Orphan perennial native species are gaining importance as sustainability in agriculture becomes crucial to mitigate climate change. Nevertheless, issues related to the undomesticated status and lack of improved germplasm impede the evolution of formal agricultural initiatives. Acrocomia aculeata - a neotropical palm with potential for oil production - is an example. Breeding efforts can aid the species to reach its full potential and increase market competitiveness. Here, we present genomic information and training set optimization as alternatives to boost orphan perennial native species breeding using Acrocomia aculeata as an example. Furthermore, we compared three SNP calling methods and, for the first time, presented the prediction accuracies of three yield-related traits. We collected data for two years from 201 wild individuals. These trees were genotyped, and three references were used for SNP calling: the oil palm genome, de novo sequencing, and the A. aculeata transcriptome. The traits analyzed were fruit dry mass (FDM), pulp dry mass (PDM), and pulp oil content (OC). We compared the predictive ability of GBLUP and BayesB models in cross- and real validation procedures. Afterwards, we tested several optimization criteria regarding consistency and the ability to provide the optimized training set that yielded less risk in both targeted and untargeted scenarios. Using the oil palm genome as a reference and GBLUP models had better results for the genomic prediction of FDM, OC, and PDM (prediction accuracies of 0.46, 0.45, and 0.39, respectively). Using the criteria PEV, r-score and core collection methodology provides risk-averse decisions. Training set optimization is an alternative to improve decision-making while leveraging genomic information as a cost-saving tool to accelerate plant domestication and breeding. The optimized training set can be used as a reference for the characterization of native species populations, aiding in decisions involving germplasm collection and construction of breeding populations.

The oilseed palm genus Acrocomia is suitable for sustainable oil production in South America. The high phenotypic diversity of wild populations poses a challenge for the delimitation of the genus. Comparing the inflorescence architecture, a first-order panicle, and staminate and pistillate flower traits could be a valuable tool in resolving the taxonomic disarray. Thus, this study aims to characterize the differences in the inflorescence architecture and floral structures of three common and economically significant Acrocomia species: A. aculeata, A. totai, and A. intumescens. Biometric traits of the inflorescence architecture and floral structures of various Acrocomia accessions in an ex situ germplasm collection in Brazil were assessed. The unweighted pair group method with arithmetic mean (UPGMA) cluster analysis based on the Gower distance was used to measure dissimilarities between the individual plants of the accessions. To our best knowledge, this study provides the first evidence of the presence of second-order rachillae in the genus Acrocomia. Evaluated traits showed a high level of variation within and between accessions, emphasizing the phenotypic diversity of the genus. The accessions of A. totai were distinguishable from those of the other two species by their inflorescence architecture and flower traits. The dissimilarities between A. aculeata and A. intumescens were not sufficient to differentiate both. In conclusion, the quantitative assessment of inflorescence and floral traits is a valuable tool for taxonomic resolution of the genus.

Although Macauba (Acrocomia aculeata) has been highlighted by its high-quality oil to fit edible and nonedible purposes, data addressing carbon and nitrogen metabolism underlying development and ripening of fruits remain scarce. In addition, accessions of Macauba exibit varied oil yield in fruits, including during the fruit development stages. Here, we monitored contents of carbohydrates, proteins, amino acids and lipids in the mesocarp and endosperm of Macauba fruits until ripening. We selected three accessions from different Brazilian regions (southeast, MG; northeast, PE; and central-west, MS) that differ in the mesocarp lipid content of ripe fruits. Despite the anatomical differences, mesocarp and endosperm exhibited similar trends of metabolite accumulation for most of the analyzed compounds. In the mesocarp, total soluble protein, free amino acids, sucrose, starch and total lipids accumulate towards ripening, while glucose and fructose declined in all accessions. Endosperm differed from mesocarp solely in the amino acid content, which decreased in ripe fruits. In the endosperm, accessions accumulated carbohydrates differently. Accession PE showed comparable fructose and starch contents in the endosperm between the beginning of fruit development and ripening, while in accessions MG and MS, both compounds decreased and increased, respectively, towards ripening. Accession MG was highlighted by its highest lipid content in the two tissues indicating its potential for energy and cosmetic industries. Our results provide novel insights into metabolic changes underlying development and ripening of Macauba fruits and variability in oil content among accessions, indicating new targets for breeding programs.

