RESUMO

A key factor contributing to resistance in immune checkpoint blockade (ICB) therapies is CD8+ T-cell tolerance in the tumor microenvironment (TME), partly resulting from upregulating coinhibitory receptors. Here, we describe the role of PGRN as a coinhibitory molecule that modulates the antitumor response of CD8+ T cells, thus presenting a novel immunosuppressive target for lung cancer. The in vivo subcutaneous transplanted lung cancer model showed that PGRN expression was elevated on CD8+ T cells that infiltrated transplanted lung cancers. Furthermore, PGRN deficiency was found to specifically encourage the infiltration of CD8+ T cells, enhance their proliferation, migration, and activation, and resist apoptosis, ultimately inhibiting tumor growth. This was achieved by PGRN knockout, increasing the production of T cell chemokine CCL3, which boosts the antitumor immune response induced by CD8+ T cells. Critically, the PD-L1 inhibitor exhibited a synergistic effect in enhancing the antitumor response in PGRN-/- mice. In summary, our findings highlight the significance of PGRN as a novel target for boosting CD8+ T cells antitumor immunity and its potential to overcome the resistance in ICB therapy.

Current clinical guidelines limit surgical intervention to patients with cT1-2N0M0 small cell lung cancer (SCLC). Our objective was to reassess the role of surgery in SCLC management, and explore novel prognostic indicators for surgically resected SCLC. We reviewed all patients diagnosed with SCLC from January 2011 to April 2021 in our institution. Survival analysis was conducted using the Kaplan-Meier method, and independent prognostic factors were assessed through the Cox proportional hazard model. In addition, immunohistochemistry (IHC) staining was performed to evaluate the predictive value of selected indicators in the prognosis of surgically resected SCLC patients. In the study, 177 SCLC patients undergoing surgical resection were ultimately included. Both univariate and multivariate Cox analysis revealed that incomplete postoperative adjuvant therapy emerged as an independent risk factor for adverse prognosis (p < 0.001, HR 2.96). Survival analysis revealed significantly superior survival among pN0-1 patients compared to pN2 patients (p < 0.0001). No significant difference in postoperative survival was observed between pN1 and pN0 patients (p = 0.062). Patients with postoperative stable disease (SD) exhibited lower levels of tumor inflammatory cells (TIC) (p = 0.0047) and IFN-γ expression in both area and intensity (p < 0.0001 and 0.0091, respectively) compared to those with postoperative progressive disease (PD). Conversely, patients with postoperative SD showed elevated levels of stromal inflammatory cells (SIC) (p = 0.0453) and increased counts of CD3+ and CD8+ cells (p = 0.0262 and 0.0330, respectively). Survival analysis indicated that high levels of SIC, along with low levels of IFN-γ+ cell area within tumor tissue, may correlate positively with improved prognosis in surgically resected SCLC (p = 0.017 and 0.012, respectively). In conclusion, the present study revealed that the patients with pT1-2N1M0 staging were a potential subgroup of SCLC patients who may benefit from surgery. Complete postoperative adjuvant therapy remains an independent factor promoting a better prognosis for SCLC patients undergoing surgical resection. Moreover, CD3, CD8, IFN-γ, TIC, and SIC may serve as potential indicators for predicting the prognosis of surgically resected SCLC.

The effects of varying fat additions (0 %, 1.0 %, 1.5 %, 2.0 %, and 2.5 %) on characteristics and interfacial properties of chicken white soup emulsion from stewing chicken skeleton were investigated. The results revealed that the chicken white soup emulsion obtained with the 2.0 % fat addition had smaller D3,2 (1.889 µm), D4,3 (2.944 µm), and higher absolute zeta potential value (23.32 mV). Viscosity values were higher for the 2.0 % fat addition compared to the other treatment groups. Techniques like scanning electron microscopy, laser confocal, and atomic force microscopy demonstrated that oil droplets and particles in the soup were smaller and more evenly dispersed with the 2.0 % fat addition. Moreover, the 2.0 % fat group exhibited higher interfacial protein concentration of 207.56 mg/m2. Lastly, low field NMR images confirmed that the stability of the soup was enhanced with a 2.0 % fat addition. This research offers a foundational understanding for producing highly stable chicken white soup.

