Hypoxia can cause a variety of diseases, including ischemic stroke and neurodegenerative diseases. Within a certain range of partial pressure of oxygen, cells can respond to changes in oxygen. Changes in oxygen concentration beyond a threshold will cause damage or even necrosis of tissues and organs, especially for the central nervous system. Therefore, it is very important to find appropriate measures to alleviate damage. MiRNAs can participate in the regulation of hypoxic responses in various types of cells. MiRNAs are involved in regulating hypoxic responses in many types of tissues by activating the hypoxia-inducible factor (HIF) to affect angiogenesis, glycolysis and other biological processes. By analyzing differentially expressed miRNAs in hypoxia and hypoxia-related studies, as well as the HT22 neuronal cell line under hypoxic stress, we found that the expression of miR-18a was changed in these models. MiR-18a could regulate glucose metabolism in HT22 cells under hypoxic stress by directly regulating the 3'UTR of the Hif1a gene. As a small molecule, miRNAs are easy to be designed into small nucleic acid drugs, so this study can provide a theoretical basis for the research and treatment of nervous system diseases caused by hypoxia.

The cultivated land area in China is approaching the red line for farmland protection. Newly reclaimed land possesses a large exploratory potential to become a reserved land resource. Identifying a fertilization strategy is vital for improving the poor properties and weak fertility of newly reclaimed land. An experiment was conducted to study the effects of traditional compound fertilizer (Fc) or bio-organic fertilizer (Ft), alone or in combination with biochar addition (6.85 t·ha-1 and 13.7 t·ha-1) on the growth, photosynthesis, yield and quality of Chinese small cabbage (CSC) plant. The results showed that compared to single compound fertilizer application, bio-organic fertilizer application promoted the plant's growth, indicated by the plant height, stem diameter and leaf area index (LAI), and significantly enhanced the yield and dry matter accumulation of CSC. In terms of the combination with biochar, the promoting effects were positively related to the biochar addition rate in the compound fertilizer group, while it was better to apply bio-organic fertilizer alone or in combination with biochar at a low rate of 6.85 t·ha-1. The highest yield was obtained under B2Fc and B1Ft with 29.41 and 37.93 t·ha-1, respectively, and the yield under B1Ft was significantly higher than that under B2Fc. The water productivity (WP) significantly improved in response to both kinds of fertilizer combined with biochar at 6.85 t·ha-1. There was a significant difference between the photosynthetic characteristics of plants treated with single-compound fertilizer and those treated with bio-organic fertilizer. The photosynthetic characteristics increased under compound fertilizer combined with biochar, while they regressed under bio-organic fertilizer combined with biochar. The quality of CSC, especially that of soluble sugars and total phenolics, improved under single bio-organic fertilizer application compared with that under single-compound fertilizer. The nitrite content of the plants increased with increasing biochar addition rate in both fertilizer groups. In conclusion, there is a significant promoting effect of applying bio-organic fertilizer to replace chemical fertilizer alone or combining compound fertilizer with low-rate biochar addition on newly reclaimed land. It is a recommended fertilization strategy to substitute or partially substitute chemical fertilizer with bio-organic fertilizer combined with biochar in newly reclaimed land, and it is of great significance to achieve fertilizer reduction.

The application of nitrification inhibitors (nitrapyrin) and urease inhibitors (N-(N-butyl) thiophosphoric triamide) under conventional water resources has been considered as an effective means to improve nitrogen utilization efficiency and mitigate soil greenhouse gas emissions. However, it is not known whether the inhibitors still have an inhibitory effect under unconventional water resources (reclaimed water and livestock wastewater) irrigation and whether their use in combination with biochar improves the mitigation effect. Therefore, unconventional water resources were used for irrigation, with groundwater (GW) control. Nitrapyrin and N-(N-butyl) thiophosphoric triamide were used alone or in combination with biochar in a pot experiment, and CO2, N2O, and CH4 emissions were measured. The results showed that irrigation of unconventional water resources exacerbated global warming potential (GWP). All exogenous substance treatments increased CO2 and CH4 emissions and suppressed N2O emissions, independent of the type of water, compared to no substances (NS). The inhibitors were ineffective in reducing the GWP whether or not in combination with biochar, and the combined application of inhibitors with biochar further increased the GWP. This study suggests that using inhibitors and biochar in combination to regulate the greenhouse effect under unconventional water resources irrigation should be done with caution.

