RESUMO

Background: Osteochondral regeneration has long been recognized as a complex and challenging project in the field of tissue engineering. In particular, reconstructing the osteochondral interface is crucial for determining the effectiveness of the repair. Although several artificial layered or gradient scaffolds have been developed recently to simulate the natural interface, the functions of this unique structure have still not been fully replicated. In this paper, we utilized laser micro-patterning technology (LMPT) to modify the natural osteochondral "plugs" for use as grafts and aimed to directly apply the functional interface unit to repair osteochondral defects in a goat model. Methods: For in vitro evaluations, the optimal combination of LMPT parameters was confirmed through mechanical testing, finite element analysis, and comparing decellularization efficiency. The structural and biological properties of the laser micro-patterned osteochondral implants (LMP-OI) were verified by measuring the permeability of the interface and assessing the recellularization processes. In the goat model for osteochondral regeneration, a conical frustum-shaped defect was specifically created in the weight-bearing area of femoral condyles using a customized trephine with a variable diameter. This unreported defect shape enabled the implant to properly self-fix as expected. Results: The micro-patterning with the suitable pore density and morphology increased the permeability of the LMP-OIs, accelerated decellularization, maintained mechanical stability, and provided two relative independent microenvironments for subsequent recellularization. The LMP-OIs with goat's autologous bone marrow stromal cells in the cartilage layer have securely integrated into the osteochondral defects. At 6 and 12 months after implantation, both imaging and histological assessments showed a significant improvement in the healing of the cartilage and subchondral bone. Conclusion: With the natural interface unit and zonal recellularization, the LMP-OI is an ideal scaffold to repair osteochondral defects especially in large animals. The translational potential of this article: These findings suggest that such a modified xenogeneic osteochondral implant could potentially be explored in clinical translation for treatment of osteochondral injuries. Furthermore, trimming a conical frustum shape to the defect region, especially for large-sized defects, may be an effective way to achieve self-fixing for the implant.

RESUMO

Constructed wetlands (CWs) are commonly used to control excessive nitrogen from farmlands; however, the interactions between vegetation and microorganisms, nitrogen removal performance, and the mechanisms involved remain unclear in subtropical areas. This study aimed to investigate the nitrogen removal performance and mechanism of CWs containing Canna indica, Acorus calamus, and Thalia dealbata. The results show that CWs with plants had significantly higher nitrogen removal efficiencies than those without, with those planted with T. dealbata having the highest efficiency. T. dealbata performed better than the other two plants due to its high biomass and excellent nitrogen uptake capacity; more importantly, CWs with it had the highest abundance of nitrogen functional genes. Microbial nitrification-denitrification, the primary process of nitrogen removal in CWs, contributed to 88 %, 91 %, and 84 % of the TN removal in the CWs with C. indica, A. calamus, and T. dealbata, respectively, 29 %-158 % higher than that in CWs without plants. Microorganisms played a crucial role in nitrogen removal in the CWs, while plants significantly stimulated microbial activity by enhancing microbial abundance and creating a suitable environment for growth and metabolism. These results can help in understanding the contribution of plants in cleaning farmland tailwater and further optimization of plant configuration and management strategies in wetland ecosystems to improve nitrogen removal efficiency.

RESUMO

Dense SiC-based composite ceramics were fabricated by means of the ex situ addition of TaC using solid-state spark plasma sintering (SPS). Commercially available ß-SiC and TaC powders were chosen as raw materials. Electron backscattered diffraction (EBSD) analysis was conducted to investigate the grain boundary mapping of SiC-TaC composite ceramics. With the increase in TaC, the misorientation angles of the α-SiC phase shifted to a relatively small range. It was deduced that the ex situ pinning stress from TaC greatly suppressed the growth of α-SiC grains. The low ß→α transformability of the specimen with the composition of SiC-20 vol.% TaC (ST-4) implied that a possible microstructure of newly nucleated α-SiC embedded within metastable ß-SiC grains, which could have been responsible for the improvement in strength and fracture toughness. The as-sintered SiC-20 vol.% TaC (ST-4) composite ceramic had a relative density of 98.0%, a bending strength of 708.8 ± 28.7 MPa, a fracture toughness of 8.3 ± 0.8 MPa·m1/2, an elastic modulus of 384.9 ± 28.3 GPa and a Vickers hardness of 17.5 ± 0.4 GPa.

