This paper reports for the first time volatile compounds, anti-nociceptive and anti-inflammatory activities of essential oils from the leaves of Waltheria indica L. (Stericullaceae) growing in Nigeria. The essential oil was hydro-distilled and characterized by gas chromatography-flame ionization detection (GC-FID) and gas chromatography coupled with mass spectrometry (GC-MS) analyses. The anti-inflammatory activity was evaluated on carrageenan induced rat paw edema while the anti-nociceptive test was based on hot plate model. The hydro-distillation afforded 0.41% (dry weight basis) of light green oil. Forty compounds representing 99.8% were identified in the oil. The main constituents of the oil were limonene (34.7%), sabinene (21.2%) and citronellal (9.7%). The anti-nociceptive property of the essential oils statically inhibited edema development (p<0.001) at a dose of 200 and 400 mg/kg independent of time of exposure. However, the 100 mg/kg Waltheria indica essential oils (WIEO) displayed a relatively low inhibition (p<0.01-p>0.5) which declines as exposure time increases. The anti-inflammatory activities shows a steady rate and non-dose dependent activity (p<0.001) up to the 3rd h of inflammation study. Conversely, a sharp reduction at the rate of p<0.5, 0.1 and 0.01 for the 100, 200 and 400 mg/kg WIEO doses respectively. Overall, the results presented sustain and establish the anti-nociceptive and anti-inflammatory properties and justifies the need for further evaluation and development of the essential oils from this plant.


Este artículo informa por primera vez de compuestos volátiles, actividades anti-nociceptivas y antiinflamatorias de aceites esenciales de las hojas de Waltheria indica L. (Stericullaceae) que crecen en Nigeria. El aceite esencial fue hidro-destilado y se caracterizó por cromatografía de gases-detección de ionización de llama (GC-FID) y cromatografía de gases junto con análisis de espectrometría de masas (GC-MS). La actividad antiinflamatoria se evaluó en el edema de pata de rata inducido por carragenano, mientras que la prueba antinociceptiva se basó en el modelo de placa caliente. La destilación hidráulica proporcionó 0,41% (en peso seco) de aceite verde claro. Cuarenta compuestos que representan el 99.8% fueron identificados en el aceite. Los principales componentes del aceite fueron el limoneno (34,7%), el sabineno (21,2%) y el citronelal (9,7%). La propiedad anti-nociceptiva de los aceites esenciales inhibió estáticamente el desarrollo del edema (p<0.001) a una dosis de 200 y 400 mg/kg independientemente del tiempo de exposición. Sin embargo, los aceites esenciales de Waltheria indica de 100 mg/kg (WIEO) mostraron una inhibición relativamente baja (p<0.01-p>0.5) que disminuye a medida que aumenta el tiempo de exposición. Las actividades antiinflamatorias muestran una tasa constante y una actividad no dependiente de la dosis (p<0.001) hasta la tercera hora del estudio de inflamación. Por el contrario, una fuerte reducción a una tasa de p<0.5, 0.1 y 0.01 para las dosis de 100, 200 y 400 mg/kg de WIEO respectivamente. En general, los resultados presentados sostienen y establecen las propiedades anti-nociceptivas y antiinflamatorias y justifican la necesidad de una mayor evaluación y desarrollo de los aceites esenciales de esta planta.

ETHNOPHARMACOLOGICAL RELEVANCE: Bougainvillea glabra is widely used in Nigeria for the treatment of inflammatory-related conditions, and as remedy for pain ailments. AIM OF THE STUDY: Considering the lack of scientific studies focused on chemical constituents and pharmacological activity of B. glabra essential oil, this work was designed to characterize the volatile compounds and evaluate the anti-inflammatory and anti-nociceptive properties ascribed to this plant species. MATERIALS AND METHODS: The essential oil was extracted from the leaf of B. glabra by hydrodistillation in an all glass Clevenger-type apparatus and characterized by gas chromatography (GC-FID) and gas chromatography-mass spectrometry (GC-MS) on HP-5 column. The anti-inflammatory property of the essential was established by measurement of carrageenan induced rat paw edema while the anti-nociceptive activity was determined by hot plate test, according to established procedures. RESULTS: The essential oil was obtained in a yield of 0.08% (v/w) calculated on dry weight basis. A total of 11 compounds representing 96.2% of the oil contents were identified by GC/MS. The main constituents of the essential oil were (E)-nerolidol (31.4%), (E)-ß-ionone (10.3%) and linalool (10.1%). The anti-nociceptive property of the essential oil was statistically significant p < 0.001 at all doses of when compared to the control while exhibiting an activity in tandem with the standard drug. For the 1st and 2nd h, at doses of 100 and 200 mg/kg, the anti-inflammatory activity was statistically very significant (p < 0.001), while at 3rd h the activity declined (p < 0.01) at a dose of 200 mg. The activity was non-significant at the 4th h experimental duration. CONCLUSIONS: This is first report on the chemical constituents and biological activity of essential oil of B. glabra from Nigeria. Overall, the results herein presented sustain and strengthen the anti-nociceptive and anti-inflammatory properties traditionally ascribed to B. glabra.

