RESUMO

DNA metabarcoding of faecal samples is being successfully used to study the foraging niche of species. We assessed the ability of two benchtop high-throughput sequencing (HTS) platforms, to identify a large taxonomic array of food items from domestic cats Felis silvestris catus, including prey and human-related food taxa (pet food and leftovers leaving undetectable solid remains in faeces). Scats from a captive feeding trial (n = 41) and from free-ranging individuals (n = 326) were collected and analysed using a cytb mini-barcode in independent PCR replicates on the Ion PGM and the MiSeq platforms. Outputs from MiSeq were more sensitive and reproducible than those from Ion PGM due to a higher sequencing depth and sequence quality on MiSeq. DNA from intact prey taxa was detected more often (82% of the expected occurrences) than DNA from pet food (54%) and raw fish and meat (31%). We assumed that this variability was linked to different degree of DNA degradation: The Ion PGM detected significantly less human-linked food, birds, field voles, murids and shrews in the field-collected samples than the MiSeq platform. Pooling the replicates from both platforms and filtering the data allowed identification of at least one food item in 87.4% of the field-collected samples. Our DNA metabarcoding approach identified 29 prey taxa, of which 25 to species level (90% of items) including 9 rodents, 3 insectivores, 12 birds and 1 reptile and 33 human-related food taxa of which 23 were identified to genus level (75% of items). Our results demonstrate that using HTS platforms such as MiSeq, which provide reads of sufficiently high quantity and quality, with sufficient numbers of technical replicates, is a robust and non-invasive approach for further dietary studies on animals foraging on a wide range of food items in anthropogenic landscapes.

Food is a major aspect of pet care; therefore, ensuring that pet foods are not only healthful but attractive to companion animals and their owners is essential. The petfood market remains active and requires ongoing evaluation of the adaptation and efficiency of the new products. Palatability-foods' characteristics enticing animals and leading them to consumption-is therefore a key element to look at. Based on the type of information needed, different pet populations (expert or naïve) can be tested to access their preference and acceptance for different food products. Classical techniques are the one-bowl and two-bowl tests, but complementary (i.e., operant conditioning) and novel (i.e., exploratory behavior) approaches are available to gather more information on the evaluation of petfood palatability.

The ontogenesis of olfactogustatory preferences has been investigated in various mammals but surprisingly not in domestic cats Felis catus. In a first experiment, we examined how prenatal exposure (25 days prepartum) to a cheese flavor via the mother's diet can influence olfactory preferences of neonatal kittens. During 2-choice tests, 2-day-old kittens oriented first toward the cheese odor experienced in utero more frequently than toward a usual pet food odor. The choice of kittens born to mothers fed with a control diet did not differ from random. In the second experiment, we assessed the role of pre- and postnatal exposure (from 25th day before to 23rd day after birth) to cheese flavor on food preferences in weaned kittens. Forty-five-day-old cats exposed to cheese flavor during uterine and postnatal life via their mothers' diet ate higher amounts of chicken supplemented with cheese flavor than food supplemented with usual pet food flavor. On the other hand, the control group did not exhibit a preference for a specific food. Our results clearly demonstrated that pre- and postnatal olfactogustatory exposure via maternal ingestion influences later olfactory and food preferences of cats.

This study investigates the consequences of interference during first suckling for subsequent mare-young attachment in horses (Equus caballus). Foals brought to their dams' teats appeared, at later ages (1-3 months), to remain closer to their dams and to play less than control foals that had been allowed to suckle spontaneously. Higher levels of play and distance initiatives from the mother are considered criteria for secure attachment in horses, humans, and other species. As this unique handling was the only event that distinguished experimental from control groups, the authors concluded that first suckling is a crucial event in the development of secure dam-young attachment. These results could have important implications for all mammal species, including humans.

RESUMO

We have established that lateral biases are characteristic of visual behaviour in 65 horses. Two breeds, Trotters and French Saddlebreds aged 2 to 3, were tested on a novel object test. The main finding was a significant correlation between emotionality index and the eye preferred to view the novel stimulus: the higher the emotionality, the more likely that the horse looked with its left eye. The less emotive French Saddlebreds, however, tended to glance at the object using the right eye, a tendency that was not found in the Trotters, although the emotive index was the same for both breeds. The youngest French Saddlebreds did not show this trend. These results are discussed in relation to the different training practices for the breeds and broader findings on lateralisation in different species.
