RESUMO

Water electrolysis to produce hydrogen (H2) using renewable energy is one of the most promising candidates for realizing carbon neutrality, but its reaction kinetics is hindered by sluggish anodic oxygen evolution reaction (OER). Ruthenium (Ru) in its high-valence state (oxide) provides one of the most active OER sites and is less costly, but thermodynamically unstable. The strong interaction between Ru nanoparticles (NPs) and nickel hydroxide (Ni(OH)2) is leveraged to directly form Ru-Ni(OH)2 on the surface of a porous nickel foam (NF) electrode via spontaneous galvanic replacement reaction. The formation of Ru─O─Ni bonds at the interface of the Ru NPs and Ni(OH)2 (Ru-Ni(OH)2) on the surface oxidized NF significantly enhance stability of the Ru-Ni(OH)2/NF electrode. In addition to OER, the catalyst is active enough for the hydrogen evolution reaction (HER). As a result, it is able to deliver overpotentials of 228 and 15 mV to reach 10 mA cm-2 for OER and HER, respectively. An industry-scale evaluation using Ru-Ni(OH)2/NF as both OER and HER electrodes demonstrates a high current density of 1500 mA cm-2 (OER: 410 mV; HER: 240 mV), surpassing commercial RuO2 (OER: 600 mV) and Pt/C based performance (HER: 265 mV).

Despite the different perspectives by diverse research sectors spanning several decades, aging research remains uncharted territory for human beings. Therefore, we investigated the transcriptomic characteristics of eight male healthy cynomolgus macaques, and the annual sampling was designed with two individuals in four age groups. As a laboratory animal, the macaques were meticulously shielded from all environmental factors except aging. The results showed recent findings of certain immune response and the age-associated network of primate immunity. Three important aging patterns were identified and each gene clusters represented a different immune response. The increased expression pattern was predominantly associated with innate immune cells, such as Neutrophils and NK cells, causing chronic inflammation with aging whereas the other two decreased patterns were associated with adaptive immunity, especially "B cell activation" affecting antibody diversity of aging. Furthermore, the hub gene network of the patterns reflected transcriptomic age and correlated with human illness status, aiding in future human disease prediction. Our macaque transcriptome profiling results offer systematic insights into the age-related immunological features of primates.

We fabricated MAPbI3 perovskite thin films with ZnO on a glass substrate, in which a passivation layer (Phenethylammonium iodide (PEAI); p-methoxyphenethylammonium iodide (CH3O-PEAI); 2-methoxyethylammonium iodide (MEAI)) was inserted between two layers. In order to understand the effect of the insertion of each passivation material on the transfer efficiency of the photo-generated electrons from MAPbI3 to ZnO, we observed the near-field heterodyne transient grating (NF-HD-TG) responses of each film and investigated the component arising from the recombination of the trapped electrons at the ZnO surface. Based on the accelerated recombination between photo-generated holes remaining in the MAPbI3 layer and surface-trapped electrons in ZnO and the increase in the number of the trapped electrons in ZnO when either CH3O-PEAI or PEAI was applied, we successfully revealed that the charge transfer efficiency was enhanced by the insertion of the passivation materials including a benzene ring stabilizing the defect states. Particularly, it was demonstrated that CH3O-PEAI showed the highest increase in the charge transfer efficiency, which could be attributed to the high electron density in the benzene ring, resulting from the existence of the electron donating group, CH3O, and its role in the effective transition from 3D to 2D perovskite phases.

Air pollution caused by the construction industry in the form of particulate matter (PM) has increased to an alarming level. The effects on the health of construction workers are found to be hazardous despite the current advancement in construction methods and practices. In particular, the efficiency of existing control measures for reducing PM from various construction activities has not been improved to the desired level. This study investigated the factors that influence the efficiency of a sprinkler system-based control measure when water spraying and dust suppressant solutions are used. The real-time PM exposure was measured during hollow-block cutting activity using Alphanese OPC-N3 sensors in dust chamber. The dust suppressant suppresses dust particles by initially forming a solidified film on the particle surface, and the high cohesion of this film enhances the suppression rate by promoting dust particle coagulation. It was observed that when using a dust suppressant, the PM concentration at 100 bar exceeded concentrations at other pressures, resulting in increased efficacy in reducing PM10. Additionally, water spraying at 115 bar was determined to be the optimal control measure for achieving a significant percentage of PM reduction in a shorter period. These findings can be highly beneficial if the water sprinkler system can be modified into a smart mobility-based sprinkler system either ground-based or drone-based at construction sites in improving PM reduction efficiency particularly on high PM emitting activities.