Agricultural utilization of reclaimed water is considered to be an effective way to solve water shortage and reduce water environmental pollution. Silicon fertilizer can improve crop yield and quality and enhance crop resistance. The effect of foliar spray with silicon fertilizer on phyllosphere microbial communities remains lacking. In this study, a pot experiment was conducted to explore the effects of different types of silicon fertilizer on the composition and diversity of a phyllosphere bacterial community and the abundances of related functional genes in rice irrigated with reclaimed water. The results showed that Firmicutes, Proteobacteria, Actinobacteriota, Bacteroidota, and Verrucomicrobiota dominated the phyllosphere bacteria of rice. The relative abundance of Bacillus was higher than that of other treatments in RIS3. Reclaimed water irrigation significantly increased the relative abundances of the potential pathogens Pantoea and Enterobacter. The unclassified bacteria were also an important part of the bacterial community in the rice phyllosphere. Bacillus, Exiguobacterium, Aeromonas, and Citrobacter were significantly enriched by silicon fertilizer treatments. Functional prediction analysis showed that indicator species were mainly involved in metabolism and degradation functions, and the predicted functional groups of phyllosphere bacteria were attributed to chemoheterotrophy, aerobic chemoheterotrophy, nitrate reduction, and fermentation. Quantitative PCR results showed that AOA, AOB, and nifH genes were at low abundance levels in all treatments, and nirK genes was not significantly different among treatments. These results contribute to the in-depth understanding of the effects of foliar spray silicon fertilizer on the bacterial community structure and diversity of rice phyllosphere and provide a theoretical basis for the application of silicon fertilizer in reclaimed water irrigation agriculture.

The effects of root exudates and irrigation with treated wastewater on heavy metal mobility and soil bacterial composition under intercropping remain poorly understood. We conducted a pot experiment with maize and soybean grown in monocultures or intercultures, irrigated with either groundwater or treated wastewater. In addition, the pre-collected root exudates from hydroponic culture with mono- or inter-cropped maize and soybean were applied to the soil at four levels (0 %, 16 %, 32 % and 64 %). The results showed that application of root exudates increased plant growth and soil nutrient content. The analysis of "Technique for Order of Preference by Similarity to Ideal Solution" for higher plant biomass and lower soil Cd and Pb concentrations indicated that the best performance of soybean under treated wastewater irrigation was recorded under intercropping applied with 64 % of exudates, with a performance score of 0.926 and 0.953 for Cd and Pb, respectively. The second-best performance of maize under treated wastewater irrigation was also observed under intercropping applied with 64 % of exudates. Root exudate application reduced heavy metals migration in the soil-plant system, with a greater impact in intercropping than in monocropping. In addition, certain soil microorganisms were also increased with root exudate application, regardless of irrigation water. This study suggests that appropriate application of root exudates could potentially improve plant growth and soil health, and reduce toxic heavy metal concentrations in soils and plants irrigated with treated wastewater.

The scarcity of freshwater resources has increased the use of nonconventional water resources such as brackish water, reclaimed water, etc., especially in water-scarce areas. Whether an irrigation cycle using reclaimed water and brackish water (RBCI) poses a risk of secondary soil salinization to crop yields needs to be studied. Aiming to find an appropriate use for different nonconventional water resources, pot experiments were conducted to study the effects of RBCI on soil microenvironments, growth, physiological characteristics and antioxidation properties of crops. The results showed the following: (1) compared to FBCI, the soil moisture content was slightly higher, without a significant difference, while the soil EC, sodium and chloride ions contents increased significantly under the RBCI treatment. With an increase in the reclaimed water irrigation frequency (Tri), the contents of EC, Na+ and Cl- in the soil decreased gradually, and the difference was significant; the soil moisture content also decreased gradually. (2) There were different effects of the RBCI regime on the soil's enzyme activities. With an increase in the Tri, the soil urease activity indicated a significant upward trend as a whole. (3) RBCI can alleviate the risk of soil salinization to some extent. The soil pH values were all below 8.5, and were without a risk of secondary soil alkalization. The ESP did not exceed 15 percent, and there was no possible risk of soil alkalization except that the ESP in soil irrigated by brackish water irrigation went beyond the limit of 15 percent. (4) Compared with FBCI, no obvious changes appeared to the aboveground and underground biomasses under the RBCI treatment. The RBCI treatment was conducive to increasing the aboveground biomass compared with pure brackish water irrigation. Therefore, short-term RBCI helps to reduce the risk of soil salinization without significantly affecting crop yield, and the irrigation cycle using reclaimed-reclaimed-brackish water at 3 g·L-1 was recommended, according to the experimental results.