The protein and fat in chicken skeleton can be emulsified in a boiling state to form milky white chicken soup. White chicken soup has a delicious taste, good nutritional value, a beautiful color, and volatile flavor compounds. However, cooking time significantly impacts the quality of white chicken soup. Herein, we investigated the influence of cooking time (30, 60, 90, 120, 150, 180, and 210 min) on the migration of nutrient substances and characteristics changes in white chicken soup from chicken skeletons. The results showed that nutrients such as total lipids, water-soluble protein, total sugars, solid matter, and oligopeptides in the chicken skeletons' tissue continuously migrated into the soup during the cooking process. The total nutrient content in the chicken soup was highest after cooking for 180 min. Simultaneously, the white chicken soup obtained after cooking for 180 min had low interfacial tension and high whiteness, viscosity, and storage stability. The high stability index was associated with increased ζ potential and decreased particle size. The contact angle analysis results also indicated that the stability of the white chicken soup was improved when the cooking time reached 180 min. This research provides basic information for the production of high-quality white chicken soup.

Super-resolving the magnetic resonance (MR) image of a target contrast under the guidance of the corresponding auxiliary contrast, which provides additional anatomical information, is a new and effective solution for fast MR imaging. However, current multi-contrast super-resolution (SR) methods tend to concatenate different contrasts directly, ignoring their relationships in different clues, e.g., in the high-and low-intensity regions. In this study, we propose a separable attention network (comprising high-intensity priority (HP) attention and low-intensity separation (LS) attention), named SANet. Our SANet could explore the areas of high-and low-intensity regions in the "forward" and "reverse" directions with the help of the auxiliary contrast while learning clearer anatomical structure and edge information for the SR of a target-contrast MR image. SANet provides three appealing benefits: First, it is the first model to explore a separable attention mechanism that uses the auxiliary contrast to predict the high-and low-intensity regions, diverting more attention to refining any uncertain details between these regions and correcting the fine areas in the reconstructed results. Second, a multistage integration module is proposed to learn the response of multi-contrast fusion at multiple stages, get the dependency between the fused representations, and boost their representation ability. Third, extensive experiments with various state-of-the-art multi-contrast SR methods on fastMRI and clinical in vivo datasets demonstrate the superiority of our model. Rosacea is a long-term inflammatory skin disease associated with the dysfunction of vascular and immunological systems. Treatment options for rosacea are difficult to implement. Oroxylin A(OA), a traditional Chinese medicine, has anti-inflammation effects in a variety of inflammatory diseases. However, it is not known that whether OA exerts protective effects against LL-37-induced rosacea. In this study, bioinformatics analyses showed that the mechanisms of rosacea and the pharmacological targets of OA were highly overlapped. Subsequently, it was shown that the administration of OA resulted in a notable amelioration of rosacea-like skin lesions, as evidenced by a reduction in immune cell infiltration, modulation of cytokine production, and inhibition of angiogenesis. Plus, it was shown that OA effectively suppressed the generation of ROS generated by LL-37, as well as the subsequent activation of NF-κB signaling pathway. To explore further, we found that OA inhibited LL-37-induced ROS production via SIRT3-SOD2 signaling pathway in keratinocytes. Based on the aforementioned evidence, it can be inferred that OA exhibits a mitigating effect on the inflammatory response in rosacea by modulating the SIRT3-SOD2-NF-κB signaling pathway.

Thanks to its powerful ability to depict high-resolution anatomical information, magnetic resonance imaging (MRI) has become an essential non-invasive scanning technique in clinical practice. However, excessive acquisition time often leads to the degradation of image quality and psychological discomfort among subjects, hindering its further popularization. Besides reconstructing images from the undersampled protocol itself, multi-contrast MRI protocols bring promising solutions by leveraging additional morphological priors for the target modality. Nevertheless, previous multi-contrast techniques mainly adopt a simple fusion mechanism that inevitably ignores valuable knowledge. In this work, we propose a novel multi-contrast complementary information aggregation network named MCCA, aiming to exploit available complementary representations fully to reconstruct the undersampled modality. Specifically, a multi-scale feature fusion mechanism has been introduced to incorporate complementary-transferable knowledge into the target modality. Moreover, a hybrid convolution transformer block was developed to extract global-local context dependencies simultaneously, which combines the advantages of CNNs while maintaining the merits of Transformers. Compared to existing MRI reconstruction methods, the proposed method has demonstrated its superiority through extensive experiments on different datasets under different acceleration factors and undersampling patterns.

