RESUMO

Coronavirus disease 2019 (COVID-19) caused by SARS-CoV-2 is characterized by acute respiratory distress syndrome (ARDS) facilitated by cytokine storm and other risk factors that increase susceptibility and complications leading to death. Emerging as a major global public health challenge, the disease has claimed more than 6 million lives and caused catastrophic global economic disruptions. However, there are concerns about the safety as well as the efficacy of drugs and vaccines presently used to control the pandemic, therefore necessitating intense global search for safe natural products that can effectively and safely combat it. This work reviews studies on lingzhi or reishi medicinal mushroom, Ganoderma lucidum and its properties that may potentially combat SARS-CoV-2 infection and the co-morbidities. Available evidence suggests that medicinal properties of the Ganoderma mushroom can combat the complications of SARS-CoV-2 infection and the co-morbidities that can aggravate the severity of the disease. Preclinical and clinical evaluation to establish dose, efficacy, and potential toxicity and possible use in the management of COVID-19 is recommended.

BACKGROUND: Brucellosis is an infectious disease caused by bacteria of the genus Brucella. Although it is the most common zoonosis worldwide, there are increasing reports of drug resistance and cases of relapse after long term treatment with the existing drugs of choice. This study therefore aims at identifying possible natural inhibitors of Brucella melitensis Methionyl-tRNA synthetase through an in-silico approach. METHODS: Using PyRx 0.8 virtual screening software, the target was docked against a library of natural compounds obtained from edible African plants. The compound, 2-({3-[(3,5-dichlorobenzyl) amino] propyl} amino) quinolin-4(1H)-one (OOU) which is a co-crystallized ligand with the target was used as the reference compound. Screening of the molecular descriptors of the compounds for bioavailability, pharmacokinetic properties, and bioactivity was performed using the SWISSADME, pkCSM, and Molinspiration web servers respectively. The Fpocket and PLIP webservers were used to perform the analyses of the binding pockets and the protein ligand interactions. Analysis of the time-resolved trajectories of the Apo and Holo forms of the target was performed using the Galaxy and MDWeb servers. RESULTS: The lead compounds, Strophanthidin and Isopteropodin are present in Corchorus olitorius and Uncaria tomentosa (Cat's-claw) plants respectively. Isopteropodin had a binding affinity score of -8.9 kcal / ml with the target and had 17 anti-correlating residues in Pocket 1 after molecular dynamics simulation. The complex formed by Isopteropodin and the target had a total RMSD of 4.408 and a total RMSF of 9.8067. However, Strophanthidin formed 3 hydrogen bonds with the target at ILE21, GLY262 and LEU294, and induced a total RMSF of 5.4541 at Pocket 1. CONCLUSION: Overall, Isopteropodin and Strophanthidin were found to be better drug candidates than OOU and they showed potentials to inhibit the Brucella melitensis Methionyl-tRNA synthetase at Pocket 1, hence abilities to treat brucellosis. In-vivo and in-vitro investigations are needed to further evaluate the efficacy and toxicity of the lead compounds.

This study evaluated the prevalence, hematological and some biochemical alterations in Brucella seropositive Muturu breed of cattle which may change the narratives of multi-disease tolerance of the breed. Sera from 33 Muturu cattle herds chosen by snow ball sampling were screened for Brucella antibodies with modified Rose Bengal test (RBT) supported with the cELISA. Eighteen (18) seropositive samples matched to18 sero-negatives, with regard to age and sex of the animals and chosen by simple random sampling, were analyzed for hematological and biochemical changes, following standard procedures. Individual seroprevalence of 38% and 10% were recorded with the RBT and cELISA respectively, while herd seroprevalence of 52% was recorded with the RBT and none with the cELISA. Seropositivity to brucellosis was significantly associated with farm origin (OR = 16.67; 95%CI = 1.56-153.85; p = 0.019). There was significantly lower packed cell volume (PCV) (p = 0.048) and absolute eosinophil count (p = 0.006), and significantly higher absolute lymphocyte count (p = 0.014) in the seropositive than the negative Muturu cattle. In addition, plasma fibrinogen (p < 0.001), serum albumin (p = 0.037), urea (p = 0.001) and cholesterol (p = 0.032) were significantly lower while serum globulin (p = 0.004), alanine aminotransferase (ALT) activity (p = 0.012) and bilirubin (p = 0.012) were significantly higher in the seropositive than sero-negative Muturu cattle. No significant variations were observed in the rest of the parameters assayed. These findings suggest that Muturu cattle are apparently susceptible to brucellosis and experience active organism-host interactions with resultant clinicopathological effects and therefore could be passive harbingers of Brucella for other animals as well as humans.