Insulin resistance is the leading cause of type 2 diabetes (T2D), and dysfunctional insulin receptor signaling is a major manifestation of this insulin resistance. In T2D, the corresponding insulin receptor levels are aberrantly down-regulated, which is one of the major factors underlying obesity-induced insulin resistance in adipose tissue. However, the precise mechanism of insulin receptor impairment in obese individuals remains unclear. In the current study, we established that immunoglobulin superfamily containing leucine-rich repeat (Islr) is highly expressed in adipocytes of mice fed a high-fat diet. We further demonstrated that Islr mediates the ubiquitin-independent proteasomal degradation of insulin receptor alpha (Insrα) by specifically interacting with proteasome subunit alpha type 4 (Psma4). Islr knockout increased the corresponding Insrα subunit levels and enhanced insulin sensitivity in adipocytes, ultimately improving systemic metabolism. Further, siRNA-mediated down-regulation of Islr expression in the white adipose tissue of obese mice increased insulin sensitivity. Overall, Islr regulates insulin sensitivity by interacting with Psma4 to control the ubiquitin-independent proteasomal degradation of Insrα in obese mice, indicating that Islr may be a potential therapeutic target for ameliorating insulin resistance.

Galloping of ice-coated transmission lines is occasionally observed under oblique wind directions. However, most current investigations on the galloping mechanisms are for flow perpendicular to the span of the transmission lines. In order to address this gap, this research studies the galloping characteristics of ice-coated transmission lines under oblique flows based on wind tunnel tests. The wind-induced displacement of an aero-elastic iced-coated transmission line model was measured with a noncontact displacement measurement equipment in a wind tunnel at different wind speeds and directions. The results show that galloping is characterized by elliptical trajectories and negative damping, which is more likely to occur under oblique flows than the direct flow (0°). At 15° wind direction, a galloping in vertical direction was observed at wind speeds above 5 m/s. At 30° wind direction, galloping was observed over the entire range of the tested wind speeds. Moreover, the galloping amplitudes under oblique flows are observed to be larger than that at the direct flows. Consequently, when the wind direction between the major winter monsoon azimuth and the lateral direction of transmission line route is between 15° and 30°, appropriate anti-galloping devices are highly recommended in practice.

Skeletal muscle mass is closely related to strength and health. Multiple genes and signaling pathways are involved in the regulation of skeletal muscle hypertrophy. miR-29 can participate in various processes of skeletal muscle development through different target genes. However, studies are needed on the function of miR-29 in skeletal muscle during mouse puberty. We used mice in which overexpression of miR-29ab1 cluster could be induced specifically within skeletal muscle, and investigated the effects of miR-29 overexpression on skeletal muscle at 1 month of age. We found that the overexpression of miR-29ab1 cluster in juvenile mice caused skeletal muscle mass and myofiber cross-sectional area to increase. The study on the mechanism of miR-29 inducing skeletal muscle hypertrophy had found that miR-29 achieved its function by inhibiting the expression of Mstn. At the same time, injured myofibers were present within miR-29ab1 cluster overexpressing skeletal muscle. The damage of skeletal muscle may be due to the inhibition of the type IV collagen by miR-29. These results indicate that although the overexpression of miR-29ab1 cluster can induce skeletal muscle hypertrophy in mouse juvenile, it simultaneously causes skeletal muscle damage.