The complexes of thiazole with CF4 and SF6 have been investigated by Fourier transform microwave spectroscopy and quantum chemical calculations. One rotational spectrum was observed for the thiazole-CF4 complex. Experiments and theoretical computations confirmed that the observed structure of thiazole-CF4 is primarily formed due to N⋯CCF4 interaction with the C atom of CF4 located in the plane of the thiazole ring. The rotational transitions of thiazole-CF4 exhibit A/E torsional splitting induced by the internal rotation of the -CF3 top. The potential barrier of the -CF3 internal rotation is 0.2411(1) kJ mol-1, consistent with the calculated value (∼0.3 kJ mol-1). For the thiazole-SF6 complex, one conformer with SF6 located above the thiazole ring is detected. The observed structure of thiazole-SF6 is mainly stabilized by van der Waals interactions. The energy decomposition analysis reveals that the electrostatics and dispersion are the dominant attractive contributions to the formation of thiazole-CF4 and thiazole-SF6 dimers, whereas the weight of the dispersion term becomes more significant in the thiazole-SF6 complex compared to that of the thiazole-CF4 complex.

The goal of Camouflaged object detection (COD) is to detect objects that are visually embedded in their surroundings. Existing COD methods only focus on detecting camouflaged objects from seen classes, while they suffer from performance degradation to detect unseen classes. However, in a real-world scenario, collecting sufficient data for seen classes is extremely difficult and labeling them requires high professional skills, thereby making these COD methods not applicable. In this paper, we propose a new zero-shot COD framework (termed as ZSCOD), which can effectively detect the never unseen classes. Specifically, our framework includes a Dynamic Graph Searching Network (DGSNet) and a Camouflaged Visual Reasoning Generator (CVRG). In details, DGSNet is proposed to adaptively capture more edge details for boosting the COD performance. CVRG is utilized to produce pseudo-features that are closer to the real features of the seen camouflaged objects, which can transfer knowledge from seen classes to unseen classes to help detect unseen objects. Besides, our graph reasoning is built on a dynamic searching strategy, which can pay more attention to the boundaries of objects for reducing the influences of background. More importantly, we construct the first zero-shot COD benchmark based on the COD10K dataset. Experimental results on public datasets show that our ZSCOD not only detects the camouflaged object of unseen classes but also achieves state-of-the-art performance in detecting seen classes.

Oil pollution can release trace metals (TMs) with cumulative toxicity into seawater, harming marine ecosystems in the long term. However, the lack of studies has inhibited our understanding of the effects and mechanisms of oil pollution on TMs in seawater. Hence, we investigated the 10-year monthly variation of TMs in Porites coral skeletons from the northern South China Sea (SCS), complemented by spatial distribution of TMs in seawater, sediments and characterization of TMs in fuel oil. The results of principal component-multivariate linear regression showed that the total contribution of oil pollution as a source to TMs in surface seawater was 77.2%, where the residence time of TMs (Ni, V, Cr, Co, Cu, Mn, Fe, and Mo) released from oil spills in surface seawater was approximately 1.4 months. Due to the geochemical nature of the metals, their seasonal variations are controlled by tropical cyclones (Ni, V, Cr, Co, Cu, Mn, Fe, and Mo), winter monsoons (Pb, Cd, Ba, and Zn) and sea surface temperature (Sr). This study shows that coral skeletons can be used as a new tool to study marine oil pollution. This provides valuable reference data for accurately identifying and quantifying the effects of oil pollution on TMs in seawater from a spatial and temporal perspective.