Good antimicrobial drug stewardship in food­producing animals boosts productivity and limits transmission of zoonotic pathogens, but the imprudent usage is counterproductive. A nation­wide survey to determine the pattern of antimicrobial drug usage in poultry was therefore conducted across 12 of the 36 States in Nigeria. The survey was conducted using structured questionnaire designed to elicit information on socio­demographics, pattern of antimicrobial drug usage and awareness of the consequences of imprudent use of antimicrobials in food­producing animals, among 2,402 randomly selected poultry farmers. Critically important antimicrobials, belonging to the WHO's lists of 'Highest' and 'High' priority antibiotics, were administered sub­therapeutically for prophylaxis and growth enhancement purposes. Many diseases that warranted the antimicrobial administration were of viral etiology. Only 64.2% of the farmers administered the drug based on veterinary doctors' prescription. Majority (62.3%) of the farmers did not observe recommended withdrawal period after the drug administration. Awareness of the respondents on consequences of non­prudent agricultural use of antimicrobials was generally poor. There is need for enlightenment of the farmers on the benefits of prudent agricultural use of antimicrobials. Immediate discontinuation of sale of essential antimicrobials as over­the­counter drugs is imperative to safeguard their therapeutic efficacy and curtail spread of antimicrobial­resistance organisms.

Using a cross-sectional survey, we determined the prevalence and risk factors associated with bovine brucellosis in herds under extensive production system in southwestern Nigeria. Antibodies to Brucella species in serum samples were tested using the Rose Bengal test (RBT) and competitive enzyme-linked immunosorbent assay (cELISA); for milk, the milk ring test (MRT) and indirect-ELISA (i-ELISA) were used. Questionnaire was administered to cattle herdsmen to determine factors predisposing the animals to bovine brucellosis. Data were analyzed using STATA 12. From 513 serum and 635 milk samples tested among 120 herds, overall animal-level prevalence of 10.1% (95% CI 7.5-12.7%) and 20.2% (95% CI 17.1-23.3%) were recorded by RBT and MRT, respectively; while 9.4% (95% CI 6.9-11.9%) and 17.8% (95% CI 14.8-20.8%) were obtained using cELISA and i-ELISA, respectively. In all, from the 120 herds tested, 29.2% and 43.3% were positive by RBT and MRT, respectively. Multivariable logistic regression revealed that herd location (OR = 8.12, 95% CI 1.68-38.90) and improper disposal of placenta/fetus (OR = 17.33, 95% CI 4.81-62.33) were predictors for a seropositive herd using RBT; while herd location (OR = 5.13, 95% CI 1.27-20.28), large herd size (OR = 2.62, 95% CI 1.15-5.85), and occurrence of abortion for a year or more (OR = 4.62, 95% CI 1.53-13.71) were predictors of seropositivity to antibodies to Brucella spp. using MRT. We found high prevalence of brucellosis in cattle herds under extensive management system in southwestern Nigeria. Urgent and coordinated control strategies are required to mitigate this problem.

INTRODUCTION: In Nigeria, there is limited information on brucellosis particularly in dogs, despite its public health implications. We undertook a sero-epidemiological survey of brucellosis in dogs to determine the prevalence of the disease and associated risk factors for its occurrence in Nigeria. METHODS: A cross-sectional study was conducted to screen dogs in south-western Nigeria for antibodies to Brucella sp using the rapid slide agglutination test (RSA) and Rose Bengal test (RBT), with positive samples confirmed respectively by serum agglutination test (SAT) and competitive enzyme linked immunosorbent assay (cELISA). Data were analyzed with STATA-12. RESULTS: From the 739 dog sera tested, 81 (10.96%) were positive by RSA and 94 (12.72%) by RBT; these were corroborated with SAT (4/81; 4.94%) and cELISA (1/94; 1.06%), respectively. Logistic regression identified location (OR=0.04; 95% CI: 0.02-0.09), breed (OR=1.71; 95% CI: 1.34-2.19), age (OR=0.10; 95% CI: 0.04-0.30) and management system (OR=8.51; 95% CI: 1.07-68.05) as risk factors for Brucella infection by RSA. However, location (OR=10.83; 95% CI: 5.48-21.39) and history of infertility (OR=2.62; 95% CI: 1.41-4.84) were identified as risk factors using RBT. CONCLUSION: Given the 10.96% to 12.72% seroprevalence of brucellosis recorded in this study, we advocate control of the disease in dogs, and public health education for those at risk of infection. Again, further studies are required to elucidate the role of dogs in the epidemiology of brucellosis in Nigeria considering the conducive human-animal interface and ecological factors responsible for the transmission of the disease.