Background: Guidelines for switching to triple combination therapy directly after monotherapy failure are limited. This study investigated the efficacy, long-term sustainability, and safety of either mono or dual add-on therapy using alogliptin and pioglitazone for patients with type 2 diabetes mellitus (T2DM) who did not achieve their target glycemic range with metformin monotherapy. Methods: The Practical Evidence of Antidiabetic Combination Therapy in Korea (PEAK) was a multicenter, placebo-controlled, double-blind, randomized trial. A total of 214 participants were randomized to receive alogliptin+pioglitazone (Alo+Pio group, n=70), alogliptin (Alo group, n=75), or pioglitazone (Pio group, n=69). The primary outcome was the difference in glycosylated hemoglobin (HbA1c) levels between the three groups at baseline to 24 weeks. For durability, the achievement of HbA1c levels <7% and <6.5% was compared in each group. The number of adverse events was investigated for safety. Results: After 24 weeks of treatment, the change of HbA1c in the Alo+Pio, Alo, and Pio groups were -1.38%±0.08%, -1.03%±0.08%, and -0.84%±0.08%, respectively. The Alo+Pio group had significantly lower HbA1c levels than the other groups (P=0.0063, P<0.0001) and had a higher proportion of patients with target HbA1c achievement. In addition, insulin sensitivity and ß-cell function, lipid profiles, and other metabolic indicators were also improved. There were no significant safety issues in patients treated with triple combination therapy. Conclusion: Early combination triple therapy showed better efficacy and durability than the single add-on (dual) therapy. Therefore, combination therapy with metformin, alogliptin, and pioglitazone is a valuable early treatment option for T2DM poorly controlled with metformin monotherapy.

The CRISPR-Cas nickase system for genome editing has attracted considerable attention owing to its safety, efficiency, and versatility. Although alternative effectors to Cas9 have the potential to expand the scope of genome editing, their application has not been optimized. Herein, we used an enhanced CRISPR-Cas12a nickase system to induce mutations by targeting genes in a human-derived cell line. The optimized CRISPR-Cas12a nickase system effectively introduced mutations into target genes under a specific directionality and distance between nickases. In particular, the single-mode Cas12a nickase system can induce the target-specific mutations with less DNA double-strand breaks. By inducing mutations in the Thymine-rich target genes in single- or dual-mode, Cas12a nickase compensates the limitations of Cas9 nickase and is expected to contribute to the development of future genome editing technologies.

Porous polymer networks (PPNs) are promising candidates as photocatalysts for hydrogen production. Constructing a donor-acceptor structure is known to be an effective approach for improving photocatalytic activity. However, the process of how a functional group of a monomer can ensure photoexcited charges transfer and improve the hydrogen evolution rate (HER) has not yet been studied on the molecular level. Herein, we design and synthesize two kinds of triazatruxene (TAT)-based PPNs: TATR-PPN with a hexyl (R) group and TAT-PPN without the hexyl group, to understand the relationship between the presence of the functional group and charge transfer. The hexyl group on the TAT unit was found to ensure the transfer of photoexcited electrons from a donor unit to an acceptor unit and endowed the TATR-PPN with stable hydrogen production.

