RESUMO

Widespread contamination of soils by neurotoxic lead ions (Pb) posed a serious risk to food security, but efficient treatment in soil remained a challenge. For the adsorption of Pb, DFT calculations were firstly performed to predict the synergistic effect of sulfhydryl-hydroxyl groups as well as the ability of sulfur ions to strengthen Pb-OH bonding. Consequently, Mn-S functionalized coffee ground biochar (MSBC) was then synthesized utilizing precipitation and impregnation methods. In the soil experiment, the removal efficiency of Pb reached 82.92%, exceeding the previous research results. In addition, it successfully restored the polluted farmland near the mining area and increased the plant height of Swiss chard by 186.23%. Subsequently, synergistic effect of sulfhydryl-hydroxyl groups was confirmed by XPS, FT-IR, and DFT calculations. Furthermore, the factors affecting the structural stability of O-Pb-S were discussed by regression analysis. These reflected that MSBC can enhance the removal efficiency of Pb in soil by mitigating the competition of impurity ions to adsorption sites. These findings may provide new insights into the development of the specific passivation materials for other heavy metals.

Objective: To reveal the efficacy and potential mechanisms of electroacupuncture (EA) in treating hypertension. Methods: Male spontaneously hypertensive rats (SHRs) were randomly assigned to the SHR group, EA group, and Sham-EA group, with Wistar-Kyoto rats (WKY) as the normal control group. SHRs in the EA group received electroacupuncture at the bilateral Taichong (LR3) acupoints for 7 consecutive days. Evaluation of systolic blood pressure (SBP), diastolic blood pressure (DBP), mean arterial pressure (MAP), and heart rate (HR) was conducted. Positron emission tomography-computed tomography (PET-CT) was employed to explore the active brain regions associated with acupuncture-induced blood pressure reduction. Furthermore, mRNA expression profiling was analyzed in the active brain regions to identify differentially expressed genes, and quantitative polymerase chain reaction (qPCR) was used to validate the mRNA expression of differentially expressed genes in the active brain region. Results: EA reduced elevated SBP, DBP, MAP and HR in SHR. PET-CT revealed that EA decreased glucose metabolism in the hypothalamus. Genomic analysis suggested that, compared to the SHR group, the differentially expressed genes in the hypothalamus of the EA group included Nr4a1, Sirt1, Trh, GPR88, Cck, and Th. EA downregulated the mRNA expression of Th, Trh, Gpr88, and Nr4a1, while upregulating the expression of Sirt1 and Cck at the mRNA level. Conclusion: EA may exert a unique antihypertensive effect in the hypothalamus of SHR, involving the modulation of sympathetic nerve activity, neuroinflammation, and oxidative stress response.