Background: A few studies found that the complement system may be involved in the onset and progression of community-acquired pneumonia (CAP). However, the role of the complement system in CAP was obscure. The goal of this study was to analyze the association of serum complement C3a with CAP severity scores based on a cross-sectional study. Methods: All 190 CAP patients and 95 control subjects were enrolled. Demographic information and clinical data were extracted. Peripheral blood samples were collected on admission. Results: Serum complement C3a on admission was elevated in CAP patients compared with healthy subjects. The level of complement C3a was gradually elevated in parallel with CAP severity scores (CURB-65, CRB-65, PSI, SMART-COP, and CURXO). Complement C3a was positively correlated with blood routine parameters, renal function markers, and inflammatory cytokines in CAP patients. Furthermore, multivariate linear and logistic regression models found that serum complement C3a on admission was positively associated with CAP severity scores. Mechanistic research suggested that complement system inhibition alleviated Streptococcus pneumoniae-induced upregulation of IL-1ß, TNF-α, IL-6, and CRP in MLE-12 cells. Conclusions: Serum complement C3a on admission is positively associated with the severity of CAP patients. Inhibiting complement system attenuates S. pneumoniae-elevated secretion of inflammatory cytokines in pulmonary epithelial cells, indicating that complement C3a is involved in the pathophysiology of CAP. Serum complement C3a may serve as an earlier diagnostic biomarker for CAP.

RESUMO

Accelerated multi-modal magnetic resonance (MR) imaging is a new and effective solution for fast MR imaging, providing superior performance in restoring the target modality from its undersampled counterpart with guidance from an auxiliary modality. However, existing works simply combine the auxiliary modality as prior information, lacking in-depth investigations on the potential mechanisms for fusing different modalities. Further, they usually rely on the convolutional neural networks (CNNs), which is limited by the intrinsic locality in capturing the long-distance dependency. To this end, we propose a multi-modal transformer (MTrans), which is capable of transferring multi-scale features from the target modality to the auxiliary modality, for accelerated MR imaging. To capture deep multi-modal information, our MTrans utilizes an improved multi-head attention mechanism, named cross attention module, which absorbs features from the auxiliary modality that contribute to the target modality. Our framework provides three appealing benefits: (i) Our MTrans use an improved transformers for multi-modal MR imaging, affording more global information compared with existing CNN-based methods. (ii) A new cross attention module is proposed to exploit the useful information in each modality at different scales. The small patch in the target modality aims to keep more fine details, the large patch in the auxiliary modality aims to obtain high-level context features from the larger region and supplement the target modality effectively. (iii) We evaluate MTrans with various accelerated multi-modal MR imaging tasks, e.g., MR image reconstruction and super-resolution, where MTrans outperforms state-of-the-art methods on fastMRI and real-world clinical datasets.

Sports industry has been playing an important role in achieving good healthcare for public. However, with the advent of COVID-19, sports industry has been influenced significantly and the industry scale is decreased considerably. In this situation, how to accurately predict the sports industry scale in terms of production and consumption is becoming a practical and valuable task, because the whole world's economy is not growing stably and users' demand to sport goods is fluctuating sharply. However, three challenges are often existing in the sports industry scale prediction. First of all, there are so many kinds of sport goods that it is hard to quickly predict their future production or consumption scales accurately. Second, for a certain sport commodity, its production or consumption scale is often related to time especially in the COVID-19 environment. Third, sports industry scale data often contain some privacy, which probably disables data stakeholders to disclose their data. In view of these three challenges, a novel sports industry scale prediction approach (named SISP) is proposed for healthcare, which is basically according to time series analysis. Through SISP approach, we can quickly and accurately predict the future production or consumption scales of sport goods, in a privacy-aware way. At last, we validate the feasibility of the proposed SISP approach in this paper.

BACKGROUND: Soybean protein isolate hydrolysates (SPIHs) produced at high hydrostatic pressure have higher bioactivity. The aim of the study was to analyze the effects of different SPIH concentrations obtained under various pressures (0.1, 100, 200, and 300 MPa) on gelling properties, structural characteristics, and main forces of myofibrillar protein (MP) in MP-SPIH plural gels. RESULTS: The MP-SPIH plural gel with 3% SPIH produced under 200 MPa had the maximum gel strength (0.42 N) and water holding capacity (53.69%). A decline in thermal stability and a rise in storage modulus (G') of MP-SPIH plural gels were found with increased SPIH pressure and concentration. Additionally, the addition of SPIHs increased the amounts of α-helix and ß-sheet, decreased random coil structural content of MP in MP-SPIH plural gels, and facilitated the generation of a denser and uniform gels network. The molecular forces in MP-SPIH plural gels were mainly hydrophobic interaction and hydrogen bond. CONCLUSION: This study showed that the interaction of MP with 3% SPIH obtained at 200 MPa improved the quality of plural gels. © 2022 Society of Chemical Industry.

