There is accumulating evidence of the toxicity of organic ultraviolet absorbers (OUVAs); however, limited information is available regarding the presence of OUVAs in terrestrial environments and organisms. Therefore, this study was conducted to investigate the occurrence of 11 OUVAs in soils and typical plant species from an industrial metropolis in China. Total OUVA concentrations in soils ranged from 1.30 to 80.3 ng g-1 DW. Based on comparison with previously reported data, OUVA contamination in soil was not severe. Benzophenone and octocrylene were the dominant OUVAs in soils, with median contributions to total concentrations of 25% and 15%, respectively. Source assessment revealed that the observed OUVA contamination primarily originated from industrial activities and the use of personal care products. The concentration of 11 OUVAs in plants ranged from 159 to 4470 ng g-1 DW, at high levels. Our findings imply that great attention should be given to the presence of these chemicals in plants because of the risk they could pose as well as the potential for biomagnification as plants are eaten by insects and birds. Our results also indicate the necessity to further study the geochemical behavior of these chemicals in urban ecosystems in order to better manage the harmfulness to terrestrial ecological health caused by their exposure through the food chains.

Sepsis-induced muscle atrophy often increases morbidity and mortality in intensive care unit (ICU) patients, yet neither therapeutic target nor optimal animal model is available for this disease. Here, by modifying the surgical strategy of cecal ligation and puncture (CLP), a novel sepsis pig model is created that for the first time recapitulates the whole course of sepsis in humans. With this model and sepsis patients, increased levels of the transcription factor zinc finger BED-type containing 6 (ZBED6) in skeletal muscle are shown. Protection against sepsis-induced muscle wasting in ZBED6-deficient pigs is further demonstrated. Mechanistically, integrated analysis of RNA-seq and ChIP-seq reveals dedicator of cytokinesis 3 (DOCK3) as the direct target of ZBED6. In septic ZBED6-deficient pigs, DOCK3 expression is increased in skeletal muscle and myocytes, activating the RAC1/PI3K/AKT pathway and protecting against sepsis-induced muscle wasting. Conversely, opposite gene expression patterns and exacerbated muscle wasting are observed in septic ZBED6-overexpressing myotubes. Notably, sepsis patients show increased ZBED6 expression along with reduced DOCK3 and downregulated RAC1/PI3K/AKT pathway. These findings suggest that ZBED6 is a potential therapeutic target for sepsis-induced muscle atrophy, and the established sepsis pig model is a valuable tool for understanding sepsis pathogenesis and developing its therapeutics.

Organic ultraviolet absorbents (OUVAs) in the environment have been of increasing concern because of their potential hazards. However, the OUVAs in waters is far from being well studied and little is known about their occurrence in small urban rivers. This study investigated the concentrations and distribution of eleven OUVAs in the sediments from five small urban rivers of Tianjin, China, and found total concentrations in the range of 11.6-189 ng/g dry weight. Relative to other rivers and lakes, no high concentrations of sediment OUVAs were observed in the small rivers. Benzophenone, homosalate and octocrylene were the dominant OUVAs, representing medians of 13.3%, 12.4% and 12.3% of the total concentrations, respectively. Our observed composition profiles of these chemicals were different from those found in most of other waters. The sediment OUVAs may originate more from industrial activities than the use of cosmetics and personal care products in this area. The risk to aquatic organisms from exposure to the sediment OUVAs in these small urban rivers was considered low, except for benzophenone. However, more researches are needed to investigate the pollution and associated risks of these chemicals in urban rivers due to the complexity of their toxicity to aquatic organisms.

The distribution of organic ultraviolet absorbers (OUVAs) in outdoor dust remains poorly understood, especially in megacities. We measured the concentrations of 11 OUVAs in street dust from Tianjin, China, by a gas chromatography-mass spectrometry, and found total concentrations in the range of 10.3-129 ng/g. These OUVAs were prevalent in the study street dust, but their concentrations were much lower than those in indoor dust reported in other areas previously. Benzophenone and octocrylene were the dominant OUVAs, representing medians of 15.5% and 13.1% of total OUVA concentrations, respectively. Total concentrations of dust OUVAs in the industrial area were higher than the residential, cultural and new urban areas. Source assessment indicated that the OUVAs likely originated mainly from the manufacture and consumption of cosmetics and personal care products, and some may have been from the production and use of OUVA-containing consumer products. The calculated non-carcinogenic risks of OUVAs in street dust were low. Our results further confirmed that the OUVAs were prevalent in the environment, provide useful information for understanding potential risks of these chemicals and developing risk management strategies. Further studies are needed to investigate the occurrence, environmental behaviors and potential risks of these emerging contaminants in outdoor environment.

Genetic variants in the asialoglycoprotein receptor 1 (ASGR1) are associated with a reduced risk of cardiovascular disease (CVD) in humans. However, the underlying molecular mechanism remains elusive. Given the cardiovascular similarities between pigs and humans, we generated ASGR1-deficient pigs using the CRISPR/Cas9 system. These pigs show age-dependent low levels of non-HDL-C under standard diet. When received an atherogenic diet for 6 months, ASGR1-deficient pigs show lower levels of non-HDL-C and less atherosclerotic lesions than that of controls. Furthermore, by analysis of hepatic transcriptome and in vivo cholesterol metabolism, we show that ASGR1 deficiency reduces hepatic de novo cholesterol synthesis by downregulating 3-hydroxy-3-methylglutaryl-CoA reductase (HMGCR), and increases cholesterol clearance by upregulating the hepatic low-density lipoprotein receptor (LDLR), which together contribute to the low levels of non-HDL-C. Despite the cardioprotective effect, we unexpectedly observed mild to moderate hepatic injury in ASGR1-deficient pigs, which has not been documented in humans with ASGR1 variants. Thus, targeting ASGR1 might be an effective strategy to reduce hypercholesterolemia and atherosclerosis, whereas further clinical evidence is required to assess its hepatic impact.

