Antarctic benthic lineages have traditionally been examined under three hypotheses: (1) high endemism and local radiation, (2) emergence of deep-sea taxa through thermohaline circulation, and (3) species migrations across the Polar Front. In this study, we investigated which hypotheses best describe benthic invertebrate origins by examining Antarctic scale worms (Polynoidae). We amassed 691 polynoid sequences from the Southern Ocean and neighboring areas: the Kerguelen and Tierra del Fuego (South America) archipelagos, the Indian Ocean, and waters around New Zealand. We performed phylogenetic reconstructions to identify lineages across geographic regions, aided by mitochondrial markers cytochrome c oxidase subunit I (Cox1) and 16S ribosomal RNA (16S). Additionally, we produced haplotype networks at the species scale to examine genetic diversity, biogeographic separations, and past demography. The Cox1 dataset provided the most illuminating insights into the evolution of polynoids, with a total of 36 lineages identified. Eunoe sp. was present at Tierra del Fuego and Kerguelen, in favor of the latter acting as a migration crossroads. Harmothoe fuligineum, widespread around the Antarctic continent, was also present but isolated at Kerguelen, possibly resulting from historical freeze-thaw cycles. The genus Polyeunoa appears to have diversified prior to colonizing the continent, leading to the co-occurrence of at least three cryptic species around the Southern and Indian Oceans. Analyses identified that nearly all populations are presently expanding following a bottleneck event, possibly caused by habitat reduction from the last glacial episodes. Findings support multiple origins for contemporary Antarctic polynoids, and some species investigated here provide information on ancestral scenarios of (re)colonization. First, it is apparent that species collected from the Antarctic continent are endemic, as the absence of closely related species in the Kerguelen and Tierra del Fuego datasets for most lineages argues in favor of Hypothesis 1 of local origin. Next, Eunoe sp. and H. fuligineum, however, support the possibility of Kerguelen and other sub-Antarctic islands acting as a crossroads for larvae of some species, in support of Hypothesis 3. Finally, the genus Polyeunoa, conversely, is found at depths greater than 150 m and may have a deep origin, in line with Hypothesis 2. These "non endemic" groups, nevertheless, have a distribution that is either north or south of the Antarctic Polar Front, indicating that there is still a barrier to dispersal, even in the deep sea.

RESUMO

The temporal dynamics of coastal planktic communities can be disclosed through DNA metabarcoding on the filters of reverse-osmosis desalination plants. Here, we describe the steps that are necessary to process the filters in order to create the subsamples used for DNA extraction and the bioinformatic pipeline to perform the first exploratory analyses on this kind of dataset.

RESUMO

DNA barcoding is a powerful and widespread method used to identify large numbers of species collected in the framework of sampling activities in the field. With the exception of research projects that may count on large teams characterized by tasks' delegation and where many activities may run in parallel, in the majority of cases the barcoding effort is handled by a limited number of persons. The guidelines here reported focus on this second case, with a special attention paid to field procedures, whose efficiency and smoothness are often overlooked.

One of the main requirements of any sound biological monitoring is the availability of long term and, possibly, temporal data with a high resolution. This is often difficult to be achieved, especially in Antarctica, due to a variety of logistic constraints, which make continuous sampling and monitoring activities generally unfeasible. Here we focus on the 5 µm filters used in the desalination plant of the Italian research base "Mario Zucchelli" in the Terra Nova Bay area (Ross Sea, Antarctica) to evaluate intra-annual coastal nanoplankton dynamics. These filters, together with others of larger mesh sizes, are used to decrease the amount of organisms and debris in the input seawater before the desalination processes take place, hence automatically collect the plankton present in the water column around the desalination system intake. We have used a DNA metabarcoding approach to characterize the communities retained by filters' sets collected in January 2012 and 2013. Intra-annual dynamics were disclosed with an unprecedented detail, that would not have been possible by using standard sampling approaches, and highlighted the importance of extreme, stochastic events such as katabatic wind pulses, which triggered dramatic, short-term shifts in coastal nanoplankton composition. This method, by combining a cost-effective sampling and molecular techniques, may represent a viable solution for long-term monitoring programs focusing on Antarctic coastal communities.

