RESUMO

The concentrations, distribution, and ecological risks of 24 typical antibiotics in Hong Kong rivers and seawater were investigated using high-performance liquid chromatography coupled with electrospray ionization tandem mass spectrometry (UHPLC-EI-MS/MS). The results showed that the select antibiotics were widely distributed in the study area. Among the target antibiotics, the detection rate of tetracyclines (TCs) was 100%, which indicated the widespread use of TCs in Hong Kong. The detection rates of sulfonamides (SAs) (57.1-100%), fluoroquinolones (FQs) (78.6-100%), roxithromycin (RTM) (50%) and novobiocin (NOV) (50%) were all above 50%. Compared with river water (7.9-114.26 ng/L, medium: 27.7 ng/L), concentrations of the most antibiotics in seawater (9.5-32.0 ng/L, medium: 13.3 ng/L) were lower; seawater concentrations were similar to those reported from other coastal cities, such as Guangzhou and Zhuhai in China, which implied that the source of marine antibiotic pollution may be the nearby rivers, and the vastness of the ocean causes environmental dilution of antibiotics. According to the ratio of the measured environmental concentration (MEC) to the predicted no-effect concentration (PNEC), ofloxacin (OFX) (average risk quotient: 1.94E-01) and ciprofloxacin (CFX) (average risk quotient: 3.53E-01) posed medium to high ecological risk in most places, whereas other antibiotics posed lower risk. In Yuen Long, where there were many livestock farms nearby, the detected concentration of antibiotics was higher, indicating that livestock wastewater may be the major reason for the increase in antibiotic levels in this area. In general, the detected concentration of antibiotics in Hong Kong was lower than that in the United States, Japan, the United Kingdom, and coastal areas of China, but the long-term existence of low concentrations of antibiotics also poses great risks. According to the risk assessment, Hong Kong should pay more attention to the use of FQs (e.g., OFX and CFX) in the future.

RESUMO

Arsenic (As) in the aquatic environment is a considerable environmental issue, previous studies have reported the toxic effects of low concentrations (≤ 150 µg/L) of As on fish. However, limited information is available regarding the impact of low levels of As on apoptosis. To evaluate this, zebrafish embryos were exposed to different concentrations (0, 25, 50, 75, and 150 µg/L) of As (arsenite [AsIII] and arsenate [AsV]) for 120 h. Our results indicated that low concentrations of AsIII exposure significantly inhibited the survival of zebrafish larvae, and significantly increased the transcription of Caspase-9 and Caspase-3, the ratio of Bax/Bcl-2 transcription, and protein levels of Caspase-3. In contrast, AsV decreased the ratios of Bax/Bcl-2 transcription and protein levels, as well as protein levels of Caspase-3. Our data demonstrated that AsIII and AsV exert different toxic effects, AsIII induced apoptosis via the mitochondrial pathway and the extrinsic pathway, while AsV induced apoptosis only via the mitochondrial pathway.

RESUMO

Gill, as the organ of fish to contact most directly with xenobiotics, suffered more threat. To evaluate the impact of arsenite (AsIII) on the gill of fish, we measured the antioxidative responses (superoxide dismutase (SOD) and catalase (CAT) activities) and oxidative damage (malondialdehyde (MDA) content), histological changes and mRNA transcriptional responses of zebrafish gill, after exposure to AsIII (0, 10, 50, 100, and 150 µg L-1) solutions for 28 days. We found that AsIII increased the activities of CAT by 46%-87%, decreased the activities of SOD and the contents of MDA by 19% and 21%-32%. Furthermore, CuZnSOD and MnSOD mRNA transcription levels were also inhibited, decreasing by 62%-82% and 70%-77%. Besides, ≥ 100 µg L-1 AsIII also caused histological changes (a loss of mucus and desquamation in the surface of the epithelial cells) on zebrafish gill. These results showed that low concentrations of AsIII influenced biochemical and physiological performances of fish gill, which probably aggravates the toxic effect of AsIII on fish.