RESUMO

In order to clarify the differences in the effects of vegetation restoration strategies on soil carbon sequestration and aggregate stability under different water-eroded environments, we collected experimental data from 91 papers and evaluated the response of soil organic carbon (SOC) stock and aggregate stability to vegetation restoration based on Meta-analysis. The results showed the following:① compared with cropland or bare land, forestland/grassland restoration was beneficial to increase SOC stock and improve aggregate stability, but the dominant functions of the two were different. The effect of forestland restoration on carbon sequestration was stronger than that of grassland reforestation, and the effect of grassland restoration on aggregate stability was stronger than that of forestland restoration. ② Multi-factor Meta-analysis showed that the factors that significantly affected SOC were restoration year, soil clay content, vegetation coverage, mean annual precipitation (MAP), mean annual temperature (MAT), and soil depth. The positive effect of vegetation restoration on SOC stock increased with the increase in vegetation coverage rate. Grassland restoration had a more significant effect on SOC stock when soil clay content was 20%-32%, it was more likely to promote the carbon sequestration effect of grassland when MAP>800 mm or MAT<15℃, and there was no significant change in SOC stock under different restoration years. However, the effect of forestland restoration on SOC stock was more significant when soil clay content was>32%. Climate conditions had no limited effect on SOC stock in forestland, and there was a positive effect between SOC stock under forestland restoration and restoration years. ③ Vegetation restoration had stronger significant positive effects on mean weight diameter (MWD) and mean geometric diameter (GMD) when the clay content was 20%-32%, and MWD and GMD increased with the increase in vegetation coverage. ④SOC stock growth could explain 25% and 24% of the variation in the effect value of MWD and GMD, respectively. These results indicated that the formation of SOC was the result of multiple factors, and soil aggregate stability was limited only by vegetation coverage and soil clay content. The increase in SOC stock could promote the improvement of water stability MWD and GMD. These results can clarify the carbon sequestration effect of different vegetation restoration measures in water-eroded environments and provide theoretical reference for the restoration and reconstruction of degraded ecosystems.

RESUMO

Purpose: There is still a lack of effective treatments for cartilage damage. Cartilage tissue engineering could be a promising treatment method. Human umbilical cord Wharton's jelly (HUCWJ) and hydrogels have received wide attention as a scaffold for tissue engineering. They have not been widely used in clinical studies as their effectiveness and safety are still controversial. This study systematically compared the ability of these two biological tissue engineering materials to carry chondrocytes to repair cartilage injury in vivo. Methods: Chondrocytes were cocultured with HUCWJ or hydrogel for in vivo transplantation. The treatments comprised the HUCWJ+cell, hydrogel+cell, and blank groups. A rabbit model with articular cartilage defect in the knee joint area was established. The defective knee cartilage of different rabbit groups was treated for 3 and 6 months. The efficacy of the various treatments on articular cartilage injury was evaluated by immunohistochemistry and biochemical indices. Results: We found that the HUCWJ+cell and hydrogel+cell groups promoted cartilage repair compared with the blank group, which had no repair effect. The treatment efficacy of each group at 6 months was significantly better than that at 3 months. HUCWJ showed accelerated cartilage repair ability than the hydrogel. Conclusion: This study showed that HUCWJ is useful in cartilage tissue engineering to enhance the efficacy of chondrocyte-based cartilage repair, providing new insights for regenerative medicine. Impact statement Human umbilical cord Wharton's jelly (HUCWJ) and hydrogel are the suitable extracellular matrix for cartilage tissue engineering. This study assessed the capacity of HUCWJ- and hydrogel-loaded chondrocytes to repair cartilage injury in vivo. The data demonstrate that both HUCWJ and hydrogel effectively facilitated cartilage repair, and the repair effects of HUCWJ were significantly better compared with hydrogel, therefore providing a potential candidate for clinical practice of cartilage regeneration therapy.

RESUMO

Mesocarbon microbead-silicon carbide (MCMB-SiC) composites were prepared by hot-press sintering (2100 °C/40 MPa/1 h) with two different graphitized MCMBs as the second phase, which exhibited good self-lubricating properties. The effects of the graphitization degree of the MCMBs on the microstructure and properties of the composites were investigated contrastively. The results showed that the composites that added raw MCMBs with a low degree of graphitization had excellent self-sintering properties, higher densities, and better mechanical properties; by comparison, the composites that added mature MCMBs with a high degree of graphitization, which has regular and orderly lamellar structures, not only had good mechanical properties but also exhibited a lower and more stable dry friction coefficient (0.35), despite the higher wear rate (2.66 × 10-6 mm3·N-1·m-1). Large amounts of mature MCMBs were peeled off during the friction process to form a uniform and flat graphite lubricating film, which was the main reason for reducing the dry friction coefficient of the self-lubricating composites and making the friction coefficient more stable.