Multiple tumor exacerbations and treatment procedures, such as extracellular matrix remodeling, metabolic reprogramming, immunological evasion, and resistance to chemotherapy and radiotherapy, are influenced by intratumoral hypoxia. It is becoming increasingly clear how hypoxia interacts with the extracellular matrix and how this affects the growth of cancer. We analyzed the published sequencing results of hypoxia-stressed mouse kidney tumor cells and found that the expression of miR-29b was significantly downregulated. There are several sites that are complementary to the miR-29 seed sequence in the 3' non-coding regions (3'UTRs) of various extracellular matrix-related genes, including collagen IV. We analyzed the sequences of the 3'UTRs of different subunits of collagen IV in different species and constructed the corresponding phylogenetic trees. We found that the 3'UTRs of Col4a1 and Col4a4 may have been subjected to particular evolutionary pressures. By cloning the 3'UTRs of collagen IV subunits into the psiCHECKTM-2 vector, we found that seven of the eight sites in the Col4a3-Col4a6 gene complementary to miR-29 were significantly repressed by miR-29a, b (except for the 7774-7781 of Col4a3 gene). The inhibitory efficiency of miR-29a, b on these seven sites was between 27% and 57%. The research on the regulation of miR-29 and extracellular matrix by hypoxia can provide a theoretical basis for tumor and fibrosis research and treatment.

Perennial ryegrass (Lolium perenne L.) is an important forage grass and has the potential to be used in phytoremediation, while little information is available regarding the transcriptome profiling of ryegrass leaves in response to high levels of Cd. To investigate and uncover the physiological responses and gene expression characteristics of perennial ryegrass under Cd stress, a pot experiment was performed to study the transcriptomic profiles of ryegrass with Cd-spiked soils. Transcriptome sequencing and comparative analysis were performed on the Illumina RNA-Seq platform at different concentrations of Cd-treated (0, 50 and 500 mg·kg−1 soil) ryegrass leaves and differentially expressed genes (DEGs) were verified by RT-qPCR. The results show that high concentrations of Cd significantly inhibited the growth of ryegrass, while the lower concentrations (5 and 25 mg·kg−1) showed minor effects. The activity levels of antioxidant enzymes such as superoxide dismutase (SOD), peroxidase (POD), catalase (CAT) and malondialdehyde (MDA) increased in Cd-treated ryegrass leaves. We identified 1103 differentially expressed genes (DEGs) and profiled the molecular regulatory pathways of ryegrass leaves with Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis in response to Cd stress. Cd stress significantly increased the membrane part, the metabolic process, the cellular process and catalytic activity. The numbers of unigenes related to signal transduction mechanisms, post-translational modification, replication, recombination and repair significantly increased. KEGG function annotation and enrichment analysis were performed based on DEGs with different treatments, indicating that the MAPK signaling pathway, the mRNA surveillance pathway and RNA transport were regulated significantly. Taken together, this study explores the effect of Cd stress on the growth physiology and gene level of ryegrass, thus highlighting significance of preventing and controlling heavy metal pollution in the future.

Brackish water has to be used to irrigate crops for harvest due to the scarcity of freshwater resources. However, brackish water irrigation may cause secondary soil salinization. Whether the combined utilization of different non-conventional water resources could relieve the risk of secondary soil salinization has not been reported. In order to explore the safe and rational utilization of brackish water in areas where freshwater resources are scarce, a pot experiment was conducted to study the risk of secondary soil mixed irrigation and rotational irrigation using brackish water and reclaimed water or freshwater. The results indicated that: (1) Short-term irrigation using reclaimed water did not cause secondary soil salinization, although increasing soil pH value, ESP, and SAR. The indices did not exceed the threshold of soil salinization. (2) Compared with mixed irrigation using brackish-freshwater, the contents of soil exchangeable Ca2+, K+, and Mg2+ increased, and the content of soil exchangeable Na+ decreased under rotational irrigation using brackish-reclaimed water. In addition, the contents of soil exchangeable Na+ and Mg2+ under mixed irrigation or rotational irrigation were significantly lower, and the exchangeable K+ content of the soil was higher compared with brackish water irrigation. The exchangeable Ca2+ content under rotational irrigation was higher than that of brackish water irrigation, while the reverse was seen under mixed irrigation. (3) For different combined utilization modes of brackish water and reclaimed water, the ESP and SAR were the lowest under rotational irrigation, followed by mixed irrigation and brackish water irrigation. The ESP under brackish water treatment exceeded 15%, indicating a certain risk of salinization, while ESPs under other treatments were below 15%. Under mixed irrigation or rational irrigation using reclaimed-brackish water, the higher the proportion or rotational times of reclaimed water, the lower the risk of secondary soil salinization. Therefore, short-term combined irrigation using brackish water and reclaimed water will not cause the risk of secondary soil salinization, but further experiments need to verify or cooperate with other agronomic measures in long-term utilization.

