RESUMO

OBJECTIVES: Anterior cruciate ligament (ACL) reconstruction after injury does not prevent post-traumatic osteoarthritis (PTOA). Circulating microRNA (miRNA) and metabolite changes emerging shortly after ACL injury and reconstruction remain insufficiently defined, potentially harbouring early cues contributing to PTOA evolution. Moreover, their differential expression between females and males also may influence PTOA's natural trajectory. This study aims to determine alterations in plasma miRNA and metabolite levels in the early stages following ACL reconstruction and between females and males. METHODS: A cohort of 43 ACL reconstruction patients was examined. Plasma was obtained at baseline, 2 weeks, and 6 weeks post-surgery (129 biospecimens in total). High-throughput miRNA sequencing and metabolomics were conducted. Differentially expressed miRNAs and metabolites were identified using negative binomial and linear regression models, respectively. Associations between miRNAs and metabolites were explored using time and sex as co-variants, (pre-surgery versus 2 and 6 weeks post-surgery). Using computational biology, miRNA-metabolite-gene interaction and pathway analyses were performed. RESULTS: Levels of 46 miRNAs were increased at 2 weeks post-surgery compared to pre-surgery (baseline) using miRNA sequencing. Levels of 13 metabolites were significantly increased while levels of 6 metabolites were significantly decreased at 2 weeks compared to baseline using metabolomics. Hsa-miR-145-5p levels were increased in female subjects at both 2 weeks (log2-fold-change 0.71, 95%CI 0.22,1.20) and 6 weeks (log2-fold-change 0.75, 95%CI 0.07,1.43) post-surgery compared to males. In addition, hsa-miR-497-5p showed increased levels in females at 2 weeks (log2-fold-change 0.77, 95%CI 0.06,1.48) and hsa-miR-143-5p at 6 weeks (log2-fold-change 0.83, 95%CI 0.07,1.59). Five metabolites were decreased at 2 weeks post-surgery in females compared to males: L-leucine (-1.44, 95%CI -1.75,-1.13), g-guanidinobutyrate (-1.27, 95%CI 1.54,-0.99), creatinine (-1.17, 95%CI -1.44,-0.90), 2-methylbutyrylcarnitine (-1.76, 95%CI -2.17,-1.35), and leu-pro (-1.13, 95%CI -1.44,-0.83). MiRNA-metabolite-gene interaction analysis revealed key signalling pathways based on post-surgical time-point and in females versus males. CONCLUSION: MiRNA and metabolite profiles were modified by time and by sex early after ACL reconstruction surgery, which could influence surgical response and ultimately risk of developing PTOA.

RESUMO

Gold tailings are characterized by low-grade, complex composition, fine embedded particle size, environmental pollution, and large land occupation. This paper describes the mineralogical properties of gold tailings, including chemical composition, phase composition, particle size distribution, and microstructure; summarizes the recycling and utilization of components such as mica, feldspar, and valuable metals in gold tailings; reviews harmless treatment measures for harmful elements in gold tailings; and adumbrated the research progress of gold tailings in the application fields of building materials, ceramics, and glass materials. Based on these discussions, a new technology roadmap that combines multistage magnetic separation and cemented filling is proposed for the clean utilization of all components of gold tailings.

As end-products of the intersection between the genome and environmental influences, metabolites represent a promising approach to the discovery of novel biomarkers for diseases. However, many potential biomarker candidates identified by metabolomics studies fail to progress beyond analytical validation for routine implementation in clinics. Awareness of the challenges present can facilitate the development and advancement of innovative strategies that allow improved and more efficient applications of metabolite-based markers in clinical settings. This minireview provides a comprehensive summary of the pre-analytical factors, required analytical validation studies, and kit development challenges that must be resolved before the successful translation of novel metabolite biomarkers originating from research. We discuss the necessity for strict protocols for sample collection, storage, and the regulatory requirements to be fulfilled for a bioanalytical method to be considered as analytically validated. We focus especially on the blood as a biological matrix and liquid chromatography coupled with tandem mass spectrometry as the analytical platform for biomarker validation. Furthermore, we examine the challenges of developing a commercially viable metabolomics kit for distribution. To bridge the gap between the research lab and clinical implementation and utility of relevant metabolites, the understanding of the translational challenges for a biomarker panel is crucial for more efficient development of metabolomics-based precision medicine.