Determining the activity of lysosomal ß-hexosaminidase in cells is of great importance for understanding the roles that these enzymes play in pathophysiological events. Herein, we designed the new fluorescent probe, ßGalNAc-Rhod-CM(NEt2), which consisted of a ßGalNAc-linked rhodol unit serving as a ß-hexosaminidase reactive fluorogenic moiety and a N,N'-diethylaminocoumarin (CM(NEt2)) group acting as a fluorescence marker for determining the degree of cell permeabilization. Treatment of ßGalNAc-Rhod-CM(NEt2) with ß-hexosaminidase promoted generation of Rhod-CM(NEt2), thereby leading to an increase in the intensity of fluorescence of Rhod. However, this probe did not respond to the functionally related glycosidase, O-GlcNAcase. The detection limit of ßGalNAc-Rhod-CM(NEt2) for ß-hexosaminidase was determined to be 0.52 nM, indicating that it has high sensitivity for this enzyme. Furthermore, the probe functioned as an excellent fluorogenic substrate for ß-hexosaminidase with kcat and Km values of 17 sec-1 and 22 µM, respectively. The results of cell studies using ßGalNAc-Rhod-CM(NEt2) showed that levels of ß-hexosaminidase activity in cells can be determined by measuring the intensity of fluorescence arising from Rhod and that the intensity of fluorescence of CM(NEt2) can be employed to determine the degree of cell permeabilization of the probe. Utilizing the new probe, we assessed ß-hexosaminidase activities in several types of cells and evaluated the effect of glucose concentrations in culture media on the activity of this enzyme.

The development of covalent organic frameworks (COFs) with efficient charge transport is of immense interest for applications in optoelectronic devices. To enhance COF charge transport properties, electroactive building blocks and dopants can be used to induce extended conduction channels. However, understanding their intricate interplay remains challenging. We designed and synthesized a tailor-made COF structure with electroactive hexaazatriphenylene (HAT) core units and planar dioxin (D) linkages, denoted as HD-COF. With the support of theoretical calculations, we found that the HAT units in the HD-COF induce strong, eclipsed π-π stacking. The unique stacking of HAT units and the weak in-plane conjugation of dioxin linkages leads to efficient anisotropic charge transport. We fabricated HD-COF films to minimize the grain boundary effect of bulk COFs, which resulted in enhanced conductivity. As a result, the HD-COF films showed an electrical conductivity as high as 1.25 S cm-1 after doping with tris(4-bromophenyl)ammoniumyl hexachloroantimonate.

Covalent organic frameworks (COFs) have emerged as a promising platform for photocatalysts. Their crystalline porous nature allows comprehensive mechanistic studies of photocatalysis, which have revealed that their general photophysical parameters, such as light absorption ability, electronic band structure, and charge separation efficiency, can be conveniently tailored by structural modifications. However, further understanding of the relationship between structure-property-activity is required from the viewpoint of charge-carrier transport, because the charge-carrier property is closely related to alleviation of the excitonic effect. In the present study, COFs composed of a fixed cobalt (Co) porphyrin (Por) centered tetraamine as an acceptor unit with differently conjugated di-carbaldehyde based donor units, such as benzodithiophene (BDT), thienothiophene (TT), or phenyl (TA), were synthesized to form Co-Por-BDT, Co-Por-TT, or Co-Por-TA, respectively. Their photocatalytic activity for reducing carbon dioxide into carbon monoxide was in the order of Co-Por-BDT>Co-Por-TT>Co-Por-TA. The results indicated that the excitonic effect, associated with their charge-carrier densities and π-conjugation lengths, was a significant factor in photocatalysis performance.

Cone-beam computed tomography (CBCT) can provide 3D images of a targeted area with the advantage of lower dosage than multidetector computed tomography (MDCT; also simply referred to as CT). However, in CBCT, due to the cone-shaped geometry of the X-ray source and the absence of post-patient collimation, the presence of more scattering rays deteriorates the image quality compared with MDCT. CBCT is commonly used in dental clinics, and image artifacts negatively affect the radiology workflow and diagnosis. Studies have attempted to eliminate image artifacts and improve image quality; however, a vast majority of that work sacrificed structural details of the image. The current study presents a novel approach to reduce image artifacts while preserving details and sharpness in the original CBCT image for precise diagnostic purposes. We used MDCT images as reference high-quality images. Pairs of CBCT and MDCT scans were collected retrospectively at a university hospital, followed by co-registration between the CBCT and MDCT images. A contextual loss-optimized multi-planar 2.5D U-Net was proposed. Images corrected using this model were evaluated quantitatively and qualitatively by dental clinicians. The quantitative metrics showed superior quality in output images compared to the original CBCT. In the qualitative evaluation, the generated images presented significantly higher scores for artifacts, noise, resolution, and overall image quality. This proposed novel approach for noise and artifact reduction with sharpness preservation in CBCT suggests the potential of this method for diagnostic imaging.

