Empathy is the capacity to feel and understand others' mental states. In some individuals, there is an imbalance between the affective and cognitive components of empathy, which can lead to deficits. This study investigated the functional connectivity of the anterior insula (AI) and dorsomedial prefrontal cortex (dmPFC), which play key roles in empathy, in covariation with the affective and cognitive subscales of the Interpersonal Reactivity Index (IRI), as a function of age and sex, as an exploratory analysis. Seed-based functional connectivity analyses were performed on 33 healthy participants that were subdivided according to their age (16 adults and 17 adolescents) and sex (16 women and 17 men). Adolescents reported lower cognitive empathy than adults and men less affective empathy than women. The connectivity of the dmPFC and AI, in covariation with the cognitive and affective subscales of empathy, respectively, differed between adolescents and adults, but was similar in men and women. Adolescents had patterns of negative covariations between the regions of interest and many brain regions associated with the default-mode and salience networks. These findings support that lower self-report levels of empathy in certain individuals could be reflected in the functional connectivity patterns of the dmPFC and AI.

This paper introduces an innovative functional magnetic resonance imaging (fMRI) protocol to study real verbal interactions while limiting the impact of speech-related movement artefacts. This protocol is based on a sparse sampling acquisition technique and allowed participants to complete a referential communication task with a real interaction partner. During verbal interactions, speakers adjust their verbal productions depending on their interlocutors' knowledge of the referents being mentioned. These adjustments have been linked to theory of mind (ToM), the ability to infer other's mental states. We thus sought to determine if the brain regions supporting ToM would also be activated during a referential communication task in which participants have to present movie characters that vary in their likelihood of being known by their interlocutor. This pilot study establishes that the sparse sampling strategy is a viable option to study the neural correlates of referential communication while minimizing movement artefacts. In addition, the brain regions supporting ToM were recruited during the task, though specifically for the conditions where participants could adjust their verbal productions to the interlocutor's likely knowledge of the referent. This study therefore demonstrates the feasibility and relevance of a sparse-sampling approach to study verbal interactions with fMRI, including referential communication.

The processing of sentences with negative quantifiers (e.g., few) is more costly than of sentences that contain their positive counterparts (e.g., many). While this polarity effect is robust and reliably replicable, its neurological bases are not well understood. In this study, we use functional magnetic resonance imaging (fMRI) paradigm for 30 participants to assess the polarity effect in sentences with polar quantifiers, and compare it with the polarity effect of polar adjectives. Both in quantifiers and in adjectives, the polarity effect manifests in the anterior insula bilaterally. The polarity effect in quantifiers, however, shows greater activation in the left hemisphere than it does for adjectives. In particular, left inferior frontal gyrus (IFG) and left superior temporal sulcus (STS) show increased activation for polarity in quantifiers than in adjectives, which is the evidence for the specific involvement of the language network in this type of polarity processing. Using the polarity effect in adjectives as a control, we provide further evidence for the linguistic complexity that negative quantifiers implicate on processing.

Speech perception can be challenging, especially for older adults. Despite the importance of speech perception in social interactions, the mechanisms underlying these difficulties remain unclear and treatment options are scarce. While several studies have suggested that decline within cortical auditory regions may be a hallmark of these difficulties, a growing number of studies have reported decline in regions beyond the auditory processing network, including regions involved in speech processing and executive control, suggesting a potentially diffuse underlying neural disruption, though no consensus exists regarding underlying dysfunctions. To address this issue, we conducted two experiments in which we investigated age differences in speech perception when background noise and talker variability are manipulated, two factors known to be detrimental to speech perception. In Experiment 1, we examined the relationship between speech perception, hearing and auditory attention in 88 healthy participants aged 19 to 87 years. In Experiment 2, we examined cortical thickness and BOLD signal using magnetic resonance imaging (MRI) and related these measures to speech perception performance using a simple mediation approach in 32 participants from Experiment 1. Our results show that, even after accounting for hearing thresholds and two measures of auditory attention, speech perception significantly declined with age. Age-related decline in speech perception in noise was associated with thinner cortex in auditory and speech processing regions (including the superior temporal cortex, ventral premotor cortex and inferior frontal gyrus) as well as in regions involved in executive control (including the dorsal anterior insula, the anterior cingulate cortex and medial frontal cortex). Further, our results show that speech perception performance was associated with reduced brain response in the right superior temporal cortex in older compared to younger adults, and to an increase in response to noise in older adults in the left anterior temporal cortex. Talker variability was not associated with different activation patterns in older compared to younger adults. Together, these results support the notion of a diffuse rather than a focal dysfunction underlying speech perception in noise difficulties in older adults.