BACKGROUND: The diagnosis of tuberculous pleurisy (TP) presents a significant challenge due to the low bacterial load in pleural effusion (PE) samples. Cell-free Mycobacterium tuberculosis DNA (cf-TB) in PE samples is considered an optimal biomarker for diagnosing TP. This study aimed to evaluate the applicability of cf-TB testing across diverse research sites with a relatively large sample size. METHODS: Patients suspected of TP and presenting with clinical symptoms and radiological evidence of PE were consecutively enrolled by treating physicians from 11 research sites across 6 provinces in China between April 2020 and August 2022. Following centrifugation, sediments obtained from PE were used for Xpert MTB/RIF (Xpert) and mycobacterial culture, while the supernatants were subjected to cf-TB testing. This study employed a composite reference standard to definite TP, which was characterized by any positive result for Mycobacterium tuberculosis (MTB) through either PE culture, PE Xpert, or pleural biopsy. RESULTS: A total of 1412 participants underwent screening, and 1344 (95.2%) were subsequently enrolled in this study. Data from 1241 (92.3%) participants were included, comprising 284 with definite TP, 677 with clinically diagnosed TP, and 280 without TP. The sensitivity of cf-TB testing in definite TP was 73.6% (95% CI 68.2-78.4), significantly higher than both Xpert (40.8%, 95% CI 35.3-46.7, P < 0.001) and mycobacterial culture (54.2%, 95% CI 48.4-59.9, P < 0.001). When clinically diagnosed TP was incorporated into the composite reference standard for sensitivity analysis, cf-TB testing showed a sensitivity of 46.8% (450/961, 95% CI 43.7-50.0), significantly higher than both Xpert (116/961, 12.1%, 95% CI 10.2-14.3, P < 0.001) and mycobacterial culture (154/961, 16.0%, 95% CI 13.8-18.5, P < 0.001). The specificities of cf-TB testing, Xpert, and mycobacterial culture were all 100.0%. CONCLUSIONS: The performance of cf-TB testing is significantly superior to that of Xpert and mycobacterial culture methods, indicating that it can be considered as the primary diagnostic approach for improving TP detection. Promoters are important cis-regulatory elements for the regulation of gene expression, and their accurate predictions are crucial for elucidating the biological functions and potential mechanisms of genes. Many previous prokaryotic promoter prediction methods are encouraging in terms of the prediction performance, but most of them focus on the recognition of promoters in only one or a few bacterial species. Moreover, due to ignoring the promoter sequence motifs, the interpretability of predictions with existing methods is limited. In this work, we present a generalized method Prompt (Promoters in multiple prokaryotes) to predict promoters in 16 prokaryotes and improve the interpretability of prediction results. Prompt integrates three methods including RSK (Regression based on Selected k-mer), CL (Contrastive Learning) and MLP (Multilayer Perception), and employs a voting strategy to divide the datasets into high-confidence and low-confidence categories. Results on the promoter prediction tasks in 16 prokaryotes show that the accuracy (Accuracy, Matthews correlation coefficient) of Prompt is greater than 80% in highly credible datasets of 16 prokaryotes, and is greater than 90% in 12 prokaryotes, and Prompt performs the best compared with other existing methods. Moreover, by identifying promoter sequence motifs, Prompt can improve the interpretability of the predictions. In this study, amino-modified graphene oxide(NGO) was prepared by introducing amino functional groups. Based on the cross-linking between Ca(II) and sodium alginate (SA), associated with dense slit-like pore resulted from the nano-sheet accumulation of NGO and montmorillonite (MMT), composite aerogels (NGM) with stable pore structure were constructed, thus it realized the selective recovery of hydrated copper ions in complex wastewater systems. Raman analysis and density functional theory calculation confirmed the construction of amino-modified defect GO and significantly improved its chemical reactivity, which laid the foundation for the construction of slit pore structure of NGM (SEM can confirm). At the same time, it proposed that the good selective adsorption of Cu(II) on NGM was related to the synergism of strong electrostatic force, ion exchange and complexation based on the characterizations of FT-IR and XPS. In order to realize the value-added utilization of NGM aerogel (NGMC) after adsorbing Cu(II), NGMC was used as a catalyst to degrade organic pollutants in wastewater. Systematic experiments shown that NGMC can degrade organic pollutants with a degradation efficiency >80 %. In summary, NGM had a broad application prospect for selective recovery of Cu(II) from complex wastewater systems without second pollution.

Phase change materials (PCMs) possess the potential to regulate temperature by utilizing their thermal properties to absorb and release heat. Nevertheless, the application of PCMs in thermal management is constrained by issues such as liquid leakage and limited flexibility. In this study, we propose a novel approach to address these challenges by incorporating a pore structure within nanofibers to confine the crystallization of phase change molecules, thereby enhancing the flexibility of the composite material. Additionally, inspired by the adaptive mechanisms observed in plants, we have developed a form stable PCM based on polyether, which effectively mitigates the issue of liquid leakage at higher temperatures. Despite being a solid-liquid PCM at its core, this material exhibits molecular-scale flow and macroscopic shape stability as a result of intermolecular forces. The composite film material possesses remarkable flexibility, efficient thermal management capabilities, adjustable phase transition temperature, and the ability to undergo repeated processing and utilization. Consequently, it holds promising potential for applications in personal thermal energy management.

