Emotion perception interacts with how we think and speak, including our concept of emotions. Body expression is an important way of emotion communication, but it is unknown whether and how its perception is modulated by conceptual knowledge. In this study, we employed representational similarity analysis and conducted three experiments combining semantic similarity, mouse-tracking task, and one-back behavioral task with electroencephalography and functional magnetic resonance imaging techniques, the results of which show that conceptual knowledge predicted the perceptual representation of body expressions. Further, this prediction effect occurred at approximately 170 ms post-stimulus. The neural encoding of body expressions in the fusiform gyrus and lingual gyrus was impacted by emotion concept knowledge. Taken together, our results indicate that conceptual knowledge of emotion categories shapes the configural representation of body expressions in the ventral visual cortex, which offers compelling evidence for the constructed emotion theory.

Economic decision-making plays a paramount role in both individual and national interests. Individuals have fairness preferences in economic decision-making, but a proposer's moral-related information may affect fairness considerations. In prior ERP studies, researchers have suggested moral identity influences fairness preferences in the Ultimatum Game (UG), but there are discrepancies in the results. Furthermore, whether role models (individuals whom someone else looks to help decide suitable behaviors), who can modulate people's moral standards, can affect fairness concerns in UG is still understudied. To address the questions, we selected the moral-related statements by eliminating those with illegal information and employed the ERP technique to explore whether the interplay of the proposer's role model and moral-related behavior influenced fairness processing in the modified UG and the corresponding neural mechanisms. We mainly found that the aforementioned interaction effect on proposal considerations in UG could be mirrored in both rejection rates and P300 variations. The results demonstrate that the interaction between the proposer's role model and moral behavior can modulate fairness concerns in UG. Our current work provides new avenues for elucidating the time course of the influencing mechanism of fair distributions in complicated social environments.

Fearful, angry, and disgusted facial expressions are evolutionarily salient and convey different types of threat signals. However, it remains unclear whether these three expressions impact sensory perception and attention in the same way. The present ERP study investigated the temporal dynamics underlying the processing of different types of threatening faces and the impact of attentional resources employed during a perceptual load task. Participants were asked to judge the length of bars superimposed over faces presented in the center of the screen. A mass univariate statistical approach was used to analyze the EEG data. Behaviorally, task accuracy was significantly reduced following exposure to fearful faces relative to neutral distractors, independent of perceptual load. The ERP results revealed that the P1 amplitude over the right hemisphere was found to be enhanced for fearful relative to disgusted faces, reflecting the rapid and coarse detection of fearful cues. The N170 responses elicited by fearful, angry, and disgusted faces were larger than those elicited by neutral faces, suggesting the largely automatic and preferential processing of threats. Furthermore, the early posterior negativity (EPN) component yielded increased responses to fearful and angry faces, indicating prioritized attention to stimuli representing acute threats. Additionally, perceptual load exerted a pronounced influence on the EPN and late positive potential (LPP), with larger responses observed in the low perceptual load condition, indicating goal-directed cognitive processing. Overall, the early sensory processing of fearful, angry, and disgusted faces is characterized by differential sensitivity in capturing attention automatically, despite the importance of these facial signals for survival. Fearful faces produce a strong interference effect and are processed with higher priority than angry and disgusted ones.

Colorectal cancer is a prevalent malignancy, with advanced and metastatic forms exhibiting poor treatment outcomes and high relapse rates. To enhance patient outcomes, a comprehensive understanding of the pathophysiological processes and the development of targeted therapies are imperative. The high heterogeneity of colorectal cancer demands precise and personalized treatment strategies. Colorectal cancer organoids, a three-dimensional in vitro model, have emerged as a valuable tool for replicating tumor biology and exhibit promise in scientific research, disease modeling, drug screening, and personalized medicine. In this review, we present an overview of colorectal cancer organoids and explore their applications in research and personalized medicine, while also discussing potential future developments in this field.