ABSTRACT: Macauba palm stands out for having favorable features to biodiesel production such as the high oil content of its fruit. Considering the great potential of the species and their applicability in the renewable energy field, it becomes indispensable to establish the right conditions for storing the seeds for propagation purpose. The aim of this research was to evaluate the effect of seed moisture content, packaging, and storage conditions such as temperature and relative humidity on the quality of seeds from Minas Gerais State, during a 12-month storage period. The research had two independent assays: (I) the seeds were stored with three moisture contents/ranges 4.0≤6.0%; 6.0≤8.0% and 8.0≤10.0% in impermeable packages, under room temperature and at 10ºC; (II) seeds with approximately 5.9% of moisture content were stored in three different types of packages: a) permeable, b) semi-permeable and c) impermeable. Three storing conditions were tested: a) room temperature and RH under laboratory conditions; b) 15ºC and 45% RH; c) 20ºC and 55% RH. Water content, germination rate and germination speed index were evaluated at 0, 4, 8 and 12 months of storing. The best germination results were obtained with the moisture range of 6.0≤8.0%, with seeds kept at room temperature; while the seeds stored at 10ºC, regardless the moisture range, did not survive. The stored seeds with 5.9% moisture content and at both 15ºC/45%RH and 20ºC/55% RH conditions, independently of the package type used, showed the best results. Thus, macaw palm seeds can be classified as intermediates seeds.
RESUMO: Considerando o grande potencial oleaginoso da macauba e a sua aplicabilidade nos setores energéticos e industriais, torna-se indispensável o conhecimento das condições adequadas de armazenamento das sementes, garantindo a manutenção da viabilidade. O objetivo do trabalho foi avaliar o efeito do teor de água da semente e das condições de armazenamento (temperatura, umidade relativa do ar e embalagem) sobre a qualidade das sementes, oriundas de Minas Gerais, durante 12 meses. O experimento foi realizado através de dois ensaios: no primeiro, as sementes foram armazenadas sob três faixas de umidade, 4,0≤6,0%; 6,0≤8,0% e 8,0≤10,0%, em embalagens impermeáveis, em temperatura ambiente e a 10°C. No segundo, as sementes foram acondicionadas em três tipos de embalagem: a) permeável; b) semipermeável; e c) impermeável. Foram comparadas três condições de armazenamento: a) temperatura e UR ambientes, em condição de laboratório; b) 15ºC e 45% UR; c) 20ºC e 55% UR. Avaliou-se o teor de água, a taxa de germinação e o índice de velocidade de germinação aos 0, 4, 8 e 12 meses de armazenamento. A faixa de umidade de 6,0≤8,0% em temperatura ambiente possibilitou a melhor conservação das sementes durante os 12 meses de armazenamento. Porém, as sementes não sobreviveram em temperatura de 10°C, independente dos teores de umidade. O teor de água de 5,9% e os ambientes de armazenamento de 15°C/45% UR e 20°C/55% UR, independente do tipo de embalagem, apresentaram os melhores resultados ao longo do armazenamento. Dessa forma, as sementes de macauba podem ser classificadas como sementes intermediárias.

Main conclusion Macauba palm fruiting is supra-annual, and the fruit growth follows a double sigmoidal trend. The prevailing compound in the mesocarp differs as the fruit ages, oil being the major storage compound. Acrocomia aculeata, macauba palm, is a conspicuous species in the tropical Americas. Because the species is highly productive in oil-rich fruits, it is the subject of domestication as an alternative vegetable oil crop, especially as a bioenergy feedstock. This detailed study first presents the macauba fruit growth and development patterns, morphological changes and accumulation of organic compounds. Fruits were monitored weekly in a natural population. The fruiting was supra-annual, and the fruit growth curve followed a double sigmoidal trend with four stages (S): SI-slow growth and negligible differentiation of the fruit inner parts; SII-first growth spurt and visible, but not complete, differentiation of the inner parts; SIII-growth slowed down and all structures attained differentiation; and SIV-second growth spurt and fruit maturation. In SII, the exocarp and endocarp were the main contributors to fruit growth, whereas the mesocarp and endosperm were responsible for most of the weight gain during SIV. In comparison with starch and oil, soluble sugars did not accumulate in the mesocarp. However, starch was transitory and fueled the oil synthesis. The protective layers, the exocarp and endocarp, fulfilling their ecological roles, were the first to reach maturity, followed by the storage tissues, the mesocarp, and endosperm. The amount and nature of organic compounds in the mesocarp varied with the fruit development and growth stages, and oil was the main and final storage material. The description of macauba fruit's transformations and their temporal order may be of importance for future ecological and agronomical references.