The chemical constituents and insecticidal activity of essential oil obtained by hydrodistillation of the leaves of Paullinia pinnata Linn (Sapindaceae) are being reported. The essential oil were analysed by using gas chromatography (GC) and gas chromatography coupled with mass spectrometry (GC-MS). Different concentrations (50, 100, 150, 200 and 250 mg/mL) of P. pinnata essential oil prepared separately and diluted in dimethyl sulfoxide (DMSO) were tested on the maize weevil, Sitophilus zeamais. The main constituents of the oil were pentadecanoic acid (17.9%), isoaromadendrene epoxide (11.5%) and wine lactone (11.2%). Other significant compounds of the essential oil were eremophilene (6.9%) and phytol (6.2%). The essential oil displayed 100% mortality (fumigant toxicity) against S. zeamais adults at tested concentration of 150 mg/mL with lethal concentrations (LC50) of 51.87 mg/mL air. This is the first report on the chemical constituents and insecticidal activity of essential oil of P. pinnata and may be explore as a potential natural herbal plant for the control of insect pest.


Se informan los componentes químicos y la actividad insecticida del aceite esencial obtenido por hidrodestilación de las hojas de Paullinia pinnata Linn (Sapindaceae). El aceite esencial se analizó mediante cromatografía de gases (GC) y cromatografía de gases acoplada con espectrometría de masas (GC-MS). Se ensayaron diferentes concentraciones (50, 100, 150, 200 y 250 mg/ml) de aceite esencial de P. pinnata preparado separadamente y diluido en dimetilsulfóxido (DMSO) en el gorgojo de maíz, Sitophilus zeamais. Los componentes principales del aceite fueron ácido pentadecanoico (17,9%), isoaromadendreno epóxido (11,5%) y vino lactona (11,2%). Otros compuestos significativos del aceite esencial fueron eremophilene (6,9%) y phytol (6,2%). El aceite esencial mostró una mortalidad del 100% (toxicidad fumigante) contra los adultos de S. zeamais a una concentración de 150 mg/ml con concentraciones letales (CL50) de 51,87 mg/ml de aire. Este es el primer informe sobre la composición del aceite esencial de P. pinnata y su actividad insecticida. Puede ser explorado como una potencial planta herbácea natural para el control de la plaga de insectos.

The chemical constituents and anti-inflammatory activity of the essential oil from the leaf of Jatropha curcas L (Euphorbiaceae) collected from Nigeria are reported. The analysis of the chemical constituents of the essential oil was achieved by using gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS). All constituents of the essential oil, namely neophytadiene (35.8%), phytol (23.1%), trans-pinane (12.7%), 6,10,14-trimethyl-2- pentadecanone (12:3%) and citronellyl propanoate (11.2%), were present in significant amounts. The anti-inflammatory activity of the leaf oil was determined on Wistar rats using egg-albumin as phlogistic agent; significant inhibition (P< 0.05) was shown at a dose of 2%, v/v. Percentage inhibition of the anti- inflammation increased steadily to 76.6% in the 4th hour.