This research addresses and resolves current challenges in meshless Lagrangian methods for simulating viscoelastic materials. A split-step scheme, or pressure Poisson reformulation of the Navier-Stokes equations, is introduced for incompressible viscoelastic flows in a Lagrangian context. The Lagrangian differencing dynamics (LDD) method, which is a thoroughly validated Lagrangian method for Newtonian and non-Newtonian incompressible flows, is extended to solve the introduced split-step scheme to simulate viscoelastic flows based on the Oldroyd-B constitutive model. To validate and evaluate the new method's capabilities, the following benchmarks were used: lid-driven cavity flow, droplet impact response, 4:1 planar sudden contraction, and die swelling. These findings highlight the LDD method's effectiveness in accurately simulating viscoelastic flows and capturing large deformations and memory effects. Even though the extra stress was directly modeled without any regularization approach, the method produced stable simulations for high Weissenberg numbers. The stability and performance of the the Lagrangian numerics for complex temporal evolution of material properties and stress responses encourage its use for industrial problems dealing with polymers.

OBJECTIVES: Carbapenem-resistant Klebsiella pneumoniae (CRKP), a superbug that can be difficult or impossible to treat, has become a worldwide problem. This study presents the first report of a CRKP strain carrying a plasmid co-harbouring blaNDM-1, blaKPC-2, and tet(A) and the subsequent analysis of its genomic features. METHODS: Isolation and identification of bacteria, antimicrobial susceptibility test, whole genome sequencing, and conjugation experiments assay were conducted in clinical epidemiological investigations and plasmid genetic characterisation analysis. RESULTS: A total of 116 strains of bacteria were isolated from patients with bloodstream infections (BSI) between 2018 and 2023. A total of 89.66% of the isolates were carbapenem-resistant Enterobacteriaceae (CRE), with the majority (75/116) being CRKP. Among these, a novel plasmid co-harbouring blaNDM-1, blaKPC-2, and tet(A) simultaneously was found in CRKP46, and the three genes mediated conjugation by IS26, ISAba125, and IS26, respectively. This plasmid conferred carbapenem resistance to E. coli J53 after conjugative transfer, which was 2 times greater than that of CRKP46. CONCLUSION: The present study identified the occurrence of a rare plasmid co-harbouring blaNDM-1, blaKPC-2, and tet(A), and the spread of these genes was mediated by the corresponding mobile elements. The increased carbapenem resistance created by this novel plasmid challenges public health security and poses a potential threat to human health; therefore, it deserves attention.

MnSO4-modified biochar (Mn-BC) was synthesized to remove berberine hydrochloride (BH) from wastewater by utilizing tea waste as raw material and MnSO4 as modifier. Brunel Emmett Taylor (BET) analysis reveals that the specific surface area (SSA) and average pore size (Dave) of Mn-BC are 1.4 and 7 times higher than those of pristine biochar apart, attributing to the dissociation effect can promote the dispersion of MnSO4 in the pores of the biochar. Meanwhile, the doping of Mn not only introduces additional oxygen-containing functional groups (OCFGs), but also modulates the π electron density. Furthermore, Response surface method (RSM) analysis reveals that Mn-BC dosage has the most significant effect on BH removal, followed by BH concentration and pH value. Kinetic and isothermal studies reveal that the BH adsorption process of Mn-BC was mainly dominated by chemical and monolayer adsorption. Meanwhile, density functional theory (DFT) calculations confirm the contribution of Mn doping to the conjugation effect in the adsorption system. Originally proposed Mn-BC is one potentially propitious material to eliminate BH from wastewater, meanwhile this also provides a newfangled conception over the sustainable utilization of tea waste resources.