A propagação in vitro via embriogênese somática é uma alternativa eficiente para a propagação em larga escala do material vegetal. No entanto, não há relatos do desenvolvimento de protocolos completos de embriogênese somática, com produção de plantas de alface. O presente trabalho teve como objetivo estabelecer a propagação in vitro de genótipos de alface, Paris White e Red Salad Bowl, avaliando a indução dos calos embriogênicos, regeneração dos embriões somáticos e posterior conversão em plantas. Para a indução de embriogênese somática, duas fontes de explante (folhas cotiledonares inteiras e seccionadas) foram cultivadas em meio MS+10,75µM de ANA+0,89µM de BA. A proliferação dos calos embriogênicos foi realizada em meio MS+24µM de AIA+0,15µM de BA. Para a maturação dos embriões somáticos e conversão em plantas, utilizou-se meio MS sem regulador de crescimento (semi-sólido sem carvão ativado e com carvão ativado e meio líquido). A fonte do explante seccionada foi estatisticamente superior apenas para o genótipo Paris White. Para a regeneração dos embriões somáticos, observou-se que, no genótipo Paris White, o meio líquido foi superior estatisticamente, quando comparado aos meios semi-sólidos e, para o genótipo Red Salad Bowl, foi o meio semi-sólido com acréscimo de carvão ativado. Tendo em vista o potencial da aplicabilidade comercial da embriogênese somática para a produção em larga escala de plântulas de alface do genótipo Paris White, os protocolos de indução, proliferação, maturação dos embriões somáticos e conversão em plantas, foram adequados, em especial, em sistema líquido.
In vitro propagation via somatic embryogenesis is an efficient alternative to large-scale propagation of plant material. Nevertheless, there are no reports of development of complete somatic embryogenesis protocols, with plant production of lettuce. This study aimed to establish lettuce in vitro propagation of Paris White and Red Salad Bowl genotypes, evaluating embryogenic callus induction, somatic embryos regeneration and subsequent conversion into plants. For somatic embryogenesis induction, two sources of explants (whole and sectioned cotyledons) were grown on MS medium containing 10.75µM NAA and 0.89µM BA. Callus proliferation occurred in MS medium supplemented with 24µM AIA and 0.15µM BA. For somatic embryos maturation and conversion into plants, we used MS medium devoid of growth regulator under three conditions: semi-solid medium with and without activated charcoal, and liquid medium. The sectioned explant source was statistically superior only for genotype Paris White. For somatic embryos regeneration, we observed that, in genotype Paris White, the liquid medium was statistically higher than semi-solid media, and for genotype Red Salad Bowl, activated charcoal-added semi-solid medium was the better. Considering the potential commercial applicability of somatic embryogenesis for seedlings mass production of lettuce Paris White genotype, somatic embryos induction, proliferation, maturation and conversion into plants protocols were adequate, especially in liquid system.