Heavy metal pollution is a serious environmental problem in the marine ecosystem. Thereinto, marine transportation activities have gradually become an important source of heavy metals in seawater. However, the lack of studies on the temporal dynamics of seawater heavy metals in marine shipping areas has hindered our understanding of the sources and transport mechanisms of heavy metals in seawater of hectic shipping waters. Therefore, we investigated the interannual resolution variation of heavy metals in Porites lutea skeletons during the past 32 years under the rapid development of the shipping sector near Weizhou Island from the northern South China Sea. Results show that most heavy metal concentrations with higher coefficients of variation (≥100 %) in the Porites coral skeletons were higher than those in the uncontaminated or less anthropogenic waters. The results of principal component analysis and multiple linear regression showed that the interannual variations of Ni, V, Cr, Co, Zn, Cu, Mn, Fe and Mo were mainly impacted by marine oil extraction and oil spills generated by shipping activities, accounting for 51.58 %. The effect of sea surface temperature accounts for 13.44 %, and controls the interannual variations of Ba and Sr. The effect of industrial pollution accounts for 13.27 %, and explains the interannual variations of Cd and Y. The fuel consumption of marine shipping accounted for 8.76 %, explaining the interannual variations of Pb. The total contribution of anthropogenic activities reached 73.61 %. The interannual variation of heavy metals indicates that hectic marine shipping activities are the dominant cause of Ni, V, Pb, Cr, Co, Zn, Cu, Mn, Fe and Mo input to surface seawater around Weizhou Island. This provided valuable data for understanding the temporal dynamics and potential sources of heavy metals in the marine environment by using coral skeletons as a high-resolution recording vehicle.

Federated learning (FL) can be used to improve data privacy and efficiency in magnetic resonance (MR) image reconstruction by enabling multiple institutions to collaborate without needing to aggregate local data. However, the domain shift caused by different MR imaging protocols can substantially degrade the performance of FL models. Recent FL techniques tend to solve this by enhancing the generalization of the global model, but they ignore the domain-specific features, which may contain important information about the device properties and be useful for local reconstruction. In this paper, we propose a specificity-preserving FL algorithm for MR image reconstruction (FedMRI). The core idea is to divide the MR reconstruction model into two parts: a globally shared encoder to obtain a generalized representation at the global level, and a client-specific decoder to preserve the domain-specific properties of each client, which is important for collaborative reconstruction when the clients have unique distribution. Such scheme is then executed in the frequency space and the image space respectively, allowing exploration of generalized representation and client-specific properties simultaneously in different spaces. Moreover, to further boost the convergence of the globally shared encoder when a domain shift is present, a weighted contrastive regularization is introduced to directly correct any deviation between the client and server during optimization. Extensive experiments demonstrate that our FedMRI's reconstructed results are the closest to the ground-truth for multi-institutional data, and that it outperforms state-of-the-art FL methods.

MET exon 14 skipping mutation (METex14m) is rare and occurs in approximately 1-4% of all non-small cell lung cancer (NSCLC) patients and approximately 2.8% of resected stage I-III NSCLC patients. Savolitinib is an oral, potent and highly selective type Ib MET inhibitor, which has been shown to be promising activity and acceptable safety profile in patients with advanced NSCLC harboring METex14m. Most recently, many studies have been probing into the feasibility and efficacy of target therapy for perioperative application in NSCLC. Interestingly, there are very few recorded cases of such treatments. Here, we presented that systemic treatment with the MET inhibitor savolitinib before surgery could provide the potential to prolong overall survival (OS) of patients with locally advanced potentially resectable NSCLC. A 49-year-old woman was diagnosed with stage IIIA (T2bN2M0) primary lung adenocarcinoma exhibiting a METex14m by real-time quantitative polymerase chain reaction (RT-qPCR). Given that the tumor load and the size of lymph nodes experienced a significant downstaging after the neoadjuvant treatment of savolitinib with 600mg once a day for 5 weeks, left lower lobectomy and systemic lymphadenectomy were successfully performed. The pathological response was 50% and the final postoperative pathological staging was pT1cN0M0, IA3 (AJCC, 8th edition). The case provides empirical basis for the neoadjuvant treatment with savolitinib in METex14m-positive locally advanced primary lung adenocarcinoma, which will offer some innovative insights and clinical evidence for more effective clinical treatment of neoadjuvant targeted therapy for METex14m-positive NSCLC.