Alzheimer's disease (AD) is a universal neurodegenerative disease with the feature of progressive dementia. Currently, there are only seven Food and Drug Administration-approved drugs for the treatment of AD, which merely offer temporary relief from symptom deterioration without reversing the underlying disease process. The identification of inhibitors capable of interacting with proteins associated with AD plays a pivotal role in the development of effective therapeutic interventions. However, a vast number of such inhibitors are dispersed throughout numerous published articles, rendering it inconvenient for researchers to explore potential drug candidates for AD. In light of this, we have manually compiled inhibitors targeting proteins associated with AD and constructed a comprehensive database known as IPAD-DB (Inhibitors of Proteins associated with Alzheimer's Disease Database). The curated inhibitors within this database encompass a diverse range of compounds, including natural compounds, synthetic compounds, drugs, natural extracts and nano-inhibitors. To date, the database has compiled >4800 entries, each representing a correspondent relationship between an inhibitor and its target protein. IPAD-DB offers a user-friendly interface that facilitates browsing, searching and downloading of its records. We firmly believe that IPAD-DB represents a valuable resource for screening potential AD drug candidates and investigating the underlying mechanisms of this debilitating disease. BACKGROUND: Non-alcoholic steatohepatitis (NASH) and liver fibrosis are progressive conditions associated with non-alcoholic fatty liver disease (NAFLD), characterized by hepatocyte pyroptosis and hepatic stellate cell (HSC) activation. Gentiopicroside (GPS) has emerged as a potential treatment for NASH, yet its underlying mechanism remains unclear. AIM: To confirm that GPS can improve NASH and liver fibrosis by blocking the NLRP3 signaling pathway STUDY DESIGN: Initially, different animal models were used to study the effects and mechanisms of GPS on NASH and fibrosis. Subsequent in vitro experiments utilized co-cultures and other techniques to delve deeper into its mechanism, followed by validation of the findings in mouse liver tissues. METHODS: C57BL/6 mice were fed high-fat, high-cholesterol (HFHC), or methionine-choline-deficient (MCD) diets to induce NASH and fibrosis. RAW264.7 cells and born marrow bone marrow-derived macrophages (BMDMs) were stimulated with LPS and ATP to induce inflammation, then co-cultured with primary hepatocytes and HSCs, treated with GPS, and its efficacy and mechanism were analyzed. RESULTS: In vivo, GPS alleviated NASH and liver fibrosis by inhibiting the NLRP3 pathway. In vitro, GPS attenuated inflammation induced by BMDMs by inhibiting TLR4 and NLRP3 signaling pathways, and Co-culture studies suggested that GPS reduced hepatocyte pyroptosis and HSC activation, which was also confirmed in liver tissues CONCLUSION: GPS improves NASH and liver fibrosis by inhibiting the TLR4 and NLRP3 signaling pathways. The specific mechanism may be related to the suppression of macrophage-mediated inflammatory responses, thereby reducing hepatocyte pyroptosis and HSC activation.

The massive burning of fossil fuels has been important for economic and social development, but the increase in the CO2 concentration has seriously affected environmental sustainability. In industrial and agricultural production, light olefins are one of the most important feedstocks. Therefore, the preparation of light olefins by CO2 hydrogenation has been intensively studied, especially for the development of efficient catalysts and for the application in industrial production. Fe-based catalysts are widely used in Fischer-Tropsch synthesis due to their high stability and activity, and they also exhibit excellent catalytic CO2 hydrogenation to light olefins. This paper systematically summarizes and analyzes the reaction mechanism of Fe-based catalysts, alkali and transition metal modifications, interactions between active sites and carriers, the synthesis process, and the effect of the byproduct H2O on catalyst performance. Meanwhile, the challenges to the development of CO2 hydrogenation for light olefin synthesis are presented, and future development opportunities are envisioned.