RESUMO

Infantile hemangioma (IH) is the most common benign tumor in infancy. Propranolol, a nonselective ß-adrenergic receptor blocker, is now the first-line therapy for IH. Recently, low sensitivity to propranolol therapy has become one major reason for the failure of IH treatment. However, the exact underlying mechanisms are yet to be fully elucidated. Here, we reported that pyruvate kinase isoform M2 (PKM2), an essential glycolytic enzyme, played a critical role in regulating the progression of IH and the therapeutic resistance of propranolol treatment. Shikonin reversed the propranolol resistance in hemangioma-derived endothelial cells and in hemangioma animal models. Moreover, shikonin combined with propranolol could induce excessive reactive oxygen species (ROS) accumulation and lead to autophagic dysfunction, which is essential for the enhanced therapeutic sensitivity of propranolol treatment. Taken together, our results indicated that PKM2 has a significant role in hemangiomas progression and therapeutic resistance; it could be a safe and effective therapeutic strategy for those hemangiomas with poor propranolol sensitivity combined with shikonin.

RESUMO

Objectives: Obstructive sleep apnea (OSA) is an independent risk factor for metabolic syndrome (MetS). Recent studies have indicated that circadian clock genes were dysregulated in OSA. In addition, it is clear that the impairment of circadian clocks drives the progression of MetS. Therefore, we hypothesized that circadian rhythm disruption links OSA with MetS. Methods: A total of 118 participants, who underwent polysomnography (PSG) and were diagnosed as healthy snorers (control, n = 29) or OSA (n = 89) patients based on the apnea-hypopnea index (AHI), were enrolled in the present study. General information, anthropometric data, blood biochemical indicators, clock gene expressions, and levels of oxidative and inflammatory indicators were collected, determined, and compared in all the participants. Results: We found that Brain and muscle aryl hydrocarbon receptor nuclear translocator-like protein 1 (Bmal1) and Differentiated embryo chondrocyte 1 (Dec1) were upregulated, while Period 1 (Per1) was reduced in OSA patients. In addition, these changing trends were closely associated with the hypoxia indicator of AHI and have a significant impact on the presence of MetS components, such as hyperglycemia (Dec1 and Per1, p < 0.05 and 0.001, respectively), hypertension (Bmal1 and Dec1, p < 0.001 and 0.01, respectively), hyperlipidemia (Dec1, p < 0.01), and obesity (Dec1, p < 0.05). Notably, expressions of Dec1 correlated with IR and predicted the presence of MetS in OSA patients. Finally, we also observed that Dec1 expression was interrelated with levels of both oxidative indicators and inflammatory biomarkers (IL-6) in OSA. Conclusion: This study concluded that circadian clock disruptions, especially Dec1, link OSA with MetS in an oxidative and inflammatory-related manner. Circadian clock Dec1 can be used as a specific biomarker (p < 0.001) and therapeutic target in OSA combined with Mets patients.

RESUMO

This study aims to reveal the buckling behavior of filament-wound composite cylindrical shells subjected to external pressure. The boundary conditions of the cylindrical shells were one end fixed and the other free. The carbon fiber stacking sequences were [±90]2/([±20]/[±90]/[±40]/[±90]/[±60]/[±90])2/[±90]. Finite element software ANSYS 16.2 was used for the numerical simulation to predict the critical buckling pressure and buckling behavior of composite cylindrical shell. External hydrostatic pressure tests were conducted, where the buckling behavior and strain response were observed. Numerical simulation accurately predicted the critical buckling pressure of carbon fiber/epoxy filament composite cylindrical shells under external pressure with 3.5% deviation from the experimental results. The buckling modes simulated by the finite element method agreed well with the deformed shape observed in the experiment, which was characterized by the uniform distribution of the three hoop waves. Comparing the axial compressive strain and hoop compressive strain of the composite shell, it was found that the circumferential stiffness of the shell was weaker than the axial stiffness. In addition, a comparative study of the strains of the fixed-end and free-end metal control sleeves was carried out. The results show that the boundary conditions have a significant influence on the strain response of control sleeves.