PURPOSE: With soft-contact-lens wear, evaporation of the pre-lens tear film affects the osmolarity of the post-lens tear film and this can introduce a hyperosmotic environment at the corneal epithelium, leading to discomfort. The purposes of the study are to ascertain whether there are differences in evaporation flux (i.e., the evaporation rate per unit area) between symptomatic and asymptomatic soft-contact-lens wearers, to assess the repeatability of a flow evaporimeter, and to assess the relationship between evaporation fluxes, tear properties, and environmental conditions. METHODS: Closed-chamber evaporimeters commonly used in ocular-surface research do not control relative humidity and airflow, and, therefore, misestimate the actual tear-evaporation flux. A recently developed flow evaporimeter overcomes these limitations and was used to measure accurate in-vivo tear-evaporation fluxes with and without soft-contact-lens wear for symptomatic and asymptomatic habitual contact-lens wearers. Concomitantly, lipid-layer thickness, ocular-surface-temperature decline rate (i.e., °C/s), non-invasive tear break-up time, tear-meniscus height, Schirmer tear test, and environmental conditions were measured in a 5 visit study. RESULTS: Twenty-one symptomatic and 21 asymptomatic soft-contact-lens wearers completed the study. A thicker lipid layer was associated with slower evaporation flux (p < 0.001); higher evaporation flux was associated with faster tear breakup irrespective of lens wear (p = 0.006). Higher evaporation flux was also associated with faster ocular-surface-temperature decline rate (p < 0.001). Symptomatic lens wearers exhibited higher evaporation flux than did asymptomatic lens wearers, however, the results did not reach statistical significance (p = 0.053). Evaporation flux with lens wear was higher than without lens wear but was also not statistically significant (p = 0.110). CONCLUSIONS: The repeatability of the Berkeley flow evaporimeter, associations between tear characteristics and evaporation flux, sample-size estimates, and near statistical significance in tear-evaporation flux between symptomatic and asymptomatic lens wearers all suggest that with sufficient sample sizes, the flow evaporimeter is a viable research tool to understand soft-contact-lens wear comfort.

We fabricated CH3NH3PbI3 (MAPbI3) thin films with and without a ZnO layer and measured the heterodyne transient grating (HD-TG) responses of each film to investigate the charge injection dynamics from MAPbI3 to ZnO, based on the component arising from the recombination of the surface trapped electrons in the ZnO layer with the remaining holes in MAPbI3. In addition, we observed the HD-TG response of the MAPbI3 thin film coated with the ZnO layer in which phenethyl ammonium iodide (PEAI) was inserted between two layers as a passivation layer and confirmed that the charge transfer was enhanced when PEAI existed, based on the increase in the amplitude of the component arising from the recombination and its acceleration.

We developed a new spectroscopic technique, which is named the charge carrier-selective heterodyne transient grating (CS-HD-TG) method, for the distinction of surface trap states existing in photocatalysts and photovoltaic materials by employing a burn laser inducing depletion of the number of surface trapped charge carriers. In addition, as a case study, we measured the heterodyne transient grating responses of hematite under bias conditions with and without the burn laser and revealed that two distinct trap states co-exist at the surface of the hematite film and only one of them could act as the reaction intermediate for the oxygen evolution reaction (OER), which is consistent with former studies.