BACKGROUND: The intricate etiology of autoimmune liver disease (AILD) involves genetic, environmental, and other factors that yet to be completely elucidated. This study comprehensively assessed the causal association between genetically predicted modifiable risk factors and AILD by employing Mendelian randomization. METHODS: Genetic variants associated with 29 exposure factors were obtained from genome-wide association studies (GWAS). Genetic association data with autoimmune hepatitis (AIH), primary biliary cholangitis (PBC) and primary sclerosing cholangitis (PSC) were also obtained from publicly available GWAS. Univariate and multivariate Mendelian randomization analyses were performed to identify potential risk factors for AILD. RESULTS: Genetically predicted rheumatoid arthritis (RA) (OR = 1.620, 95%CI 1.423-1.843, P = 2.506 × 10- 13) was significantly associated with an increased risk of AIH. Genetically predicted smoking initiation (OR = 1.637, 95%CI 1.055-2.540, P = 0.028), lower coffee intake (OR = 0.359, 95%CI 0.131-0.985, P = 0.047), cholelithiasis (OR = 1.134, 95%CI 1.023-1.257, P = 0.017) and higher C-reactive protein (CRP) (OR = 1.397, 95%CI 1.094-1.784, P = 0.007) were suggestively associated with an increased risk of AIH. Genetically predicted inflammatory bowel disease (IBD) (OR = 1.212, 95%CI 1.127-1.303, P = 2.015 × 10- 7) and RA (OR = 1.417, 95%CI 1.193-1.683, P = 7.193 × 10- 5) were significantly associated with increased risk of PBC. Genetically predicted smoking initiation (OR = 1.167, 95%CI 1.005-1.355, P = 0.043), systemic lupus erythematosus (SLE) (OR = 1.086, 95%CI 1.017-1.160, P = 0.014) and higher CRP (OR = 1.199, 95%CI 1.019-1.410, P = 0.028) were suggestively associated with an increased risk of PBC. Higher vitamin D3 (OR = 0.741, 95%CI 0.560-0.980, P = 0.036) and calcium (OR = 0.834, 95%CI 0.699-0.995, P = 0.044) levels were suggestive protective factors for PBC. Genetically predicted smoking initiation (OR = 0.630, 95%CI 0.462-0.860, P = 0.004) was suggestively associated with a decreased risk of PSC. Genetically predicted IBD (OR = 1.252, 95%CI 1.164-1.346, P = 1.394 × 10- 9), RA (OR = 1.543, 95%CI 1.279-1.861, P = 5.728 × 10- 6) and lower glycosylated hemoglobin (HbA1c) (OR = 0.268, 95%CI 0.141-0.510, P = 6.172 × 10- 5) were positively associated with an increased risk of PSC. CONCLUSIONS: Evidence on the causal relationship between 29 genetically predicted modifiable risk factors and the risk of AIH, PBC, and PSC is provided by this study. These findings provide fresh perspectives on the management and prevention strategies for AILD.

In this study, electrokinetically-delivered persulfate (PS) coupled with thermal conductive heating (TCH) method was proposed for the remediation of petroleum hydrocarbons (PHs) contaminated low-permeability soil, based on the investigation of PS injection and activation by different electric field form, effective heating radius of TCH to activate PS, and their influencing factors. The uniform delivery and effective activation of PS were unrealizable by one-dimensional electric field (1 V/cm) with the operation of cathode injection, anode injection, bipolar injection, polarity-reversal, or bipolar injection coupled polarity-reversal, which would result in large spatial difference of soil pH and PHs residual. Similar results were obtained under the two-dimensional symmetric electric field (TEF) due to the large spatial difference in electric field intensity. Superimposed electric field (SEF, 1 V/cm) that based on the intermittent worked electrode groups coupled with polarity-reversal (every 3 h) and bipolar injection (10% PS solution) operation could achieve homogenized mass transfer of PS (53.8-65.7 g/kg, average 60.0 g/kg) in 15 days, due to the positive correlation between electric field intensity and transport of ionic substance. Meanwhile, the difference in decontamination efficiency caused by difference in PS activation efficiency could be reduced, since the heating rod was placed at the position where the concentrations of PS was the lowest, whereat the removal of PHs could not rely on alkali activated PS (cathode), anodic oxidation (anode), and electrochemical activated PS (cathode and anode). The residual concentration of PHs in soil remediated by SEF/PS-TCH was in the range of 640.7-763.8 mg/kg (average 701.5 mg/kg), and the corresponding removal efficiency was 73.3%-77.6% (average75.4%). The research can provide an in-situ remediation method for organic contaminants in low permeability soil featured with more uniform PS injection and activation, and small spatial differences in remediation efficiency.

