RESUMO

Although polyethylene terephthalate (PET) fibers are a representative form of plastic pollutants, studies on their toxicity are currently limited compared to other plastic types. Moreover, the effect of natural organic matter (NOM) on their toxicity has not been investigated. In this study, female and male adult zebrafish were exposed to synthesized PET fibers at concentrations of 0.1, 1, 10, and 100 mg/L in the presence and absence of 10 mg/L of NOM for 10 d. Bioaccumulation of PET fibers in zebrafish intestine, liver, and gills was identified and expression levels of reactive oxygen species (ROS) generation, sex hormones, and oxidative stress and sex hormone-related genes were measured. In addition, the developmental stages of gonadal cells were examined through histological analysis. We found that PET fibers bioaccumulated in the intestine and liver of zebrafish. ROS generation significantly increased at 100 mg/L of PET fibers, the expression of oxidative stress-related genes decreased in female and increased in male zebrafish. Exposure to 100 mg/L of PET fibers did not affect 17-beta estradiol, but significantly decreased the testosterone levels in male zebrafish. Sex hormone-related genes significantly decreased in both female and male zebrafish, except for androgen receptor in female zebrafish. However, these changes were exacerbated by the removal of NOM, suggesting a protective effect of NOM against PET fibers toxicity. We demonstrated that the accumulated PET fibers may lead to oxidative stress and sex hormone alteration, and disrupt the development of gonadal cells. Additionally, the NOM coating did not alter bioaccumulation considerably, but mitigated the adverse effects at the hormone level in PET fiber-exposed zebrafish. Thus, this study provides a basis for further research on the toxicity assessment of PET fibers and interactions between NOM and PET fiber-related toxicity.

Regulatory decisions for skin sensitization are now based on adverse outcome pathway (AOP) and integrated approaches to testing and assessment (IATA). Based on these, Organisation for Economic Co-operation and Development (OECD) guidelines on defined approaches for skin sensitization were adopted with a fixed data interpretation procedure (DIP). In the guidelines, "Defined Approaches" (DA) on skin sensitization uses the results from multiple information sources of in chemico, in vitro, and in silico data to achieve an equivalent predictive capacity as those of the animal tests. In this review, we evaluated the skin sensitization of eleven isothiazolinone compounds including 4,5-Dichloro-2-octyl-3(2H)-isothiazolone (DCOIT), 2-n-Octyl-4-isothiazolin-3-one (OIT), 2-Methyl-4-isothiazolin-3-one (MIT), 1,2-Benzisothiazolin-3-one (BIT), 1,2-Benzisothiazolin-3-one, 2-butyl (BBIT), 5-Chloro-2-methyl-4-isothiazolin-3-one (CMIT), 2-methyl-4,5-trimethylene-4-isothiazolin-3-one (MTMIT), 2-methyl-1,2-benzothiazol-3-one (MBIT), 2-methyl-1,2-benzothiazole-3-thione (MBIT-S), 1,2-benzisothiazolin-3-one, 2-methyl-, 1,1-dioxide (BBIT-O), and a mixture of CMIT/MIT. Data from direct peptide reactivity assay (DPRA), human cell line activation (h-CLAT) test, and quantitative structure activity relationship (QSAR) Toolbox were evaluated and were applied to the DIP to derive a prediction of hazard identification and a potency classification. Among the evaluated chemicals, six isothiazolinone compounds were classified to be UN GHS 1A, one compound to be UN GHS 1, and four compounds could not be classified due to lack of data. The results of sensitizer chemicals were found to coincide well with those of in vivo test.

BACKGROUND: There has been an increasing need to update the recommended values of Korean exposure factors for adults aged 19 and older, as using exposure factors developed over a decade ago could reduce risk assessment reliability. OBJECTIVE: Exposure factor data have been compiled and standardized using the latest national statistical reports and academic literature, as well as studies conducted from 2016 to 2018. METHODS: The updated data contained anthropometric parameters, inhalation rates, food and drinking water ingestion rates, and time-activity patterns and provided technical information on Koreans' exposure factors classified by sex, age group, per capita and general population, and doer-only for various exposure assessments. RESULTS: Although the average life expectancy, body weight, body surface area, and inhalation rate increased slightly compared to the 2007 Korean Exposure Factor Handbook, differences various in food consumption were remarkable. Because of Asians' similar food preferences, the intake rate of grain products and vegetables in Koreans, Chinese, and Japanese contributed much toward total intake. Koreans spent half their times outdoors compared to Americans and Chinese. SIGNIFICANCE: This study provided the currently updated exposure factor information for Koreans and could be compared with recommendations provided by exposure factor resources in various countries. IMPACT STATEMENT: Exposure to environmental pollutants may significantly vary depending on the exposure factors related to human behaviors and characteristics. Therefore the exposure factors need to be continuously updated along with more extensive survey areas and improved measurement methods. We utilized the existing data with the aim to develop general exposure factors for risk assessment in Korean aged ≥19 years. Measurements and questionnaire surveys were also performed if there were no existing data. This study provided the currently updated exposure factor information for Koreans and could be compared to those of other countries.

