RESUMO

Nucleic acid amplification methods are increasingly being used to detect trace quantities of DNA in samples for various diagnostic applications. However, quantifying the amount of DNA from such methods often requires time consuming purification, washing or labeling steps. Here, we report a novel microfluidic centrifugation assisted precipitation (µCAP) method for single-step DNA quantification. The method is based on formation of a visible precipitate, which can be quantified, when an intercalating dye (GelRed) is added to the DNA sample and centrifuged for a few seconds. We describe the mechanism leading to the precipitation phenomenon. We utilize centrifugal microfluidics to precisely control the formation of the visible and quantifiable mass. Using a standard CMOS sensor for imaging, we report a detection limit of 45 ng µl-1. Furthermore, using an integrated lab-on-DVD platform we recently developed, the detection limit is lowered to 10 ng µl-1, which is comparable to those of current commercially available instruments for DNA quantification. As a proof of principle, we demonstrate the quantification of LAMP products for a HIV-1B type genome containing plasmid on the lab-on-DVD platform. The simple DNA quantification system could facilitate advanced point of care molecular diagnostics.

RESUMO

This report reviews two cases of multiple localized (agminated) intradermal neurilemomas that clinically mimicked a granulomatous dermatitis. The cutaneous lesions occurred in the second decade of life as firm papules on the chin and the lower part of the leg, respectively. Features of neurofibromatosis and neurilemomatosis were absent.

RESUMO

Keratoacanthoma of the lower eyelid developed in two patients. In the first case, conventional excision biopsy necessitated a large eyelid reconstruction procedure. In the second case, 5-fluorouracil was injected into the base of the keratoacanthoma. Both treatments resulted in complete resolution of the lesion with excellent cosmesis. Thus, intralesional 5-fluorouracil can be useful for keratoacanthomas of the eyelid in patients who are not good candidates for surgery.

RESUMO

For the past 2 years, we have been treating a teenage girl for a disfiguring, extensive, inflammatory linear verrucous epidermal nevus (ILVEN). In this one patient, we examined the efficacy of numerous treatment modalities and compared our results with those reported in the literature. In our experience, cryotherapy using liquid nitrogen proved to be the most successful therapy, and we have subsequently used it to treat almost all of this patient's lesions. Although cryotherapy does not provide an easy or dramatic cure, we believe it does produce reasonably good cosmetic results.

RESUMO

White piedra, a fungal infection caused by Trichosporon cutaneum, has been reported in Europe, South America, and the Orient. A few cases have also been reported in the United States, all of them acquired as well in this country. A patient whose condition was diagnosed in California apparently contracted white piedra while visiting Panama, demonstrating that tropical diseases may occasionally be found at great distances from their point of origin. Therapy with topical clotrimazole and removal of infected hairs by shaving failed to produce a clinical or culturally confirmed cure in this patient.

RESUMO

A case of profuse yet distinct telangiectasia of the nose was successfully treated by light dermabrasion. Dermabrasion may be useful for treating superficial telangiectasia that are numerous and occupy a considerable area. A small test patch is recommended.

RESUMO

A cutaneous horn is a keratotic, usually conical tumor, consisting of a spire of accumulated, cohesive stratum corneum. A number of primary lesions underlying the horny material are known to cause this condition. We present an unusual case of pyogenic granuloma clinically mimicking a cutaneous horn and suggest that it be included in the initial differential diagnosis as a rare cause of this entity. We also review various known causes of cutaneous horn.

RESUMO

A 35-year-old man had the insidious onset of an asymptomatic cyst-like swelling of the auricle. Serous fluid was evacuated but continued to reaccumulate. Application of a pressure dressing resulted in resolution without deformity. Distinguishing features of seroma, pseudocyst, and hematoma are discussed.

RESUMO

In 18 examples of keratoacanthoma, the proliferating squamous epithelium histologically invaded the perineural space. The invasion did not adversely affect the biologic behavior and the prognosis.

Among forty-three cases of lichen nitidus studied, twenty-one patients were Caucasian, twenty-one were Negro, and one was Spanish-American, ranging in age from five to fifty-eight years. The arm, forearm, trunk, and genitalia were sites most commonly involved. Clinically, the eruption usually appeared as tiny papules. Histologically, the papules showed a parakeratotoic "cap," epidermal atrophy, liquefaction degeneration of the basal layer, and a dermal infiltrate of lymphocytes, epithelioid cells, and sometimes giant cells.