Background and objective: Cognitive-behavioral therapy (CBT) for somatoform disorders (SFDs) is understudied in China. Western findings may not be applicable to Chinese culture. This preliminary study evaluated the efficacy of CBT for patients in China, relative to treatment-as-usual (TAU). Methods: Seventy patients with SFDs randomly received either combined CBT and TAU (CBT + TAU), or TAU alone between January 2018 to May 2019. The CBT + TAU group received 12 weekly individual 50-minute CBT sessions. Participants were blindly assessed at 4 timepoints (baseline, week 6, end of treatment: week 12; 12 weeks post-treatment: week 24) using the following outcome measures: SQSS (Self-screening Questionnaire for Somatic Symptoms); PHQ-15 (Patient-Health-Questionnaire-15) and the WI (Whiteley Index); GAD-7 (General Anxiety Disorder-7); HAMD-17 (Hamilton Depression Rating Scale-17); Family Burden Interview Schedule (FBIS); Sheehan Disability Scale (SDS); and the Short Form of Quality-of-Life Enjoyment and Satisfaction Questionnaire (Q-LES-Q-SF). The primary endpoint was the difference between the SQSS total score at week 24 and the baseline. A mixed model for repeated measures was used to analyze inter- and intra-group changes from the baseline. Results: At week 24, The least-squares mean (LSM) change of the total score on the SQSS was -18.87 points and -9.69 points, respectively in the CBT + TAU group and in the TAU group (LSM difference, -9.18 points; 95% confidence interval, -15.72 to -2.64; P = 0.0068). At week 24, the LSM changes from baseline in the WI, HAMD, PHQ15, FBIS and SDS total scores were significantly different between the two groups, however, there was no significant difference in the Q-LES-Q-SF. The SQSS of group effect sizes were 0.63 at 24 weeks. The dropout rates of the CBT + TAU and TAU groups were comparable (22.9% and 19.3%). Conclusions: These preliminary findings suggest that CBT may be helpful for improving the symptoms of patients with SFDs in China.

Multi-view subspace clustering (MVSC) has drawn significant attention in recent study. In this paper, we propose a novel approach to MVSC. First, the new method is capable of preserving high-order neighbor information of the data, which provides essential and complicated underlying relationships of the data that is not straightforwardly preserved by the first-order neighbors. Second, we design log-based nonconvex approximations to both tensor rank and tensor sparsity, which are effective and more accurate than the convex approximations. For the associated shrinkage problems, we provide elegant theoretical results for the closed-form solutions, for which the convergence is guaranteed by theoretical analysis. Moreover, the new approximations have some interesting properties of shrinkage effects, which are guaranteed by elegant theoretical results. Extensive experimental results confirm the effectiveness of the proposed method.