Developing an effective and efficient recycling process for marine debris (MD) is one of the most urgent issues to maintain environmental sustainability on Earth. However, restricted storage capacities and secondary pollution (e.g., microbial adhesion, putrefaction) limit the proper MD recycling. Here, we proposed a complete eco-friendly low-temperature MD pulverizing system that utilizes excessive liquefied natural gas (LNG) cold energy (LCE) in an LNG propulsion ship to improve the efficiency and effectiveness of MD recycling. The prototype design of the low-temperature pulverization (LTP) system showed that consumable refrigerant (liquid nitrogen) up to 2831 kg per hour could be substituted. Furthermore, with a 20% ship output, 1250 kg of MD could be treated with 363 kg of additional refrigerant. In addition, LTP systems utilizing LCE could increase the storage capacity by more than 10 times compared to bulk MD while minimizing the required energy consumption. To determine the feasibility of LTP for MD recycling, four types of plastics obtained from actual MD from a coastal area in Busan, Korea were classified and tested.

Commercial spray products are commonly used in daily life and airborne particles generated by these products may cause adverse health effects. Our study was aimed to characterize the behaviors of airborne particles from spray products and to determine the deposition loss rate. Four categories of spray products with highly frequent use - air fresheners, fabric deodorants, window cleaners, and a bathroom cleaner - were selected for the study. The products were applied in a cleanroom according to the instructions for use. Airborne particles (10-10,000 nm) were measured within the breathing zone of a user with a scanning mobility particle sizer and an optical particle spectrometer. Additionally, filter sampling was performed to examine the morphological characteristics of the particles using a field emission-scanning electron microscope (FE-SEM). The initial concentration and particle size distribution varied among different spray types and products. Two propellant-type air fresheners that we tested showed a high initial concentration of smaller sized particles. However, one of these and all hand-pressure type propellants showed a low initial concentration in all size ranges. We observed that particles in nucleation mode (10-31.6 nm) decreased and aggregated particles shifted to accumulation mode (100-1,000 nm) over time. The FS-SEM analysis confirmed the aggregation of nano-sized particles for all products. The deposition loss rates of various particle sizes depended on the initial concentration and distribution of particle sizes. For two air fresheners with high initial concentrations, the loss rate of small-sized particles was higher than that of the other products whereas the particle loss rate of large-sized particles was higher, regardless of initial concentration. The results of this study can give us useful information in the behaviors of airborne particles in the consumer spray products and resulting exposure assessment especially in the application to the exposure modeling of spray products.

There has been an increasing need for the risk assessment of external environmental hazards in children because they are more sensitive to hazardous chemical exposure than adults. Therefore, the development of general exposure factors is required for appropriate risk assessment in Korean children. This study aimed to determine the general exposure factors among Korean children aged ≤18 years. We developed the recommended exposure factors across five categories: physiological variables, inhalation rates, food and drinking water intake, time-activity patterns, and soil and dust ingestion. National databases were used, and direct measurements and questionnaire surveys of representative samples were performed to calculate the inhalation rate, water intake, and soil ingestion rate. With regard to the time-activity patterns, the daily inhalation rates ranged from 9.49 m3/day for children aged 0-2 years to 14.98 m3/day for those aged 16-18 years. This study found that Korean children spent an average of 22.64 h indoors, 0.63 h outdoors, and 0.73 h in-transit on weekdays. The general exposure factors of Korean children were studied for the first time, and these results could be used to assess children's exposure and risk. They also suggest the differences compared with the results of international results.