CONTEXT: The current rapid increase in the incidence of cardiovascular events indicates a need for the discovery of more effective cardioprotective agents. OBJECTIVE: This study evaluated the cardioprotective potential of a lanosteryl triterpene from Protorhus longifolia (Benrh.) Engl. stem bark. MATERIALS AND METHODS: Spectroscopic data analysis was used to confirm the structure of methyl-3ß-hydroxylanosta-9, 24-dien-21-oate (RA-3). The cardioprotective effect of RA-3 in isoproterenol-induced myocardial injury in hyperlipidemic rats was investigated. Rats were divided into the normal diet (ND) fed and high fat diet (HFD) fed groups. The HFD rats were further subdivided into three groups. The experimental group was orally administered with RA-3 (100 mg/kg) for 15 days. The rats were then injected with isoproterenol (85 mg/kg) to induce myocardial injury. At the end of the experiment, hearts and blood tissues were collected and used for histology and biochemical assays, respectively. RESULTS: RA-3 exhibited a cardioprotective effect as it minimized myocardial injury in HFD rats. Few lesions of acute hyaline degeneration and reduced fat deposition were observed in the heart tissue of the triterpene pretreated rats. Lactate dehydrogenase (LDH) activity was decreased in the blood of the RA-3 pretreated rats (44.1 mU/mL) compared to the untreated group (64.8 mU/mL). Increased glutathione (GSH) content and catalase (CAT) activity along with lower levels of malondialdehyde (MDA) in the triterpene pretreated animals (120.8 nmol/µL) than in the non-treated HFD fed rats (143.6 nmol/µL) were also observed. DISCUSSION AND CONCLUSION: The cardioprotective effect exhibited by RA-3 indicates its potential use in the management of cardiovascular diseases (CVD) and related health problems.

The essential oil constituents of the leaves of Jasminum subtriplinerve (Oleaceae) and Vitex quinata (Verbanaceae) cultivated in Vietnam were analysed by gas chromatography--flame ionisation detector (GC-FID) and gas chromatography--mass spectrometry (GC-MS) techniques. The main constituents identified in J. subtriplinerve were mainly oxygenated monoterpenes represented by linalool (44.2%), α-terpineol (15.5%), geraniol (19.4%) and cis-linalool oxide (8.8%). The quantitative significant components of V. quinata were terpene hydrocarbons comprising of ß-pinene (30.1%), ß-caryophyllene (26.9%) and ß-elemene (7.4%). The chemical compositions of the essential oils are being reported for the first time.

The volatile constituents and insecticidal activity of the essential oil obtained by hydrodistillation of pulverized air-dried leaves of Dehinbolliapinnata Schum. & Thonn (Sapindaceae) growing in Nigeria were studied. The constituents of the oil were analyzed by gas chromatography (GC) aid gas chromatography coupled with mass spectrometry (GC-MS). The percentage yield of the oil was 0.04% (v/w). Fourteen compounds were characterized, representing 94.0% of the total volatile component of the oil. The major constituents of the oil were hexahydrofarnesyl acetone (37.5%), fainesyl acetone (17.2%) and geranyl acetone (14.6%). The essential oil, at a concentration of 120 mg/mL, displayed 100% mortality against Sitophilus zeamais at 72 h with a LC(50) > 40.00 mg/mL air.

This paper reports on the compounds identified in the leaf and flower essential oils obtained by hydrodistillation of Plumeria alba L. (Apocynaceae) grown in Nigeria. The chemical analysis of the essential oils was achieved by means of gas chromatography (GC) and gas chromatography coupled with mass spectrometry (GC-MS). Linalool (13.2%), n-nonanal (9.6%), phenyl acetaldehyde (8.5%), neryl acetone (5.3%) and n-decanal (5.1%) were the main constituents of the leaf oil. On the other hand, the flower oil comprised mainly of limonene (9.1%), linalool (7.9%), α-cedrene (8.0%), caryophyllene oxide (7.9%) and (E, E)-α-farnesene (6.6%). This is the first report on the essential oil constituents of P. alba.

RESUMO

The leaf essential oil of Parquetina nigrescens collected from Badary, Nigeria, was obtained by hydrodistillation and analyzed by gas chromatography - mass spectrometry. The essential oil had a simple composition with only five identified components. The oil was dominated by citral (35.0% neral and 53.7% geranial). The high concentration of citral in the essential oil likely contributes to the ethnomedicinal utility and bioactivities associated with this medicinal plant.

The chemical composition of essential oil obtained by hydrodistillation of the leaves of Pachira glabra Pasq., (PgEO) has been studied by Gas Chromatography (GC) and Gas Chromatography coupled with Mass Spectrometry (GC/MS). Thirty three constituents representing 98.4% of total contents were identified from the essential oil. The major constituents of oil were limonene (23.2%), ß-caryophyllene (14.5%), phtyol (8.5%) and ß-bisabolene (6.3%). The antimicrobial activity of the PgEO was evaluated against a panel of ten bacteria and three fungal strain using agar diffusion and broth microdilution methods. Results have shown that the PgEO exhibited moderate to strong antimicrobial activity against the tested microorganisms except Citrobacter youagae, Micrococcus spp. and Proteus spp. The activity zones of inhibition (ZI) and minimum inhibitory concentrations (MIC) ranged between 13.7 mm-24.0 mm and 0.3 mg/mL-2.5 mg/mL, respectively. The insecticidal activity of PgEO was assayed against the adult Sitophilus zeamais. The lethal concentrations (LC50 and LC90) of the PgEO showed it to be toxic against adult S. zeamais at 32.2 and 53.7 mg/mL, respectively. This is the first report on the chemical composition and in vitro biological activities of essential oil of P. glabra growing in Nigeria.

