RESUMO

Immunotherapy and associated immune regulation strategies gained huge attraction in order to be utilized for treatment and prevention of respiratory diseases. Engineering specifically nanomedicines can be used to regulate host immunity in lungs in the case of respiratory diseases including coronavirus disease 2019 (COVID-19) infection. COVID-19 causes pulmonary embolisms, thus new therapeutic options are required to target thrombosis, as conventional treatment options are either not effective due to the complexity of the immune-thrombosis pathophysiology. In this review, we discuss regulation of immune response in respiratory diseases especially COVID-19. We further discuss thrombosis and provide an overview of some antithrombotic nanoparticles, which can be used to develop nanomedicine against thrombo-inflammation induced by COVID-19 and other respiratory infectious diseases. We also elaborate the importance of immunomodulatory nanomedicines that can block pro-inflammatory signalling pathways, and thus can be recommended to treat respiratory infectious diseases.

BACKGROUND: Ischemic stroke (IS) is a fearful disease that can cause a variety of immune events. Nevertheless, precise immune-related mechanisms have yet to be systematically elucidated. This study aimed to identify immune-related signatures using machine learning and to validate them with animal experiments and single cell analysis. METHODS: In this study, we screened 24 differentially expressed genes (DEGs) while identifying immune-related signatures that may play a key role in IS development through a comprehensive strategy between least absolute shrinkage and selection operation (LASSO) regression, support vector machine (SVM) and immune-related genes. In addition, we explored immune infiltration using the CIBERSORT algorithm. Finally, we performed validation in mouse brain tissue and single cell analysis. RESULTS: We identified 24 DEGs for follow-up analysis. ID3 and SLC22A4 were finally identified as the better immune-related signatures through a comprehensive strategy among DEGs, LASSO, SVM and immune-related genes. RT-qPCR, western blot, and immunofluorescence revealed a significant decrease in ID3 and a significant increase in SLC22A4 in the middle cerebral artery occlusion group. Single cell analysis revealed that ID3 was mainly concentrated in endothelial_2 cells and SLC22A4 in astrocytes in the MCAO group. A CIBERSORT finds significantly altered levels of immune infiltration in IS patients. CONCLUSIONS: This study focused on immune-related signatures after stroke and ID3 and SLC22A4 may be new therapeutic targets to promote functional recovery after stroke. Furthermore, the association of ID3 and SLC22A4 with immune cells may be a new direction for post-stroke immunotherapy.

Orbital observations suggest that Mars underwent a recent 'ice age' (roughly 0.4-2.1 million years ago), during which a latitude-dependent ice-dust mantle (LDM)1,2 was emplaced. A subsequent decrease in obliquity amplitude resulted in the emergence of an 'interglacial period'1,3 during which the lowermost latitude LDM ice4-6 was etched and removed, returning it to the polar cap. These observations are consistent with polar cap stratigraphy1,7, but lower- to mid-latitude in situ surface observations in support of a glacial-interglacial transition that can be reconciled with mesoscale and global atmospheric circulation models8 is lacking. Here we present a suite of measurements obtained by the Zhurong rover during its traverse across the southern LDM region in Utopia Planitia, Mars. We find evidence for a stratigraphic sequence involving initial barchan dune formation, indicative of north-easterly winds, cementation of dune sediments, followed by their erosion by north-westerly winds, eroding the barchan dunes and producing distinctive longitudinal dunes, with the transition in wind regime consistent with the end of the ice age. The results are compatible with the Martian polar stratigraphic record and will help improve our understanding of the ancient climate history of Mars9.

Landforms on the Martian surface are critical to understanding the nature of surface processes in the recent past. However, modern hydroclimatic conditions on Mars remain enigmatic, as explanations for the formation of observed landforms are ambiguous. We report crusts, cracks, aggregates, and bright polygonal ridges on the surfaces of hydrated salt-rich dunes of southern Utopia Planitia (~25°N) from in situ exploration by the Zhurong rover. These surface features were inferred to form after 1.4 to 0.4 million years ago. Wind and CO2 frost processes can be ruled out as potential mechanisms. Instead, involvement of saline water from thawed frost/snow is the most likely cause. This discovery sheds light on more humid conditions of the modern Martian climate and provides critical clues to future exploration missions searching for signs of extant life, particularly at low latitudes with comparatively warmer, more amenable surface temperatures.

On 3 January 2019, China's Chang'E-4 (CE-4) successfully landed on the eastern floor of Von Kármán crater within the South Pole-Aitken Basin, becoming the first spacecraft in history to land on the Moon's farside. Here, we report the observations made by the Lunar Penetrating Radar (LPR) onboard the Yutu-2 rover during the first two lunar days. We found a signal penetration at the CE-4 landing site that is much greater than that at the CE-3 site. The CE-4 LPR images provide clear information about the structure of the subsurface, which is primarily made of low-loss, highly porous, granular materials with embedded boulders of different sizes; the images also indicate that the top of the mare basal layer should be deeper than 40 m. These results represent the first high-resolution image of a lunar ejecta sequence ever produced and the first direct measurement of its thickness and internal architecture.