Distributional data on planktic copepods (Crustacea, Copepoda) collected in the framework of the IIIrd, Vth, and Xth Expeditions of the Italian National Antarctic Program (PNRA) to the Ross Sea sector from 1987 to 1995 are here provided. Sampling was performed with BIONESS and WP2 nets at 94 sampling stations at depths of 0-1,000 m, with a special focus on the Terra Nova Bay area. Altogether, this dataset comprises 6,027 distributional records, out of which 5,306 were obtained by digitizing original data reports and 721 are based on physical museum vouchers curated by the Italian National Antarctic Museum (MNA, Section of Genoa). The MNA samples include 8,224 individual specimens that were identified to the lowest possible taxonomic level. They belong to four orders, 25 families, 52 genera, and 82 morphological units (out of which 17 could be determined at the genus level only). A variety of environmental data were also recorded at each of the sampling stations, and we report original abundances (ind/m3) to enable future species distribution modelling. From a biogeographic point of view, the distributional data here reported represented new records for the Global Biogeographic Information Facility (GBIF) registry. In particular, 62% of the total number of species are new records for the Ross Sea sector and another 28% new records for the Antarctic region.

This study provides taxonomic and distributional data of bryozoan species from the Ross Sea area, mainly around Terra Nova Bay, based on specimens curated at the Italian National Antarctic Museum (MNA, Section of Genoa). Bryozoan specimens were collected at 75 different sampling stations in the Ross Sea and in the Magellan Strait, in a bathymetric range of 18-711 meters, during 13 expeditions of the Italian National Antarctic Research Program (PNRA) conducted between 1988 and 2014. A total of 282 MNA vouchers corresponding to 311 specimens and 127 morphospecies have been identified and included in the present dataset. 62% of the species were already reported for the Terra Nova Bay area, where most of the Italian samples come from, with a 35% of samples representing new records classified at the specific level, and 3% classified at the genus level. These new additions increase to 124 the total number of species known to occur in Terra Nova Bay. Four 3D-models of Antarctic bryozoans from the Ross Sea are also presented and will be released for research and educational purposes on the Museum website.

This new dataset presents occurrence data for Porifera collected in the Ross Sea, mainly in the Terra Nova Bay area, and curated at the Italian National Antarctic Museum (MNA, section of Genoa). Specimens were collected in 331 different sampling stations at depths ranging from 17 to 1,100 meters in the framework of 17 different Italian Antarctic expeditions funded by the Italian National Antarctic Research Program (PNRA). A total of 807 specimens, belonging to 144 morphospecies (i.e., 95 taxa identified at species level and 49 classified at least at the genus level) is included in the dataset. Nearly half (45%) of the species reported here correspond to species already known for Terra Nova Bay. Out of the remaining 55% previously unknown records, under a third (~29%) were classified at the species level, while over a quarter (~26%) were ascribed to the genus level only and these would require further study. All vouchers are permanently curated at the MNA and are available for study to the scientific community. A 3D model of an uncommon species from the Ross Sea, i.e. Tethyopsisbrondstedi (Burton, 1929), is also presented and will be made available for outreach purposes.

The distributional records of Ophiuroidea stored at the Italian National Antarctic Museum (MNA, Section of Genoa) are presented, corresponding to 1595 individuals that belong to 35 species and 17 genera. Specimens were collected in 106 different sampling stations at depths ranging from 21 to 1652 m in the framework of 14 Antarctic expeditions to the Ross Sea, one to the Antarctic Peninsula, and one to the Falkland Islands (Islas Malvinas). Three species, Amphiura joubini Koehler, 1912, Amphiura (Amphiura) angularis Lyman, 1879, and Ophiura flexibilis (Koehler, 1911), are reported as new records for the Terra Nova Bay area, whose check-list of species increases from 15 to 18 species. The determination of these three new records was based both on morphological identification and molecular analyses (COI barcoding). Some of the genetically characterised specimens were also documented through photogrammetry and micro-computed tomography and represent the first bulk of 3D models that will be available through the MNA and Sketchfab websites, both for research and educational purposes.