O-GlcNAc modification of proteins often has crosstalk with protein phosphorylation. These posttranslational modifications are highly dynamic events that modulate a wide range of cellular processes. Owing to the physiological and pathological significance of protein O-GlcNAcylation and phosphorylation, we designed the fluorescent probe, ßGlcNAc-CM-Rhod-P, to differentially detect activities of O-GlcNAcase (OGA) and phosphatase, enzymes that are responsible for these modifications. ßGlcNAc-CM-Rhod-P was comprised of a ßGlcNAc-conjugated coumarin (ßGlcNAc-CM) acting as an OGA substrate, a phosphorylated rhodol (Rhod-P) as a phosphatase substrate and a piperazine bridge. Because the emission wavelength maxima of CM and Rhod liberated from the probe are greatly different (100 nm), spectral interference is avoided. The results of this study revealed that treatment of ßGlcNAc-CM-Rhod-P with OGA promotes formation of the GlcNAc-cleaved probe, CM-Rhod-P, and a consequent increase in the intensity of fluorescence associated with free CM. Also, it was found that exposure of the probe to phosphatase produces a dephosphorylated probe, ßGlcNAc-CM-Rhod, which displays strong fluorescence arising from free Rhod. On the other hand, when incubated with both OGA and phosphatase, ßGlcNAc-CM-Rhod-P was converted to CM-Rhod which lacked both ßGlcNAc and phosphoryl groups, in conjunction with increases in the intensities of fluorescence arising from both free CM and Rhod. This probe was employed to detect activities of OGA and phosphatase in cell lysates and to fluorescently image both enzymes in cells. Collectively, the findings indicate that ßGlcNAc-CM-Rhod-P can be utilized as a chemical tool to simultaneously determine activities of OGA and phosphatase.

Legal age estimation of living individuals is a critically important issue, and radiomics is an emerging research field that extracts quantitative data from medical images. However, no reports have proposed age-related radiomics features of the condylar head or an age classification model using those features. This study aimed to introduce a radiomics approach for various classifications of legal age (18, 19, 20, and 21 years old) based on cone-beam computed tomography (CBCT) images of the mandibular condylar head, and to evaluate the usefulness of the radiomics features selected by machine learning models as imaging biomarkers. CBCT images from 85 subjects were divided into eight age groups for four legal age classifications: ≤17 and ≥18 years old groups (18-year age classification), ≤18 and ≥19 years old groups (19-year age classification), ≤19 and ≥20 years old groups (20-year age classification) and ≤20 and ≥21 years old groups (21-year age classification). The condylar heads were manually segmented by an expert. In total, 127 radiomics features were extracted from the segmented area of each condylar head. The random forest (RF) method was utilized to select features and develop the age classification model for four legal ages. After sorting features in descending order of importance, the top 10 extracted features were used. The 21-year age classification model showed the best performance, with an accuracy of 91.18%, sensitivity of 80%, and specificity of 95.83%. Radiomics features of the condylar head using CBCT showed the possibility of age estimation, and the selected features were useful as imaging biomarkers.

A number of flavored capsule heat-not-burn (FC-HNB) tobacco products such as IQOS, Lil, and Glo have been introduced as a new generation of cigarettes. As they can release various types of volatile organic compounds (VOCs), it is important to assess the harmfulness associated with their use. Thus, the composition of VOCs in HNB cigarette vapor was evaluated to investigate the interactive roles of key variables controlling the relationships between VOC composition and capsule breaking, particularly the compositional changes induced by capsule breaking and release of flavor from FC-HNB cigarettes relative to regular products. As the capsules of FC-HNB cigarettes were broken, the total VOC concentrations increased by as high as eight times from 60.3 ± 0.48 to 488 ± 21.8 µg cig-1. The key VOC components released after breaking the flavored capsules were identified as ethyl butyrate (157 ± 13.6 µg cig-1; Lil), isoamyl acetate (76.9 ± 1.98 µg cig-1; Lil), and limonene (52.3 ± 3.29 µg cig-1; Glo). If the primary health risks of FC-HNB cigarette vapor are assessed using National Institute for Occupational Safety & Health (NIOSH) guidelines, 2,3-butanedinone exceeds the maximum daily intake limit (i.e., 0.05 mg day-1). Our study is expected to offer valuable insights into the harmful effects of direct and indirect exposure to various VOCs in FC-HNB products.