It has been proposed that the maintenance of phonological information in verbal working memory (vWM) is carried by a domain-specific short-term storage center-the phonological loop-which is composed of a phonological store and an articulatory rehearsal system. Several brain regions including the left posterior inferior frontal gyrus (pIFG) and anterior supramarginal gyri (aSMG) are thought to support these processes. However, recent behavioral evidence suggests that verbal and non-verbal auditory information may be processed as part of a unique domain general short-term storage center instead of through specialized subsystems such as the phonological loop. In the current study, we used a single-pulse transcranial magnetic stimulation (TMS)-delayed priming paradigm with speech (syllables) and acoustically complex non-speech sounds (bird songs) to examine whether the pIFG and aSMG are involved in the processing of verbal information or, alternatively, in the processing of any complex auditory information. Our results demonstrate that TMS delivered to both regions had an effect on performance for speech and non-speech stimuli, but the nature of the effect was different. That is, priming was reduced for the speech sounds because TMS facilitated the detection of different but not identical stimuli, and accuracy was decreased for non-speech sounds. Since TMS interfered with both speech and non-speech sounds, these findings support the existence of an auditory short-term storage center located within the dorsal auditory stream.

RESUMO

The COVID-19 pandemic is a stark reminder of the heavy toll that emerging infectious diseases (EIDs) with epidemic and pandemic potential can inflict. Vaccine development, scale-up, and commercialization is a long, expensive, and risky enterprise that requires substantial upfront planning and offers no guarantee of success. EIDs are a particularly challenging target for global health preparedness, including for vaccine development. Insufficient attention has been given to challenges, lessons learned, and potential solutions to support and sustain vaccine industry engagement in vaccine development for EIDs. Drawing from lessons from the most recent Ebola epidemic in the Democratic Republic of the Congo, as well as the 2009 H1N1 influenza, 2014-2016 Ebola, and 2015-16 Zika outbreaks preceding it, we offer our perspective on challenges facing EID vaccine development and recommend additional solutions to prioritize in the near term. The 6 recommendations focus on reducing vaccine development timelines and increasing business certainty to reduce risks for companies. The global health security community has an opportunity to build on the current momentum to design a sustainable model for EID vaccines.

High-level cognitive capacities that serve communication, reasoning, and calculation are essential for finding our way in the world. But whether and to what extent these complex behaviors share the same neuronal substrate are still unresolved questions. The present study separated the aspects of logic from language and numerosity-mental faculties whose distinctness has been debated for centuries-and identified a new cytoarchitectonic area as correlate for an operation involving logical negation. A novel experimental paradigm that was implemented here in an RT/fMRI study showed a single cluster of activity that pertains to logical negation. It was distinct from clusters that were activated by numerical comparison and from the traditional language regions. The localization of this cluster was described by a newly identified cytoarchitectonic area in the left anterior insula, ventro-medial to Broca's region. We provide evidence for the congruence between the histologically and functionally defined regions on multiple measures. Its position in the left anterior insula suggests that it functions as a mediator between language and reasoning areas.

In this article, we used High Angular Resolution Diffusion Imaging (HARDI) with advanced anatomically constrained particle filtering tractography to investigate the role of the arcuate fasciculus (AF) and the middle longitudinal fasciculus (MdLF) in speech perception in noise in younger and older adults. Fourteen young and 15 elderly adults completed a syllable discrimination task in the presence of broadband masking noise. Mediation analyses revealed few effects of age on white matter (WM) in these fascicles but broad effects of WM on speech perception, independently of age, especially in terms of sensitivity and criterion (response bias), after controlling for individual differences in hearing sensitivity and head size. Indirect (mediated) effects of age on speech perception through WM microstructure were also found, after controlling for individual differences in hearing sensitivity and head size, with AF microstructure related to sensitivity, response bias and phonological priming, and MdLF microstructure more strongly related to response bias. These findings suggest that pathways of the perisylvian region contribute to speech processing abilities, with relatively distinct contributions for the AF (sensitivity) and MdLF (response bias), indicative of a complex contribution of both phonological and cognitive processes to age-related speech perception decline. These results provide new and important insights into the roles of these pathways as well as the factors that may contribute to elderly speech perception deficits. They also highlight the need for a greater focus to be placed on studying the role of WM microstructure to understand cognitive aging.