Exposure assessment, a fundamental element of the risk assessment process, is the estimation of what consumers may be exposed to by using household products. Reliable exposure factors were investigated to understand the pattern of household products usage. Six household product categories were selected and divided by application types. Two surveys were carried out and 6397 participants completed the web-based questionnaires, and laboratory evaluations were also conducted to measure the actual level of exposure (g/use). We focused more on inhalation exposure from products (and/or their application types) that affect indoor air such as diffusible types. To estimate actual exposure amounts, the time spent to apply the products was investigated from the beginning to end of the product use. For spraying type products, the consumer could be exposed to the maximum amount of used product while using the triggering action and pressing the button on the aerosol spray product, and during cleaning, adhesion, and coating target, consumer could be exposed continuously. For diffuser type products, exposure duration was the time for which consumers were exposed to the diffused product. The exposure amount for trigger and aerosol spray type products were measured using the time taken to trigger or spray the product and the mass generated from the same. This study provides valuable information on the individual exposure factors that may be useful to conduct accurate exposure assessments and to manage household products.

In Korea, humidifiers that include biocidal ingredients have caused serious lung injuries and deaths. After these incidents, public concern regarding the use of chemicals in products (i.e., chemical phobia) increased. Frequent health risk assessments and stringent management of consumer products are, therefore, of paramount importance to reduce these serious occurrences. In this study, the irritative and respiratory health effects of deodorants were assessed in relation to dermal and inhalation exposure. In total, 64 deodorants were divided into 5 groups by application type, and health risk assessments were conducted on each group. In total, 26 fragrance ingredients and 27 biocidal ingredients were analyzed and assessed according to their risk to human health. Exposure assessment was performed in two steps. In the tiered 1 assessment (screening), the 95th exposure factor values were used to estimate exposure to assume the worst-case scenario. The maximum concentration in the deodorants was used without considering the application type. In the tiered 2 assessment (detail assessment), the 75th exposure factor values were used to estimate the assumed reasonable exposure to ingredients. In these assessments, the maximum concentration used in the exposure models was determined by the product purpose and application type. The values input into the exposure algorithms were developed via the exposure route. Of the selected fragrance and biocidal active ingredients, 18 fragrance and 13 biocidal ingredients were detected in the deodorants that were assessed. From the results of the tiered 1 assessment, it was necessary for tiered 2 risk assessments to be conducted for 6 ingredients for the inhalation route, and 13 ingredients for the dermal route. The inhalation margin of exposure of ingredients in deodorants of gel/trigger/spray types for home/car and fabric/air usage was above the target margin of exposure. The health risk of 6 evaluated ingredients was relatively low for the inhalation route of exposure. This study showed that the assessed ingredients have no health risks at their maximum concentrations in deodorants. The approach discussed in this study should be used to establish improved guidelines for specific ingredients in consumer products, and for setting limits for newly developed raw materials that may pose dermal and inhalation hazard.

Ingredient chemicals like fragrances may cause adverse health effects. Frequent health risk assessments and stringent management of consumer products are of paramount importance to reduce these serious occurrences. In this study, the respiratory and dermal health effects were assessed in relation to air fresheners. Twenty six fragrance ingredients, thirty four biocidal ingredients, and sixteen hazardous ingredients were analyzed and assessed according to their risk to human health on five groups by application type in eighty two air fresheners. For hazard characterization of ingredients, toxicological information on the intrinsic properties of the ingredients was collected, and reference values were determined as chronic NOAEL. Exposure assessment was performed in two steps. The 95th exposure factor values were used to estimate exposure to assume the worst-case scenario and the maximum concentration determined by the product purpose and application type was used type in tiered 1 assessment. The values input into the exposure algorithms were developed via the exposure route. In the tiered 2 assessment, the 75th exposure factor values were used to estimate the assumed reasonable exposure to ingredients. Six ingredients for the inhalation and twelve ingredients for the dermal route were conducted for tiered 2 assessment. This study showed that the assessed ingredients have no health risks at their maximum concentrations in air fresheners. The approach should be used to establish improved guidelines for specific ingredients that may pose inhalation and dermal hazard.