RESUMO

Low concentrations of arsenic (As) contamination in aquatic environment is a worldwide issue, which is of great concern. To evaluate the impact of low concentrations of As on zebrafish, we measured the growth, antioxidant enzymes including superoxide dismutase (SOD) and catalase (CAT), oxidative damage (malondialdehyde, MDA) and apoptosis-related genes (nrf2, p53 and c-jun) of adult zebrafish after exposing to different AsIII concentrations (0, 10, 50, 100 or 150 µg L-1) for 28 d. Results indicated that exposure to low AsIII concentrations decreased the zebrafish weight by 14%, increased the activities of SOD and CAT by 23-41% and 31-59%, decreased the contents of MDA by 29-54%, and modulated transcription of apoptosis related genes. Our study showed that chronic exposure to AsIII concentrations <150 µg L-1 generated oxidative stress and damage on zebrafish, and altered apoptosis-related genes in zebrafish.

RESUMO

The major objective of the present study was to investigate the contribution of major biomolecules, including protein, carbohydrates and lipids, in predicting DBPs formation upon chlorination of algal cells. Three model compounds, including bovine serum albumin (BSA), starch and fish oil, as surrogates of algal-derived proteins, carbohydrates and lipids, and cells of three algae species, representing blue-green algae, green algae, and diatoms, were chlorinated in the laboratory. The results showed that BSA (27 microg mg(-1) C) and fish oil (50 microg mg(-1) C) produced more than nine times higher levels of chloroform than starch (3 microg mg(-1) C). For the formation of HAAs, BSA was shown to have higher reactivity (49 microg mg(-1) C) than fish oil and starch (5 microg mg(-1) C). For the algal cells, Nitzschia sp. (diatom) showed higher chloroform yields (48 microg mg(-1) C) but lower HAA yields (43 microg mg(-1) C) than Chlamydomonas sp. (green algae) (chloroform: 34 microg mg(-1) C; HAA: 62 microg mg(-1) C) and Oscillatoria sp. (blue-green algae) (chloroform: 26 microg mg(-1) C; HAA: 72 microg mg(-1) C). The calculated chloroform formation of cells from the three algal groups, based on their biochemical compositions, was generally consistent with the experimental data, while the predicted values for HAAs were significantly lower than the observed ones. As compared to humic substances, such as humic and fulvic acids, the algal cells appeared to be important precursors of dichloroacetic acid.

RESUMO

The main objective of this study was to investigate occurrence of polycyclic aromatic hydrocarbons (PAHs) in the sources of the drinking water supply of Hong Kong. The main emphasis was on the Dongjiang River in mainland China which is the major source, supplying 80% of the total consumption in Hong Kong (the remaining 20% is obtained from rain water). Sediments were collected from four sites along the Dongjiang River and four reservoirs in Hong Kong during both the dry and wet weather seasons. The concentrations of total PAHs in the sediments ranged between 36 and 539 microg/kg dry wt. The lower levels were detected at the upstream site on the Dongjiang River and at the reservoirs in Hong Kong (44-85 microg/kg dry wt), while the mid- and downstream sites on the Dongjiang River were more polluted (588-658 microg/kg dry wt). Examination of the PAH profiles revealed that the mid- and downstream sections of the Dongjiang River contained high percentages of 4,5,6-ring PAHs, similar to the amounts of atmospheric particulate matter and road dust collected during the dry weather season from the Pearl River Delta region as reported in the literature. Seasonal changes were revealed in the reservoirs of Hong Kong, with higher PAH levels in the wet weather season than in the dry weather season. For those reservoirs in Hong Kong that store water from the Dongjiang River, a distinct seasonal pattern was also observed, namely, that under dry weather season conditions the PAHs found in the sediments were primarily from petrogenic source, while under wet weather season conditions they were from pyrolytic sources. No such pattern was detected in the reservoirs which stored only rain water.