ZBED6 (zinc finger BED domain containing protein 6) is a transcription factor unique to placental mammals and its interaction with the IGF2 (insulin-like growth factor 2) locus plays a prominent role in the regulation of postnatal skeletal muscle growth. Here, we generated lean Bama miniature pigs by generating ZBED6-knockout (ZBED6-/-) and investigated the mechanism underlying ZBED6 in growth of muscle and internal organs of placental mammals. ZBED6-/- pigs show markedly higher lean mass, lean mass rate, larger muscle fiber area and heavier internal organs (heart and liver) than wild-type (WT) pigs. The striking phenotypic changes of ZBED6-/- pigs coincided with remarkable upregulation of IGF2 mRNA and protein expression across three tissues (gastrocnemius muscle, longissimus dorsi, heart). Despite a significant increase in liver weight, ZBED6-/- pigs show comparable levels of IGF2 expression to those of WT controls. A mechanistic study revealed that elevated methylation in the liver abrogates ZBED6 binding at the IGF2 locus, explaining the unaltered hepatic IGF2 expression in ZBED6-/- pigs. These results indicate that a ZBED6-IGF2-independent regulatory pathway exists in the liver. Transcriptome analysis and ChIP-PCR revealed new ZBED6 target genes other than IGF2, including cyclin dependent kinase inhibitor 1A (CDKN1A) and tsukushi, small leucine rich proteoglycan (TSKU), that regulates growth of muscle and liver, respectively.

Available information is still insufficient for a comprehensive understanding of the global distribution of polybrominated diphenyl ethers (PBDEs) in the environment. In particular, little is known about the changing trend of their distribution in urban soils. We conducted a survey of 21 PBDEs in urban soils from Tianjin, China. The chemicals were widely present in the area and summed concentrations ranged from 0.65 to 108 ng/g in soil, indicating low to moderate levels of pollution relative to other areas. BDE-209 was the predominant congener, contributing 88.9% of the concentrations of total soil PBDEs. Source assessment indicated that soil PBDEs in the area were mainly derived from the release of commercial deca-BDE from local industrial production processes and consumer products. We found that the soil concentrations of PBDEs appear to have declined in recent years, compared with other previous reports in this region. However, more studies are needed on this possible change trend of PBDE pollution, especially its impact on human health, although their calculated non-carcinogenic health risks in this study were low.

Vehicle exhaust seriously pollutes urban air and harms human health. Photocatalytic technology can effectively degrade automobile exhaust. This work prepared g-C3N4/CeO2 photocatalytic material by constructing heterojunctions. Four kinds of g-C3N4/CeO2 composite photocatalytic materials with different mass ratios were prepared. An indoor exhaust gas purification test was carried out under natural light and ultraviolet light irradiations. The optimum mass ratio of g-C3N4 material and CeO2 material was determined by evaluating the exhaust gas degradation effective. Moreover, the structure and morphology of the g-C3N4/CeO2 composite were investigated with microscopic characterization experiments (including XRD, TG-DSC, FT-IR, UV-Vis, SEM and XPS). The results obtained were that the optimum mass ratio of g-C3N4 material to CeO2 material was 0.75. The degradation efficiencies under ultraviolet irradiation in 60 min for HC, CO, CO2, NOX were 7.59%, 12.10%, 8.25% and 36.82%, respectively. Under visible light conditions, the degradation efficiency in 60 min for HC, CO, CO2 and NOX were 15.88%, 16.22%, 10.45% and 40.58%, respectively. This work is useful for purifying automobile exhaust in the future.

In adult mammals, neurogenesis is limited to specific niches in the brain, but considerable adult neurogenesis occurs in many brain regions in non-mammalian vertebrates. Non-mammalian vertebrates provide invaluable comparative material for understanding the core mechanisms of adult neural stem cell maintenance and fate, but phylogenetic differences in adult neurogenesis remain poorly understood. Here we examine cell proliferation seasonality in the telencephalon of adult female tsinling dwarf skinks (Scincella tsinlingensis) by injecting wild animals caught in summer, autumn and spring, and animals caught in autumn and raised under winter conditions, with 5-Bromo-2'-deoxyuridine (BrdU). Then, 24h, 7d and 28d after BrdU administration we examined brain tissue and quantified BrdU-labeled cells as a marker of neuronal proliferation. The highest number of labeled cells in the telencephalon was found in the 7d group. BrdU-positive cells were widely distributed in the anterior olfactory nucleus (AON), medial cortex (MC), dorsal cortex (DC), lateral cortex (LC), dorsal ventricular ridge (DVR), septum (SP), striatum (STR) and nucleus sphericus (NS). No BrdU-positive cells were detected in olfactory bulbs or elsewhere in the telencephalon. The highest proliferative levels were found in the AON in autumn. The NS exhibited relatively high levels of cell proliferation. The proliferative rate in the AON fluctuated seasonally as autumn>summer>spring>winter. Glial fibrillary acidic protein-positive cells were widely distributed in the telencephalon and their fibrous processes extended into brain parenchyma and anchored in the meninges. Doublecortin-positive newborn neurons of the subventricular zone appeared to migrate into the cerebral cortex via the radial migratory stream. Cell proliferation in the telencephalon of adult female S. tsinlingensis fluctuates seasonally, especially in regions related to olfactory memory. This is the first demonstration of proliferative activity in the telencephalon of a skink.
