RESUMO

Ecitonides constanceae sp. nov., a new species of the myrmecophilous genus Ecitonides Wasmann, 1894, which is associated with army ants (Hymenoptera: Formicidae: Ecitoninae), is described from Peru. The holotype, which is the only known specimen, was collected at light at Los Amigos Biological Station, Madre de Dios department, Peru. The army ant host species is unknown. The new species is distinguished from congeners and an updated key of the species, as well as a checklist of Ecitonides and their associated host ants are provided. Based on the holotype we non-destructively extracted DNA and sequenced two genes which were incorportated into an existing molecular phylogenetic dataset and analysed using Bayesian Inference. Ecitonides is recovered as sister to recently described thermitophilous genus Ruptor ya et al. 2022 and placed in the so-called Medonina and allied taxa clade. Beyond Bayesian Inference phylogenetic analysis and their associated nodal support methods, we also explored the position of the Ecitonides within the posterior sample of trees produced in the Bayesian analyses, which further supported its placement.

RESUMO

Solitary aculeate wasps are abundant and diverse hymenopteran insects that disable prey using venom. The venom may possess neuromodulation, immunomodulatory, metabolic-modulatory and antimicrobial functions. Venom analysis of transcriptomes and proteomes has been previously performed in social and parasitoid wasp species. We develop methodologies including mass spectrometry-based shotgun proteomics to analyse the protein constituents from venom sacs of the solitary aculeate wasp Cerceris rybyensis. The venom sac constituents of C. rybyensis are discussed with respect to other wasp species.

Spatial and temporal collections provide important data on the distribution and dispersal of species. Regional-scale monitoring invariably involves hundreds of thousands of samples, the identification of which is costly in both time and money. In this respect, metabarcoding is increasingly seen as a viable alternative to traditional morphological identification, as it eliminates the taxonomic bottleneck previously impeding such work. Here, we assess whether terrestrial arthropods collected from 12 pitfall traps in two farms of a coffee (Coffea arabica L.) growing region of Sao Paulo State, Brazil could differentiate the two locations. We sequenced a portion of the cytochrome oxidase 1 region from minimally processed pools of samples and assessed inter- and intraspecific parameters across the two locations. Our sequencing was sufficient to circumscribe the overall diversity, which was characterized by few dominant taxa, principally small Coleoptera species and Collembola. Thirty-four operational taxonomic units were detected and of these, eight were present in significantly different quantities between the two farms. Analysis of community-wide Beta diversity grouped collections based on farm provenance. Moreover, haplotype-based analyses for a species of Xyleborus beetle showed that there is significant population genetic structuring between the two farms, suggesting limited dispersal. We conclude that metabarcoding can provide important management input and, considering the rapidly declining cost of sequencing, suggest that large-scale monitoring is now feasible and can identify both the taxa present as well as contribute information about genetic diversity of focal species.

BACKGROUND: Falls are associated with morbidity, loss of independence, and mortality. While land-based group exercise and Tai Chi programs reduce the risk of falls, aquatic therapy may allow patients to complete balance exercises with less pain and fear of falling; however, limited data exist. OBJECTIVE: The objective of the study was to pilot the implementation of an aquatic group based on Ai Chi principles (Aquabalance) and to evaluate the safety, intervention acceptability, and intervention effect sizes. DESIGN: Pilot observational cohort study. METHODS: Forty-two outpatients underwent a single 45-minute weekly group aquatic Ai Chi-based session for eight weeks (Aquabalance). Safety was monitored using organizational reporting systems. Patient attendance, satisfaction, and self-reported falls were also recorded. Balance measures included the Timed Up and Go (TUG) test, the Four Square Step Test (FSST), and the unilateral Step Tests. RESULTS: Forty-two patients completed the program. It was feasible to deliver Aquabalance, as evidenced by the median (IQR) attendance rate of 8.0 (7.8, 8.0) out of 8. No adverse events occurred and participants reported high satisfaction levels. Improvements were noted on the TUG, 10-meter walk test, the Functional Reach Test, the FSST, and the unilateral step tests (p < 0.05). The proportion of patients defined as high falls risk reduced from 38% to 21%. The study was limited by its small sample size, single-center nature, and the absence of a control group. CONCLUSIONS: Aquabalance was safe, well-attended, and acceptable to participants. A randomized controlled assessor-blinded trial is required.