Ensemble coding allows observers to form an average to represent a set of elements. However, it is unclear whether observers can extract an average from a cross-category set. Previous investigations on this issue using low-level stimuli yielded contradictory results. The current study addressed this issue by presenting high-level stimuli (i.e., a crowd of facial expressions) simultaneously (Experiment 1) or sequentially (Experiment 2), and asked participants to complete a member judgment task. The results showed that participants could extract average information from a group of cross-category facial expressions with a short perceptual distance. These findings demonstrate cross-category ensemble coding of high-level stimuli, contributing to the understanding of ensemble coding and providing inspiration for future research.

Objective: This cross-sectional study aimed to explore the association of overweight and inflammatory indicators with breast cancer risk in Chinese patients. Methods: Weight, height, and peripheral blood inflammatory indicators, including white blood cell count (WBC), neutrophil count (NE), lymphocyte count (LY), platelet count (PLT) and the concentration of hypersensitivity C-reactive protein (hsCRP), were collected in 383 patients with benign breast lumps (non-cancer) and 358 patients with malignant breast tumors (cancer) at the First Affiliated Hospital of Soochow University, China, from March 2018 to July 2020. Body mass index (BMI), neutrophil-lymphocyte ratio (NLR), platelet-lymphocyte ratio (PLR) and systemic immune-inflammation index (SII) were determined according to the ratio equation. The correlations among overweight, inflammatory indicators, and the proportion of non-cancer or cancer cases were analyzed. Results: BMI is associated with an increased breast cancer risk. Compared with non-cancer patients, the average WBC count, NE count, NLR, and level of hsCRP were significantly higher in cancer patients. The level of hsCRP was closely associated with the size of malignant breast tumors. Conclusion: We conclude that overweight and high levels of hsCRP may serve as putative risk factors for malignant breast tumors in Chinese women.

The impact of emotional words on the recognition of body expression and the underlying neurodynamic mechanisms remain poorly understood. This study used a classic supraliminal priming paradigm and event related potential (ERP) to investigate the effect of emotion-label words (experiment 1) and emotional verbs (experiment 2) on the recognition of body expressions. The behavioral results revealed that individuals exhibited a higher accuracy in recognizing happy expressions when presented with a happy-label word condition, in contrast to neutral expressions. Furthermore, it was observed that the accuracy of recognizing happy body expressions was reduced when preceded by angry verb priming, compared to happy and neutral priming conditions. Conversely, the accuracy of recognizing angry body expressions was higher in response to angry verb priming than happy and neutral primings. The ERP results showed that, in the recognition of happy body expressions, the P300 amplitude elicited by angry-label words was more positive, while a congruent verb-expression condition elicited more positive P300 amplitude than an incongruent condition in the left hemisphere and midline. However, in the recognition of angry body expressions, the N400 amplitude elicited by a congruent verb-expression condition was smaller than that elicited by an incongruent condition. These results suggest that both abstract emotion-label words and specific emotional verbs influence the recognition of body expressions. In addition, integrating happy semantic context and body expression might occur at the P300 stage, whereas integrating angry semantic context and body expression might occur at the N400 stage. These findings present novel evidence regarding the criticality of emotional context in the recognition of emotions.

Economic decision-making is pivotal to both human private interests and the national economy. People pursue fairness in economic decision-making, but a proposer's moral identity can influence fairness processing. Previous ERP studies have revealed that moral identity has an effect on fairness considerations in the Ultimatum Game (UG), but the findings are inconsistent. To address the issue, we revised the moral-related sentences and used the ERP technique to measure the corresponding neural mechanism. We have observed that the fairness effect in UG can be mirrored in both MFN and P300 changes, whereas the moral identity effect on fairness in UG can be reflected by MFN but not P300 changes. These findings indicate that the moral identity of the proposer can modulate fairness processing in UG. The current study opens new avenues for clarifying the temporal course of the relationship between the proposer's moral identity and fairness in economic decision-making, which is beneficial for understanding the influencing mechanism of fairness processing and fair allocations in complex social contexts.