A qualidade da uva pode responder ao manejo da irrigação e ao ajuste da adubação nitrogenada. Assim, este trabalho teve como objetivo estudar o efeito do manejo de irrigação e doses de nitrogênio sobre a qualidade da uva 'Niagara Rosada' no município de Colatina-ES. Foram avaliadas três doses de nitrogênio, aplicadas via fertirrigação: 35, 70 e 140g planta-1 e quatro estratégias de manejo de irrigação. No início do experimento até a fase de maturação das bagas, todos os tratamentos receberam a mesma lâmina de irrigação, elevando-se a umidade do solo à capacidade de campo (M1=100 por cento). Após este período, foram avaliados mais três estratégias de manejo de irrigação: m²=67 por cento, M3=33 por cento e M4=0 por cento da lâmina correspondente ao tratamento M1. Foram avaliados os teores de sólidos solúveis (SS), pH e acidez titulável (AT). Os resultados indicaram que os tratamentos sob déficit hídrico (M3 e M4), nas doses de 70 e 140g planta-1 apresentaram maiores teores de SS e pH das bagas e diminuição dos valores de AT.
The quality of the grape can respond to irrigation and N fertilization adjustment. So, this research aimed to study the effect of irrigation management and doses of nitrogen on 'Niagara Rosada' grape quality in Colatina (ES) municipality. It was evaluated three doses of nitrogen applied by fertigation: 35, 70 and 140g plant-1 and four strategies of irrigation management. From the beginning of the experiment until the stage of maturation, all treatments received the same water depth, increasing soil moisture at field capacity (M1=100 percent). After this period, it was evaluated three strategies for irrigation management: m²=67 percent, M3=33 percent and M4=0 percent of the depth corresponding to the M1 treatment. It was appraised the content of soluble solids (SS), pH and acidity (AT). The results indicated that the treatments under water stress, in the rates of 70 and 140g plant-1 presented larger rates of SS and pH of the berries and decreasing values of AT.

Estimativas do coeficiente de repetibilidade têm sido utilizadas no estudo de caracteres de várias espécies de palmeiras, auxiliando na definição do número e período adequado de avaliações dos genótipos para maior eficiência dos programas de melhoramento. Para macaúba, não há relatos de estudos dessa natureza. Nesse contexto, faz-se importante a determinação do coeficiente de repetibilidade das seguintes características biométricas dos frutos: peso do fruto, volume do fruto, diâmetro equatorial do fruto, peso da amêndoa, volume da amêndoa, diâmetro da amêndoa e espessura do endocarpo; e a determinação do número mínimo de avaliações para um eficiente processo de seleção e avaliação de subamostras de macaúba. O trabalho foi desenvolvido no Departamento de Fitotecnia da Universidade Federal de Viçosa, analisando-se 145 subamostras de macaúba, provenientes de coletas realizadas nos estados de Minas Gerais, São Paulo, Mato Grosso do Sul, Pará, Maranhão e Pernambuco. Houve diferença significativa para todas as características analisadas e observou-se concordância nas magnitudes dos coeficientes de repetibilidade de cada característica, obtidos pelos diferentes métodos. No entanto, os coeficientes de repetibilidade variam entre as oito características, de 0,68 a 0,99, observados para a característica espessura do endocarpo e volume da amêndoa, respectivamente. As estimativas dos coeficientes de determinação obtidas a partir das 10 repetições para as oito características e pelos quatro métodos foram superiores a 95 por cento. A avaliação de quatro frutos é suficiente para compreensão das características estudadas das subamostras de macaúba com 90 por cento de acurácia.
Estimates of repeatability coefficient have been used in character study of several species of palms, helping to define the number and appropriate period of assessment of genotypes to increase the efficiency of breeding programs. For macaw palm there are no reports of such studies. In this context it is important to determine the repeatability coefficient of the following biometric characteristics of fruits: fruit weight, fruit volume, equatorial diameter of the fruit, kernel weight, kernel volume, equatorial diameter of de kernel, and thickness of the endocarp, and the determination of the minimum number of ratings for an efficient selection process and evaluation of macaw palm subsamples. The study was conducted at the Department of Plant Science, Federal University of Viçosa, and 145 macaw palm subsamples were analyzed, from collections made in the states of Minas Gerais, São Paulo, Mato Grosso do Sul, Pará, Maranhão and Pernambuco. There were significant differences for all traits analyzed, and there was agreement on the magnitude of repeatability coefficients of each feature obtained by different methods. However, the repeatability coefficients ranging between eight characteristics, from 0.68 to 0.99, and were observed for feature endocarp thickness and volume of kernel, respectively. The coefficient of determination obtained from 10 repetitions for the eight features and the four methods were superior to 95 percent. The evaluation of four fruits is sufficient to assess the traits of macaw palm subsamples with 90 percent accuracy.