Background: Acute respiratory distress syndrome (ARDS) is a serious respiratory disease, caused by severe infection, trauma, shock, inhalation of harmful gases and poisons and presented with acute-onset and high mortality. Timely and accurate identification will be helpful to the treatment and prognosis of ARDS cases. Herein, we report a case of ARDS caused by occupational exposure to waterproofing spray. To our knowledge, inhalation of waterproofing spray is an uncommon cause of ARDS, and what makes our case special is that we ruled out concurrent infections with some pathogens by using metagenomic next-generation sequencing (mNGS) as an auxiliary diagnosis, which presents the most comprehensive etiological examination of similar reports. Case Presentation: A previously healthy 25 years old delivery man developed hyperpyrexia, chest tightness, cough and expectoration. The symptoms occurred and gradually exacerbated after exposure to a waterproofing spray. The chest computed tomography (CT) finding showed diffuse ground glass and infiltrative shadows in both lungs. The diagnosis of ARDS related to waterproofing spray was established on the basis of comprehensive differential diagnosis and etiological examination. The patient achieved good curative effect after proper systemic glucocorticoid therapy. Conclusions: The diagnosis and differential diagnosis of acute respiratory failure for outdoor workers, such as delivery drivers or hikers, should be considered whether toxic aerosol exposure exists from daily contacts. The case can educate the public that more attention should be paid to avoid exposure to these chemicals by aerosols/ingestion mode and some preventive strategies should be taken in occupational environment. The treatment effect of glucocorticoids is significant in ARDS patients with general chemical damage caused by inhaling toxic gases and substances.

Background: The previous studies have revealed that IL-27 was involved in the pathophysiology of pulmonary inflammatory diseases. However, the role of IL-27 in community-acquired pneumonia (CAP) was unclear. The goal of this research was to explore the associations of serum IL-27 with the severity and prognosis among CAP patients through a prospective cohort study. Methods: The whole of 239 healthy population and 239 CAP patients were enrolled. Fasting blood samples were collected. Inflammatory cytokines were detected using enzyme linked immunosorbent assay (ELISA). Demographic characteristics and clinical information were analyzed. Results: Serum IL-27 on admission was significantly risen in CAP patients compared with control subjects. Besides, serum IL-27 was gradually increased in line with CAP severity scores. Correlative analysis suggested that serum IL-27 was associated with blood routine indices, renal function, liver function, myocardial function and inflammatory cytokines. Linear and logistic regression analyses revealed that serum IL-27 was positively correlated with CAP severity scores. Logistic regression analysis demonstrated that serum higher IL-27 on admission elevated the risks of vasoactive agent usage and longer hospital stay during hospitalization among CAP patients. Conclusions: Serum IL-27 is markedly and positively associated with the severity and poor prognosis among CAP patients, indicating that IL-27 may involve in the pathophysiological process of CAP. Serum IL-27 may be used as a biomarker for diagnosis and prognosis in CAP patients.

As one of the most abundant metals in heavy oils, Ni has suffered so notably increasing impacts from industrial and traffic activities that anthropogenic Ni emissions have altered natural geochemical processes. The coral Ni/Ca has become a reliable proxy for characterizing marine pollution, but this potential has been unexploited for highlighting oil pollution. Here, we utilized a high-resolution record of geochemical parameters (Ni/Ca, δ18O, and δ13C) in a Porites coral of an offshore island in the northern South China Sea to reconstruct of Ni distribution patterns in surface seawater from 1984 to 2015. The coral Ni/Ca ratios exhibit minor fluctuations, except for multiple mutation peaks (0.20 ± 0.42 µmol/mol) during the period from 1984 to 1993. The ratio was low and stable (0.10 ± 0.09 µmol/mol) from 1994 to 2008, and then increased rapidly with significant variations (1.60 ± 4.56 µmol/mol) from 2009 to 2015. The coral Ni/Ca ratios captured all significant Ni discharges, and this demonstrates its potential for recording oil spill episodes. The historical variations in the contributions of Ni indicate that industrial and traffic activities should be responsible for changes in the anthropogenic input. The leaks and consumptions of petroleum likely account for the primary Ni emission sources.