The detection of maxillary sinus wall is important in dental fields such as implant surgery, tooth extraction, and odontogenic disease diagnosis. The accurate segmentation of the maxillary sinus is required as a cornerstone for diagnosis and treatment planning. This study proposes a deep learning-based method for fully automatic segmentation of the maxillary sinus, including clear or hazy states, on cone-beam computed tomographic (CBCT) images. A model for segmentation of the maxillary sinuses was developed using U-Net, a convolutional neural network, and a total of 19,350 CBCT images were used from 90 maxillary sinuses (34 clear sinuses, 56 hazy sinuses). Post-processing to eliminate prediction errors of the U-Net segmentation results increased the accuracy. The average prediction results of U-Net were a dice similarity coefficient (DSC) of 0.9090 ± 0.1921 and a Hausdorff distance (HD) of 2.7013 ± 4.6154. After post-processing, the average results improved to a DSC of 0.9099 ± 0.1914 and an HD of 2.1470 ± 2.2790. The proposed deep learning model with post-processing showed good performance for clear and hazy maxillary sinus segmentation. This model has the potential to help dental clinicians with maxillary sinus segmentation, yielding equivalent accuracy in a variety of cases.

Uptake and release kinetics are investigated of a dilute aqueous polymeric-surfactant wetting agent, (ethylene oxide)45-(butylene oxide)10 copolymer, also referred to as poly(oxyethylene)-co-poly(oxybutylene), impregnated into a newly designed silicone-hydrogel lens material. Transient scanning concentration profiles of the fluorescently tagged polymeric surfactant follow Fick's second law with a diffusion coefficient near 10-11 cm2/s, a value 3-4 orders smaller than that of the free surfactant in bulk water. The Nernst partition coefficient of the tagged polymeric wetting agent, determined by fluorescence microscopy and by methanol extraction, is near 350, a very large value. Back-extraction of the polymeric-surfactant wetting agent releases only ∼20% of the loaded amount after soaking the fully loaded lens for over 7 days. The remaining ∼80% is irreversibly bound in the lens matrix. Reverse-phase liquid chromatography of the lens-loaded and lens-extracted surfactant demonstrates that the released wetting agent is more hydrophilic with a higher polarity. Aqueous poly(oxyethylene)-co-poly(oxybutylene) is hypothesized to attach strongly to the lens matrix, most likely to the lens silicone domains. Strong binding leads to slow transient diffusion, to large uptake, and to significant irreversible retention. These characteristics indicate the suitability of using a poly(oxyethylene)-co-poly(oxybutylene) nonionic polymeric surfactant to maintain enhanced lens wettability over time. Methodology and findings from this study provide useful insights for designing sustained-release contact-lens wetting agents and materials.

Purpose: This study aimed to evaluate the performance of transfer learning in a deep convolutional neural network for classifying implant fixtures. Materials and Methods: Periapical radiographs of implant fixtures obtained using the Superline (Dentium Co. Ltd., Seoul, Korea), TS III (Osstem Implant Co. Ltd., Seoul, Korea), and Bone Level Implant (Institut Straumann AG, Basel, Switzerland) systems were selected from patients who underwent dental implant treatment. All 355 implant fixtures comprised the total dataset and were annotated with the name of the system. The total dataset was split into a training dataset and a test dataset at a ratio of 8 to 2, respectively. YOLOv3 (You Only Look Once version 3, available at https://pjreddie.com/darknet/yolo/), a deep convolutional neural network that has been pretrained with a large image dataset of objects, was used to train the model to classify fixtures in periapical images, in a process called transfer learning. This network was trained with the training dataset for 100, 200, and 300 epochs. Using the test dataset, the performance of the network was evaluated in terms of sensitivity, specificity, and accuracy. Results: When YOLOv3 was trained for 200 epochs, the sensitivity, specificity, accuracy, and confidence score were the highest for all systems, with overall results of 94.4%, 97.9%, 96.7%, and 0.75, respectively. The network showed the best performance in classifying Bone Level Implant fixtures, with 100.0% sensitivity, specificity, and accuracy. Conclusion: Through transfer learning, high performance could be achieved with YOLOv3, even using a small amount of data.