Chang'E-4 (CE-4) was the first mission to accomplish the goal of a successful soft landing on the lunar farside. The landing trajectory and the location of the landing site can be effectively reconstructed and determined using series of images obtained during descent when there were no Earth-based radio tracking and the telemetry data. Here we reconstructed the powered descent trajectory of CE-4 using photogrammetrically processed images of the CE-4 landing camera, navigation camera, and terrain data of Chang'E-2. We confirmed that the precise location of the landing site is 177.5991°E, 45.4446°S with an elevation of -5935 m. The landing location was accurately identified with lunar imagery and terrain data with spatial resolutions of 7 m/p, 5 m/p, 1 m/p, 10 cm/p and 5 cm/p. These results will provide geodetic data for the study of lunar control points, high-precision lunar mapping, and subsequent lunar exploration, such as by the Yutu-2 rover.

Over 60 years of spacecraft exploration has revealed that the Earth's Moon is characterized by a lunar crust1 dominated by the mineral plagioclase, overlying a more mafic (richer in iron and magnesium) mantle of uncertain composition. Both crust and mantle formed during the earliest stages of lunar evolution when late-stage accretional energy caused a molten rock (magma) ocean, flotation of the light plagioclase, sinking of the denser iron-rich minerals, such as olivine and pyroxene, and eventually solidification2. Very large impact craters can potentially penetrate through the crust and sample the lunar mantle. The largest of these craters is the approximately 2,500-kilometre-diameter South Pole-Aitken (SPA) basin3 on the lunar far side. Evidence obtained from orbiting spacecraft shows that the floor of the SPA basin is rich in mafic minerals4, but their mantle origin is controversial and their in situ geologic settings are poorly known. China's Chang'E-4 lunar far-side lander recently touched down in the Von Kármán crater5,6 to explore the floor of the huge SPA basin and deployed its rover, Yutu-2. Here we report on the initial spectral observations of the Visible and Near Infrared Spectrometer (VNIS)7 onboard Yutu-2, which we interpret to represent the presence of low-calcium (ortho)pyroxene and olivine, materials that may originate from the lunar mantle. Geological context6 suggests that these materials were excavated from below the SPA floor by the nearby 72-km-diameter Finsen impact crater event, and transported to the landing site. Continued exploration by Yutu-2 will target these materials on the floor of the Von Kármán crater to understand their geologic context, origin and abundance, and to assess the possibility of sample-return scenarios.

For the Time Delay Integration (TDI) staggered line-scanning thermal infrared imager, a Computational Imaging (CI) approach is developed to achieve higher spatial resolution images. After a thorough analysis of the causes of non-uniform image displacement and degradation for multi-channel staggered TDI arrays, the study aims to approach one-dimensional (1D) sub-pixel displacement estimation and superposition of images from time-division multiplexing scanning lines. Under the assumption that a thermal image is 2D piecewise C(2) smooth, a sparse-and-smooth deconvolution algorithm with L1-norm regularization terms combining the first and second order derivative operators is proposed to restore high frequency components and to suppress aliasing simultaneously. It is theoretically and experimentally demonstrated, with simulation and airborne thermal infrared images, that this is a state-of-the-art practical CI method to reconstruct clear images with higher frequency components from raw thermal images that are subject to instantaneous distortion and blurring.

Based on incoherent optical synthetic aperture, a super-resolution reconstruction method is proposed with pixel-wise phase correlation superposition and sparse a priori constraints deconvolution. A "full spatial-frequency" image is synthesized from multiview low resolution images captured from successive exposures of a single focus plane array with shift-motion over a short range. Owing to a displacement matrix of 1/30th pixel accuracy and high-order differential L1 norm sparse regularization, the experimental results show that this approach achieves a state-of-the-art result with a resolution gain factor of about 3.0 times. It is demonstrated theoretically and experimentally to be a good and practical solution to improve spatial resolution beyond the resolving power limitation of conventional imaging systems and at low cost.

It has been recognized that other than habitat loss, degradation and fragmentation, the infection of the roundworm Baylisascaris schroederi (B. schroederi) is one of the major causes of death in wild giant pandas. However, the prevalence and intensity of the parasite infection has been inconsistently reported through a method that uses sedimentation-floatation followed by a microscope examination. This method fails to accurately determine infection because there are many bamboo residues and/or few B. schroederi eggs in the examined fecal samples. In the present study, we adopted a method that uses PCR and capillary electrophoresis combined with a single-strand conformation polymorphism analysis (PCR/CE-SSCP) to detect B. schroederi infection in wild giant pandas at a nature reserve, and compared it to the traditional microscope approach. The PCR specifically amplified a single band of 279-bp from both fecal samples and positive controls, which was confirmed by sequence analysis to correspond to the mitochondrial COII gene of B. schroederi. Moreover, it was demonstrated that the amount of genomic DNA was linearly correlated with the peak area of the CE-SSCP analysis. Thus, our adopted method can reliably detect the infectious prevalence and intensity of B. schroederi in wild giant pandas. The prevalence of B. schroederi was found to be 54% in the 91 fecal samples examined, and 48% in the fecal samples of 31 identified individual giant pandas. Infectious intensities of the 91 fecal samples were detected to range from 2.8 to 959.2 units/gram, and from 4.8 to 959.2 units/gram in the fecal samples of the 31 identified giant pandas. For comparison, by using the traditional microscope method, the prevalence of B. schroederi was found to be only 33% in the 91 fecal samples, 32% in the fecal samples of the 31 identified giant pandas, and no reliable infectious intensity was observed.