Due to reclaimed water, irrigation can cause human health and environmental risks. Soil amendments are applied to reveal the abundance of pathogens and antibiotic resistance genes in rhizosphere soil irrigated by reclaimed water and to better understand the effects of environmental factors on the rhizosphere soil bacterial composition, which has guiding significance for the reasonable use of soil amendments. In this study, the effects of biochar, bioorganic fertilizer, humic acid, loosening soil essence, and corn vinasse on bacterial community diversity and certain gene abundances in rhizosphere soil under drip irrigation with reclaimed water were studied using high-throughput assays and quantitative PCR. The results showed that biochar significantly increased pH, organic matter, and total nitrogen contents in the rhizosphere soil. The corn vinasse significantly decreased soil pH and increased the contents of total nitrogen and total phosphorus but significantly increased the soil EC value (P<0.05). The effects of the five soil amendments on the α-diversity of rhizosphere bacteria were not significantly different. The bacterial community structure and diversity of rhizosphere bacteria were similar at different taxonomic levels, but their relative abundance was different. α-Proteobacteria, γ-Proteobacteria, Bacteroidia, Actinobacteria, Acidimicrobiia, and Anaerolineae were the dominant bacteria in all treatments. The dominant genera consisted of Pseudomonas, Sphingobium, Sphingomonas, Cellvibrio, Allorhizobium-Neorhizobium-Pararhizobium-Rhizobium, Flavobacterium, and Algoriphagus (relative abundance>1%). Correlation analysis of environmental factors showed that the composition of the rhizosphere bacterial community was strongly correlated with pH, EC, total nitrogen, and total phosphorus content. The abundances of pathogenic bacteria and antibiotic resistance genes were 103-107 copies·g-1 and 104-108 copies·g-1, respectively. There were significant differences in the detection levels of pathogens and antibiotic resistance genes. Bioorganic fertilizer, loosening soil essence, and corn vinasse significantly increased the abundances of some antibiotic resistance genes, whereas humic acid and corn vinasse significantly decreased the abundances of Pseudomonas syringae, Ralstonia solanacearum, and total coliforms (P<0.05). A significant correlation was found between pathogens (Arcobacter, Bacillus cereus, Pantoea agglomerans, and Fecal bacteroidetes) and antibiotic resistance genes (tetA, tetB, tetO, tetQ, sul1, ermB, and ermC). In conclusion, while monitoring pathogens and antibiotic resistance genes in the agricultural environment under reclaimed water irrigation, attention should be paid to the rational application of soil amendments to avoid exacerbating the spread of biological contamination.

Objective: Currently, percutaneous endovascular creation of arteriovenous fistula (AVF) shows excellent outcomes. However, few systematic research evidence to support clinical decision making on the benefit of endovascular AVF is available. The purpose of this study was to evaluate the efficacy and safety of endovascular AVF (endoAVF) in patients with renal failure. Methods: We searched the Medline, Embase, Cochrane Library, and ClinicalTrials.gov databases for studies on endovascular or endovascular versus surgery for the creation of AVF. Two reviewers independently selected studies and extracted data. A systematic review and meta-analysis were performed by Review Manager 5.4 software (Revman, The Cochrane Collaboration, Oxford, United Kingdom) and Stata 15.0 (Stata Corp, College Station, TX, United States). Results: A total of 14 case series and 5 cohort studies, with 1,929 patients, were included in this study. The technique success was 98.00% for endoAVF (95% CI, 0.97-0.99; I 2 = 16.25%). There was no statistically significant difference in 3 cohort studies between endovascular and surgical AVF for procedural success (OR = 0.69; 95% CI, 0.04-11.98; P = 0.80; I 2 = 53%). The maturation rates of endoAVF were 87.00% (95% CI, 0.79-0.93; I 2 = 83.96%), and no significant difference was observed in 3 cohort studies between the 2 groups (OR = 0.73; 95% CI, 0.20-2.63; P = 0.63; I 2 = 88%). Procedure-related complications for endoAVF was 7% (95% CI, 0.04-0.17; I 2 = 78.31%), and it did not show significant difference in 4 cohort studies between the 2 groups (OR = 1.85; 95% CI, 0.37-9.16; P = 0.45; I 2 = 59%). Conclusion: The endovascular creation of AVF is potentially effective and safe. These important data may provide evidence to support clinicians and patients in making decisions with endovascular AVF. But further research is great necessary due to lack of randomized controlled studies.

