RESUMO

Macleaya cordata was a kind of traditional herbal medicine, which may a potential substitute for antibiotics. However, the effects of Macleaya cordata on neonatal piglets have rarely been reported. In this study, three groups were designed, including normal saline (Control group, CON), 8 mg/mL Macleaya cordata extract (MCE group, MCE) and 5 mg/mL Chlortetracycline Hydrochloride (CCH group, CCH), to investigate the effects of MCE on growth performance, blood parameters, inflammatory cytokines, regenerating islet-derived 3 gamma (REG3γ) expression and the transcriptomes of neonatal piglets. The results showed that, compared with the control group, MCE significantly increased the average daily gain (p < 0.01); spleen index (p < 0.05) contents of IL-10, TGF-ß, IgG in serum and sIgA in the ileum mucus of neonatal piglets at 7 d and 21 d (p < 0.01). The diarrhoea incidence and serum TNF-α and IFN-γ contents of neonatal piglets at 7 d and 21 d were significantly decreased (p < 0.01). In addition, MCE significantly increased the mRNA expression of TGF-ß, IL-10, and REG3γ (p < 0.01) and significantly decreased the mRNA expression of IL-33, TNF-α and IFN-γ in the ileal mucosa of neonatal piglets at 21 d (p < 0.01). The differentially expressed genes and the signal pathways, related to cytokine generation and regulation, immunoregulation and inflammation were identified. In conclusion, MCE can significantly improve growth performance, reduce diarrhoea incidence, relieve inflammation, improve immune function, and improve disease resistance in neonatal piglets. MCE can be used as a potential substitute for antibiotics in neonatal piglets.

Z-scheme heterostructure-based photocatalysts consist of a reduction photocatalyst and an oxidation photocatalyst, enabling them to possess a high capacity for both reduction and oxidation. However, the coupling reaction between photocatalytic H2 generation through water reduction and sterilization using Z-scheme systems has been rarely reported. Herein, 1D W18O49 nanowires embedded over 2D g-C3N4 nanosheets are well-constructed as an integrated Z-scheme heterojunction. Experimental results and density functional theory calculations not only demonstrate the achievement of efficient interfacial charge separation and transport, leading to prolonged lifetime of photogenerated charge carriers, but also directly confirm the mechanism of Z-scheme charge transfer. As expected, the optimized W18O49/g-C3N4 nanostructure exhibits superior photocatalytic sterilization activity against Staphylococcus aureus as well as excellent H2 generation performance under visible-light irradiation (λ ≥ 420 nm). Due to its nontoxic nature, W18O49/g-C3N4 holds great potential in eradicating bacterial infections in living organisms.

Iron levels are closely related to animals' growth performance and anti-oxidant function. Lactoferrin (LF) is an iron-binding glycoprotein, which can promote the absorption of iron and regulate immune function. This study aimed to clarify the effect of maternal LF supplementation on the iron metabolism of Dahe piglets. Sixty sows (Dahe black, parity 3-4, no significant differences in body weight) were randomly assigned to five groups: control (basal diet with no iron supplementation), supplemented 100 (LF1 group), 200 (LF2 group), or 300 (LF3 group) mg LF/kg in the basal diet, and the basal diet supplemented with 100 (Fe-Gly group) mg Fe/kg as ferrous glycine (Fe-Gly). The serum anti-oxidant parameters of the sows and neonatal piglets were determined. The iron contents, anti-oxidant gene expression levels, and Fe-acquisition genes were detected in the liver, heart, spleen, and other neonatal organs. The results indicated that (1) the LF3 group of sows had the highest serum and colostrum iron contents (P < 0.05). The maternal LF significantly promoted the iron stores in the heart, liver, spleen, and lung of piglets compared with Fe-Gly. (2) The maternal LF increased serum glutathione peroxidase (GSH-Px) and total superoxide dismutase (T-SOD) activities of sows. Compared with other groups, the total anti-oxidant capacity (T-AOC) activity of LF2 groups increased significantly (P < 0.05). (3) LF significantly increased piglet serum GSH-Px, T-SOD, and T-AOC activities (P < 0.05). (4) Gene expression levels of GSH-Px, and SOD in the duodenum and jejunum of the LF2 group were significantly higher than in the Fe-Gly group (P < 0.05), while the expression levels in the liver and heart were lower (P < 0.05). (5) The expression levels of hepcidin and LF in the liver and duodenum of the LF2 group were significantly higher than in the Fe-Gly group (P < 0.05). In conclusion, maternal LF supplementation showed remarkable effects on iron storage in neonatal piglets, and exhibited strong antioxidant activities, it is helpful to prevent the occurrence of iron deficiency, and improves the immune function of animals.

beta-Defensins are cysteine-rich endogenously produced antimicrobial peptides that play an important role in innate immunity. In this study, the expressions of genes porcine beta-defensins-1(pBD-1), pBD-2 and pBD-3 were determined using real-time PCR for Chinese Meishan pigs and Crossbred (Duroc x Yorkshire x Landrace) pigs of 7 days old in various tissues. The results showed that expressions of pBD-1, 2 and 3 of Meishan pigs in most tissues were higher than those of crossbred pigs and main expression sites for pBD-1 and pBD-3 were tongue and oral mucosa in two varieties of pigs, whereas pBD-2 of crossbred pig was mainly expressed in kidney and liver, and pBD-2 of Meishan pigs mainly in tongue and oral mucosa. The higher expression of pBDs might be the reason of Meishan pigs has higher immunity and disease resistance. The mechanisms of this need a further research.