RESUMO

The normal anatomical structure of articular cartilage determines its limited ability to regenerate and repair. Once damaged, it is difficult to repair it by itself. How to realize the regeneration and repair of articular cartilage has always been a big problem for clinicians and researchers. Here, we conducted a comprehensive analysis of the physical properties and cytocompatibility of hydrogels, and evaluated their feasibility as cell carriers for Adipose-derived mesenchymal stem cell (ADSC) transplantation. Concentration-matched hydrogels were co-cultured with ADSCs to confirm ADSC growth in the hydrogel and provide data supporting in vivo experiments, which comprised the hydrogel/ADSCs, pure-hydrogel, defect-placement, and positive-control groups. Rat models of articular cartilage defect in the knee joint region was generated, and each treatment was administered on the knee joint cartilage area for each group; in the positive-control group, the joint cavity was surgically opened, without inducing a cartilage defect. The reparative effect of injectable glycol chitosan/dibenzaldehyde-terminated polyethylene glycol (GCS/DF-PEG) hydrogel on injured articular cartilage was evaluated by measuring gross scores and histological score of knee joint articular-cartilage injury in rats after 8 weeks. The 1.5% GCS/2% DF-PEG hydrogels degraded quickly in vitro. Then, We perform in vivo and in vitro experiments to evaluate the feasibility of this material for cartilage repair in vivo and in vitro.

RESUMO

Laser additive manufacturing is a promising technique for the preparation of complex-shaped SiC composites. High-quality powders are critical for high-precision laser printing. In this work, core-shell Cf @phenolic resin (PR) composites for selective laser sintering of carbon fiber reinforced silicon carbide (Cf/SiC) composites were fabricated by surface modification using 3-aminopropyltriethoxy silane coupling agent (KH550) in combination with planetary ball milling. PR coated uniformly on the fiber surface to form a core-shell structure. The effects of PR on the morphology, elemental composition, interfacial interactions, and laser absorption of the core-shell composite powder were investigated in detail. Results indicated that the composite powder exhibited good laser absorption within the infrared band.

RESUMO

In this work, a (SiC-AlN)/ZrB2 composite with outstanding mechanical properties was prepared by using polymer-derived ceramics (PDCs) and hot-pressing technique. Flexural strength reached up to 460 ± 41 MPa, while AlN and ZrB2 contents were 10 wt%, and 15 wt%, respectively, under a hot-pressing temperature of 2000 °C. XRD pattern-evidenced SiC generated by pyrolysis of polycarbosilane (PCS) was mainly composed by 2H-SiC and 4H-SiC, both belonging to α-SiC. Micron-level ZrB2 secondary phase was observed inside the (SiC-AlN)/ZrB2 composite, while the mean grain size (MGS) of SiC-AlN matrix was approximately 97 nm. This unique nano-micron hybrid microstructure enhanced the mechanical properties. The present investigation provided a feasible tactic for strengthening ceramics from PDCs raw materials.

RESUMO

The purpose of this study was to analyze the acute effect of ambient temperature on hospitalization due to acute exacerbation of chronic obstructive pulmonary disease (AECOPD), stroke, and myocardial infarction (MI) in Shenyang, China. We used the distributed delayed nonlinear model to evaluate the impact of ambient temperature on respiratory and cardiovascular diseases. The study population was divided into four groups: < 65 group and ≥ 65 age groups, female and male groups. The < 65 age group of AECOPD patients was more likely to be affected by high ambient temperature, while the ≥ 65 age group of AECOPD patients was more sensitive to low ambient temperature. The hospitalization risk of MI admission increased in the ≥ 65 age group at 1-8 days delay under low ambient temperature conditions.

RESUMO

BACKGROUND: Myocardial infarction (MI) is the most terrible appearance of cardiovascular disease. The incidence of heart failure, one of the complications of MI, has increased in the past few decades. Therefore, the identification of MI from angina patients and the determination of new diagnoses and therapies of MI are increasingly important. The present study was aimed at identifying differentially expressed genes and miRNAs as biomarkers for the clinical and prognosis factors of MI compared with angina using microarray data analysis. METHODS: Differentially expressed miRNAs and genes were manifested by GEO2R. The biological function of differentially expressed genes (DEGs) was examined by GO and KEGG. The construction of a protein-protein network was explored by STRING. cytoHubba was utilized to screen hub genes. Analysis of miRNA-gene pairs was executed by the miRWalk 3.0 database. The miRNA-target pairs overlapped with hub genes were seen as key genes. Logistic regressive analysis was performed by SPSS. RESULTS: A number of 779 DEGs were recorded. The biological function containing extracellular components, signaling pathways, and cell adhesion was enriched. Twenty-four hub genes and three differentially expressed miRNAs were noted. Eight key genes were demonstrated, and 6 out of these 8 key genes were significantly related to clinical and prognosis factors following MI. CONCLUSIONS: CALCA, CDK6, MDM2, NRXN1, SOCS3, VEGFA, SMAD4, NCAM1, and hsa-miR-127-5p were thought to be potential diagnosis biomarkers for MI. Meanwhile, CALCA, CDK6, NRXN1, SMAD4, SOCS3, and NCAM1 were further identified to be potential diagnosis and therapy targets for MI.