RESUMO

We trace the crafting of expert narratives during the initial months of the COVID-19 pandemic in Mainland China, Hong Kong, and the United States. By expert narratives, we refer to how experts drew different lessons from past disease experiences to guide policymakers and the public amidst uncertainty. These expert narratives were mobilized in different sociopolitical contexts, resulting in varying configurations of expertise networks and allies that helped contain and mitigate COVID-19. In Mainland China, experts carefully advanced a managed narrative, emphasizing the new pandemic akin to the 2003 SARS outbreak can be managed while destressing the similar mistakes the government made during the two crises. In Hong Kong, experts invoked a distrust narrative, pointing to a potential coverup of COVID-19 similar to SARS, activating allies in civil society to pressure policymakers to act. In the United States, experts were mired in a contested narrative and COVID-19 was compared to different diseases; varying interpretations of COVID-19's consequences was exacerbated by political polarization. In expert narratives, the resonance of the past is emergent: the past becomes a site of struggle and a cultural object that is presented as potentially useful in solving problems of the present.

Background: In intensive and large-scale farms, abnormal estradiol levels in sows can cause reproductive disorders. The high incidence rate of reproductive disturbance will induce the elimination of productive sows in large quantities, and the poor management will bring great losses to the pig farms. The change in estradiol level has an important effect on follicular development and estrus of sows. To solve this practical problem and improve the productive capacity of sows, it is significant to further clarify the regulatory mechanism of estradiol synthesis in porcine granulosa cells (GCs). The most important function of granulosa cells is to synthesize estradiol. Thus, the studies about the complex transcriptome in porcine GCs are significant. As for precursor-messenger RNAs (pre-mRNAs), their post-transcriptional modification, such as alternative polyadenylation (APA) and alternative splicing (AS), together with long non-coding RNAs (lncRNAs), may regulate the functions of granulosa cells. However, the above modification events and their function are unclear within pig granulosa cells. Methods: Combined PacBio long-read isoform sequencing (Iso-Seq) was conducted in this work for generating porcine granulosa cells' transcriptomic data. We discovered new transcripts and possible gene loci via comparison against reference genome. Later, combined Iso-Seq data were adopted to uncover those post-transcriptional modifications such as APA or AS, together with lncRNA within porcine granulosa cells. For confirming that the Iso-Seq data were reliable, we chose four AS genes and analyzed them through RT-PCR. Results: The present article illustrated that pig GCs had a complex transcriptome, which gave rise to 8,793 APA, 3,465 AS events, 703 candidate new gene loci, as well as 92 lncRNAs. The results of this study revealed the complex transcriptome in pig GCs. It provided a basis for the interpretation of the molecular mechanism in GCs.

The production of valuable aromatics and the rapid catalyst deactivation due to coking are intimately related in the zeolite-catalyzed aromatization reactions. Here, we demonstrate that these two processes can be decoupled by promoting the Ga/HZSM-5 aromatization catalyst with Ca. The resulting bimetallic catalysts combine high selectivity to light aromatics with extended catalyst lifetime in the methanol-to-aromatics process. Evaluation of the catalytic performance combined with detailed catalyst characterization suggests that the added Ca interacts with the Ga-LAS, with a strong effect on the aromatization processes. A genetic algorithm approach complemented by ab initio thermodynamic analysis is used to elucidate the possible structures of bimetallic extraframework species formed under reaction conditions. The promotion effect of minute amounts of Ca is attributed to the stabilization of the intra-zeolite extraframework gallium oxide clusters with moderated dehydrogenation activity.