Orthophytum grossiorum is a typical bromeliad from Atlantic forestry threatened of extinction. The objectives of this research were to select O. grossiorum clones with ornamental values easy to propagate in vitro, and establish in vitro propagation protocols for these clones. The project was developed in three steps: germination and in vitro selection of seedlings responsive to BAP (6-benzylaminopurine), selection of clones with ornamental values, and establishment of protocol for in vitro propagation of the selected clones. In the first step only 18.33 percent of plantlets germinated in vitro were responsive to BAP. These plantlets were selected and replicated in vitro several times, each replicated plantlet constituting a clone. In the second step these clones were established ex vitro and surveyed for ornamental attributes. Five out of 11 clones were selected in this step. These clones presented distinct phenotypic traits and were considered of high ornamental quality. In the third step a protocol for in vitro propagation was developed for each selected clone.
Orthophytum grossiorum é uma bromélia ameaçada de extinção típica de Mata Atlântica. Os objetivos deste trabalho foram selecionar clones de O. grossiorum com potencial ornamental e de fácil propagação in vitro e estabelecer protocolo de propagação in vitro para esses clones. O trabalho foi desenvolvido em três etapas: germinação e em seleção in vitro de plântulas responsivas a BAP (6-benzylaminopurine), seleção de clones com valores ornamentais e estabelecimento de protocolo para propagação in vitro dos clones selecionados. Na primeira etapa, foi observado que apenas 18.33 por cento das plântulas germinadas in vitro eram responsivas a BAP. Essas plântulas foram selecionadas e reproduzidas em in vitro, e cada plântula selecionada e reproduzida constituiu um clone. Na segunda etapa, esses clones foram estabelecidos ex vitro e selecionados em relação aos atributos ornamentais. Nessa etapa, foram selecionados cinco entre 11 clones. Esses apresentaram características fenotípicas distintas, sendo considerados de alta qualidade ornamental. Na terceira etapa, o protocolo para em propagação in vitro foi desenvolvido para cada clone selecionado.

A micropropagação de bananeira (Musa spp.) via ápices caulinares já está bem estabelecida para diversas cultivares. Entretanto, podem ser feitos aperfeiçoamentos nos protocolos visando a obtenção de mudas mais vigorosas. Objetivou-se, neste trabalho, verificar a adição de nitrogênio na forma de uréia em diferentes concentrações (0, 100, 200, 400 e 800 mg L-1), no alongamento e enraizamento de brotos das cultivares Grande Naine, Prata Anã, Maçã e Nanicão. Houve efeito positivo da uréia no crescimento dos explantes, na produção de raízes e de massa seca para a cultivar Maçã, até 200 mg L-1 de uréia. As cultivares Prata Anã e Grande Naine não foram beneficiadas com a adição de uréia, e a concentração de 800 mg L-1 gerou morte das microplantas dessas cultivares e da cultivar Maçã. 'Nanicão' foi mais tolerante às altas concentrações de uréia, e a concentração de 800 mg L-1 foi ótima para seu desenvolvimento. Além disso, a uréia induziu brotações em 'Nanicão' nas concentrações mais baixas.
Banana (Musa spp.) micropropagation via shoot tips has already been established for many cultivars. However, protocol adjustments aiming to obtain more vigorous plants can be made. The aim of this work was to verify the nitrogen addition in the form of urea on different concentrations (0, 100, 200, 400 and 800 mg L-1) on shoot growth and rooting of Grande Naine, Prata Anã, Maçã and Nanicão cultivars. There was a positive urea effect on Maçã shoot growth, dry matter production and rooting up to 200 mg L-1 concentrations. Prata Anã and Grande Naine cultivars were not benefited by urea addition, while 800 mg L-1 concentration lead to death of microplants. However, Nanicão cultivar was more tolerant to higher urea concentrations, and 800 mg L-1 was optimal for its development. Moreover, urea induced shoot growth on 'Nanicão' at lower concentrations.