Implicit emotion regulation provides an effective means of controlling emotions triggered by a single face without conscious awareness and effort. Crowd emotion has been proposed to be perceived as more intense than it actually is, but it is still unclear how to regulate it implicitly. In this study, participants viewed sets of faces of varying emotionality (e.g. happy to angry) and estimated the mean emotion of each set after being primed with an expressive suppression goal, a cognitive reappraisal goal, or a neutral goal. Faster discrimination for happy than angry crowds was observed. After induction of the expressive suppression goal instead of the cognitive reappraisal goal, augmented N170 and early posterior negativity (EPN) amplitudes, as well as attenuated late positive potential (LPP) amplitudes, were observed in response to happy crowds compared to the neutral goal. Differential processing of angry crowds was not observed after the induction of both regulatory goals compared to the neutral goal. Our findings thus reveal the happy-superiority effect and that implicit induction of expressive suppression improves happy crowd emotion recognition, promotes selective coding, and successfully downregulates the neural response to happy crowds.

Intestinal epithelia express two long myosin light-chain kinase (MLCK) splice variants, MLCK1 and MLCK2, which differ by the absence of a complete immunoglobulin (Ig)-like domain 3 within MLCK2. MLCK1 is preferentially associated with the perijunctional actomyosin ring at steady state, and this localization is enhanced by inflammatory stimuli including tumor necrosis factor (TNF). Here, we sought to identify MLCK1 domains that direct perijunctional MLCK1 localization and their relevance to disease. Ileal biopsies from Crohn's disease patients demonstrated preferential increases in MLCK1 expression and perijunctional localization relative to healthy controls. In contrast to MLCK1, MLCK2 expressed in intestinal epithelia is predominantly associated with basal stress fibers, and the two isoforms have distinct effects on epithelial migration and barrier regulation. MLCK1(Ig1-4) and MLCK1(Ig1-3), but not MLCK2(Ig1-4) or MLCK1(Ig3), directly bind to F-actin in vitro and direct perijunctional recruitment in intestinal epithelial cells. Further study showed that Ig1 is unnecessary, but that, like Ig3, the unstructured linker between Ig1 and Ig2 (Ig1/2us) is essential for recruitment. Despite being unable to bind F-actin or direct recruitment independently, Ig3 does have dominant negative functions that allow it to displace perijunctional MLCK1, increase steady-state barrier function, prevent TNF-induced MLCK1 recruitment, and attenuate TNF-induced barrier loss. These data define the minimal domain required for MLCK1 localization and provide mechanistic insight into the MLCK1 recruitment process. Overall, the results create a foundation for development of molecularly targeted therapies that target key domains to prevent MLCK1 recruitment, restore barrier function, and limit inflammatory bowel disease progression.

Recognizing the emotions of faces in a crowd is crucial for understanding overall behavior and intention as well as for smooth and friendly social interactions. However, it is unclear whether the spatial frequency of faces affects the discrimination of crowd emotion. Although high- and low-spatial-frequency information for individual faces is processed by distinct neural channels, there is a lack of evidence on how this applies to crowd faces. Here, we used functional magnetic resonance imaging (fMRI) to investigate neural representations of crowd faces at different spatial frequencies. Thirty-three participants were asked to compare whether a test face was happy or more fearful than a crowd face that varied in high, low, and broad spatial frequencies. Our findings revealed that fearful faces with low spatial frequencies were easier to recognize in terms of accuracy (78.9%) and response time (927 ms). Brain regions, such as the fusiform gyrus, located in the ventral visual stream, were preferentially activated in high spatial frequency crowds, which, however, were the most difficult to recognize behaviorally (68.9%). Finally, the right inferior frontal gyrus was found to be better activated in the broad spatial frequency crowds. Our study suggests that people are more sensitive to fearful crowd faces with low spatial frequency and that high spatial frequency does not promote crowd face recognition.

The eye region conveys considerable information regarding an individual's emotions, motivations, and intentions during interpersonal communication. Evidence suggests that the eye regions of an individual expressing emotions can capture attention more rapidly than the eye regions of an individual in a neutral affective state. However, how attentional resources affect the processing of emotions conveyed by the eye regions remains unclear. Accordingly, the present study employed a dual-target rapid serial visual presentation task: happy, neutral, or fearful eye regions were presented as the second target, with a temporal lag between two targets of 232 or 696 ms. Participants completed two tasks successively: Task 1 was to identify which species the upright eye region they had seen belonged to, and Task 2 was to identify what emotion was conveyed in the upright eye region. The behavioral results showed that the accuracy for fearful eye regions was lower than that for neutral eye regions under the condition of limited attentional resources; however, accuracy differences across the three types of eye regions did not reach significance under the condition of adequate attentional resources. These findings indicate that preferential processing of fearful expressions is not automatic but is modulated by available attentional resources.