Gentiopicroside (GPS) is a highly water-soluble small-molecule drug and the main bioactive secoiridoid glycoside of Gentiana scabra that has been shown to have hepatoprotective effects against non-alcoholic steatohepatitis (NASH), a form of non-alcoholic fatty liver disease (NAFLD) that can progress to cirrhosis and hepatocellular carcinoma. However, the effects of GPS on NASH and the underlying mechanisms remain obscure. Firstly, a high-fat, high-cholesterol (HFHC) diet and a high-sugar solution containing d-fructose and d-glucose were used to establish a non-alcoholic steatohepatitis (NASH) mice model. Secondly, we confirmed GPS supplementation improve metabolic abnormalities and reduce inflammation in NASH mice induced by HFHC and high-sugar solution. Then we used metabolomics to investigate the mechanisms of GPS in NASH mice. Metabolomics analysis showed GPS may work through the Peroxisome Proliferator-Activated Receptor (PPAR) signaling pathway and glycine, serine, and threonine metabolism. Functional metabolites restored by GPS included serine, glycine, eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA). Western blot and qRT-PCR analysis confirmed GPS improve NASH by regulating PPARα and Hypoxia-Inducible Factor-1α (HIF-1α) signaling pathways. In vitro, studies further demonstrated EPA and DHA enhance fatty acid oxidation through the PPARα pathway, while serine and glycine inhibit oxidative stress through the HIF-1α pathway in palmitic acid-stimulated HepG2 cells. Our results suggest GPS's anti-inflammatory and anti-steatosis effects in NASH progression are related to the suppression of HIF-1α through the restoration of L-serine and glycine and the activation of PPARα through increased EPA and DHA.

The metal supported catalysts are emerging catalysts that are receiving a lot of attention in CO2 hydrogenation to C1 products. Numerous experiments have demonstrated that the support (usually an oxide) is crucial for the catalytic performance. The support metal oxides are used to aid in the homogeneous dispersion of metal particles, prevent agglomeration, and control morphology owing to the metal support interaction (MSI). MSI can efficiently optimize the structural and electronic properties of catalysts and tune the conversion of key reaction intermediates involved in CO2 hydrogenation, thereby enhancing the catalytic performance. There is an increasing attention is being paid to the promotion effects in the catalytic CO2 hydrogenation process. However, a systematically understanding about the effects of MSI on CO2 hydrogenation to C1 products catalytic performance has not been fully studied yet due to the diversities in catalysts and reaction conditions. Hence, the characteristics and modes of MSI in CO2 hydrogenation to C1 products are elaborated in detail in our work.

RESUMO

Plastics have revolutionized our lives; however, the exponential growth of their usage has led to a global crisis. More sustainable strategies are needed to address this dilemma and transform the plastics economy from a linearity to a circular model. Herein, we systematically summarize the recent progress in renewable energy-driven plastic conversion strategies, including photocatalysis, electrocatalysis, and their integration. By introducing the significant works, the design principles, mechanisms, and system regulations, we decipher and compare the various aspects of plastic conversion. These approaches show high reactivity and selectivity under environmentally benign conditions and provide alternative reaction pathways for plastic conversion. Plastic upcycling as a chemical feedstock can yield value-added chemicals and fuels, contributing to the establishment of a sustainable and circular economy. Additionally, several innovations in reaction engineering and system designs are presented. Finally, the challenges and perspectives of sustainable energy-driven plastic conversion technologies are comprehensively discussed.

BACKGROUND: Neurofibrillary tangles (NFTs) are one of the most common pathological characteristics of Alzheimer's disease. The NFTs are mainly composed of hyperphosphorylated microtubule-associated tau. Thus, recombinant tau is urgently required for the study of its fibrillogenesis and its associated cytotoxicity. METHODS AND RESULTS: Heterologous expression, purification, and fibrillation of the microtubule-binding domain (MBD) of tau (tauMBD) were performed. The tauMBD was heterologously expressed in E. coli. Ni-chelating affinity chromatography was then performed to purify the target protein. Thereafter, tauMBD was systematically identified using the SDS-PAGE, western blot and MALDI-TOF MS methods. The aggregation propensity of the tauMBD was explored by both the thioflavin T fluorescence and atomic force microscopy experiments. CONCLUSIONS: The final yield of the recombinant tauMBD was ~ 20 mg L-1. It is shown that TauMBD, in the absence of an inducer, self-assembled into the typical fibrils at a faster rate than wild-type tau. Finally, the in vitro cytotoxicity of tauMBD aggregates was validated using PC12 cells. The heterologously expressed tau in this study can be further used in the investigation of the biophysical and cellular cytotoxic properties of tau.
