RESUMO

The study involved 287 patients with chronic recurrent urethroprostatitis. In 83 (28.9%) of them the bacterial microflora in prostatic secretions was detected. The remaining 204 patients with nonbacterial prostatitis underwent in-depth laboratory testing. Cytological and immunological methods, electron microscopy, PCR, McCoy cell culture and developed by the authors the pre-culture pathogen accumulation technique were used to investigate urethral and prostatic smears. As a result C. trachomatis was detected in 84.8% of patients, T. vaginalis--75.5%, and viruses of the Herpesviridae family--in 68.6%. Co-infection was diagnosed in 82.4% of patients, no infection was found in 6.9% of patients. The highest diagnostic sensitivity and specificity were observed in the pre-culture pathogen accumulation technique. According to electron microscopy, T. vaginalis was presented in several morphological shapes: flagellated, spherical and amoeboid. Thus, in 93.1% of patients who had previously been diagnosed with nonbacterial prostatitis by standard bacterial culture, the in-depth microbiological examination identified atypical infections (chlamydia, herpes virus, trichomonas), revealing that the rate of real nonbacterial (non-infectious) prostatitis did not exceed 6.9%.

UNLABELLED: AIM. Study of antibody genesis after introduction of native and inactivated Chlamydia trachomatis MT-2A strain into Balb/c mice and analysis of molecular-genetic features of omp A gene that is responsible for its immunogenicity. MATERIALS AND METHODS: Chlamydia infection was modeled in Balb/c mice by using human isolated C. trachomatis serovariant D. RESULTS: After the introduction of this strain, 5 days after the infection of mice A, M and G class antibodies including G class antibodies against chlamydia heat shock protein are induced. Studies of molecular-genetic features of the isolated C. trachomatis MT-2A strain showed presence of 2 nucleotide mutations in ompAgene. A synonymous substitution of cytosine to thymine in position 574 (CD) that does not result in amino acid substitution in the protein was registered. A non-synonymous substitution of thymine to guanine was present in position 843 (VDIII), that is most probably one of the main reasons of the registered higher immunogenicity of C. trachomatis MT-2A serovariant D strain. CONCLUSION: Further studies of C. trachomatis MT-2A serovariant D strain are promising, but do not exclude search of other immunogenic strains for the generation of agents for specific prophylaxis and immunotherapy of chlamydiosis.

RESUMO

Biological characteristics of C. trachomatis author's strain MT-2A (serovar D) is presented. Stages of development on its basis the experimental formalin-inactivated vaccine against Chlamydia were described. Humoral and cellular immune response to the vaccine administered on 3-dose immunization schedule in conjunction with polyoxidonium as adjuvant was studied. Significant immunological efficacy of the vaccine was shown. T- and B-cell immune responses were characterized. Titer of IgG antibodies against Chlamydia in blood serum after 3rd dose of the vaccine was 10,880+/-1,817.76. Assessment of T-cell response showed that reaction of delayed hypersensitivity with formation of granuloma presented in 60% of animals. Proportion of immunoblasts in reaction of blast-transformation was 29.3+/-2.8%. Perspectives of further studies of the developed corpuscular vaccine against Chlamydia are discussed.