Despite the huge importance of spoken language production in everyday life, little is known about the manner and extent to which the motor aspects of speech production evolve with advancing age, as well as the nature of the underlying senescence mechanisms. In this cross-sectional group study, we examined the relationship between age and speech production performance using a nonlexical speech production task in which spoken syllable frequency and phonological complexity were systematically varied to test hypotheses about underlying mechanisms. A nonprobabilistic sample of 60 cognitively healthy adults (18-83 years) produced meaningless nonwords aloud as quickly and accurately as possible. Error rate, vocal reaction time (RT), vocal RT variability, vocal response duration, and vocal response duration variability were used as dependent variables to characterize speech production performance. The results showed an overall increase in error rate, which occurred mainly in the final syllable position (coda). There was also an increase in vocal response duration and in duration variability with age, which was moderated by phonological complexity and syllable frequency. Finally, we also found an age-related change in the relationship between vocal RT and vocal response duration. Together, these findings were interpreted as reflecting an age-related decline in the planning and execution of speech movements in cognitively healthy adults. Purpose: The factors that influence the evaluation of voice in adulthood, as well as the consequences of such evaluation on social interactions, are not well understood. Here, we examined the effect of listeners' age and the effect of talker age, sex, and smoking status on the auditory-perceptual evaluation of voice, voice-related psychosocial attributions, and perceived speech tempo. We also examined the voice dimensions affecting the propensity to engage in social interactions. Method: Twenty-five younger (age 19-37 years) and 25 older (age 51-74 years) healthy adults participated in this cross-sectional study. Their task was to evaluate the voice of 80 talkers. Results: Statistical analyses revealed limited effects of the age of the listener on voice evaluation. Specifically, older listeners provided relatively more favorable voice ratings than younger listeners, mainly in terms of roughness. In contrast, the age of the talker had a broader impact on voice evaluation, affecting auditory-perceptual evaluations, psychosocial attributions, and perceived speech tempo. Some of these talker differences were dependent upon the sex of the talker and his or her smoking status. Finally, the results also show that voice-related psychosocial attribution was more strongly associated with the propensity of the listener to engage in social interactions with a person than auditory-perceptual dimensions and perceived speech tempo, especially for the younger adults. Conclusions: These results suggest that age has a broad influence on voice evaluation, with a stronger impact for talker age compared with listener age. While voice-related psychosocial attributions may be an important determinant of social interactions, perceived voice quality and speech tempo appear to be less influential. Healthy aging is associated with a decline in cognitive, executive, and motor processes that are concomitant with changes in brain activation patterns, particularly at high complexity levels. While speech production relies on all these processes, and is known to decline with age, the mechanisms that underlie these changes remain poorly understood, despite the importance of communication on everyday life. In this cross-sectional group study, we investigated age differences in the neuromotor control of speech production by combining behavioral and functional magnetic resonance imaging (fMRI) data. Twenty-seven healthy adults underwent fMRI while performing a speech production task consisting in the articulation of nonwords of different sequential and motor complexity. Results demonstrate strong age differences in movement time (MT), with longer and more variable MT in older adults. The fMRI results revealed extensive age differences in the relationship between BOLD signal and MT, within and outside the sensorimotor system. Moreover, age differences were also found in relation to sequential complexity within the motor and attentional systems, reflecting both compensatory and de-differentiation mechanisms. At very high complexity level (high motor complexity and high sequence complexity), age differences were found in both MT data and BOLD response, which increased in several sensorimotor and executive control areas. Together, these results suggest that aging of motor and executive control mechanisms may contribute to age differences in speech production. These findings highlight the importance of studying functionally relevant behavior such as speech to understand the mechanisms of human brain aging. Many factors affect our ability to decode the speech signal, including its quality, the complexity of the elements that compose it, as well as their frequency of occurrence and co-occurrence in a language. Syllable frequency effects have been described in the behavioral literature, including facilitatory effects during speech production and inhibitory effects during word recognition, but the neural mechanisms underlying these effects remain largely unknown. The objective of this study was to examine, using functional neuroimaging, the neurobiological correlates of three different distributional statistics in simple 2-syllable nonwords: the frequency of the first and second syllables, and the mutual information between the syllables. We examined these statistics during nonword perception and production using a powerful single-trial analytical approach. We found that repetition accuracy was higher for nonwords in which the frequency of the first syllable was high. In addition, brain responses to distributional statistics were widespread and almost exclusively cortical. Importantly, brain activity was modulated in a distinct manner for each statistic, with the strongest facilitatory effects associated with the frequency of the first syllable and mutual information. These findings show that distributional statistics modulate nonword perception and production. We discuss the common and unique impact of each distributional statistic on brain activity, as well as task differences.