The inhalation of a water aerosol from a humidifier containing disinfectants has led to serious lung injuries in Korea. To promote the safe use of products, the Korean government enacted regulations on the chemicals in various consumer products that could have adverse health effects. Given the concern over the potential health risks associated with the hazardous ingredients in deodorizing consumer products, 17 ingredients were analyzed and assessed according to their health risk on 3 groups by the application type in 47 deodorizing products. The risk assessment study followed a stepwise procedure (e.g., collecting toxicological information, hazard identification/exposure assessment, and screening and detailed assessment for inhalation and dermal routes). The worst-case scenario and maximum concentration determined by the product purpose and application type were used as the screening assessment. In a detailed assessment, the 75th exposure factor values were used to estimate the assumed reasonable exposure to ingredients. The exposed concentrations of seven ingredients were calculated. Due to limitation of toxicity information, butylated hydroxyl toluene for a consumer’s exposure via the dermal route only was conducted for a detailed assessment. This study showed that the assessed ingredients have no health risks at their maximum concentrations in deodorizing products. This approach can be used to establish guidelines for ingredients that may pose inhalation and dermal hazards.

Reliable exposure factors are essential to determine health risks posed by chemicals in consumer products. We analyzed five risk-concerned product categories (anti-fogging, dye, disinfectant, repellent, and preservative products) for 13 products (three car anti-fogging products, a lens anti-fogging product, two car dye products, two drain disinfectants, an air conditioner disinfectant, a chlorine-based disinfectant, a fabric repellent, an insect repellent for food, and a wood preservative) considered to be of high risk in order to determine exposure factors via web surveys and estimation of amount of product. Among the 3000 participants (1482 (49%) men) aged ≥19 years, drain disinfectants were used most frequently (38.2%); the rate of usage of the other products ranged between 1.1-24.0%. The usage rates for the consumer products differed by sex, age, income, and education. Some consumer products such as car and lens anti-fogging products, chlorine-based disinfectants, fabric repellents, and drain disinfectants were regularly used more than once a month, while car dye products, air conditioner disinfectants, insect repellents for food, and wood preservatives were not regularly used owing to the specific product purposes and seasonal needs. Our results could be used for managing or controlling chemical substances in consumer products and conducting accurate exposure assessments.

OBJECTIVES: Rapid increase in engineered nanoparticles (ENPs) in many goods has raised significant concern about their environmental safety. Proper methodologies are therefore needed to conduct toxicity and exposure assessment of nanoparticles in the environment. This study reviews several analytical techniques for nanoparticles and summarizes their principles, advantages and disadvantages, reviews the state of the art, and offers the perspectives of nanometrology in relation to ENP studies. METHODS: Nanometrology is divided into five techniques with regard to the instrumental principle: microscopy, light scattering, spectroscopy, separation, and single particle inductively coupled plasma-mass spectrometry. RESULTS: Each analytical method has its own drawbacks, such as detection limit, ability to quantify or qualify ENPs, and matrix effects. More than two different analytical methods should be used to better characterize ENPs. CONCLUSIONS: In characterizing ENPs, the researchers should understand the nanometrology and its demerits, as well as its merits, to properly interpret their experimental results. Challenges lie in the nanometrology and pretreatment of ENPs from various matrices; in the extraction without dissolution or aggregation, and concentration of ENPs to satisfy the instrumental detection limit.

Environmental factors are important in that they might interact with toxicants and could affect the concentration-response relationship of the toxicants in the water environment. Microcystins (MCs) produced by algal blooming are natural toxins that exert severe impacts on aquatic organisms. Despite the possibility of interaction effects between environmental factors and MCs, very few studies have been carried out to date. In the present study, the authors evaluated the effect of 3 environmental factors on chronic toxicity of MC-leucine-arginine (MC-LR; 0 µg/L, 0.8 µg/L, 4 µg/L, 20 µg/L, 100 µg/L, 500 µg/L) to the freshwater invertebrate Moina macrocopa. Three environmental factors were determined to reflect the reasonable worst conditions of the water body in South Korea: high water temperature (25 °C), the highest concentration of nutrients (ammonia-nitrogen [NH3 -N] = 3.8 mg/L; nitrate-nitrogen [NO3 -N] = 8.5 mg/L) during the occurrence of algal bloom, and 2 pH conditions of 7.0 and 9.0, which satisfy the test acceptance criteria for the Daphnia test. Among the various environmental factors being tested, high water temperature and NO3 -N elevated chronic toxicity of MC-LR, whereas NH3 -N reduced toxicity. Water pH did not influence chronic toxicity of MC-LR to M. macrocopa. This observation suggests that those environmental factors are responsible for changing the trend of MC-LR toxicity.