The volatile oils extracted from leaves of eight cultivars of Citrus sinensis (L) Osbeck were comprehensively analysed by a combination of GC and GC-MS. Fifty four constituents accounting for 82.3-98.2% were identified. Sabinene (20.9-49.1%), delta-3-carene (0.3-14.3%), (E)-beta-ocimene (4.4-12.6%), linalool (3.7-11.1%) and terpinen-4-ol (1.7-12.5%) were the major constituents that are common to all the volatile oils. In addition, a cluster analysis was carried out and indicated at least four different chemotypes for the C. sinensis cultivars.

RESUMO

The hydrodistilled essential oils of the leaves of Senecio pterophorus collected from three different localities in KwaZulu-Natal Province, South Africa have been analyzed by GC and GC/MS. Thirteen - eighteen compounds were identified, accounting for over 98% of the total oil composition. The major components of the oils were limonene (10.3 - 32.3%), myrcene (14.4-19.7%), sabinene (13.0-18.0%), alpha-phellandrene (3.4-16.9%) and p-cymene (15.6-16.7%).

RESUMO

The essential oil from the rhizomes of Cyperus distans L.f. obtained by hydrodistillation was analyzed by capillary GC and GC/MS techniques. Eight constituents were identified, representing 99.6% of the total oil. The major components of the oil were cyperene (47.6%), alpha-pinene (18.8%), 1,8-cineole (14.5%) and caryophyllene oxide (7.3%).

The essential oils from the rhizomes of Cyperus rotundus L. collected from two different locations (Empangeni-A and KwaDlangezwa-B; both in the Kwa-Zulu Natal Province of South Africa) were obtained by hydrodistillation and analyzed by capillary GC and GC/MS. Forty-one and 43 components were identified, representing 89.9% and 92.0% of sample A and sample B, respectively. Alpha-cyperone (11.0%), myrtenol (7.9%), caryophyllene oxide (5.4%) and beta-pinene (5.3%) were major compounds in the oil of sample A. The main constituents of the oil of sample B were beta-pinene (11.3%), alpha-pinene (10.8%), alpha-cyperone (7.9%), myrtenol (7.1%) and alpha-selinene (6.6%).

The chemical composition and the antibacterial activity of the essential oils obtained by hydrodistillation from the leaves of Callistemon citrinus and Callistemon viminalis were analyzed by GC and GC/MS. Twenty-four and twelve components were identified for C. citrinus and C. viminalis, representing 92.0% and 98.3% of the total oils. The major components of C. citrinus and C. viminalis were 1,8-cineole (61.2% and 83.2%) and alpha-pinene (13.4% and 6.4%), respectively. The in vitro antibacterial activity of the essential oils was studied against 12 bacteria strains using disc diffusion and broth microdilution methods. The oils exhibited strong zone of inhibitions against some bacteria such as S. faecalis (20.3-24.0 mm), both strains of S. aureus (23.0-26.3 mm), B. cereus (17.3-19.0 mm) and S. macrcesens (11.3-23.7 mm) when compared to standard antibiotics gentamycin and tetracycline used as controls. Expect for P. aeruginosa and S. macrcescens, the MIC values of both essential oils ranged from 0.31-2.50 mg/mL.

In vivo clinical trials involving the administration of crude extracts of Cissus populnea to male subjects (normospermic, oligospermic, and azoopermic) in a 72-day study revealed that continuous exposure of the subjects to the extracts over this period did not significantly (p < or = 0.05) alter sperm count, morphology, motility, or volume. Antimicrobial screening of the extract against some selected microbial isolates secondarily implicated in male infertility revealed total inactivity against the microbial isolates screened, i.e., Staphylococcus aureus, Salmonella paratyphi, Escherichia coli, Proteus mirabilis, Pseudomonas aeruginosa, Candida albicans, and Klebsiella sp. Phytochemistry revealed the presence of tannins, flavonoids, saponins, and steroids. The presence of these secondary metabolites was confirmed by thin layer chromatography. We conclude that oral administration of aqueous extracts of the stem bark of Cisssus populnea over a 72-day period to human subjects apparently had no fertility enhancement effects on sperm parameters monitored in this study.