The processing of continuous and complex auditory signals such as speech relies on the ability to use statistical cues (e.g. transitional probabilities). In this study, participants heard short auditory sequences composed either of Italian syllables or bird songs and completed a regularity-rating task. Behaviorally, participants were better at differentiating between levels of regularity in the syllable sequences than in the bird song sequences. Inter-individual differences in sensitivity to regularity for speech stimuli were correlated with variations in surface-based cortical thickness (CT). These correlations were found in several cortical areas including regions previously associated with statistical structure processing (e.g. bilateral superior temporal sulcus, left precentral sulcus and inferior frontal gyrus), as well other regions (e.g. left insula, bilateral superior frontal gyrus/sulcus and supramarginal gyrus). In all regions, this correlation was positive suggesting that thicker cortex is related to higher sensitivity to variations in the statistical structure of auditory sequences. Overall, these results suggest that inter-individual differences in CT within a distributed network of cortical regions involved in statistical structure processing, attention and memory is predictive of the ability to detect structural structure in auditory speech sequences.

While there has been a growing number of studies examining the neurofunctional correlates of speech production over the past decade, the neurostructural correlates of this immensely important human behaviour remain less well understood, despite the fact that previous studies have established links between brain structure and behaviour, including speech and language. In the present study, we thus examined, for the first time, the relationship between surface-based cortical thickness (CT) and three different behavioural indexes of sublexical speech production: response duration, reaction times and articulatory accuracy, in healthy young and older adults during the production of simple and complex meaningless sequences of syllables (e.g., /pa-pa-pa/ vs. /pa-ta-ka/). The results show that each behavioural speech measure was sensitive to the complexity of the sequences, as indicated by slower reaction times, longer response durations and decreased articulatory accuracy in both groups for the complex sequences. Older adults produced longer speech responses, particularly during the production of complex sequence. Unique age-independent and age-dependent relationships between brain structure and each of these behavioural measures were found in several cortical and subcortical regions known for their involvement in speech production, including the bilateral anterior insula, the left primary motor area, the rostral supramarginal gyrus, the right inferior frontal sulcus, the bilateral putamen and caudate, and in some region less typically associated with speech production, such as the posterior cingulate cortex.

BACKGROUND: Technological innovations in mining equipment have led to increased productivity and occupational health and safety (OHS) performance, but their introduction also brings new risks for workers. OBJECTIVE: The aim of this study is to provide support for mining industry managers who are required to reconcile equipment choices with OHS and productivity. METHODS: Examination of the literature through interdisciplinary digital databases. Databases were searched using specific combinations of keywords and limited to studies dating back no farther than 1992. The ``snowball'' technique was also used to examining the references listed in research articles initially identified with the databases. RESULTS: A total of 19 contextual factors were identified as having the potential to influence the OHS and productivity leverage of equipment innovations. The most often cited among these factors are the level of training provided to the equipment operators, operator experience and age, supervisor leadership abilities, and maintaining good relations within work crews. CONCLUSIONS: Interactions between these factors are not discussed in mining innovation literature. It would be helpful to use a systems thinking approach which incorporates interaction between relevant actors and factors to define properly the most sensitive aspects of innovation management as it applies to mining equipment.

Previous research has associated the inferior frontal and posterior temporal brain regions with a number of phonological processes. In order to identify how these specific brain regions contribute to phonological processing, we manipulated subsyllabic phonological complexity and stimulus modality during speech perception using fMRI. Subjects passively attended to visual or auditory pseudowords. Similar to previous studies, a bilateral network of cortical regions was recruited during the presentation of visual and auditory stimuli. Moreover, pseudowords recruited a similar network of regions as words and letters. Few regions in the whole-brain results revealed neural processing differences associated with phonological complexity independent of modality of presentation. In an ROI analysis, the only region sensitive to phonological complexity was the posterior part of the inferior frontal gyrus (IFGpo), with the complexity effect only present for print. In sum, the sensitivity of phonological brain areas depends on the modality of stimulus presentation and task demands.