Although the eye region has been found to convey sufficient information for emotional recognition and interpersonal communication, little is known regarding the extent to which the prioritized processing of emotional eye regions relies on available attentional resources. To address this issue, the present study used a dual-target rapid serial visual presentation task in which the perceptual load levels of the first target (T1), as well as the valence of the second target (T2), were manipulated. In addition to the traditional event-related potential (ERP) analysis method, the mass univariate statistics approach was employed. Behaviorally, both happy and fearful eye regions were recognized more accurately than neutral eye regions, regardless of the T1 perceptual load. ERP findings revealed an enhanced N170 amplitude for fearful eye regions compared to neutral eye regions, confirming the preferential and automatic processing of fearful signals at the early sensory stage. The late positive potential component exhibited enhanced responses to fearful and happy eye regions, suggesting the amplified representation consolidation in working memory. Collectively, these findings indicate that isolated eye regions are processed automatically to a higher degree owing to their perceptual and motivational significance.

In recent years, the processing mechanism of group expression has gradually gained the attention of researchers owing to its high ecological validity. However, research on the relationship between ensemble coding and individual representation is still in the early stage of the investigation, with many studies remaining at the behavioral level and findings varying widely. Based on our behavioral research (Experiment 1), we used EEG measures (Experiments 2A and 2B) to investigate the relationship between summary and object representations by manipulating the exposure time of crowd emotions. The behavioral results indicated that participants performed better in judging emotions of multiple faces compared to a single face during the shorter exposure time, whereas the reverse occurred during the long exposure time. Furthermore, ERP results revealed that the N2pc effect was not affected by the number of faces in the short exposure time; however, as the exposure time increased, the N2pc increased as a function of the number of faces. The findings of the current investigation align with time-dependent assumption, indicating that during short time of visual processing, although individual representations may not be fully developed, ensemble representations are initially established. With longer processing times, detailed individual representations become complete and take precedence.

Observational evidences have suggested that the surface scums of cyanobacterial harmful blooms (CyanoHABs) are highly patchy, and their spatial patterns can vary significantly within hours. This stresses the need for the capacity to monitor and predict their occurrence with better spatiotemporal continuity, in order to understand and mitigate their causes and impacts. Although polar-orbiting satellites have long been used to monitor CyanoHABs, these sensors cannot be used to capture the diurnal variability of the bloom patchiness due to their long revisit periods. In this study, we use the Himawari-8 geostationary satellite to generate high-frequency time-series observations of CyanoHABs on a sub-daily basis not possible from previous satellites. On top of that, we introduce a spatiotemporal deep learning method (ConvLSTM) to predict the dynamics of bloom patchiness at a lead time of 10 min. Our results show that the bloom scums were highly patchy and dynamic, and the diurnal variability was assumed to be largely associated with the migratory behavior of cyanobacteria. We also show that, ConvLSTM displayed fairly satisfactory performance with promising predictive capability, with Root Mean Square Error (RMSE) and determination coefficient (R2) varying between 0.66∼1.84 µg/L and 0.71∼0.94, respectively. This suggests that, by adequately capturing spatiotemporal features, the diurnal variability of CyanoHABs can be well learned and inferred by ConvLSTM. These results may have important practical implications, because they suggest that spatiotemporal deep learning integrated with high-frequency satellite observations could provide a new methodological paradigm in nowcasting of CyanoHABs.