The transfer of antibiotic resistance genes (ARGs) in soil under reclaimed water irrigation poses a potential environmental risk. Regulation of NPK fertilizer could influence the behavior of bacterial communities, mobile genetic elements (MGEs), and soil properties, which determine the fate of ARGs. To identify the key element in NPK fertilizer and realize efficient regulation, we explored the effect of individual N, P, K fertilization on ARG variation in tomato rhizosphere and bulk soils. Compared with an unfertilized treatment, N fertilization resulted in greater decreases in the abundance of ARGs (decreases of 24.06%-73.09%) than did either P fertilization (increases of up to 35.84%, decreases of up to 58.80%) or K fertilization (decreases of 13.47%-72.47%). The influence of different forms of N (CO(NH2)2, NaNO3, and NH4HCO3), P (Ca(H2PO4)2 and CaMgO4P+), and K (KCl and K2(SO4)) fertilizers was also investigated in this study, and showed the influence of NaNO3, CaMgO4P+, and K2(SO4) on reducing ARGs abundance was greater in different types of N, P, K fertilizers. Bacterial communities showed the strongest response to N fertilization. The reduced bacterial diversity and abundance of ARG-host and non-host organisms explained the decline of total ARG abundance in soil. In soils fertilized with either P or K, the effect of soil properties, especially total nitrogen and pH, on ARG variation was greater than that of bacterial community and MGEs. These results suggest that N regulation of in NPK fertilizer may be an effective way to reduce the risks of ARGs in soil associated with reclaimed water irrigation.

Skeletal muscle has great plasticity. An increase in protein degradation can cause muscle atrophy. Atrogin-1 and muscle ring finger-1 (MuRF1) are dramatically upregulated in various muscle atrophy. Inhibition of Atrogin-1 and MuRF1 protects against muscle atrophy. MiR-29 plays an important regulatory role in skeletal muscle development. However, the function of miR-29 in skeletal muscle protein metabolism is not clear. To investigate the function of miR-29, we generated miR-29 knockout mice and the miR-29ab1 cluster overexpression mice. The disruption of miR-29 led to severe atrophy of skeletal muscle during puberty, and the muscle-specific overexpression of the miR-29ab1 cluster protected against denervation-induced and fasting-induced muscle atrophy. Furthermore, the overexpression of miR-29a, b mimics in myotubes resisted the muscle atrophy. MuRF1 was the direct target gene of miR-29a, b. These results demonstrate that miR-29ab1 cluster plays a critical role in the maintenance of skeletal muscle. MiR-29ab1 cluster is the excellent inhibitor of MuRF1, ultimately indicating that miR-29ab1 cluster is good therapeutic molecule candidate for adulthood.

Mg-containing high-silicon aluminum alloy is a heat-treatable aluminum alloy that is now widely used in the aerospace and automotive industries because of its high specific strength, high wear resistance and corrosion resistance, low thermal expansion coefficient, and low cost. More attention has been paid to optimizing the microstructure to increase the performance of this type of aluminum alloy. In the present work, the solidification processes of Mg-free and Mg-containing (0.33-1.32%) Al-8Si alloys were analyzed by the experimental results combined with the thermodynamic calculation. The results showed that α-Al, Si, and Al5FeSi were in the Mg-free Al-8Si alloy ingots, while the Al5FeSi phases in alloys with Mg additions were transformed into π phases (Al8Mg3FeSi6) by the reaction L+Al5FeSi→α-Al+Si+Al8Mg3FeSi6. There was a binary eutectic reaction of L→α-Al+Al5FeSi when the Mg content exceeded 0.51% and the Fe content was higher than 0.17%. With the increase of Mg content, the volume of Mg2Si was gradually increased while the divorced eutectic phenomenon of the quaternary eutectic structure (α-Al+Si+Mg2Si+Al8Mg3FeSi6) was weakened and the eutectic structure was significantly refined.