We investigated the course of language processing in the context of a verification task that required numerical estimation and comparison. Participants listened to sentences with complex quantifiers that contrasted in Polarity, a logical property (e.g., more-than-half, less-than-half), and then performed speeded verification on visual scenarios that displayed a proportion between 2 discrete quantities. We varied systematically not only the sentences, but also the visual materials, in order to study their effect on the verification process. Next, we used the same visual scenarios with analogous non-verbal probes that featured arithmetical inequality symbols (<, >). This manipulation enabled us to measure not only Polarity effects, but also, to compare the effect of different probe types (linguistic, non-linguistic) on processing. Like many previous studies, our results demonstrate that perceptual difficulty affects error rate and reaction time in keeping with Weber's Law. Interestingly, these performance parameters are also affected by the Polarity of the quantifiers used, despite the fact that sentences had the exact same meaning, sentence structure, number of words, syllables, and temporal structure. Moreover, an analogous contrast between the non-linguistic probes (<, >) had no effect on performance. Finally, we observed no interaction between performance parameters governed by Weber's Law and those affected by Polarity. We consider 4 possible accounts of the results (syntactic, semantic, pragmatic, frequency-based), and discuss their relative merit.

The processing of fluent speech involves complex computational steps that begin with the segmentation of the continuous flow of speech sounds into syllables and words. One question that naturally arises pertains to the type of syllabic information that speech processes act upon. Here, we used functional magnetic resonance imaging to profile regions, using a combination of whole-brain and exploratory anatomical region-of-interest (ROI) approaches, that were sensitive to syllabic information during speech perception by parametrically manipulating syllabic complexity along two dimensions: (1) individual syllable complexity, and (2) sequence complexity (supra-syllabic). We manipulated the complexity of the syllable by using the simplest syllable template-a consonant and vowel (CV)-and inserting an additional consonant to create a complex onset (CCV). The supra-syllabic complexity was manipulated by creating sequences composed of the same syllable repeated six times (e.g., /pa-pa-pa-pa-pa-pa/) and sequences of three different syllables each repeated twice (e.g., /pa-ta-ka-pa-ta-ka/). This parametrical design allowed us to identify brain regions sensitive to (1) syllabic complexity independent of supra-syllabic complexity, (2) supra-syllabic complexity independent of syllabic complexity and, (3) both syllabic and supra-syllabic complexity. High-resolution scans were acquired for 15 healthy adults. An exploratory anatomical ROI analysis of the supratemporal plane (STP) identified bilateral regions within the anterior two-third of the planum temporale, the primary auditory cortices as well as the anterior two-third of the superior temporal gyrus that showed different patterns of sensitivity to syllabic and supra-syllabic information. These findings demonstrate that during passive listening of syllable sequences, sublexical information is processed automatically, and sensitivity to syllabic and supra-syllabic information is localized almost exclusively within the STP.

The supramarginal gyrus (SMG) is activated for phonological processing during both language and verbal working memory tasks. Using rTMS, we investigated whether the contribution of the SMG to phonological processing is domain specific (specific to phonology) or more domain general (specific to verbal working memory). A measure of phonological complexity was developed based on sonority differences and subjects were tested after low frequency rTMS on a same/different judgment task and an n-back verbal memory task. It was reasoned that if the phonological processing in the SMG is more domain general, i.e., related to verbal working memory demands, performance would be more affected by the rTMS during the n-back task than during the same/different judgment task. Two auditory experiments were conducted. The first experiment demonstrated that under conditions where working memory demands are minimized (i.e. same/different judgment), repetitive stimulation had no effect on performance although performance varied as a function of phonological complexity. The second experiment demonstrated that during a verbal working memory task (n-back task), where phonological complexity was also manipulated, subjects were less accurate and slower at performing the task after stimulation but the effect of phonology was not affected. The results confirm that the SMG is involved in verbal working memory but not in the encoding of sonority differences.

INTRODUCTION: The role of the left planum temporale (PT) in auditory language processing has been a central theme in cognitive neuroscience since the first descriptions of its leftward neuroanatomical asymmetry. While it is clear that PT contributes to auditory language processing there is still some uncertainty about its role in spoken language production. METHODS: Here we examine activation patterns of the PT for speech production, speech perception and single word reading to address potential hemispheric and regional functional specialization in the human PT. To this aim, we manually segmented the left and right PT in three non-overlapping regions (medial, lateral and caudal PT) and examined, in two complementary experiments, the contribution of exogenous and endogenous auditory input on PT activation under different speech processing and production conditions. RESULTS: Our results demonstrate that different speech tasks are associated with different regional functional activation patterns of the medial, lateral and caudal PT. These patterns are similar across hemispheres, suggesting bilateral processing of the auditory signal for speech at the level of PT. CONCLUSIONS: Results of the present studies stress the importance of considering the anatomical complexity of the PT in interpreting fMRI data.
