Ensiling of high-moisture agro-industrial wastes with dry roughage as animal feeds may be a secure way to reduce the feeding cost. The objective of this study was to investigate the effects of either sugarbeet or citrus pulp ensiled with ground corncobs and used as a replacement of maize silage or maize fodder in total mixed ration (TMR) for fattening of buffalo calves. Twenty male calves of Nili-Ravi buffalo (aged 14 ± 2 months weighing 133 ± 5 kg) were randomly assigned to one of four iso-nutritious (12% CP and 69% TDN) experimental diets with 50:50 forage to concentrate ratio (dry matter basis) as: (1) TMR maize silage, (2) TMR sugarbeet pulp ensiled with corncobs, (3) TMR citrus pulp ensiled with corncobs, and (4) TMR green maize fodder with wheat straw. All calves received TMRs for ad libitum intake twice daily for 80-day period. Daily feed intake, fortnightly body weights, and digestibility and N balance study during last 15 days were recorded of individual animals. The calves fed TMR sugarbeet pulp silage got highest daily gain (856 g/d, P < 0.05) than citrus pulp silage (776 g/d) or green maize fodder (704 g/d). Average intake of nutrients was same among all the calves. Feed conversion ratio and digestibility of DM, NDF, and ADF were higher (P < 0.05) with TMR sugarbeet pulp silage compared to other TMRs. Economic benefit (output/input) with TMR sugarbeet pulp silage had 15.46, 34.27, and 37.36% higher effect for fattening of buffalo calves compared to citrus pulp silage, green maize fodder, and maize silage-based TMRs, respectively. In conclusion, sugarbeet pulp ensiled with ground corncobs is simple and appropriate method for silage making and its inclusion up to 50% of DM in TMR had pronounced economic impact to the production of buffalo calves.

Spirulina has been used as a food source from many years for human and animals because of its superior profile of nutrition. Spirulina (SPA) having antiviral activities can boost immune system, and better nutritional profile can increase growth performance by improving gut morphology. This study was conducted to investigate the impact of spirulina supplementation in ration on the humoral and cellular immune response, gut histomorphology, and economics of broiler production. A total of 120 day-old chicks were assigned to 4 dietary treatment; SPA0, SPA1, SPA2, and SPA3, each with 3 replicates having 10 birds in each replicate until 42 days of age. Treatment SPA0 was kept as control, and the birds were fed on basal ration, while the other treatments SPA1, SPA2, and SPA3 were supplemented with 1, 1.5, and 2 g spirulina/kg feed, respectively. Dietary supplementation of SPA3 significantly (P < 0.05) improved feed intake (8.95%), weight gain (12.5%), feed conversion ratio, and dressing percentage than that of control. Dietary treatment of SPA had comparatively low level of heterophil and higher level of lymphocytes than the control treatment. Spirulina supplementation had significant (P < 0.05) effect over antibody titer, against ND vaccine. The supplementation of SPA in diet at all levels has a significant effect on lengths of villi, and it increased number of goblet cells. The SPA3 showed significant increase in net and gross return of birds followed by SPA2, SPA1, and SPA0. It was concluded that spirulina improved growth performance, gut integrity, and immunity with better economics in broiler production.

Aflatoxin M1 contamination of milk in Pakistan, like many developing countries, is poorly understood. The present study was therefore conducted to determine AFM1 contamination of milk and its contributory factors in Pakistan. We sampled milk and feedstuffs from 450 peri-urban dairy farms in seven major cities following a cross-sectional study design. Analysis of milk using ELISA revealed high contamination with an overall average of 3164.5 ng of AFM1/L, and significant differences (p < 0.001) between cities. The milk sampled from Gilgit, in northern hilly areas, had an average AFM1 level of 92.5 ng/L. Milk from other cities had 3529.7 ng/L average contamination, with only 5.7% samples qualifying the maximum tolerable limit of 500 ng of AFM1/L. Heavy mean aflatoxin contamination was found in bakery waste (724.6 µg/kg), and cottonseed cake (600.8 µg/kg). Rest of the other feedstuffs had moderate to low mean aflatoxin contamination, ranging from 66.0 µg/kg in maize stover to 3.4 µg/kg in wheat bran. The mean aflatoxin level in commercial dairy concentrates was 32.7 µg/kg. About 80% of the total aflatoxin intake of dairy animals was contributed by cottonseed cake alone due to its high aflatoxin contamination and proportion in dairy rations. On-farm storage time of oilseed cakes varied (p < 0.01) in different cities but was not associated with aflatoxin contamination. The exceptionally high AFM1 contamination suggests that milk from peri-urban dairy farms is a serious public health threat in Pakistan. This situation can be mitigated by reducing aflatoxin contamination in cottonseed cake and promoting the use of commercial concentrates and other feedstuffs with low contamination.

Conformational changes in the light illuminated intermediate (pB) of photoactive yellow protein (PYP) were studied from a viewpoint of the diffusion coefficient (D) change of several N-truncated PYPs, which lacked the N-terminal 6, 15, or 23 amino acid residues (T6, T15, and T23, respectively). For intact PYP (i-PYP), D of pB (D(pB)) was approximately 11% lower than that (D(pG)) of the ground state (pG) species. The difference in D (D(pG) - D(pB)) decreased upon cleavage of the N-terminal region in the order of i-PYP>T6>T15>T23. This trend clearly showed that conformational change in the N-terminal group is the main reason for the slower diffusion of pB. This slower diffusion was interpreted in terms of the unfolding of the two alpha-helices in the N-terminal region, increasing the intermolecular interactions due to hydrogen bonding with water molecules. The increase in friction per one residue by the unfolding of the alpha-helix was estimated to be 0.3 x 10(-12) kg/s. The conformational change in the N-terminal group upon photoillumination is discussed.

Heat capacity changes of short-lived transient species in different time ranges were measured for the first time by using the thermal component of the transient grating and transient lens signals at various temperatures. This method was applied to the transient intermediates of Photoactive Yellow Protein (PYP). The temperature dependence of the enthalpy change shows that the heat capacity of the short-lived intermediate pR2 (also called I1 or PYP(L)) species is the same as that of the ground state (pG) species within our experimental accuracy, whereas that of the long-lived intermediate pB (I2 or PYP(M)) is much larger (2.7 +/- 0.4 kJ/mol K) than that of pG. The larger heat capacity is interpreted in terms of the conformational change of the pB species such as melted conformation and/or exposure of the nonpolar residues to the aqueous phase. This technique can be used for photochemical reactions in general to investigate the conformational change and the hydrophobic interaction in a time domain.

RESUMO

A new type of biosensor is presented for the first time. This method is based on the diffusion measurement in the time domain by the transient grating method. As the first demonstration of this new method, various intermolecular interactions with a reaction intermediate of photoactive yellow protein, such as the protein-protein interaction, protein-DNA interaction, and protein-small molecule binding are detected. The characteristic advantages and limitations are summarized.