Psoriatic arthritis (PsA) is a chronic, systemic, immune-mediated inflammatory disease causing cutaneous and musculoskeletal inflammation that affects 25% of patients with psoriasis. Current methods for evaluating PsA disease activity are not accurate enough for precision medicine. A metabolomics-based approach can elucidate psoriatic disease pathogenesis, providing potential objective biomarkers. With the hypothesis that serum metabolites are associated with skin disease activity, we aimed to identify serum metabolites associated with skin activity in PsA patients. We obtained serum samples from patients with PsA (n = 150) who were classified into mild, moderate and high disease activity groups based on the Psoriasis Area Severity Index. We used solid-phase microextraction (SPME) for sample preparation, followed by data acquisition via an untargeted liquid chromatography-mass spectrometry (LC-MS) approach. Disease activity levels were predicted using identified metabolites and machine learning algorithms. Some metabolites tentatively identified include eicosanoids with anti- or pro-inflammatory properties, like 12-Hydroxyeicosatetraenoic acid, which was previously implicated in joint disease activity in PsA. Other metabolites of interest were associated with dysregulation of fatty acid metabolism and belonged to classes such as bile acids, oxidized phospholipids, and long-chain fatty acids. We have identified potential metabolites associated with skin disease activity in PsA patients.

The main disease that decreases the manufacturing of natural rubber is tapping panel dryness (TPD). To solve this problem faced by a large number of rubber trees, it is recommended to observe TPD images and make early diagnosis. Multi-level thresholding image segmentation can extract regions of interest from TPD images for improving the diagnosis process and increasing the efficiency. In this study, we investigate TPD image properties and enhance Otsu's approach. For a multi-level thresholding problem, we combine the snake optimizer with the improved Otsu's method and propose SO-Otsu. SO-Otsu is compared with five other methods: fruit fly optimization algorithm, sparrow search algorithm, grey wolf optimizer, whale optimization algorithm, Harris hawks optimization and the original Otsu's method. The performance of the SO-Otsu is measured using detail review and indicator reviews. According to experimental findings, SO-Otsu performs better than the competition in terms of running duration, detail effect and degree of fidelity. SO-Otsu is an efficient image segmentation method for TPD images.

The world is undergoing an unprecedented trend of fast urbanization, which causes a range of socio-environmental consequences, one of which is shrinking cities and towns (SCT). SCT refer to the cities or towns that are experiencing population decline and economic downturn. In the existing literature, there have been numerous studies on SCT; however, there is a lack of study which investigates its knowledge domains. Therefore, this paper aims to conduct a scientometric analysis to achieve an outline of the SCT research status. Through the procedures of literature search and screening, a total of 716 SCT-related studies were extracted from the Scopus. The VOSviewer software system program was then utilized to visualize the present SCT-related studies. The visualization results revealed that the journal of Sustainability made significant contributions to the SCT research in terms of relevant publications. In addition, Haase, Annegret received the most co-citations, and was also the most productive author in this field. Furthermore, it was identified that current SCT research is mainly conducted in developed countries. Through the analysis of keywords, the emerging research topics were revealed. Discussions were further made from the perspectives of prevailing research methods, evaluation criteria, and solutions for SCT problems.