Decision-making under time pressure may better reflect an individual's response preference, but few studies have examined whether individuals choose to be more selfish or altruistic in a scenario where third-party punishment is essential for maintaining social norms. This study used a third-party punishment paradigm to investigate how time pressure impacts on individuals' maintenance of behavior that follows social norms. Thirty-one participants observed a Dictator Game and had to decide whether to punish someone who made what was categorized as a high unfair offer by spending their own Monetary units to reduce that person's payoff. The experiment was conducted across different offer conditions. The study results demonstrated that reaction times were faster under time pressure compared with no time pressure. Time pressure was also correlated with less severe punishment. Specifically, participants were less likely to punish the dictator under time pressure compared with no time pressure when the offer was categorized as a high unfair. The findings suggested that individuals in these game conditions and under time pressure do not overcome their pro-selves and that time pressure weakens an individual's willingness to punish high unfair offers.

Osteosarcoma is the most common primary malignant bone tumour in children and adolescents. Chemoresistance leads to poor responses to conventional therapy in patients with osteosarcoma. The discovery of novel effective therapeutic targets and drugs is still the main focus of osteosarcoma research. Nuclear receptors (NRs) have shown substantial promise as novel therapeutic targets for various cancers. In the present study, we performed a drug screen using 29 chemicals that specifically target 17 NRs in several different human osteosarcoma and osteoblast cell lines. The retinoic acid receptor beta (RARb) antagonist LE135, peroxisome proliferator activated receptor gamma (PPARg) antagonist T0070907, liver X receptor (LXR) agonist T0901317 and Rev-Erba agonist SR9011 significantly inhibited the proliferation of malignant osteosarcoma cells (U2OS, HOS-MNNG and Saos-2 cells) but did not inhibit the growth of normal osteoblasts. The effects of these NR modulators on osteosarcoma cells occurred in a dose-dependent manner and were not observed in NR-knockout osteosarcoma cells. These NR modulators also significantly inhibited osteosarcoma growth in vivo and enhanced the antitumour effect of doxorubicin (DOX). Transcriptomic and immunoblotting results showed that these NR modulators may inhibit the growth of osteosarcoma cells by regulating the PI3K/AKT/mTOR and ERK/mTOR pathways. DDIT4, which blocks mTOR activation, was identified as one of the common downstream target genes of these NRs. DDIT4 knockout significantly attenuated the inhibitory effects of these NR modulators on osteosarcoma cell growth. Together, our results revealed that modulators of RARb, PPARg, LXRs and Rev-Erba inhibit osteosarcoma growth both in vitro and in vivo through the mTOR signaling pathway, suggesting that treatment with these NR modulators is a novel potential therapeutic strategy.

In daily life, individuals need to recognize and update emotional information from others' changing body expressions. However, whether emotional bodies can enhance working memory (WM) remains unknown. In the present study, participants completed a modified n-back task, in which they were required to indicate whether a presented image of an emotional body matched that of an item displayed before each block (0-back) or two positions previously (2-back). Each block comprised only fear, happiness, or neutral. We found that in the 0-back trials, when compared with neutral body expressions, the participants took less time and showed comparable ceiling effects for accuracy in happy bodies followed by fearful bodies. When WM load increased to 2-back, both fearful and happy bodies significantly facilitated WM performance (i.e., faster reaction time and higher accuracy) relative to neutral conditions. In summary, the current findings reveal the enhancement effect of emotional body expressions on WM and highlight the importance of emotional action information in WM.

Although emotional expressions conveyed by the eye regions are processed efficiently, little is known regarding the relationship between emotional processing of isolated eye regions and temporal attention. In this study, we conducted three rapid serial visual presentation (RSVP) experiments with varying task demands (emotion discrimination, eye detection, eyes ignored) related to the first target (T1) to investigate how the perception of emotional valence in the eye region (T1: happy, neutral, fearful) impacts the identification of a second target (T2: neutral houses). Event-related potential (ERP) findings indicated that fearful stimuli reliably increased N170 amplitude regardless of the emotional relevance of task demands. The P3 component exhibited enhanced responses to happy and fearful stimuli in the emotion discrimination task and to happy eye regions in the eye detection task. Analysis of T2-related ERPs within the attentional blink period revealed that T2 houses preceded by fearful and happy stimuli elicited larger N2 and P3 amplitudes than those preceded by neutral stimuli only in the emotion discrimination task. Together, these findings indicate that attention to affective content conveyed by the eyes can not only amplify the perceptual analysis of emotional eye regions but also facilitate the processing of a subsequent target.
