RESUMO

The effect of buildup factors on absorbed dose rate in air and the effective dose from gamma rays of primordial radionuclides in building materials, was investigated in the article. Specific absorbed dose rates were calculated for the standard concrete room, as well as, for rooms where brick and covering building materials were used. For all room models the Harima (G-P) buildup factors were applied, while for the standard room the Berger's and Taylor's buildup factors were used, too. The contribution of the radiation buildup to absorbed dose rate and effective dose was determined as large as 41%.

RESUMO

The spindle assembly checkpoint (SAC) is a surveillance mechanism contributing to the preservation of genomic stability by monitoring the microtubule attachment to, and/or the tension status of, each kinetochore during mitosis. The SAC halts metaphase to anaphase transition in the presence of unattached and/or untensed kinetochore(s) by releasing the mitotic checkpoint complex (MCC) from these improperly-oriented kinetochores to inhibit the anaphase-promoting complex/cyclosome (APC/C). The reversible phosphorylation of a variety of substrates at the kinetochore by antagonistic kinases and phosphatases is one major signaling mechanism for promptly turning on or turning off the SAC. In such a complex network, some kinases act at the apex of the SAC cascade by either generating (monopolar spindle 1, MPS1/TTK and likely polo-like kinase 1, PLK1), or contributing to generate (Aurora kinase B) kinetochore phospho-docking sites for the hierarchical recruitment of the SAC proteins. Aurora kinase B, MPS1 and budding uninhibited by benzimidazoles 1 (BUB1) also promote sister chromatid biorientation by modulating kinetochore microtubule stability. Moreover, MPS1, BUB1, and PLK1 seem to play key roles in APC/C inhibition by mechanisms dependent and/or independent on MCC assembly. The protein phosphatase 1 and 2A (PP1 and PP2A) are recruited to kinetochores to oppose kinase activity. These phosphatases reverse the phosphorylation of kinetochore targets promoting the microtubule attachment stabilization, sister kinetochore biorientation and SAC silencing. The kinase-phosphatase network is crucial as it renders the SAC a dynamic, graded-signaling, high responsive, and robust process thereby ensuring timely anaphase onset and preventing the generation of proneoplastic aneuploidy.

Promyelomonocytic leukemia (PML) is a prominent oncosuppressor whose inactivation is involved in the pathogenesis of hematological and epithelial cancers. Here, we report that PML aggregated in nuclear bodies in syncytia elicited by the envelope glycoprotein complex (Env) of human immunodeficiency virus-1 (HIV-1) in vitro. PML aggregation occurred after the fusion of nuclei (karyogamy) within syncytia but before the apoptotic program was activated. The aggregation of PML was detectable in syncytia present in the brain or lymph nodes from patients with HIV-1 infection, as well as in a fraction of blood leukocytes, correlating with viral status. Using a range of specific inhibitors of PML (the oncogenic PML/RARalpha fusion product or specific small interfering RNAs), we demonstrated that, in Env-elicited syncytia, PML was required for activating phosphorylation of ataxia telangiectasia mutated (ATM), which colocalized with PML in nuclear bodies, in a molecular complex that also involved topoisomerase IIbeta-binding protein 1. PML knockdown thus inhibited the ATM-dependent DNA damage response that culminates in the activation of p53, p53-dependent transcription of pro-apoptotic genes and cell death. Infection of CD4-expressing cells with HIV-1 also induced syncytial apoptosis, which could be suppressed by inhibiting PML. Altogether, these data indicate that PML activation is a critical early event that participates in the apoptotic demise of HIV-1-elicited syncytia.

RESUMO

The optimization of the Benchmark Simulation Model 1 (BSM1) through a multi objective genetic algorithm (MOGA) is studied in this paper. First, the optimization of the set points of the two Proportional Integral (PI) controllers proposed in BSM1 is performed. Then, a new controller layout composed of three PI controllers is proposed and the set points are also optimized. Among all performance indexes proposed in BSM1, only the effluent quality and the energy consumption for pumping and aeration were taken into account in both optimization problems. Since these two objectives are conflicting, the use of the MOGA allows in both cases a direct visualization of the possible trade-offs through a Pareto curve. These two case studies showed the feasibility of such optimizations even when dealing with computing intensive model like the full scale waste water treatment plant (WWTP) model.

RESUMO

Managing sewerage systems is a highly complex task due to the dynamic nature of the facilities. Their performance strongly depends on the know-how applied by the operators. In order to define optimal operational settings, two decision support tools based on mathematical models have been developed. Moreover, easy-to-use interfaces have been created as well, aiding operators who presumably do not have the necessary skills to use modelling software. The two developed programs simulate the behaviour of both wastewater treatment plants (WWTP) and sewer network systems, respectively. They have essentially the same structure, including raw data management and statistical analysis, a simulation layer using the application programming interface of the applied software and a layer responsible for the representation of the obtained results. Four user modes are provided in the two software including the simulation of historical data using the applied and novel operational settings, as well as modes concerning prediction of possible operation periods and updates. Concerning the WWTP software, it was successfully installed in Nantes (France) in June 2004. Moreover, the one managing sewer networks has been deployed in Saint-Malo (France) in January 2005. This paper presents the structure of the developed software and the first results obtained during the commissioning phase.

The paper presents the results of indoor radon concentration survey in 201 homes and offices in Niska Banja (the Spa of Nis), a well-known health resort and a spa in the South-East of Serbia. Radon indoor concentrations were determined by active charcoal method, according to standard EPA procedure. The indoor radon concentrations were in the range of up to 200 Bq/m(3) (47%), from 200-600 Bq/m(3) (26%) and over 600 Bq/m(3) (27%). Three areas of extremely high average radon concentrations were found (1,340-4,340 Bq/m(3)), with a maximum above 13,000 Bq/m(3). The content of natural radionuclides ((226)Ra, (214)Pb, (214)Bi, (235)U, (228)Ac, (212)Pb, (212)Bi, (208)Tl, (40)K) and (137)Cs, as well as the content of total uranium, thorium and potassium in mud used in peloidotherapy in the Health Institute "Niska Banja" was determined, too. The activities of the radionuclides were determined on an HPGe detector, by standard gamma spectroscopy. The results indicated considerably high amounts of total uranium and thorium (0.021 g/kg mud and 0.003 g/kg mud, respectively), due to the karsts origin of the soil.