RESUMO

Silicon carbide (SiC) ceramics with Y2O3-Er2O3 as sintering additives were prepared by spark plasma sintering (SPS). The effects of sintering temperatures and Y2O3-Er2O3 contents on the microstructure, thermal conductivity, electrical, and mechanical properties were investigated. The increasing of sintering temperatures promoted the densification of SiC ceramics, thus increasing the thermal conductivity and electrical resistivity. With the increase of the sintering additive contents, the electrical resistivity increased due to the formation of the electrical insulating network; and the thermal conductivity first increased and then decreased, which was related to the content and distribution of the secondary phase among the SiC grains. The SiC ceramics sintered at 2000 °C with 9 wt.% Y2O3-Er2O3 exhibited higher electrical resistivity and thermal conductivity, which were 4.28 × 109 Ω·cm and 96.68 W/m·K, respectively.

RESUMO

A laser-diode-based white lighting module is fabricated via spectral component optimization, which can achieve both high luminous flux and high color rendering index (CRI). In this work, the laser module is constituted by blue laser diodes (LDs) which excite YAG:Ce-Al2O3 and red LDs that can compensate for the lack of red spectrum to improve the CRI of the light source. To fulfill the requirements of flexibility and compactness of light source, the blue and red LDs beams are combined by optical fiber coupling. A simulation framework is employed to optimize the dominant wavelength of red LDs and the power ratio of red to blue LDs. According to the results of the integrating sphere, high luminous flux of 1102 lm and high CRI of 77.8 are achieved simultaneously, which is consistent with the simulation results. The tunable correlated color temperature (CCT) varying from 4000 K to 2800 K and high angular color uniformity (ACU) can be obtained.

RESUMO

This study aims to analyze the acute effects of PM2.5, PM10, SO2, NO2, and O3 on hospitalizations for acute exacerbation of chronic obstructive pulmonary disease (AECOPD), stroke, and myocardial infarction (MI) from 2014 to 2017 in Shenyang, China. Hospitalization records for AECOPD (17,655), stroke (276,736) and MI (26,235) and air pollutions concentration data (PM2.5, PM10, SO2, NO2, and O3) were collected. A generalized additive model (GAM) was utilized to determine the impact of air pollutants on the relative risk (RR) of hospitalization for AECOPD, stroke, and MI. Stratified analysis for AECOPD was based on gender and age. It was based on gender, age, hypertension, and diabetes for stroke, and for MI it was based on gender, age, and coronary atherosclerosis. The lag effect for AECOPD in terms of gender analysis occurred at lag3-lag5. The hospitalization risk for stroke with hypertension due to SO2 and NO2 was greater than that of stroke without hypertension. The risk of hospitalization for stroke with hypertension as a comorbidity due to O3 was lower than without hypertension. The risk of hospitalization for MI combined with coronary atherosclerosis due to PM2.5, PM10, or NO2 was higher than that of hospitalizations for MI without coronary atherosclerosis. Air pollution increased the rate of hospitalization for AECOPD. SO2 and O3 appeared protective for stroke patients with coronary atherosclerosis. PM2.5, PM10, and NO2 had no influence on total hospitalization for myocardial infarction.

RESUMO

Mesocarbon microbead-silicon carbide (MCMB-SiC) composites with 0-30 wt % MCMBs were prepared by pressureless sintering (PLS) method at 2200 °C in Ar. The microstructure and tribological properties of the prepared composites were investigated. The results show that there was a finer grain size of SiC with the increase in MCMB content because MCMBs hinder the growth of SiC grains. The hardness of the composites decreased with increasing MCMB content, whereas the fracture toughness fluctuated showing a complex trend. The tribological properties of the composites under dry friction conditions were evaluated using the pin-on-disk method against a SiC counterpart. We found that the tribological properties of the samples were influenced by the oxide film or lubricating film that formed during the wear process on wear surfaces. Different wear mechanisms were found to be associated with differing MCMB contents.