The notorious growth of lithium (Li) dendrites and the instability of the solid electrolyte interface (SEI) during cycling make Li metal anodes unsuitable for use in commercial Li-ion batteries. Herein, the use of simple sugar coating (α-d-glucose) is demonstrated on top of Li metal to halt the growth of Li dendrites and stabilize the SEI. The α-d-glucose layer possesses high surface and adhesive energies toward Li, which promote the homogenous stripping and plating of Li ions on top of the Li metal. Density functional theory reveals that Li-ion diffusion within the α-d-glucose layer is governed by hopping around the bare sides of the O atoms and along the apparent passages formed by the glucose molecules. Stable cycling performance is achieved when combining α-d-glucose-coated Li (G|Li) anodes with sulfur- and LiFePO4 -based cathodes in both LiTFSI (ether) and LiPF6 (carbonate) electrolyte systems. A G|Li-based symmetrical cell operates at a current density of 1 mA cm-2 and areal capacity of 1 mAh cm-2 displays a stable overpotential profile for over 9 months (7000 h) of continuous charge/discharge cycling.

Public participation is an important procedure of the environmental impact assessment. Effective public participation is essential to the Public-Private Partnership (PPP) projects as such projects usually exert tremendous impacts on the environment and society. However, in literature, there are few studies investigating the driving factors of public participation in PPP projects, especially in the context of China. To bridge this research gap, this study proposed a theoretical model, which incorporates contextual factors (i.e., perceived benefit and perceived risk) into the classical Theory of Planned Behavior model, to explore the determinants. The initial proposed model was tested using structural equation modeling. Analysis results indicated that attitude towards behavior, subjective norm, perceived risk and perceived behavioral control were the four significant driving factors of public participation in PPP projects, whereas perceived benefit had limited impact. Furthermore, this study evaluated eight public participation approaches in PPP projects. Results revealed that the public were more willing to participate in public decisions through the internet platform, followed by the information disclosure or consultation provided by the government. The research findings derived in this study can provide valuable reference for the government to promulgate proper policies to attract more public participation in PPP projects. Moreover, the research idea and methods used in this study can be popularized in other countries to enhance the public participation in PPP projects.

In recent years, with the development of green building and the increase of health awareness, the concept of healthy building has been proposed. Recently, studies have been made on developing healthy residential buildings; however, few attentions have been paid to the development of healthy dormitories. To bridge this research gap, this paper aims to investigate the crucial aspects of developing a healthy dormitory. Based on the Maslow's hierarchy of needs, three influencing aspects which include 17 measurement indicators are identified. Questionnaire surveys are subsequently conducted to collect students' perceptions on the identified indicators. After a structural equation modeling (SEM) analysis, the relationships between the three influencing aspects are analyzed. The research findings show that building performance, bodily sensation, and humanistic environment must be taken into account in the development of a healthy dormitory. In addition, it is revealed that building performance has a significant impact on bodily sensation, while bodily sensation has a significant impact on humanistic environment. However, building performance is found having little impact on humanistic environment. The findings of this study could provide useful information for the construction of healthy dormitories.

With the rapid development of the economy, people are paying more and more attention to the environmental problems. In this circumstance, the concept of a circular economy is proposed for making efficient use of resources and minimizing the production of waste and other emissions. Each year, the construction sector consumes a vast volume of resources and makes impacts on the environment. To align with the development of the circular economy, the concept of green building is proposed. In China, though the concept of green building has been promoted for decades, the development status is far from optimistic. Thus, this paper aims to investigate the barriers that hinder green building development (GBD) in China. Through a systematic review and semi-structured interviews with experienced industrial practitioners, 24 potential barriers of GBD in China were identified. A questionnaire survey was then conducted for data collection. After descriptive and inferential statistical analyses, a partial least squares structural equation model (PLS-SEM) was established to investigate the impacts of different barriers on GBD. Results showed that the lack of policy and industry guidance, the immature market environment, and the lack of environmental awareness are the most important GBD barriers in China. This research can assist stakeholders in better understanding the status of GBD in China and enable decision-makers to formulate appropriate strategies to promote green building.

RESUMO

A novel solution-based approach is presented to process earth-abundant Cu(2)ZnSn(S,Se)(4) absorbers using fully dissolved CZTS precursors in which each of the elemental constituents intermix on a molecular scale. This method enables the low-temperature processing of chemically clean kesterite films with excellent homogeneity. The high performance of resulting optoelectronic devices represents a chance to extend the impact of CZTS into the next chapter of thin-film solar cells.
